Herein we describe the in situ inhibitory activity of three hybrid films (FL1, FL2, and FL3) against two wild strains of Colletotrichum gloeosporioides and Penicillium digitatum as causal agents of rot in Persian limes. The films FL2 and FL3 contained 1.0 and 1.3 % weight/volume Litsea glaucescens essential oil (LgEO) and significantly (p<0.05) delayed rot emergence in Persian limes caused by both pathogens up to 10 days. The physicochemical properties of LgEO and hybrid films were obtained, whereas detailed HPLC profiling revealed that fruit covered with these films significantly (p<0.01) preserved reducing sugars (sucrose, fructose, and glucose), organic acids (citric acid, ascorbic acid, malic acid, and oxalic acid), and flavonoids with nutraceutical activity (hesperidin, eriocitrin, naruritin, neohesperidin, diosmin, vitexin, rutin, and quercetin). This evidence sustains that the composites generated in this investigation improve the shelf life of Persian limes and conserve their nutraceutical content.

OBJETIVO: Analizar la expresión diferencial de proteínas de Aedes aegypti infectados con Wolbachia y su asociación con el ciclo viral del virus dengue (DENV). Material y métodos. Se revisó una base de datos de proteínas de Ae. aegypti infec-tados y no infectados con Wolbachia, cepa wMel y se buscaron estas en revistas indizadas, que hablaran de la proteína y el ciclo viral de DENV. RESULTADOS: La expresión diferencial de proteínas de los mosquitos durante la infección con Wolbachia intervienen en los procesos de entrada, replicación y salida del DENV. CONCLUSIONES: Existen cambios en la expresión de proteínas de células infectadas con Wolbachia, que son necesarias para el ciclo de replicación de DENV, explicando porque algunos mosquitos infectados con Wolbachia son refractarios a la infección por DENV.

OBJECTIVE: To evaluate the presence of Elizabethkingia anophelis infection in Aedes albopictus wild populations of Southern Mexico. MATERIALS AND METHODS: Eight sites were selected to collect Aedes albopictus in the Soconusco region, Chiapas, females were analyzed to amplify the Gyrase B gene by PCR, the minimum infection rate of E. anopheliswas calculated and its species was determined by sequencing and phylogeny. RESULTS: The presence of E. anophelis was only observed in Huehuetán with a minimum infection rate of 37.8%. CONCLUSION: A local strain of E. anophelis was detected for the first time in Ae. albopictus from Chiapas and this bacterium could be considered a candidate for study as a probable control agent or as a vehicle for transgenesis.

Mayaro virus (MAYV) is an arthropod-borne virus (arbovirus) belonging to the family Togaviridae, genus Alphavirus. In recent years, the geographic distribution of MAYV may have expanded north from South and Central America into the Caribbean Islands. Although Haemagogus janthinomys is considered the main vector for MAYV, the virus has also been isolated from other mosquitoes, including Aedes aegypti, a widespread species that serves as the main vector for highly epidemic viruses. Given the possible expansion and outbreaks of MAYV in Latin America, it is possible that MAYV might be adapting to be efficiently transmitted by urban vectors. Therefore, to investigate this possibility, we evaluated the vector competence of Ae. aegypti and Ae. albopictus mosquitoes to transmit MAYV isolated during a year of low or high MAYV transmission. Adult Ae. aegypti and Ae. albopictus were orally infected with the MAYV strains, and the infection, dissemination, and transmission rates were calculated to evaluate their vector competence. Overall, we found higher infection, dissemination, and transmission rates in both Ae. aegypti and Ae. albopictus mosquitoes infected with the strain isolated during a MAYV outbreak, whereas low/no transmission was detected with the strain isolated during a year of low MAYV activity. Our results confirmed that both Ae. aegypti and Ae. albopictus are competent vectors for the emergent MAYV. Our data suggest that strains isolated during MAYV outbreaks might be better fit to infect and be transmitted by urban vectors, raising serious concern about the epidemic potential of MAYV.

Lepechinia meyenii is a medicinal plant specialized in the biosynthesis of different types of antioxidants including the diterpenes carnosic (CA) acid and carnosol (CS). Herein we present the results of plant tissue culture approaches performed in this medicinal plant with particular emphasis on the generation and evaluation of a cell suspension system for CA and CS production. The effect of sucrose concentration, temperature, pH, and UV-light exposure was explored. In addition, diverse concentrations of microbial elicitors (salicylic acid, pyocyanin, Glucanex, and chitin), simulators of abiotic elicitors (polyethylene glycol and NaCl), and biosynthetic precursors (mevalonolactone, geranylgeraniol, and miltiradiene/abietatriene) were evaluated on batch cultures for 20 days. Miltiradiene/abietatriene obtainment was achieved through a metabolic engineering approach using a recombinant strain of Saccharomyces cerevisiae. Our results suggested that the maximum accumulation (Accmax ) of CA and CS was mainly conferred to stimuli associated with oxidative stress such as UV-light exposure (Accmax , 6.2 mg L-1 ) polyethylene glycol (Accmax , 6.5 mg L-1 ) NaCl (Accmax , 5.9 mg L-1 ) which simulated drought and saline stress, respectively. Nevertheless the bacterial elicitor pyocyanin was also effective to increase the production of both diterpenes (Accmax , 6.4 mg L-1 ). Outstandingly, the incorporation of upstream biosynthetic precursors such as geranylgeraniol and miltiradiene/abietatriene, generated the best results with Accmax of 8.6 and 16.7 mg L-1 , respectively. Optimized batch cultures containing 100 mg L-1 geranylgeraniol, 50 mg L-1 miltiradiene/abietatriene (95 : 5 %) and 5 g L-1 polyethylene glycol treated with 6 min UV light pulse during 30 days resulted in Accmax of 26.7 mg L-1 for CA and 17.3 mg L-1 for CS on days 18-24. This strategy allowed to increase seven folds the amounts of CA and CS in comparison with batch cultures without elicitation (Accmax , 4.3 mg L-1 ).

Avocadoes are a rich source of nutrients and nutraceuticals that preserve human health. Nevertheless, this fruit is susceptible to phytopathogen infection during the postharvest period causing severe economic losses. Herein, we report on the in situ antifungal assessment of biodegradable films impregnated with the essential oil of Cinnamomum verum (CvEO) as natural fungistatic coatings to extend postharvest quality of Hass avocadoes (Persea americana cv. Hass). These coatings were evaluated on fruits previously infected with a native strain of Fusarium verticillioides. The cytotoxic assessment of CvEO on F. verticillioides revealed a minimum inhibitory concentration of 0.3±0.0 g L-1 whereas its chemical profiling showed (E)-cinnamaldehyde (45.9 %) 1,3,8-p-menthatriene (7.9 %) and linalool (6.8 %) as the major bioactive compounds. Four coatings (FC1-FC4) were made using chitosan (1 %) and a diverse quantity of CvEO (0.4-1.3 % w/v). The physicochemical properties demonstrated that the films FC3 and FC4 have the optimal characteristics of a food coating. Avocadoes preserved with the films FC3 and FC4 substantially kept (p<0.01) fruit firmness and the nutraceutical content of infected avocados at least for 21 d. The results of the present investigation suggest that our hybrid materials can conserve basic nutritional parameters such as fiber, protein, reducing sugars, as well as the content of unsaturated fatty acids which are the main nutraceuticals of this fruit.

Cera pepper (Capsicum pubescens) is an exotic fruit considered as a rich source of nutraceuticals with known benefits for human health and also an economic resource for local producers in Mexico. The present investigation reports on the in vitro and in situ antifungal activity of the essential oil from Mentha piperita and its two major volatiles (menthol and menthone) against Fusarium sambucinum, which is a causal agent of soft rot in cera pepper. The application of these components in pepper fruits previously infected with F. sambucinum caused a significant delay (p<0.05) in the emergence of soft rot symptoms. This effect was reflected in the maintenance of pH and fruit firmness during a period of 10 days. The nutrimental content of the fruits (protein, fiber, fat and other proximate parameters) was conserved in the same period of time. The nutraceutical content of these fruits was estimated by the quantification of seven carotenoids (violaxanthin, cis-violaxanthin, luteoxanthin, antheraxanthin, lutein, zeaxanthin and ß-carotene), ascorbic acid and capsaicinoids (capsaicin and dihydrocapsaicin). According to our results, the essential oil from M. Piperita and its major volatiles exerted a preservative effect on these metabolites. Our findings demonstrated that the essential oil of M. Piperita and its major volatiles represent an ecological alternative for the control of fusariosis caused by F. sambucinum in cera peppers under postharvest conditions.

The Asian tiger mosquito Aedes albopictus is currently the most invasive vector species, with a widespread global distribution. Aedes albopictus is the potential vector of diverse arboviruses, including Zika and dengue. This study updated the ecological niche model of Ae. albopictus and inferred the potential distribution of natural Wolbachia infections in Ae. albopictus in México. The ecological niche models were constructed based on diverse model settings to better estimate the potential distributions and uncertainty indices of both Ae. albopictus and its natural Wolbachia infections in México. The distribution of Ae. albopictus covered the states across Northern México, the Gulf of México, the Pacific Coast of México, Central México, and the southeast of México. The ecological niche model of the natural Wolbachia infections in Ae. albopictus populations anticipated the occurrence of natural Wolbachia infections in the southeast of México, the Chiapas border with Guatemala, and Veracruz. These results can be used to prioritize vector surveillance and control programs in México for strategic and future decision-making; however, it is still necessary to establish active surveillance programs to assess model predictions based on the independent sampling of Ae. albopictus from different invasion zones in México. Finally, vector surveillance should also screen the natural Wolbachia infections in Ae. albopictus to validate Wolbachia predictions across México, particularly in the southeast of México.

INTRODUCTION: On the eleventh of March 2020, the World Health Organization (WHO) declared the novel coronavirus disease 2019 (COVID-19) a pandemic by the number of cases and deaths worldwide: more than 91.1 million confirmed cases and approx. 1.9 million deaths globally, as of date. The aims of this systematic review were to identify and to evaluate the reports associated on Knowledge, Attitude and Practices (KAP) towards COVID-19 pandemic in America. METHODOLOGY: The Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines were adopted for searching reports published from December 2019 to September 2020, regarding "COVID-19 KAP" across six electronic databases. Inclusion and exclusion criteria were taken up to select the articles and focus to the topic. RESULTS: Thirteen scientific papers were finally eligible and included in this systematic review. The surveys were directed to common citizens, healthcare workers and patients with chronic conditions. General public acquired information about COVID-19 mainly through social media; several misconceptions due to falsehoods circulating on-line were identified. The pandemic COVID-19 has severely affected the physical and emotional health of many people in America. Nevertheless, many American citizens do not recognize or have poor knowledge about COVID-19 risks. CONCLUSIONS: This systematic review brings information for governments and scientific community that may be useful in the development of official awareness and prevention campaigns aiming mainly at marginated groups of the society.

The essential oil from the annual plant Lepidium virginicum L. was chemically characterized in three consecutive years (2018-2020). The essential oils were evaluated in vitro and in situ on the causal agent of anthracnose in tamarillo fruits (Solanum betaceum). The main volatile constituents were phenylacetonitrile (>60 %), linalool (>10 %), limonene (>7 %) and α-terpineol (>5 %). The essential oil (MIC, 19-30 µg mL-1 ), phenylacetonitrile (MIC, 45 µg mL-1 ) and α-terpineol (MIC, 73 µg mL-1 ) caused a significant inhibition in the conidial viability from a wild strain of Colletotrichum acutatum, which was isolated and identified as a causal agent of anthracnose. The inoculation of conidia from C. acutatum in non-symptomatic tamarillo fruits, followed by the in situ treatment with different concentrations of the essential oil (>30 µg mL-1 ), phenylacetonitrile and α-terpineol, significantly (p<0.01) avoided the degradation of anthocyanins (delphinidin 3-O-rutinoside, cyanidin 3-O-rutinoside and pelargonidin 3-O-rutinoside) and carotenoids (ß-cryptoxanthin and ß-carotene) as well as retarded yellowing and necrosis triggered by anthracnose at least for 10 days. Our results suggest the potential use of the essential oil from L. virginicum as a natural component to preserve the nutraceutical content of tamarillo fruits against C. acutatum infection.

BACKGROUND: Aedes aegypti and Aedes albopictus are the main mosquito species responsible for dengue virus (DENV) transmission to humans in the tropical and subtropical regions of the world. The role of vertical transmission in the epidemiology of dengue and the maintenance of this arbovirus in nature during interepidemic periods remain poorly understood, and DENV vertical transmission could sustain the existence of virus reservoirs within Aedes populations. METHODS: Between April 2011 and October 2012, we monitored vertical transmission of DENV in Ae. aegypti and Ae. albopictus in 9 cities of 4 Mexican states. Aedes eggs were collected in ovitraps, then adults were reared under laboratory conditions and their heads were used to infect C6/36 cells. The presence of flavivirus was detected by immunofluorescence assays (IFA), and DENV infection was confirmed by RT-PCR. RESULTS: About 96% of reared adults were Ae. aegypti and 4.0% were Ae. albopictus. No infection was detected in Ae. albopictus, whereas 54 of 713 (7.8%) of Ae. aegypti pools tested positive. A minimum infection rate (MIR) of 2.52 per 1000 mosquitoes was estimated for Ae. aegypti. DENV-1, DENV-2 & DENV-3 serotypes were detected even during interepidemic periods. CONCLUSIONS: This study reports the evidence of vertical transmission of dengue virus with viral isolation and molecular confirmation in Ae. aegypti eggs collected in four endemic regions of Central and Southern Mexico. Vertical transmission may play a role as a reservoir mechanism during mosquito dormancy in interepidemic periods but with minor participation in transmission during epidemic periods.


TRANSMISSION VERTICALE DU VIRUS DE LA DENGUE CHEZ AEDES AEGYPTI ET SON RÔLE DANS LA PERSISTANCE ÉPIDÉMIOLOGIQUE DE LA DENGUE DANS LE CENTRE ET LE SUD DU MEXIQUE: OBJECTIF: Aedes aegypti et Aedes albopictus sont les principales espèces de moustiques responsables de la transmission du virus de la dengue (DENV) à l'homme dans les régions tropicales et subtropicales du monde. Le rôle de la transmission verticale dans l'épidémiologie de la dengue et le maintien de cet arbovirus dans la nature pendant les périodes d'inter-épidémiques restent mal compris, et la transmission verticale du DENV pourrait maintenir l'existence de réservoirs de virus au sein des populations d'Aedes. Notre objectif était d'évaluer la transmission verticale du DENV au Mexique. MÉTHODES: Entre avril 2011 et octobre 2012, nous avons surveillé la transmission verticale du DENV chez Ae. aegypti et Ae. albopictus dans 9 villes de 4 états mexicains. Les œufs d'Aedes ont été collectés dans des ovitraps, puis les adultes ont été élevés dans des conditions de laboratoire et leur tête a été utilisée pour infecter les cellules C6/36. La présence de flavivirus a été détectée par des tests d'immunofluorescence (IFA) et l'infection par DENV a été confirmée par RT-PCR. RÉSULTATS: 96% des adultes élevés étaient Ae. aegypti et 4,0% étaient Ae. albopictus. Aucune infection n'a été détectée chez Ae. albopictus, alors que 54 des 713 (7,8%) des pools d'Ae. aegypti ont été testés positifs. Un taux d'infection minimum (MIR) de 2,52 pour 1000 moustiques a été estimé pour Ae. aegypti. Les sérotypes DENV-1, DENV-2 et DENV-3 ont été détectés même pendant les périodes inter-épidémiques. CONCLUSIONS: Cette étude rapporte les preuves de transmission verticale du virus de la dengue avec isolement viral et confirmation moléculaire dans les œufs d'Ae. Aegypti collectés dans quatre régions d'endémie du centre et du sud du Mexique. La transmission verticale pourrait jouer un rôle de mécanisme réservoir lors de la dormance des moustiques en période inter-épidémique, mais avec une participation mineure à la transmission en période d'épidémie.

El locus g.37190613 en 7p14.2-14.1 del gen ELMO1 presenta un polimorfismo el cual consiste en un cambio G>A(rs1345365). Esta variante, entre otras, de ELMO1, ha sido asociada con nefropatía diabética en diferentes poblaciones. En México son limitados los estudios de asociación de diabetes mellitus con genes candidatos. Por ello, el objetivo del presente trabajo fue determinar la asociación del SNP rs1345365 del gen ELMO1 con el desarrollo de diabetes mellitus tipo 2 (DM2). Para ello se analizaron 148 pacientes con DM2, 156 individuos sin diabetes pero con factores de riesgo cardiovascular y 269 personas sanas sin DM2. El polimorfismo se identificó por PASA (PCR AMPLIFICATION ALLELE SPECIFIC) y PAGE teñida con nitrato de plata. La asociación se estableció por diferentes modelos de epidemiología genética; el modelo dominante mostró una asociación positiva (p=0,0006) como factor protector y el para-dominante mostró al estado heterocigoto como factor de riesgo. En conclusión el estudio reveló la asociación del SNP rs1345365 del gen ELMO1 con DM2 en una población mestiza Mexicana.

The g.37190613 locus on 7p14.2-14.1 in the ELMO1 gene has a G>A polymorphism (SNP rs1345365) that has been associated with diabetic nephropathy in different populations. The genetic-association studies in type 2 diabetes mellitus (DM2) on the Mexican population are limited. The aim of this study was to estimate whether the polymorphism G>A of ELMO1 gene is associated with the development of DM2. We included 148 DM2 individuals, 156 individuals with cardiovascular risk factors without diabetes and 269 healthy proband without DM2. The polymorphism was identified by PCR amplification specific allele (PASA), PAGE and silver staining. The association was established by genetic epidemiological models; the dominant model showed a positive association (p=0.0006) as a protective factor, and the para-dominant model to heterozygous, as risk factor. RESUMEN

BACKGROUND: ELMO1 is a gene located at locus 7p14.2-14.1 that codifies a regulatory protein involved in fibrogenesis, cell migration, phagocytosis and apoptosis. This gene presents a single nucleotide polymorphism, which appears to be linked with the development of diabetic nephropathy. OBJECTIVES: Currently, there are no studies in regard to the presence of such polymorphism in the Mexican population. Therefore, the aim of this work was to estimate the frequency rate of alleles and genotypes of polymorphism rs1345365 from ELMO1 in Mexican mestizos who inhabit the west and the southeast regions of Mexico in order to generate reliable data for further association studies. METHODS: There were 322 individuals who were screened for the identification of polymorphism rs1345365 using genomic DNA from leucocytes as a template for PCR-PASA reactions. Amplicons were separated in 7% PAGE and visualized after staining with silver nitrate. RESULTS: The reference allele (A) was the most frequent in both western and southeastern populations of Mexico. In addition, a different genotype distribution was found with respect to other populations. The results of this study indicate that both populations are in Hardy-Weinberg equilibrium. CONCLUSION: This study also reveals a low frequency rate of the ancestral genotype for the polymorphism rs1345365 in mestizos from the western and southeastern regions of Mexico.


ANTECEDENTES: el gen ELMO1 tiene su locus en 7p14.2-14.1 y codifica para una proteína que regula la fibrogénesis, migración celular, fagocitosis, y apoptosis. Tiene diferentes variaciones de cambio de una sola base asociadas con nefropatía diabética. En la población mexicana no hay estudios de variantes de este gen, y es necesario conocerlas porque pueden ser polimorfismos que posteriormente se usen en estudios de asociación. OBJETIVOS: estimar la frecuencia de alelos y genotipos del polimorfismo rs1345365 del gen ELMO1 en población mestiza del Occidente y de la región sureste de México. MATERIAL Y MÉTODOS: estudio descriptivo de alelos y genotipos del polimorfismo rs1345365 que se identificó con PCR-PASA y PAGE 7% teñida con nitrato de plata. RESULTADOS: se incluyeron 322 individuos y el alelo más frecuente en las dos poblaciones es el de referencia (A). Se encontró una distribución diferente a otras poblaciones respecto a los genotipos. En ambas poblaciones está en equilibrio de Hardy-Weinberg p< 0.05. CONCLUSIONES: el estudio revela una baja frecuencia del genotipo ancestral en población mexicana del polimorfismo rs1345365.

Introducción: la diabetes mellitus tipo 2 es uno de los principales padecimientos crónicos degenerativos en México. Diversos tratamientos se usan para atender a la población oaxaqueña de este país, incluido el empleo de plantas medicinales. Objetivo: efectuar una revisión bibliográfica exhaustiva a través de plataformas electrónicas científicas, con el objetivo de hallar datos sobre plantas antidiabéticas y sus extractos con efecto hipoglucemiante, que son usadas empíricamente en Oaxaca, México. Métodos: se realizó una búsqueda bibliográfica exhaustiva de estudios científicos concernientes a la evaluación de la actividad biológica (que incluyen pruebas in vitro e in vivo) y la química de plantas comúnmente utilizadas para el tratamiento de la diabetes en el estado de Oaxaca, México. Resultados: se presentan 35 plantas de 22 familias botánicas que se usan para el tratamiento de la diabetes mellitus tipo 2, entre ellas se cuenta con un número mayor de especies de las familias Lamiaceae, Cecropiaceae y Equisetaceae. De las 35 especies, 23 sustentan su acción antidiabética en modelos animales y solo en 19 se lograron identificar los compuestos con actividad hipoglucemiante. Conclusión: aproximadamente 77 % de estas plantas cuenta con estudios sobre el efecto de sus extractos o bien acerca de sus metabolitos secundarios individuales. Algunas plantas usadas empíricamente en el estado de Oaxaca, México, contienen flavonoides y terpenos bien identificados con actividad antioxidante e inhibitoria de enzimas del metabolismo de carbohidratos. Miembros específicos de estas familias de metabolitos poseen efecto evidente en la regulación de la glucemia bajo previos bioensayos in vivo o in vitro. Estos hallazgos sustentan de modo coherente el uso milenario de las especies aquí tratadas.


Introduction: type 2 diabetes mellitus (DM2) is one of the most common chronic degenerative diseases in Mexico. Medicinal plants are one of the treatments provided to the Oaxacan population suffering from this disease. Objective: carry out an exhaustive bibliographic review in electronic scientific databases with the purpose of collecting information about antidiabetic plants and hypoglycemic extracts empirically used in Oaxaca, Mexico. Methods: an exhaustive bibliographic search was conducted for scientific studies dealing with the biological activity (in vitro and in vivo tests) and chemical characteristics of plants commonly used to treat diabetes in the state of Oaxaca, Mexico. Results: a presentation is made of 35 plants from 22 botanical families which are used to treat type 2 diabetes mellitus. Most of these species are from the families Lamiaceae, Cecropiaceae and Equisetaceae. In 23 of the 35 species, antidiabetic action was tested in animal models, and only in 19 of them could compounds with hypoglycemic activity be identified. Conclusion: about 77 % of these plants have reports on the effect of their extracts or their individual secondary metabolites. Some of the plants used empirically in the state of Oaxaca, Mexico, contain clearly identified flavonoids and terpenes with antioxidant activity and inhibitory action on enzymes involved in carbohydrate metabolism. Previous in vitro and in vivo assays have shown that certain members of these metabolite families have an obvious effect on glycemia regulation. These findings consistently support the millenarian use of the species herein dealt with.
