RESUMEN

Spinal cord injury (SCI) is a serious central nervous system (CNS) injury disease related to hypoxia-ischemia and inflammation. It is characterized by excessive reactive oxygen species (ROS) production, oxidative damage to nerve cells, and mitochondrial dysfunction. Mitochondria serve as the primary cellular origin of ROS, wherein the electron transfer chain complexes within oxidative phosphorylation frequently encounter electron leakage. These leaked electrons react with molecular oxygen, engendering the production of ROS, which culminates in the occurrence of oxidative stress. Oxidative stress is one of the common forms of secondary injury after SCI. Mitochondrial oxidative stress can lead to impaired mitochondrial function and disrupt cellular signal transduction pathways. Hence, restoring mitochondrial electron transport chain (ETC), reducing ROS production and enhancing mitochondrial function may be potential strategies for the treatment of SCI. This article focuses on the pathophysiological role of mitochondrial oxidative stress in SCI and evaluates in detail the neuroprotective effects of various mitochondrial-targeted antioxidant therapies in SCI, including both drug and non-drug therapy. The objective is to provide valuable insights and serve as a valuable reference for future research in the field of SCI.

OBJECTIVE: To investigate the presence of transforming growth factor-beta 1 (TGF-ß1) and connective tissue growth factor (CTGF) in women with pelvic organ prolapse (POP). METHODS: This study was conducted from May to December 2009. Fifty patients with POP that underwent vaginal hysterectomy in the Department of Gynecology, Renmin Hospital of Wuhan University, Hubei, Wuhan, China were enrolled in this study. They were divided into: Group 1 (n=10); Group 2 (n=10); and Group 3 (n=10) according to Pelvic Organ Prolapse Quantitation (POP-Q). Meanwhile, 20 cases treated by vaginal hysterectomy for other benign gynecological diseases were selected as the control group. Immunohistochemical staining and Western blot were performed to detect the expression of TGF-ß1 and CTGF. RESULTS: Immunohistochemical staining of TGF-ß1 and CTGF were mainly expressed in the cytoplasm of fibroblast cells. The expression of TGF-ß1 and CTGF protein was significantly negatively correlated with POP-Q stage. There were significantly positive correlations between the expression of TGF-ß1 and CTGF protein. The expression of TGF-ß1 protein among the 3 POP groups were all significantly lower than that of the control group, while there was no significant differences in the expression of TGF-ß1 protein among the POP groups, excluding the comparison between Groups 1 and 3. The expression of CTGF protein in the 3 POP groups were all significantly lower than that of the control group, and significant differences were also detected among the 3 POP groups. CONCLUSION: In this study, we found that the TGF-ß1 and CTGF protein expression may be associated with POP, especially in POP-Q stages.

OBJECTIVE: To report our experience on the erosion of polypropylene mesh used in pelvic reconstructive surgeries, and to discuss the pathological changes of mesh erosion. METHODS: We conducted a retrospective study of 128 patients receiving pelvic reconstructive surgeries with polypropylene mesh from May 2006 to May 2009 in the Department of Gynecology and Obstetrics, Renmin Hospital of Wuhan University, Hubei, China. Data regarding patient demographics, operation notes, and follow up were collected. RESULTS: The mean follow-up time for the 128 patients was 15.2 (1.3-60) months. Seven patients were diagnosed with vaginal mesh erosion (Prolene mesh), and one of them suffered from anaphylactoid breakout related to the mesh. The mean (+/- standard deviation) time occurring in the mesh erosion was 9.1 +/- 7.6 months. All the 7 patients were treated with surgery. Chronic inflammation was the main pathological manifestations in the eroded tissue. CONCLUSION: Most cases of polypropylene mesh erosions occur within one year postoperatively. Removal of the mesh could be the best therapy for mesh erosion. Further study is needed to determine if the polypropylene mesh induces supersensitivity.

RESUMEN

Three new topology-varied rod-coil block copolymers, comprising the same oligo(p-phenyleneethynylene) (OPE) rod components and the same coil components, were synthesized by atom-transfer radical polymerization. Their photophysical properties were systematically studied and compared in consideration of their solid-state structures and self-assembly abilities. These copolymers have similar intrinsic photophysical properties to the OPE rods, as reflected in dilute solution. However, their photophysical properties in the solid state are manipulated to be dissimilar by supramolecular organization. Wide-angle X-ray diffraction (WAXD) and atomic force microscopy (AFM) data demonstrate that these copolymers possess different self-assembly abilities due to the molecular-architecture-dependent pi-pi interactions of the rods. Hence, the aggregates in the solid state are formed with a different mechanism for these copolymers, bringing about the discrepancy in the solid-state luminescent properties.

By multiparameter flow cytometry, the T-cell-associated markers CD4 and CD7 were aberrantly coexpressed on a case of B-cell chronic lymphocytic leukemia (CLL). The CLL had an immunophenotype: CD19+, CD20+, CD79b+, CD5+, CD23+, FMC7+, kappa+, CD4+, and CD7+. Molecular analysis confirmed clonal rearrangement of the immunoglobin heavy chain genes and a germline configuration of the T-cell receptor genes. Fluorescence in situ hybridization showed trisomy 12 anomaly. There was no evidence of deletion 17p (p53), 11q23 (ATM), or 13q14 or translocation t(11:14). The patent was clinically stable without treatment. Although the pathogenesis of such aberrant immunophenotype remains to be determined, the current case illustrates the phenotypic heterogeneity of CLL, and emphasizes the importance of a comprehensive diagnostic approach including clinical, morphologic, immunophenotypic, and molecular cytogenetic studies.

RESUMEN

A facile strategy affording an intimate nanocomplex of an amine-containing rod-coil triblock copolymer poly(2-(dimethylamino)ethylmethacrylate)-poly(fluorene)-poly(2-(dimethyl amino) ethylmethacrylate) and CdSe nanocrystals is presented. Ligand exchange is observed by (31)P NMR. TEM and UV-vis absorption results indicate the CdSe NCs have a good dispersion in the conjugated polymer. Also, the PL spectra show photoinduced charge transfer has been facilitated by the complex formation.

Clonal plasma cells from patients with multiple myeloma (MM), plasma cell leukemia (PCL) and human myeloma cell lines (HMCLs) were analyzed for deletions/mutations of the tumor suppressor gene PTEN. By interphase-FISH, hemizygous PTEN deletions were detected in 4 (5.6%) of 71 MM patients, 2 (20%) of 10 PCLs, and 2 (20%) of 10 HMCLs. PTEN deletions were detected in 4 MM patients at diagnosis with stage III disease (Durie-Salmon). Of the six cases with PTEN deletions, 1 MM had a 13q deletion, 1 PCL had a t(11;14), and the other PCL had a t(14;16), a 13q deletion and a p53 deletion. Sequencing analysis did not detect PTEN mutations in 11 primary MM and 5 PCL cases. Our results indicate that alterations of PTEN are uncommon in MM patients, and PTEN deletions tend to occur in advanced disease suggesting that they are secondary, rather than primary, events in the pathogenesis of MM.

Bone marrow cells from five patients with acute promyelocytic leukemia (APL) with t(15;17) were studied by a combination of fluorescence activated cell sorting and fluorescence in situ hybridization (FISH) to establish the cell lineage involvement of t(15;17). Interphase FISH demonstrated that the fusion gene (PML/RARA) was present in almost all abnormal promyelocytes. In one case, the translocation was demonstrated in both CD34+ and CD34- APL cells. The t(15;17) abnormality was not detectable in erythroblasts nor in T- or B-lymphoid cells. These results suggest that lymphocytes and erythroblasts are not clonally involved in APL, and that malignant transformation in some cases of APL may occur at the level of CD34+ cells.

The t(4;14) translocation detected by fluorescence in situ hybridization (FISH) is an independent prognostic factor for an adverse outcome of multiple myeloma (MM). Because t(4;14) uniquely results in fibroblast growth factor receptor 3 (FGFR3) expression, decalcified, paraffin-embedded bone marrow biopsies were immunostained for FGFR3, and its expression was correlated with the t(4;14) status. FISH detected t(4;14) in 16 (19%) of 85 MM patient specimens, and immunocytochemistry detected aberrant FGFR3 expression in 13 (15%). Twelve (75%) t(4;14)-positive cases expressed FGFR3, and 12 (92%) FGFR3-positive cases harbored a t(4;14). FGFR3 expression and t(4;14) were strongly correlated (P < .001). FGFR3 expression by immunohistochemistry was associated with the immunoglobulin A (IgA) isotype (P < .001), a shorter progression-free survival (median, 11.5 versus 25.8 months; P < .001), and a shorter overall survival (median, 19.2 versus 46.3 months; P < .001).

In a series of poly[di(alkyl) vinylterephthalates] (PDAVTs) synthesized via radical polymerization, fine-tuning the size and shape of the side groups manipulated the supramolecular self-assembly and led to control over the formations between amorphous and 2D ordered hexagonal phases. To introduce the 2D long-range ordered structure, the size of the ester side groups at the 2- and 5-positions of the phenyl rings laterally attached to the backbones had to be in the range of propyl/isopropyl to hexyl. The relatively extended backbones observed in these polymers were attributed to steric effects from the side groups. When the n-alkyl groups were larger than hexyl, the ability to form the liquid crystalline phase gradually decreased. A completely disordered phase could be observed by substituting dodecyl groups as side groups.
