Herbal spices are an agricultural commodity, economically very important and beneficial in primary healthcare in the food and medicine sectors. Herbal spices are used as food flavoring agents as well as in phytotherapies throughout the world and have nutritive benefits. The food and medicine industries widely employ artificial or natural adulteration to retard the deterioration and utilization of these adulterants in food and medicine products has given rise to significant apprehension among consumers, primarily stemming from the potential health risks that they pose. Thus, their characterization for the purpose of identification, origin, and quality assurance is mandatory for safe human consumption. Here, we studied 22 samples of commonly traded herbal spices that belong to 20 different genera and 21 species comprising 14 families, investigated macroscopically or organoleptically as well as histologically under microscopic examination. In this study, we provide details on organoleptic features including appearance, taste, odor, color, shape, size, fractures, types of trichomes, and the presence of lenticels among the examined herbal spices and these features have great significance in the detection of both natural as well as artificial deterioration. In terms of microscopic characterization, each examined plant part comprising different anatomical characteristics has taxonomic importance and also provides useful information for authentication from natural adulterants. Furthermore, the studied taxa were also described with nutritive and therapeutic properties. For condiments, herbal beverages and medicinal purposes, different herbal parts such as leaves, floral buds, seeds, fruit, and accessory parts like mericarp, rhizome, bulbs, and bark were used and commercially traded. Similarly, in this study, the leaves of Cinnamomum tamala and Mentha spicata, the floral buds of Syzygium aromaticum, the seeds of Amomum subulatum, Brassica nigra, Punica granatum, Myristica fragrans, Phyllanthus emblica, and Elettaria cardamomum, the mericarp of Coriandrum sativum, and Cuminum cyminum were observed. As a result, we show the potential of herbal spices as a source of many valuable phytochemicals and essential nutrients for food, nutraceutical, and homoeopathic medicine.

Due to the diverse and rich taxa, Asteraceae needs to explore from the untouched area. In this study, the pollen study was intended to assess the taxonomic value for Asteraceous taxa growing on the Sikaram Mountain Pak-Afghan border. Both microscopic techniques (LM and SEM) play a significant role in the identification and classification of herbaceous species of Asteraceae for their taxonomic and systematic implication. Pollen was observed and measured for the 15 species of Asteraceae. For investigated taxa 15 pollen characters with size, shape, polar view, pollen type, aperture orientation, and exine sculpturing in the eurypalynous pollen. Consequently, the pollen grains are usually Tricolporate, triangular to circular in polar view, while, the shape of pollen varies from subulate, oblate, and prolate along with prolate to spheroidal whereas, sculpturing also varies from Scabrate to micro reticulate, echino-perforate, Scabrate to echinate, echinate to granulate and echinate were observed. Similarly, among the quantitative data minimum values of polar and equatorial were 15.8 ± 0.74 µm in Filago pyramidata and 17.85 ± 0.39 µm in Heteropappus altaicus was measured respectively, while; the length of the spine with the least value of 2.45 ± 0.31 µm in Hertia intermedia and highest with 7.55 ± 0.31 µm was observed in Cirsium wallichii. The Exine thickness is a minimum of 1.70 ± 0.35 µm in Launaea nudicaulis and a maximum of 5.65 ± 3.59 µm in Cirssium vulgare. In addition, the highest pollen fertility (87%) in Centaurea iberica while the highest pollen sterility (32%) was recorded in Cirsium verutum. Furthermore, clustering via UPGMA, PCA, and PCoA analysis was performed for the separation of closely related taxa. It is concluded from this research work that palynological study has a crucial role in taxonomic, pure, and applied sciences. This study can be further authenticated and improved by a Phylogenetic study based on chloroplast DNA analysis & whole genome sequencing. RESEARCH HIGHLIGHTS: Pollen ultrastructure among 15 Asteraceous taxa. Micromorphological attributes measured via LM and SEM. Exine sculpture elements patterns provide accurate identification. Taxonomic keys were developed for its systematics implications.

Opuntia is the most diverse and widely distributed drought resistant promising genus of family Cactaceae. The cladodes were utilized to quantify the chemical composition of these plants helpful in lignocellulose conversion and their application towards biofuel production. The present study was aimed to evaluate and compare the taxonomic relationship based on morphology, stem anatomy and palynology of important desert cacti including Opuntia dillenii and Opuntia monacantha. This study also evaluates the potential usefulness of morphological, anatomical and pollen traits using light and scanning electron microscopy. The obtained microcharacters of stem and flowers are considered diagnostic at the generic and specific levels. Some distinguishing morphological features observed were elliptical to obovate cladodes, 1-7 spines per areole and presence of glochidia in O. dillenii. Pollen and stem anatomical characters of the studied taxa are considered highly diagnostic at the generic and species levels. The epidermis has irregular and wavy cells, with straight to sinuate wall pattern and paracytic stomata. Pollen grains appear as pantoporate and prolate spheroidal having reticulate to perforate-reticulate sculpturing while exine semi-tectate to tectate. The taxonomic features studied could be valuable to elaborate and helpful in correctly identification of Opuntia species. The methods of diverse microscopic examination also providing sufficient evidence about the taxonomy of the Opuntia species. RESEARCH HIGHLIGHTS: Description and illustration of desert cacti Opuntia. Morpho-anatomy and palynology were studied with LM and SEM. Highly variation in taxonomic qualitative and quantitative features. Systematic significance based on taxonomic characters was presented.

Biodiesel is a promising, bio-based, renewable, nontoxic, environment friendly, and alternative fuel for petroleum derived fuels which helps to reduce dependency on conventional fossil fuels. In this study, six novel, nonedible seed oil producing feedstock were explored for their potential for sustainable production of biodiesel. It is very important to correctly identify oil yielding plant species. Scanning electron microscopy (SEM) was used as reliable tool for authentic identification of oil yielding seeds. Macromorphological characters of seeds were studied with light microscopy (LM). Outcomes of LM of seeds exposed distinctive variation in seed size from 16.3 to 3.2 mm in length and 12.4 to 0.9 mm in width, shape varied from oval to triangular, and color from black to light brown. Oil content of nonedible seed ranged from 25 to 30% (w/w). Free fatty acid content of seed oil varied from 0.32 to 2.5 mg KOH/g. Moreover, ultra structural study of seeds via SEM showed variation in surface sculpturing, cell arrangement, cell shape, periclinal wall shape, margins, protuberances, and anticlinal wall shape. Surface sculpturing varied from rugged, reticulate, varrucose, papillate, and striate. Periclinal wall arrangements confirmed variation from rough, wavy, raised, depressed, smooth, and elevated whereas, anticlinal walls pattern showed variation from profuse undulating, smooth, raised, grooved, deep, curved, and depressed. It was concluded that SEM could be a latent and advanced technique in unveiling hidden micromorphological characters of nonedible oil yielding seeds which delivers valuable information to researchers and indigenous people for precise and authentic identification and recognition.

In this study, foliar anatomy and pollen morphology of 10 species of Acanthaceae has been investigated using light and scanning electron microscopy. The study was aimed to highlight the role of microscopy in microteaching at community for proper characterization of plants using palyno-anatomical characters including pollen type, exine sculpturing, shape of epidermal cells, pattern of anticlinal wall, type and size of stomata, and trichome. Most of the species have polygonal cell shapes but some species have irregular, tetragonal, and pentagonal shape of epidermal cells. The largest epidermal cell length on adaxial and abaxial surface were observed in Asystasia gangetica 66.95 and 87.40 µm whereas least was observed on adaxial surface in Justicia adhatoda 36.9 µm and on abaxial surface in Barleria cristata 35.65 µm. In anatomy, species have diacytic type of stomata, whereas stomata of paracytic type observed in two species, while in A. gangetica cyclocytic type of stomata are present. Quantitively on abaxial surface, largest stomata length 29.9 µm and width 24.30 µm was noted in B. cristata. While shortest stomata length was observed in Ruellia prostrata 25.95 µm whereas minimum width of stomata was examined in Barleria acanthoides 2.05 µm. The diversity of trichomes are present in all species except in Ruellia brittoniana. Acanthaceae can be characterized by exhibiting different pollen morphology having five types of pollen shapes, prolate, spheroidal, perprolate, subprolate, and oblate spheroidal. Exine peculiarities showing variations such as reticulate, granulate, coarsely reticulate, lophoreticulate, perforate tectate, and granulate surface were examined.

The family Cactaceae is the diversified group of angiosperm plants whose pollen statistics has been used for taxonomic identification. In this article, we present the pollen morphology of eight species belong to seven taxonomically complex genera of Cactaceae including Astrophytum, Cylindropuntia, Echinocereus, Echinopsis, Mammillaria, Opuntia, and Thelocactus using light and scanning electron microscopy. The pollen grains were acetolyzed, measured, described, and electron photomicrographs were taken. Cactaceae can be characterized by presenting different palynomorphological features including pollen type, sculpturing, polar and equatorial diameter, aperture orientation, exine thickness, P/E ratio, and echini features. Four types of pollen shapes, that is, prolate spheroidal (three species), subprolate (two species), prolate (two species), and oblate spheroidal in Echinocereus reichenbachii were observed. The polar and equatorial diameter observed maximum in O. ficus indica 116.95 and 112.27 µm while minimum in M. compressa 38.42 and 21.05 µm. Pollen of two types, tricolpate in members of subfamily Cactioideae and pantoporate in the Opuntioideae were examined. The fertility percentage has been observed maximum in Opuntia macrocentra (83.84%) and minimum in Opuntia ficus-indica (57.89%). Exine sculpturing showing great variations such as granulate, reticulate, granulate perforate and micro-echinate foveolate ornamentation was examined only in Echinopsis eyriesii. A key to species, based on pollen micromorphological attributes, has been constructed for correct identification of complex cactus species.

The current study deals with the detailed morphology investigation of eight Cypsela species belonging to tribe Cichoreae. The different Cypsela types were described, explained, compared, and their taxonomic significance is discussed in detail. Light microscopy (LM) and scanning electron microscopy (SEM) have been used to highlight quantitative and qualitative characters of underestudied species. Cypsela exhibit great diversity in macro and micromorphological features such as shape, color, length, width, anticlinal and periclinal wall patterns, surface patterns, epicuticular projections. Majority of Cypsela species were brownish in color and their size ranges from 2.16 to 3.98 mm in length and 1.16 to 0.82 mm in breadth. A great diversity in Cypsela shapes like oblanceolate to obovate, obovoid to cylindrical, obvate, narrowly lanceolate were observed. Most of the platelets having epicuticular projections were observed. The surface pattern on the cypsela surface varied from rugose papillate, verrucose papillate, and striated. On the basis of considerable variations observed, the present study can assist as useful constraints at various taxonomic levels. The aim of the present study is to provide a comprehensive description of the Cypsela morphology and to determine the extent to which these micro morphological data can be used as a taxonomic character to delineate various taxa belonging to the tribe Cichoreae.
