RESUMEN

BACKGROUND: In the past decade, the field of tumour immunotherapy has made a great progress. However, the efficacy of immune checkpoint blocking (ICB) in the treatment of hepatocellular carcinoma (HCC) remains limited. Cytotoxic lymphocyte trafficking into tumours is critical for the success of ICB. Therefore, additional strategies that increase cytotoxic lymphocyte trafficking into tumours are urgently needed to improve patient immune responses. METHODS: Paired adjacent tissue and cancerous lesions with HBV-associated HCC were subjected to RNA-seq analysis. Bone morphogenetic protein (BMP9), which reflects vessel normalisation, was identified through Cytoscape software, clinical specimens and Gene Expression Omnibus (GEO) datasets for HCC. The functional effects and mechanism of BMP9 on the tumour vasculature were evaluated in cells and animals. An ultrasound-targeted microbubble destruction (UTMD)-mediated BMP9 delivery strategy was used to normalise the vasculature and evaluate therapeutic efficacy mediated by cytotoxic lymphocytes (NK cells) in combination with a PD-L1 antibody in human cancer xenografts of immune-deficient mice. RESULTS: We discovered that hepatitis B virus (HBV) infection-induced downregulation of BMP9 expression correlated with a poor prognosis and pathological vascular abnormalities in patients with HCC. BMP9 overexpression in HBV-infected HCC cells promoted intra-tumoural cytotoxic lymphocyte infiltration via vascular normalisation by inhibiting the Rho-ROCK-myosin light chain (MLC) signalling cascade, resulting in enhanced efficacy of immunotherapy. Furthermore, UTMD-mediated BMP9 delivery restored the anti-tumour function of cytotoxic lymphocytes (NK cells) and showed therapeutic efficacy in combination with a PD-L1 antibody in human cancer xenografts of immune-deficient mice. CONCLUSIONS: HBV-induced BMP9 downregulation causes vascular abnormalities that inhibit intra-tumoural cytotoxic lymphocyte infiltration, providing a rationale for developing and combining immunotherapy with BMP9-based therapy to treat HBV-associated HCC.

In this study, samples were taken of the surface dust of main roads in Xinxiang City, and the concentrations of five heavy metals (Cd, Pb, Cr, Cu and Zn) and fifteen polycyclic aromatic hydrocarbons (PAHs) were determined by inductively coupled plasma mass spectrometry (ICP-MS) and gas chromatography-mass spectrometry (GC-MS), respectively. Meanwhile, the effects of vehicle emissions on the pollution characteristics were investigated. The results showed that the concentrations of heavy metals and PAHs ranged from 2.58 to 1560 mg·kg-1 and ND to 1.30 mg·kg-1, respectively. Overall, the concentrations of heavy metals and PAHs increased with a decrease in dust particle size. In terms of composition, the heavy metals were dominated by Zn while the high-molecular-weight PAHs were mainly homologous. In spatial distribution, the concentrations of heavy metals and PAHs were different. The total concentrations of heavy metals in road dust near Renmin Road, Xiaodian Industrial Park, and Cement Plant were the highest, while the high concentrations of PAHs appeared in the dust of Renmin Road, Upper Expressway, and 107 National Highway. Pearson correlation analysis showed that there was no positive correlation between the five heavy metals and fifteen PAHs. Then cluster analysis and factor analysis indicated that the PAHs were greatly affected by vehicle emissions, while the heavy metals were basically unaffected.

Glutathione (GSH) plays crucial roles in various physiological and pathological processes. The fluctuation of the GSH level is closely associated with a variety of diseases and cellular functions. Hence, it is important to real-time monitor the fluctuation of GSH in living cells. In this work, we presented a rhodol-hybridized hemicyanine fluorophore (RdH) as a selective, rapid-response, ratiometric, and reversible fluorescent probe for intracellular GSH (t1/2 = 89 ms, Kd = 1.42 mM). The imaging assays in living cells revealed that RdH could be used to real-time monitor GSH dynamics in A549 cells under a laser scanning confocal microscope by ratiometric fluorescence changes.

Although strategies based on π-conjugation expansion and one-atom replacement have routinely been used to extend the emission wavelengths of rhodamine dyes from the visible region to the biologically more favorable red to near-infrared (NIR) region, the strategy of introducing electron-withdrawing groups to the meso-position of pyronin dyes for the same goal is still in its infant phase. In this work, we present a benzothiazole-functionalized pyronin dye BTP as a red-emission fluorescent dye platform for bioimaging applications. Due to the electron-withdrawing nature of the meso-substituted benzothiazole unit, BTP exhibited a large red-shift in absorption and emission wavelengths compared to classic rhodamines. Interestingly, BTP could not only behave like a molecular rotor to fluorescently respond to viscosity changes, but also specifically target lysosomes and light up them assisted by a lysosomal viscous microenvironment. Furthermore, based on the BTP platform, we developed its "dihydro" derivative, i.e., HBTP, and evaluated its sensing performance to reactive oxygen species (ROS). The obtained results showed that HBTP is a highly selective fluorescent probe for sensing endogenous peroxynitrite (ONOO-) with quite rapid fluorescence off-on response and high sensitivity. It is greatly expected that the present study could stimulate research interest in exploiting various rhodamine-inspired fluorescent dyes or probes for bioimaging applications.
