RESUMEN

Poly(N-isopropylacrylamide) thermosensitive hydrogel tube tip solid-phase extraction/ultra-high liquid chromatography-mass spectrometry (UPLC-MS/MS) method was developed for analysis of methylimidazoles in beverages. Thermosensitive poly(N-isopropylacrylamide) (PNIPA) hydrogel solid-phase extraction (SPE) medium was prepared on the surface of highly internal phase emulsion (HIPE) porous polymer by thermally initiated polymerization in a tube tip. The temperature sensitive SPE medium has the characteristics of high porosity and high specific surface area. When the temperature is higher than 30.0℃, it can well adsorb polar molecular, and could quickly desorb polar molecular when the temperature was less than 20.0℃. The tube tip SPE coupled with UPLC-MS/MS method was established for the determination of three polar molecules including 1-methylimidazole, 4-methylimidazole and 2-methylimidazole, with linear ranges of 2.50 - 240 µg/L, and detection limits of 1.20, 1.20 and 0.65 µg/L, respectively. The method was applied to the determination of three methylimidazoles in beverages with the spiked recoveries of 81.5%-115.5% and the RSD of 0.6%-5.0%, and the relative errors of the results with the national standard UPLC-MS/MS method were in the range of -8.5%-8.9%.

In this work, Ag nanoparticles (AgNPs) were embedded into a poly(N-isopropylacrylamide)-laponite (Ag/PNIP-LAP) hydrogel membrane for highly sensitive surface-enhancement Raman scattering (SERS) detection. In situ polymerization was initiated by UV light to encapsulate AgNPs in a PNIP-LAP hydrogel to prepare a highly active SERS membrane with a three-dimensional structure. Due to the surface plasmon resonance and high swelling/shrinkage ratio of the Ag/PNIP-LAP hydrogel SERS membrane, its network structure has a "sieving" effect, which makes it easier for hydrophilic small-molecule targets to enter the sterically confined hydrogel, and the AgNPs are close to each other to form a Raman "hot spot" through the shrinkage of the hydrogel, at the same time, the analyte is enriched in the confined space and close to the AgNPs to form a stronger SERS signal. The characterization of the SERS activity of the Ag/PNIP-LAP hydrogel showed that the prepared three-dimensional membrane had high detection sensitivity for urotropine, 2,5-dimethylpyrazine, pyrazinamide, and pyrazine; the detection limits (S/N = 3) were 17.4, 31.0, 53.1, and 1.11 µg/L, and the analytical time was 35 min. Due to the hydrophilicity of the Ag/PNIP-LAP hydrogel membrane, the small molecules can easily enter the SERS membrane, and the hydrophobic macromolecules are blocked outside the SERS membrane. The SERS method has good selectivity, stability, and reproducibility. The SERS method was applied to the detection of urotropine in dried bean curd sticks, 2,5-dimethylpyrazine in nuts and potato chips, and pyrazinamide in human plasma with recoveries of 81.8-116.8% and the relative standard deviation within 4.9-9.9%. The results matched well with that found by the corresponding chromatographic methods. The proposed method has the advantages of simple sample pretreatment, speediness, high sensitivity, and good selectivity to hydrophilic compounds and has potential application in the rapid on-site detection.

It is challenging to capture glycoprotein from complicated samples with both high specificity and high recovery. Herein, we synthesized temperature-responsive poly (N-isopropylacrylamide-4-vinylbenzeneboronic acid) (PNIPAM-VPBA) microgels having enhanced affinity capability towards glycoprotein, resulting signal amplification in colorimetric analysis subsequently. Through temperature control, the target glycoprotein can be captured and released by the reported microgels unbiasedly. The microgels fabricated were characterized in terms of particle size, phase transition temperature, and toxicity. It was revealed that obvious changes in the particle diameter ranged from 600 to 323 nm between 25 °C and 55 °C, the volume transition temperature was close to human physiological temperature, and low toxicity towards HeLa cells after 24 h incubation. Using glucose oxidase as a model target, the microgels were applied as adsorbent materials for preconcentration of glucose oxidase, resulting 22-fold signal enhancement and a limit of detection (LOD) as low as 16.7 U/L. In beer sample analysis, satisfied recoveries of 78.7%-101.6% with the relative standard deviations (RSDs) less than 7.2% was observed. Subsequently, the microgels were combined with enzyme-linked immunosorbent assay (ELISA) for alpha fetoprotein (AFP) determination, by immobilizing horseradish peroxidase (HRP) and anti-AFP antibodies on the surface of microgels. A meaningful low concentration range of 2.5 × 10-2 to 1.0 µg/L was obtained for AFP detection using the microgels based colorimetric method with a LOD of 8.4 ng/L. In human serum sample analysis, good accuracy was achieved from method comparing with commercial ELISA kit, satisfied recoveries of 86.1%-96.4% with the RSDs less than 6.0% was observed. The proposed PNIPAM-VPBA microgels have great potential applications in the fields of food and clinical analysis.

A large-scale high-efficient enzyme reactor based on polymerized high internal phase emulsion monolith (polyHIPE) was prepared. First, a porous cross-linked polyHIPE monolith was prepared by in-situ thermal polymerization of a high internal phase emulsion containing styrene, divinylbenzene and polyglutaraldehyde. The enzyme of TPCK-Trypsin was then immobilized on the monolithic polyHIPE. The performance of the resultant enzyme reactor was assessed according to the conversion ability of Nα-benzoyl-l-arginine ethyl ester to Nα-benzoyl-l-arginine, and the protein digestibility of bovine serum albumin (BSA) and cytochrome (Cyt-C). The results showed that the prepared enzyme reactor exhibited high enzyme immobilization efficiency and fast and easy-control protein digestibility. BSA and Cyt-C could be digested in 10 min with sequence coverage of 59% and 78%, respectively. The peptides and residual protein could be easily rinsed out from reactor and the reactor could be regenerated easily with 4 M HCl without any structure destruction. Properties of multiple interconnected chambers with good permeability, fast digestion facility and easily reproducibility indicated that the polyHIPE enzyme reactor was a good selector potentially applied in proteomics and catalysis areas.

RESUMEN

Traditional Chinese herbal medicines (TCHMs) are promising approach for the treatment of various diseases which have attracted increasing attention all over the world. Chemometrics in quality control of TCHMs are great useful tools that harnessing mathematics, statistics and other methods to acquire information maximally from the data obtained from various analytical approaches. This feature article focuses on the recent studies which evaluating the pharmacological efficacy and quality of TCHMs by determining, identifying and discriminating the bioactive or marker components in different samples with the help of chemometric techniques. In this work, the application of chemometric techniques in the classification of TCHMs based on their efficacy and usage was introduced. The recent advances of chemometrics applied in the chemical analysis of TCHMs were reviewed in detail.

A new class of solid-phase extraction column prepared with grafted mercapto-silica polymerized high internal phase emulsion particles was used for the preconcentration of trace lead. First, mercapto-silica polymerized high internal phase emulsion particles were synthesized by using high internal phase emulsion polymerization and carefully assembled in a polyethylene syringe column. The influences of various parameters including adsorption pH value, adsorption and desorption solvents, flow rate of the adsorption and desorption procedure were optimized, respectively, and the suitable uploading sample volumes, adsorption capacity, and reusability of solid phase extraction column were also investigated. Under the optimum conditions, Pb(2+) could be preconcentrated quantitatively over a wide pH range (2.0-5.0). In the presence of foreign ions, such as Na(+) , K(+) , Ca(2+) , Zn(2+) , Mg(2+) , Cu(2+) , Fe(2+) , Cd(2+) , Cl(-) and NO3 (-) , Pb(2+) could be recovered successfully. The prepared solid-phase extraction column performed with high stability and desirable durability, which allowed more than 100 replicate extractions without measurable changes of performance. The feasibility of the developed method was further validated by the extraction of Pb(2+) in rice samples. At three spiked levels of 40.0, 200 and 800 µg/kg, the average recoveries for Pb(2+) in rice samples ranged from 87.3 to 105.2%.

In this work, polymerized high internal phase emulsion (polyHIPE) monoliths were prepared and applied as monolithic adsorbent materials for proconcentration of trace polycyclic aromatic hydrocarbons (PAHs) from large-volume water samples. The monolithic polyHIPE columns were prepared by in situ polymerization of the continuous phase of a high internal phase emulsion (HIPE) containing styrene (STY), divinylbenzene (DVB) and glycidyl methacrylate (GMA) in pipette tips, and the resulting STY/DVB/GMA polyHIPE monoliths exhibited highly interconnected porosity and large surface areas, making them excellent candidates as adsorbents for enrichment of trace aromatic compounds. The prepared STY/DVB/GMA polyHIPE monoliths were applied to the determination of trace PAHs in environmental water samples by combing with high performance liquid chromatography-fluorescence detection (HPLC-FLD). Under the optimized experimental conditions, the polyHIPE monoliths could effectively enrich trace 13 PAHs from 500mL of water samples, the mean recoveries at four spiked levels were ranged from 80.7% to 115.0% with the relative standard deviations (RSDs) lower than 14%, and the detection limits (LODs) were ranged from 4.0 to 228pg/L. In addition, the prepared polyHIPE monolith was stable enough for more than 200 replicate extraction cycles without measurable loss of performance on the enrichment of PAHs, and good column-to-column repeatability was obtained with RSD less than 13%. The proposed method was applied to simultaneous analysis of 13 PAHs in water samples with satisfactory recoveries.

A novel large-volume immobilized enzyme reactor (IMER) on small column was prepared with organic-inorganic hybrid silica particles and applied for fast (10 min) and oriented digestion of protein. At first, a thin enzyme support layer was formed in the bottom of the small column by polymerization with α-methacrylic acid and dimethacrylate. After that, amino SiO2 particles was prepared by the sol-gel method with tetraethoxysilane and 3-aminopropyltriethoxysilane. Subsequently, the amino SiO2 particles were activated by glutaraldehyde for covalent immobilization of trypsin. Digestive capability of large-volume IMER for proteins was investigated by using bovine serum albumin (BSA), cytochrome c (Cyt-c) as model proteins. Results showed that although the sequence coverage of the BSA (20%) and Cyt-c (19%) was low, the large-volume IMER could produce peptides with stable specific sequence at 101-105, 156-160, 205-209, 212-218, 229-232, 257-263 and 473-451 of the amino sequence of BSA when digesting 1mg/mL BSA. Eight of common peptides were observed during each of the ten runs of large-volume IMER. Besides, the IMER could be easily regenerated by reactivating with GA and cross-linking with trypsin after breaking the -C=N- bond by 0.01 M HCl. The sequence coverage of BSA from regenerated IMER increased to 25% comparing the non-regenerated IMER (17%). 14 common peptides. accounting for 87.5% of first use of IMER, were produced both with IMER and regenerated IMER. When the IMER was applied for ginkgo albumin digestion, the sequence coverage of two main proteins of ginkgo, ginnacin and legumin, was 56% and 55%, respectively. (Reviewer 2) Above all, the fast and selective digestion property of the large-volume IMER indicated that the regenerative IMER could be tentatively used for the production of potential bioactive peptides and the study of oriented protein digestion.
