RESUMEN

A copper-catalyzed thiocyanation of cycloketone oxime esters with ammonium thiocyanate has been developed for the first time. This innovative approach allows access to cyano and thiocyano bifunctionally substituted alkanes, which can be further transformed into their respective trifluoromethylthiol-substituted or difluoromethylthiol-substituted alkylnitriles, alkynyl sulfides, and phosphorothioate esters. The readily available nature of ammonium thiocyanate and the cost-effectiveness of the copper catalyst make this method a promising strategy for the synthesis of sulfur-containing alkylnitriles.

RESUMEN

A novel visible-light induced sulfonylation/ipso-cyclization of N-arylpropiolamides with cyclobutanone oxime esters and Na2S2O5 was reported. This protocol proceeded via a radical process. The wide substrate scope, sustainable metal-free conditions and readily accessible reagents make this protocol an efficient and green strategy for the synthesis of cyanoalkyl sulfonated spiro[4,5]trienones.

RESUMEN

We report the visible-light-promoted selenocyanation of cyclobutanone oxime esters using potassium selenocyanate in the presence of a fac-Ir(ppy)3 catalyst for the first time. Because of the mild conditions employed and use of readily accessible potassium selenocyanate, this method is an effective and green strategy for the synthesis of cyano and selenocyano bifunctional substituted alkanes.

RESUMEN

Mammalian patatin-like phospholipase domain containing proteins (PNPLAs) play critical roles in triglyceride hydrolysis, phospholipids metabolism, and lipid droplet (LD) homeostasis. PNPLA7 is a lysophosphatidylcholine hydrolase anchored on the endoplasmic reticulum which associates with LDs through its catalytic region (PNPLA7-C) in response to increased cyclic nucleotide levels. However, the interaction of PNPLA7 with LDs through its catalytic region is unknown. Herein, we demonstrate that PNPLA7-C localizes to the mature LDs ex vivo and also colocalizes with pre-existing LDs. Localization of PNPLA7-C with LDs induces LDs clustering via non-enzymatic intermolecular associations, while PNPLA7 alone does not induce LD clustering. Residues 742-1016 contains four putative transmembrane domains which act as a LD targeting motif and are required for the localization of PNPLA7-C to LDs. Furthermore, the N-terminal flanking region of the LD targeting motif, residues 681-741, contributes to the LD targeting, whereas the C-terminal flanking region (1169-1326) has an anti-LD targeting effect. Interestingly, the LD targeting motif does not exhibit lysophosphatidylcholine hydrolase activity even though it associates with LDs phospholipid membranes. These findings characterize the specific functional domains of PNPLA7 mediating subcellular positioning and interactions with LDs, as wells as providing critical insights into the structure of this evolutionarily conserved phospholipid-metabolizing enzyme family.

Due to rapid urbanization, industrialization and population growth, wetland area in China has shrunk rapidly and many wetland ecosystems have been reported to degrade during recent decades. Wetland health assessment could raise the public awareness of the wetland condition and guide policy makers to make reasonable and sustainable policies or strategies to protect and restore wetland ecosystems. This study assessed the health levels of wetland ecosystem at the Hangzhou Bay, China using the pressure-state-response (PSR) model through synthesizing remote sensing and statistical data. Ten ecological and social-economic indicators were selected to build the wetland health assessment system. Weights of these indicators and PSR model components as well as the normalized wetland health score were assigned and calculated based on the analytic hierarchy process (AHP) method. We analyzed the spatio-temporal changes in wetland ecosystem health status during the past 20years (1990-2010) from the perspectives of ecosystem pressure, state and response. The results showed that the overall wetland health score was in a fair health level, but displayed large spatial variability in 2010. The wetland health score declined from good health level to fair health level from 1990 to 2000, then restored slightly from 2000 to 2010. Overall, wetland health levels showed a decline from 1990 to 2010 for most administrative units. The temporal change patterns in wetland ecosystem health varied significantly among administrative units. Our results could help to clarify the administrative responsibilities and obligations and provide scientific guides not only for wetland protection but also for restoration and city development planning at the Hangzhou Bay area.
