Silver and silver ions have a long history of antimicrobial activity and medical applications. Nevertheless, the activity of Ag+ against bacteria, how it enters a cell, has not yet been established. The K+ channel, a membrane protein, is a possible route. The addition of a channel inhibitor (4-aminopyridine) to modulate the Ag+ uptake could support this view. However, the inhibitor enhances the uptake of Ag+, the opposite result. We have applied cold ion trap infrared laser spectroscopy to complexes of Ag+ and Ac-Tyr-NHMe (a model for GYG) which is a portion of the selectivity filter in the K+ channel to consider the question of permeation. With support from quantum chemical calculations, we have determined the stable conformations of the complex. The conformations strongly suggest that Ag+ would not readily permeate the K+ channel. The mechanism of the unexpected enhancement by the inhibitor is discussed.

The results of several epidemiological studies and clinical trials investigating the effects of n-3 polyunsaturated fatty acids (PUFAs) on antenatal and postnatal depression remain controversial. In a previous case-control study of early pregnancy in Japan, we found an inverse association between eicosapentaenoic acid and risk of psychological distress after adjusting for possible confounders. Here, in a 1:2 matched case-control study, we further investigated the possible relationship between serum n-3 PUFAs and risk of psychological distress in the second and third trimesters of pregnancy. The psychological distress group (n=71) consisted of subjects with a score of ≥13 on the Kessler Psychological Distress Scale. The control group (n=142) was matched for age, educational level, and family income. Fatty acid composition of total lipid was determined from serum samples by gas chromatography. Associations between fatty acid levels and incidence of psychological distress were evaluated by logistic regression. Sixty-six percent of blood samples were collected in the second trimester and the remainder in the third. There were no significant differences in any of the n-3 PUFAs between the two groups. After adjustment for possible confounders, none of the n-3 PUFAs showed an association with risk of psychological distress. Peripheral n-3 PUFA levels might not influence the risk of psychological distress in later pregnancy. Further research is warranted to clarify this finding.

We have developed a new orthodontic bracket with three slots with lubricative properties on the working surfaces and proposed a new orthodontic treatment system employing 0.012-0.014-inch Ni-Ti arch wires. We recruited 54 patients, of which 27 received treatment with the new zirconia bracket with multiple slots system (M group), and the others received treatment with standard edge-wise appliances (control group [C group]). We compared the (1) tooth movement rate at the early stage of leveling; (2) changes in the dental arch morphology before and after leveling; and (3) pain caused by orthodontic treatment. Student's t-test was used in all assessments. The tooth movement rate in the maxillomandibular dentition was higher in the M group. The basal arch width, anterior length, and the intercanine width in the maxillary dentition were not significantly different in the two groups; however, the intercanine width in the mandibular dentition was higher in the C group. In assessments of treatment-related pain, the visual analogue pain score was 56.0 mm and 22.6 mm in the C and M groups, respectively. A new zirconia bracket with multiple slots system provided better outcomes with respect to tooth movement rate, treatment period, and postoperative pain, thus indicating its effectiveness over conventional orthodontic systems.

In this study, we examined the relationship between upper limb joint movements and horizontal racket head velocity to clarify joint movements for developing racket head speed during tennis serving. Sixty-six male tennis players were videotaped at 200 Hz using two high-speed video cameras while hitting high-speed serves. The contributions of each joint rotation to horizontal racket velocity were calculated using vector cross-products between the angular velocity vectors of each joint movement and relative position vectors from each joint to the racket head. Major contributors to horizontal racket head velocity at ball impact were shoulder internal rotation (41.1%) and wrist palmar flexion (31.7%). The contribution of internal rotation showed a significant positive correlation with horizontal racket head velocity at impact (r = 0.490, P < 0.001), while the contribution of palmar flexion showed a significant negative correlation (r = - 0.431, P < 0.001). The joint movement producing the difference in horizontal racket head velocity between fast and slow servers was shoulder internal rotation, and angular velocity of shoulder internal rotation must be developed to produce a high racket speed.

To estimate energy cost of a gymnastic-like exercise performed by an astronaut during spaceflight (cosmic exercise), energy expenditure was determined by measuring mechanical work done around the center of mass (COM) of the body. The cosmic exercise, which consisted of whole-body flexion and extension, was performed during a spaceflight and recorded with a video camera. By analyzing the videotape, the internal mechanical work (W(int)) against inertia load of the body segments was calculated. To compare how human muscles work on Earth, a motion similar to the cosmic exercise was performed by a control subject who had a physique similar to that of the astronaut. The total mechanical power of the astronaut was determined to be about 119 W; although the control subject showed a similar total power value, half of the power was external work (W(ext)) against gravitational load. By assuming a mechanical efficiency of 0.25, the energy expenditure was estimated to be 476 W or 7.7 W/kg, which is equivalent to that expended during fast walking and half of that used during moderate-speed running. Our results suggest that this form of cosmic exercise is appropriate for astronauts in space and can be performed safely, as there are no COM shifts while floating in a spacecraft and no vibratory disturbance.
