Background: Intervertebral disc degeneration (IVDD) is the main cause of low back pain (LBP), but the specific regulatory factors, pathways and specific molecular mechanisms remain unclear. Methods: We identified and quantitatively analyzed Pfirrmann Grade II (n=3) and Pfirrmann Grade IV (n=3) pulposus samples via MRI. The differential abundance of proteins in the samples was determined and quantitatively analyzed by relative and absolute quantitative analysis of the isotope marker levels combined with the liquid chromatography-tandem mass spectrometry (LC‒MSMS/MS). Results: A total of 70 proteins (30 significantly increased proteins (> 1.2-fold change) and 40 significantly decreased proteins (< 0.8-fold change)) showed different levels among the groups. Kyoto Encyclopedia of Genes and Genomes and Gene Ontology (GO) enrichment analyses and Western blot analysis showed that CYCS, RAC1, and PSMD14 may play important roles in IVDD and that Epstein‒Barr virus infection, viral myocarditis, colorectal cancer, nonalcoholic fatty liver disease (NAFLD) and amyotrophic lateral sclerosis (ALS) are the main pathways involved in IVDD. Conclusion: CYCS, RAC1 and PSMD14 may play important roles in IVDD, and Epstein‒Barr virus infection, viral myocarditis, colorectal cancer, NAFLD and ALS may be the main pathways involved in IVDD.

Adolescent idiopathic scoliosis is the deformity of the human spine in three-dimensional space. It is a disease with a long course and difficult to recover in a short time. Currently, spinal orthotic braces are an effective non-surgical treatment for this condition. However, existing spinal orthotic braces are still deficient. For example, existing spinal orthotics braces have no way of knowing how correct, effective, and comfortable a patient is wearing the orthotics. Based on distributed pressure monitoring technology, this paper conducts pressure acquisition and compensation research on spinal orthotics used by adolescent patients. After analyzing the principles of orthopedics and selecting monitoring points, this paper selects appropriate sensors and calibrates them. An intelligent wearable system for adolescent spinal orthotics was developed based on these findings. The experimental results show that the system can effectively monitor the process of patients wearing spinal orthotic braces. In addition, the system can compensate and visualize the pressure in real-time, so that doctors and patients can know the process and make judgments and adjustments.

We measured the levels of High-Mobility Group Box 1 (HMGB1), Receptor for Advanced Glycation Endproducts (RAGE), T Helper 17 cells (Th17), Regulatory T cells (Treg), and related cytokines in the peripheral blood of patients with severe preeclampsia (SPE) complicated with acute heart failure (AHF) to explore the expression changes in these indicators. In total, 96 patients with SPE admitted to Gansu Provincial Maternity and Child-care Hospital between June 2020 and June 2022 were included in the study. The patients were divided into SPE+AHF (40 patients) and SPE (56 patients) groups based on whether they suffered from AHF. Additionally, 56 healthy pregnant women who either received prenatal examinations or were admitted to our hospital for delivery during the same period were selected as the healthy control group. An enzyme-linked immunosorbent assay was performed to detect the expression levels of HMGB1, RAGE, interleukin (IL)-17, IL-6, transforming growth factor ß (TGF-ß), IL-10, and NT-proBNP in plasma. Flow cytometry was employed to determine the percentages of Th17 and Treg cells. Compared to the healthy control group, the SPE+AHF and SPE groups had higher plasma levels of HMGB1 and RAGE expression, higher Th17 percentage and Th17/Treg ratio, and lower Treg percentage. Compared to the SPE group, the SPE+AHF group had higher plasma levels of HMGB1 and RAGE expression, higher Th17 percentage and Th17/Treg ratio, and lower Treg percentage (P < .05). In patients with SPE with AHF, plasma HMGB1 was positively correlated with RAGE, Th17, Th17/Treg, IL-17, and IL-6 and was negatively correlated with TGF-ß and IL-10 (P < .05). Our findings revealed that patients with SPE with AHF had elevated levels of HMGB1 and RAGE while exhibiting Th17/Treg immune imbalance, suggesting that the abnormal expression of these indicators may be involved in the pathogenesis of SPE with AHF.

Ultralow-field magnetic resonance imaging (ULF-MRI) has broad application prospects because of its portable hardware system and low cost. However, the low B0 magnitude of ULF-MRI results in a reduced signal-to-noise ratio in qualitative images compared with that of commercial high-field MRI, which can affect the visibility and delineation of tissues and lesions. In this work, a magnetic resonance fingerprinting (MRF) approach is applied to a homemade 50-mT ULF-MRI scanner to achieve efficient quantitative brain imaging, which is an original and promising disease-diagnosis approach for portable MRI systems. An inversion recovery fast imaging with steady-state precession-based sequence is utilized for MRF through Cartesian acquisition. A microdictionary analysis method is proposed to select the optimal repetition time and flip angle variation schedule and ensure the best possible tissue discriminative ability of MRF. The T1 and T2 relaxation properties and the B1 + distribution are considered for estimation, and the results are compared with those of gold standard (GS) quantitative imaging or qualitative imaging methods. The phantom experiment indicates that the quantitative values obtained by schedule-optimized MRF show good agreement, and the bias from the GS results is acceptable. The in vivo experiment shows that the relaxation times of white and gray matter estimated by MRF are slightly lower than the reference data, and the relaxation times of lipid are within the range of the reference data. Compared with qualitative MRI under ULF, MRF can intuitively reflect various items of brain tissue information in a single scan, so it is a valuable addition to point-of-care imaging approaches.

OBJECTIVE: To investigate the clinical efficacy of acrylic cement (PMMA) mixed with calcium sulfate combined with percutaneous kyphoplasty (PKP) in the treatment of osteoporotic fracture (OVCF). METHODS: The clinical data of 191 patients with OVCF treated with PKP from January 2020 to March 2021 were retrospectively analyzed. Among them, 82 patients with 94 vertebral bodies were treated with PMMA mixed with calcium sulfate as the observation group, and 109 patients with 125 vertebral bodies were treated with pure PMMA as the control group. Among the 82 patients in the observation group, there were 16 males and 66 females, with a mean age of (75.35±11.22) years old, including 36 thoracic vertebrae and 58 lumbar vertebrae. In the control group, there were 109 patients, 22 males and 87 females, with an average age of (74.51±9.21) years old, including 63 thoracic vertebrae and 62 lumbar vertebrae. The visual analog scale (VAS) before operation and 1 day, 3 months and 1 year after operation were calculated. The Oswestry disability index (ODI), Cobb's angle, vertebral body height and the probability of postoperative bone cement leakage were used to analyze the efficacy of the two groups. RESULTS: All the patients were followed up for more than one year. Compared with the control group, there was no significant difference in operation time, bleeding volume and bone cement injection volume between the two groups(P>0.05), while the leakage rate of bone cement was significantly lower in the observation group (P<0.05). In addition, there was no significant difference in VAS, ODI, Cobb angle, and vertebral body height between the two groups before operation, and 1 day, 3 months, and 1 year after operation (P>0.05), but each index was improved compared with that before operation (P<0.05). CONCLUSION: PMMA mixed with calcium sulfate has equivalent efficacy in treating OVCF than PMMA alone, but can effectively reduce the probability of cement leakage.

Ultralow-field (ULF) magnetic resonance imaging (MRI) can suffer from inferior image quality because of low signal-to-noise ratio (SNR). As an efficient way to cover the k-space, the spiral acquisition technique has shown great potential in improving imaging SNR efficiency at ULF. The current study aimed to address the problems of noise and blurring cancelation in the ULF case with spiral trajectory, and we proposed a spiral-out sequence for brain imaging using a portable 50-mT MRI system. The proposed sequence consisted of three modules: noise calibration, field map acquisition, and imaging. In the calibration step, transfer coefficients were obtained between signals from primary and noise-pick-up coils to perform electromagnetic interference (EMI) cancelation. Embedded field map acquisition was performed to correct accumulated phase error due to main field inhomogeneity. Considering imaging SNR, a lower bandwidth for data sampling was adopted in the sequence design because the 50-mT scanner is in a low SNR regime. Image reconstruction proceeded with sampled data by leveraging system imperfections, such as gradient delays and concomitant fields. The proposed method can provide images with higher SNR efficiency compared with its Cartesian counterparts. An improvement in temporal SNR of approximately 23%-44% was measured via phantom and in vivo experiments. Distortion-free images with a noise suppression rate of nearly 80% were obtained by the proposed technique. A comparison was also made with a state-of-the-art EMI cancelation algorithm used in the ULF-MRI system. SNR efficiency-enhanced spiral acquisitions were investigated for ULF-MR scanners and future studies could focus on various image contrasts based on our proposed approach to widen ULF applications.

OBJECTIVE: To explore the clinical outcomes of anteromedial combined with lateral locking plate for complex proximal humeral fractures in the eldery. METHODS: From June 2018 to October 2020, 30 patients who underwent surgical treatment for Neer grade 3 to 4 proximal humeral fractures, including 8 males and 22 females, aged from 51 to 78 years old with an average of (61.5±7.5) years old. Of them, 15 patients had fractures fixed with anteromedial combined with lateral locking plate(ALLP group), whereas 15 received internal fixation with proximal humerus locking plate only(PHLP group). The clinical data, simple shoulder test (SST), humeral head height loss, varus angle and shoulder range of motion were compared between the two groups. RESULTS: All fractures were healed. The follow-up time ranged from 12 to 24 months, with an average of(14.3±2.9) months. The operation time of ALLP group was longer than that of PHLP group (P<0.05). There was no significant difference in intraoperative blood loss between the two groups (P>0.05). There was no significant difference in SST score between the two groups at 1, 3 and 12 months after operation (P>0.05). In terms of radiographic measurement, there was no significant difference in humeral head height loss and varus angle between the two groups at 1 and 3 months after operation (P>0.05). At 12 months after operation, the height loss and varus angle of humeral head in ALLP group were lower than those in PHLP group (P<0.05). In shoulder range of motion, the range of forward elevation in ALLP group was larger than that in PHLP group 1 year after operation(P<0.05). There was no significant difference in external rotation between the two groups. CONCLUSION: Anteromedial combined with lateral locking plate in the treatment of complex proximal humeral fractures in the elderly can increase the stability of the medial column and obtain a good fracture prognosis. But there are also disadvantages such as longer operation time, so it should be individualized according to the fracture type of the patient.

Objective:To analyze the influencing factors of the disease cognition level in main caregivers of children with congenital heart disease (CHD).Methods:In this prospective study, the main caregivers of 150 children with CHD admitted to the Seventh People′s Hospital of Zhengzhou from January 2018 to January 2020 were selected as the research objects with convenient sampling method, and a self-designed questionnaire on awareness of CHD disease was used for face-to-face investigation. A total of 150 questionnaires were issued, and 150 valid questionnaires were reclaimed. According to the total score of the questionnaire (>45 or ≤45), the subjects were divided into high score group (90 cases) and low score group (60 cases). The general data of the main caregivers of children with CHD in the two groups were collected, the χ2 test and logistic regression were used to analyze the influencing factors of disease cognition level in those caregivers. Results:The disease cognition score of the main caregivers of children with CHD in the two groups was (58.69±7.33). The proportions of people with high school education or above, living in cities and towns and obtaining disease knowledge from hospitals in the high score group were all significantly higher than those in the low score group (66.7% vs 36.7%, 72.2% vs 55.0%, 92.2% vs 78.3%) (all P<0.05). Low education level (primary school, OR=1.394, 95% CI: 1.131-1.657; junior high school/technical secondary school, OR=1.406, 95% CI: 1.052-1.761), living in rural area ( OR=1.343, 95% CI: 1.214-1.472), obtaining disease knowledge from outside the hospital ( OR=1.328, 95% CI: 1.037-1.620) were all positively related to the decline of disease cognition scores of the main caregivers of CHD children (all P<0.05). Conclusion:The main caregivers of children with CHD have a moderate to high level of disease cognition, and the education level, residence and access to disease knowledge are the main influencing factors.

Cholestatic liver disease is a common disease that causes bile flow dysfunction due to various reasons. The etiology of cholestatic liver disease is complexed, and therapeutic drugs are extremely limited. To date, ursodeoxycholic acid is the only FDA-approved drug for treating primary biliary cirrhosis, whereas its efficacy is limited to early stage of the disease, therefore novel drugs are urgently needed. Nuclear receptors become therapeutic hotspot target in cholestasis since these receptors play a key role in regulating bile acid homeostasis. Peroxisome proliferator-activated receptor (PPAR) is an important nuclear receptor involved in regulating multiple mechanisms of cholestasis in vivo. It can improve intrahepatic cholestasis by inhibiting bile acid synthesis, reducing bile acid toxicity, affecting the expression of bile acid metabolic enzymes and transporters, and can play an anti-inflammatory, anti-oxidation and anti-fibrosis role. A number of studies have shown that PPAR agonists represented by fibrates alone or in combination can improve liver function indexes, inflammatory factors and fibrosis markers in patients with cholestasis. This review analyzes and summarizes the lastest advances in the molecular mechanism of PPAR as a therapeutic target for cholestasis and drug treatment in development or have been used in clinical.

Objective: Chronic low back pain (CLBP) after percutaneous endoscopic transforaminal discectomy (PTED) surgery may be caused by preoperative lumbar microinstability (MI). However, there is a paucity of research on the relationship between lumbar microinstability and chronic low back pain. The purpose of this article is to assess the preoperative radiographic characteristics of patients and evaluate the effects of lumbar microinstability on patient-reported outcomes among single-level lumbar disc herniation (LDH) patients who underwent PTED. Methods: This study retrospectively reviewed the radiographic characteristics of a consecutive series of 127 patients with low back pain and leg pain caused by single-level LDH underwent PTED from August 2018 to March 2021. They were divided into three groups according to the radiographic parameters: the stable group (Group S), the dysfunctional group (Group D), and the microinstability group (Group M). The visual analogue scale (VAS) scores for leg and low back pain and Oswestry Disability Index (ODI) were evaluated preoperatively and postoperatively. Logistic regression analysis was used to identify independent risk factors for CLBP. Results: Compared with Group D and Group S, Group M had the highest ODI scores (P < 0.01) and VAS scores (low back pain) (P < 0.01) after 1 year, while there were no significant differences in the VAS scores for leg pain at different time points after surgery (P > 0.05). In addition, the logistic regression analysis results regarding CLBP revealed that muscle fatty degeneration on MRI (95% CI, 1.20-8.51, P = 0.02), and facet tropism (95% CI, 1.39 -11.37, P = 0.01) may be independent risk factors. Conclusion: Patients with lumbar microinstability may have CLBP after PTED, so patients with lumbar microinstability may need to take internal fixation surgery to solve their symptoms.

Ni3S2 nanowires were synthesized in situ using a one-pot hydrothermal reaction on Ni foam (NF) for use in supercapacitors as a positive electrode, and various contents (0.3-0.6 mmol) of Co(OH)2 shells were coated onto the surfaces of the Ni3S2 nanowire cores to improve the electrochemical properties. The Ni3S2 nanowires were uniformly formed on the smooth NF surface, and the Co(OH)2 shell was formed on the Ni3S2 nanowire surface. By direct NF participation as a reactant without adding any other Ni source, Ni3S2 was formed more closely to the NF surface, and the Co(OH)2 shell suppressed the loss of active material during charging-discharging, yielding excellent electrochemical properties. The Co(OH)2-Ni3S2/Ni electrode produced using 0.5 mmol Co(OH)2 (Co0.5-Ni3S2/Ni) exhibited a high specific capacitance of 1837 F g-1 (16.07 F cm-2) at a current density of 5 mA cm-2, and maintained a capacitance of 583 F g-1 (16.07 F cm-2) at a much higher current density of 50 mA cm-2. An asymmetric supercapacitor (ASC) with Co(OH)2-Ni3S2 and active carbon displayed a high-power density of 1036 kW kg-1 at an energy density of 43 W h kg-1 with good cycling stability, indicating its suitability for use in energy storage applications. Thus, the newly developed core-shell structure, Co(OH)2-Ni3S2, was shown to be efficient at improving the electrochemical performance.

Global plastics production has been increasing year by year. Due to the large quantity of plastics and the difficulty of their degradation, plastics are continuously accumulated in the environment. Therefore, plastic waste has become one of the most serious threats to the global environment. Microplastics can be absorbed into organisms through the mouth, respiratory tract and skin, causing organ(intestine, liver) toxicity, reproductive and developmental toxicity, and neurotoxicity. Moreover, microplastics can also take up other pollutants distributed in the surrounding environment, such as heavy metals and organic pollutants, jointly exerting combined toxic effects. The extracts of microplastics, including microplastics unstable polymers and additives, also have toxic effects. The molecular mechanisms involved in the toxic effects induced by microplastics include oxidative stress, inflammation, disturbance of intestinal flora, disturbance of gene expression, and others.

【Objective】 To analyze the data of non-small cell lung cancer (NSCLC) gene chip using the bioinformatics method, screen differential expression genes (DEGs), and explore the biomarkers related to the prognosis of NSCLC so as to provide a new target for the treatment of NSCLC. 【Methods】 The NSCLC gene chip data were downloaded from the GEO database and the common DEGs in the two datasets were screened by GEO2R tool and FunRich3.1.3 software. The DAVID database was used in GO analysis and KEGG analysis of the DEGs. The protein-protein interaction (PPI) network was constructed using the STRING database; Cytoscape 3.8.0 software was used to select the top 20 hub genes. Then Kaplan-Meier plotter was used to analyze the prognosis of the identified hub genes, and multiple external databases were used to verify the expressions of the hub genes and their relationship with prognosis. 【Results】 A total of 159 intersect DEGs were screened from the two datasets. A total of 20 hub genes were identified via PPI network. Survival analysis and validation results from multiple external databases showed that SPP1 was highly expressed in NSCLC tumor tissues and was significantly correlated with the patients’ poor prognosis (P<0.05). The subgroup analysis showed that SPP1 might cause the poor prognosis by affecting lymph node metastasis. 【Conclusion】 SPP1 may be a biomarker for evaluating the prognosis of NSCLC patients, providing a new idea for the targeted therapy of NSCLC.

In the last decade, research studies on parking planning mainly focused on path planning rather than trajectory planning. The results of trajectory planning are more instructive for a practical parking process. Therefore, this paper proposes a trajectory planning method in which the optimal autonomous valet parking (AVP) trajectory is obtained by solving an optimal control problem. Additionally, a vehicle kinematics model is established with the consideration of dynamic obstacle avoidance and terminal constraints. Then the parking trajectory planning problem is modeled as an optimal control problem, while the parking time and driving distance are set as the cost function. The homotopic method is used for the expansion of obstacle boundaries, and the Gauss pseudospectral method (GPM) is utilized to discretize this optimal control problem into a nonlinear programming (NLP) problem. In order to solve this NLP problem, sequential quadratic programming is applied. Considering that the GPM is insensitive to the initial guess, an online calculation method of vertical parking trajectory is proposed. In this approach, the offline vertical parking trajectory, which is calculated and stored in advance, is taken as the initial guess of the online calculation. The selection of an appropriate initial guess is based on the actual starting position of parking. A small parking lot is selected as the verification scenario of the AVP. In the validation of the algorithm, the parking trajectory planning is divided into two phases, which are simulated and analyzed. Simulation results show that the proposed algorithm is efficient in solving a parking trajectory planning problem. The online calculation time of the vertical parking trajectory is less than 2 s, which meets the real-time requirement.

Docetaxel is an important anti-microtubule agent used to treat a variety of solid tumors, including breast cancer; notably, docetaxel-containing regimens improve outcomes for patients in metastatic, adjuvant, and neoadjuvant settings. However, the effectiveness of docetaxel in clinical practice can be compromised by suboptimal management of side effects. Here, we report two cases of docetaxel-based chemotherapy regimens in patients who exhibited invasive ductal breast cancer and underwent two different clinical treatment approaches. A 58-year-old postmenopausal female received salvage treatment with 8 cycles of docetaxel (67 mg/m2), and a 74-year-old female received 1 cycle of docetaxel (100 mg/m2). The two patients exhibited considerable hearing loss two days later. Of note, both patients had no hearing loss symptoms prior to docetaxel. Thus, ototoxicity may be a side effect of docetaxel that should be considered during treatment.

BACKGROUND: The function of miR-20a-5p in pulmonary artery smooth muscle cells (PASMCs) and the underlying mechanism remains largely unknown. METHODS: C57BL/6J mice and PASMCs were used for constructing pulmonary artery hypertension (PAH) animal and cell models, respectively. Reverse transcription polymerase chain reaction (RT-PCR) was employed to detect miR-20a-5p and ATP-binding cassette subfamily A member 1 (ABCA1) messenger RNA expression. CCK-8, Transwell, and TUNEL experiments were used to determine PASMCs proliferation, migration, and apoptosis. The relationship between miR-20a-5p and ABCA1 was detected by luciferase reporter experiment, Western blot analysis, and qRT-PCR. RESULTS: miR-20a-5p was remarkably elevated in PASMCs of PAH mice and human PASMCs treated by hypoxia, while ABCA1 was remarkably decreased. After transfection of miR-20a-5p mimics, PASMCs proliferation and migration were promoted and PASMCs apoptosis was suppressed. ABCA1 was confirmed to be a target of miR-20a-5p and restoration of ABCA1 reversed the function of miR-20a-5p. CONCLUSION: miR-20a-5p enhances the proliferation and migration of PASMCs to promote the development of PAH via targeting ABCA1.

To decipher the Cd hyperaccumulation and tolerance mechanisms of plants and increase phytoremediation efficiency, in this study, the physiological effects induced by environmentally relevant concentrations (0, 25 and 200 mg/kg) of Cd were characterized in Amaranthus hypochondriacus (K472) at three growth stages using LC/MS-based metabolomics. Metabolomic analysis identified 31, 29 and 30 significantly differential metabolites (SDMs) in K472 exposed to Cd at the early, intermediate and late stages of vegetative growth, respectively. These SDMs are involved in nine metabolic pathways responsible for antioxidation, osmotic balance regulation, energy supplementation and the promotion of metabolites that participate in phytochelatin (PC) synthesis. K472 at the intermediate stage of vegetative growth had the strongest tolerance to Cd with the combined action of Ala, Asp and Glu metabolism, purine metabolism, Gly, Ser and Thr metabolism and Pro and Arg metabolism. Among these crucial metabolic biomarkers, purine metabolism could be the primary regulatory target for increasing the Cd absorption of K472 for the restoration of Cd-contaminated soil.

PURPOSE: Various reports found a relationship between lymphocyte-related blood parameters (LRBP), including absolute lymphocyte counts (ALC), neutrophil-to-lymphocyte ratio (NLR), and platelet-to-lymphocyte ratio (PLR) and prognosis of breast cancer. Most of the studies focused on LRBP pre-treatment. Seldom have studies focused on LRBP during radiotherapy. We intended to perform a retrospective cohort study on the prognostic value of LRBP at the time point of lowest ALC during radiotherapy for breast cancer. PATIENTS AND METHODS: A total of 158 female patients were included in radiotherapy group because of the strict limitation standards of complete routine blood test results at pre-treatment and pre-operation, and at least once a week during radiotherapy. Besides 221 patients, including the 158 patients of radiotherapy group, were adopted in pre-treatment group and pre-operation group. RESULTS: ALC and PLR at the time point of lowest ALC during radiotherapy are prognostic predictors of breast cancer, and lower ALC and higher PLR are independent significant predictors of poor DFS. Besides, lower ALC, higher NLR and higher PLR at both pre-treatment and pre-operation were found to be independent variables for predicting poor DFS. CONCLUSION: LRBP at pre-treatment, pre-operation, and during radiotherapy may serve as predictors of outcomes of breast cancer.
