BACKGROUND: The clinical incidence of spinal infection is gradually increasing, and its onset is insidious, easily leading to missed diagnosis and misdiagnosis, which may lead to serious complications such as nervous system dysfunction, spinal instability and/or deformity, and cause a huge burden on society and families. Early identification of the causative agent and precision medicine will greatly reduce the suffering of patients. At present, the main pathogenic bacteria that cause spinal infection are Staphylococcus aureus, Streptococcus, Pneumococcus, Escherichia coli, and Klebsiella. There are no reports of spinal infection caused by Pseudomonas fluorescens. CASE SUMMARY: We report a 32-year-old female patient with spinal infection. She presented with flank pain, initially thought to be bone metastases or bone tuberculosis, and had a family background of tumors. Her clinical features and changes in imaging and laboratory tests led to the suspicion of thoracic spine infection. Histopathology of the lesion showed inflammation, tissue culture of the lesion was negative several times, and the possible pathogen - Pseudomonas fluorescens was found after gene sequencing of the lesion. The patient recovered completely after a full course of antibiotic treatment. CONCLUSION: This report increases the range of pathogens involved in spinal infections, highlights the unique advantages of gene sequencing technology in difficult-to-diagnose diseases, and validates conservative treatment with a full course of antibiotics for spinal infections without complications.

Soil microplastics (MPs) have attracted widespread attention recently. Most studies have explored how soil MPs affect the soil's physicochemical parameters, matter circulation, and soil microbial community assembly. Similarly, a key concern in agricultural development has been the use of phosphorus (P) fertiliser, which is essential for plant health and development. However, the relationship between MPs and phosphate fertilisers and their effects on the soil environment and plant growth remains elusive. This study assessed the influence of adding low-density polyethylene MPs (1%) with different phosphate fertiliser application rates on microbial communities and rice biomass. Our results showed that MPs changed the structure of soil bacterial and phoD-harbouring microbial communities in the treatment with P fertiliser at the same level and suppressed the interactions of phoD-harbouring microorganisms. In addition, we found that MPs contamination inhibited rice growth; however, the inclusion of P fertiliser in MP-contaminated soils reduced the inhibitory action of MPs on rice growth, probably because the presence with P fertiliser promoted the uptake of NO3--N by rice in MP-contaminated soils. Our results provide further insights into guiding agricultural production, improving agricultural management, and rationally applying phosphate fertilisers in the context of widespread MPs pollution and global P resource constraints.

OBJECTIVE: Numerous schizophrenic patients are suffering from obesity primarily attributed to antipsychotic medication and poor dietary habits. This study investigated the progressive deterioration of olanzapine-induced metabolic disorders in the presence of a high-fat diet (HFD) and explored the involvement of endoplasmic reticulum (ER) stress. METHODS: Female Sprague-Dawley rats fed on a standard chow diet or HFD were treated with olanzapine (3 mg/kg/day) and the ER stress inhibitor 4-phenylbutyric acid (4-PBA, 1 and 0.5 g/kg/day) for 8 days. Changes in body weight, food intake, and plasma lipids were assessed. Hepatic fat accumulation was evaluated using oil red O staining. Western blotting and immunofluorescence assays were employed to examine the expression of ER stress markers, NOD-like receptor pyrin domain-containing protein 3 (NLRP3), and proopiomelanocortin (POMC) in the hypothalamus or liver. RESULTS: Compared to olanzapine alone, olanzapine+HFD induced greater weight gain, increased hyperlipidemia, and enhanced hepatic fat accumulation (P<0.05). Co-treatment with 4-PBA exhibited a dose-dependent inhibition of these effects (P<0.05). Further mechanistic investigations revealed that olanzapine alone activated ER stress, upregulated NLRP3 expression in the hypothalamus and liver, and downregulated hypothalamic POMC expression. The HFD exacerbated these effects by 50%-100%. Moreover, co-administration of 4-PBA dose-dependently attenuated the olanzapine+HFD-induced alterations in ER stress, NLRP3, and POMC expression in the hypothalamus and liver (P<0.05). CONCLUSION: HFD worsened olanzapine-induced weight gain and lipid metabolic disorders, possibly through ER stress-POMC and ER stress-NLRP3 signaling. ER stress inhibitors could be effective in preventing olanzapine+HFD-induced metabolic disorders.

Biochar as an agricultural soil amendment plays vital roles in mediating methane (CH4) and nitrous oxide (N2O) emissions in soils. The link between different types of biochar, bulk soil, and rhizosphere microbial communities in relation to CH4 and N2O emissions is being investigated in this study. The rice pot experiment was conducted using biochar at two temperatures (300°C and 500°C) in combination with three biochar levels (0, 2, 10% w/w). Soil properties and the abundance of genes associated with CH4 and N2O emissions from both rhizosphere and bulk soils were investigated. The study also aimed to examine the structure of microbial communities (pmoA, nosZ) in rhizosphere and bulk soils whereas CH4 and N2O emissions were monitored while growing rice. Results showed that biochar at 300°C and 10% incorporation significantly increased the CH4 emissions by up to 59% rise compared to the control group. Random Forest analysis revealed that the ratio of mcrA/pmoA along with the abundance of mcrA from both rhizosphere and bulk soils, the abundance of AOA, TN, DOC, and the community composition of pmoA-harboring microorganisms from both bulk and rhizosphere soils were important predictors of CH4 emissions. Therefore, the ratio of mcrA/pmoA in rhizosphere soil and the abundance of AOA in bulk soil were the main factors influencing CH4 emissions. Variation Partitioning Analysis (VPA) results indicated that the effects of these factors on bulk soil were 9% of CH4 emissions variations in different treatments, which contributed more than rhizosphere soils' factors. Moreover, random forest analysis results indicated that the abundance of AOB in bulk soil was the most important predictor influencing N2O emissions. The VPA result revealed that the factors in rhizosphere soil could explain more than 28% of the variations in N2O emissions. Our study highlights that rhizosphere soil has a more significant effect than bulk soil on N2O production. Our findings further the understanding of the link between bulk and rhizosphere attributes, and their impact on CH4 and N2O emissions in paddy soils. In summary, we recommend the application of biochar at 500°C and 2% incorporation rate for agricultural production in the area.

Vascular hypo-responsiveness to vasopressors in patients with obstructive jaundice (OJ) is a common anesthetic event, which leads to perioperative complications and increased mortality. The cause of this clinical issue remains unclear. In this study, we estimated the actin cytoskeleton and arterial protein level in the artery of OJ patients by proteomic analysis. Ten patients with OJ due to bile duct diseases or pancreatic head carcinoma were enrolled, while another ten non-jaundice patients with chronic cholecystitis or liver hemangioma as the control group. Vascular reactivity to noradrenaline was measured before anesthesia on the day of surgery. Artery samples in adjacent tissues of removed tumor were collected and evaluated by 2-dimensional electrophoresis. Proteins with differential expression were detected by MALDI-TOF mass spectrometry with immunoblot confirmation. The results confirmed the phenomenon of vascular hypo-reactivity in OJ patients as suppressed aortic response to noradrenaline were existed in these patients. We also found that actin cytoskeleton and several actin-binding proteins were up- or down-regulated in the artery of OJ patients. These proteins changed in OJ patents might be the basic mechanism of vascular hypo-reactivity, further studies to uncover the role of these proteins in OJ is critical for clinical treatment of these patients.

Background: Hepatocellular carcinoma (HCC) is a lethal disease with high relapse and dismal survival rates. Alternative splicing (AS) plays a crucial role in tumor progression. Herein, we aim to integratedly analyze the relapse-associated AS events and construct a signature predicting tumor relapse in stage I-III HCC. Methods: AS events of stage I-III HCC with tumor relapse or long-term relapse-free survival were profiled to identify the relapse-associated AS events. A splicing network was set up to analyze the correlation between the relapse-associated AS events and splicing factors. Cox regression analysis and receiver operating characteristic curve were performed to develop and validate the relapse-predictive AS signature. Single-sample gene set enrichment analysis (ssGSEA) and the ESTIMATE algorithm were used to assess the immune infiltration status of the HCC microenvironment between different risk subgroups. Unsupervised cluster analysis was conducted to assess the relationship between molecular subtypes and local immune status and clinicopathological features. Results: In total, 2441 ASs derived from 1634 mRNA were identified as relapse-associated AS events. By analyzing the proteins involved in the relapse-associated AS events, 1573 proteins with 11590 interactions were included in the protein-protein interaction (PPI) network. In total, 16 splicing factors and 61 relapse-associated AS events with 85 interactions were involved in the splicing network. The relevant genes involved in the PPI network and splicing network were also analyzed by Gene Ontology enrichment analysis. Finally, we established a robust 16-gene AS signature for predicting tumor relapse in stage I-III HCC with considerable AUC values in all of the training cohort, testing cohort, and entire cohort. The ssGSEA and ESTIMATE analyses showed that the AS signature was significantly associated with the immune status of the HCC microenvironment. Moreover, four molecular subgroups with distinguishing tumor relapse modes and local immune status were also revealed. Conclusion: Our study built a novel 16-gene AS signature that robustly predicts tumor relapse and indicates immune activity in stage I-III HCC, which may facilitate the deep mining of the mechanisms associated with tumor relapse and tumor immunity and the development of novel individualized treatment targets for HCC.

Magmatic liquids, including silicate and carbonate melts, are principal agents of mass and heat transfer in the Earth and terrestrial planets, and they play a crucial role in various geodynamic processes and in Earth's evolution. Electrical conductivity data of these melts elucidate the cause of electrical anomalies in Earth's interior and shed light on the melt structure. With the improvement in high-pressure experimental techniques and theoretical simulations, major progress has been made on this front in the past several decades. This review aims to summarize recent advances in experimental and theoretical studies on the electrical conductivity of silicate and carbonate melts of different compositions and volatile contents under high temperature and pressure. The electrical conductivity of silicate melts depends strongly on temperature, pressure, water content and the ratio of non-bridging oxygens to tetrahedral cations (NBO/T). By contrast, the electrical conductivity of carbonate melts exhibits a weak dependence on temperature and pressure due to their fully depolymerized structure. The electrical conductivity of carbonate melts is higher than that of silicate melts by at least two orders of magnitude. Water can increase electrical conductivity significantly and reduce the activation energy of silicate melts. Conversely, this effect is weak for carbonate melts. In addition, the replacement of alkali-earth elements (Ca2+ or Mg2+) with alkali elements causes a significant decrease in the electrical conductivity of carbonate melts. A distinct compensation trend is revealed for the electrical conductivity of silicate and carbonate melts under anhydrous and hydrous conditions. Several important applications of laboratory-based melt conductivity are introduced in order to understand the origin of high-conductivity anomalies in the Earth's mantle. Perspectives for future studies are also provided.

Controlled-release pesticide formulations using natural polymers as carriers are highly desirable owing to their good biocompatibility, biodegradability, and improved pesticide utilization. In this study, the application potential of our previously prepared spinosad/chitosan controlled-release suspension (SCCS) was evaluated through both toxicity and dissipation tests. A comparison with the spinosad suspension concentrate and the commercial spinosad emulsion in water showed that the insecticidal activity of SCCS against Plutella xylostella larvae displayed the best quick-acting performance as well as long-term efficacy of more than 20 days. The 48 h LC50 for a 20-day efficacy was calculated to be 29.36 mg/L. The dissipation behavior of spinosad in the spinosad/chitosan microparticles in soil was found to follow the first-order kinetics, with a relatively shorter half-life (2.1 days) than that observed for the unformulated spinosad (3.1 days). This work showed the positive effect of chitosan on spinosad in improving insecticidal activity and reducing environmental risks in soil, which provided useful information on the application potential of pesticide-carrier systems based on natural polymer materials in crop protection and food safety.

BACKGROUND: Gastric cancer is a prevalent malignant cancer with a high incidence and significantly affects the health of modern people globally. Cisplatin (DDP) is one of the most common and effective chemotherapies for patients with gastric cancer, but DDP resistance remains a severe clinical challenge. AIM: To explore the function of M2 polarized macrophages-derived exosomal microRNA (miR)-588 in the modulation of DDP resistance of gastric cancer cells. METHODS: M2 polarized macrophages were isolated and identified by specific markers using flow cytometry analysis. The exosomes from M2 macrophages were identified by transmission electron microscopy and related markers. The uptake of the PKH67-labelled M2 macrophages-derived exosomes was detected in SGC7901 cells. The function and mechanism of exosomal miR-588 from M2 macrophages in the modulation of DDP resistance of gastric cancer cells was analyzed by CCK-8 assay, apoptosis analysis, colony formation assay, Western blot analysis, qPCR analysis, and luciferase reporter assay in SGC7901 and SGC7901/DDP cells, and by tumorigenicity analysis in nude mice. RESULTS: M2 polarized macrophages were isolated from mouse bone marrow stimulated with interleukin (IL)-13 and IL-4. Co-cultivation of gastric cancer cells with M2 polarized macrophages promoted DDP resistance. M2 polarized macrophages-derived exosomes could transfer in gastric cancer cells to enhance DDP resistance. Exosomal miR-588 from M2 macrophages contributed to DDP resistance of gastric cancer cells. miR-588 promoted DDP-resistant gastric cancer cell growth in vivo. miR-588 was able to target cylindromatosis (CYLD) in gastric cancer cells. The depletion of CYLD reversed miR-588 inhibition-regulated cell proliferation and apoptosis of gastric cancer cells exposed to DDP. CONCLUSION: In conclusion, we uncovered that exosomal miR-588 from M2 macrophages contributes to DDP resistance of gastric cancer cells by partly targeting CYLD. miR-588 may be applied as a potential therapeutic target for the treatment of gastric cancer.

OBJECTIVE: Antipsychotics, in particular olanzapine, are first-line medications for schizophrenia. The prefrontal cortex (PFC) is an important region for antipsychotics' therapeutic effects. The PFC inflammatory and immune pathways are associated with schizophrenia pathogenesis. However, the effect of antipsychotics on the inflammatory and immune pathways in the PFC remains unclear. We aimed to examined the time-dependent effect of olanzapine on inflammatory and immune markers in the PFC of rats. Since the inflammatory and immune pathways are related to endoplasmic reticulum (ER) stress, we further investigated whether or not olanzapine-induced inflammation and immune responses were related to ER stress. METHODS: Expression of pro-inflammatory markers including IkappaB kinase ß (IKKß), nuclear factor kappa B (NFκB), tumor necrosis factor α (TNF-α), interleukin-6 (IL-6) and IL-1ß, and immune-related proteins including inducible nitric oxide synthase (iNOS), toll-like receptor 2 (TLR2) and cluster of differentiation 14 (CD14) were examined by Western blotting. RESULTS: Olanzapine treatments for 1, 8 and 36 days significantly activated the inflammatory IKKß/NFκB signaling, and increased the expression of TNF-α, IL-6, IL-1ß and immune-related proteins such as iNOS, TLR4 and CD14. Olanzapine treatment for 1 day, 8 and 36 days also induced ER stress in the PFC. Co-treatment with an ER stress inhibitor, 4-phenylbutyrate, inhibited olanzapine-induced inflammation and the immune response in the PFC. CONCLUSION: These results suggested olanzapine exposure could be a factor that induces central inflammation and immunological abnormities in schizophrenia subjects. Olanzapine induces PFC inflammation and immune response, possibly via activating ER stress signaling.

Pollution related to pesticides has become a global problem due to their low utilization and non-targeting application, and nanotechnology has shown great potential in promoting sustainable agriculture. Nowadays, mesoporous silica-based nanomaterials have garnered immense attention for improving the efficacy and safety of pesticides due to their distinctive advantages of low toxicity, high thermal and chemical stability, and particularly size tunability and versatile functionality. Based on the introduction of the structure and synthesis of different types of mesoporous silica nanoparticles (MSNs), the multiple roles of mesoporous silica in safe pesticide application using nanotechnology are discussed in this Review: (i) as nanocarrier for sustained/controlled delivery of pesticides, (ii) as adsorbent for enrichment or removal of pesticides in aqueous media, (iii) as support of catalysts for degradation of pesticide contaminants, and (iv) as support of sensors for detection of pesticides. Several scientific issues, strategies, and mechanisms regarding the application of MSNs in the pesticide field are presented, with their future directions discussed in terms of their environmental risk assessment, in-depth mechanism exploration, and cost-benefit consideration for their continuous development. This Review will provide critical information to related researchers and may open up their minds to develop new advances in pesticide application.

PURPOSES: This study was designed to evaluate the effect of preoperative jaundice on long-term prognosis of gallbladder carcinoma (GBC) after radical resection (R0). METHODS: A total of 267 GBC patients who underwent R0 resection from January 2004 to December 2014 were enrolled, including 54 patients with preoperative jaundice and 213 patients without jaundice. The clinicopathological parameters between the two groups were compared, and the correlation between preoperative jaundice and the long-term prognosis was furtherly analyzed. RESULTS: Unilateral and multivariate analyses of 267 GBC patients showed that the depth of tumor invasion (pT stage), lymphatic metastasis, and hepatic invasion were independent prognostic factors. The univariate and multivariate analysis of 54 GBC patients with preoperative jaundice showed that only pT stage was an independent factor for prognosis. Furthermore, the intraoperative blood transfusion and pT stage were significant different between long-term survival (survive for more than 3 years) and those who died within 3 years (P < 0.05). CONCLUSION: Preoperative jaundice was not the independent factor resulting in the poor long-term prognosis of gallbladder carcinoma after R0 resection. The pT stage was the only long-term prognostic factor in all GBC patients regardless of preoperative jaundice.

The study recruited 168 patients diagnosed with major depressive disorder (MDD). The nine-item Patient Health Questionnaire (PHQ-9) and Perceived Deficits Questionnaire for Depression (PDQ-D) were lower and the Digit Symbol Substitution Test (DSST) was higher in the community volunteers than those in MDD patients. Depression-related scores (17-item Hamilton Depression Rating Scale [HAMD-17], Clinical Global Impressions-Severity of Illness Scale [CGI-S], and PHQ-9), functioning-related scores (Sheehan Disability Scale [SDS]), and Work Efficiency and Activity Damage-Specific Health Problems questionnaire work productivity loss were decreased, and the quality of life-related scores (European Quality of life-5 Dimensions [EQ-5D] utility score) were increased in the MDD patients. PDQ-D was decreased and DSST was increased with the increase of follow-up time. Linear regression indicated that cognitive symptoms (PDQ-D and DSST) improved more slowly than depressive symptoms (PHQ-9). At baseline, PDQ-D was related with functioning (SDS and work productivity loss). PDQ-D and DSST were related with EQ-5D utility score. In addition, at month 6, PDQ-D was related with functioning (SDS and work productivity loss) and EQ-5D utility score. Cognitive impairment might be a risk for MDD and MDD-related changes in the functioning and quality of life.

BACKGROUND: Transarterial chemoembolization (TACE) and percutaneous microwave coagulation therapy (PMCT) are commonly used to treat intrahepatic recurrent liver cancers. However, there is no information regarding their effectiveness in patients with recurrent intrahepatic cholangiocarcinoma (ICC) after resection. METHODS: A total of 275 patients with localized recurrent ICC who received either TACE (n = 183) or PMCT (n = 92) were studied. A propensity score matching analysis was performed to compare prognostic impact of TACE and PMCT. Prognostic factors for TACE and PMCT were identified respectively. Predictive nomograms for each TACE and PMCT were developed using the Cox independent prognostic factors and were validated in independent patient groups by receiver operating characteristic curves and area under curve values. RESULTS: Both TACE and PMCT provided curativeness in partial patients (5-year overall survival: 21.4% and 6.1%, respectively), but TACE provided better survival benefit in both overall patients (hazard ratio [HR] = 0.71; 95% confidence interval [CI]: 0.50-0.97; P = 0.034) and propensity score matching analysis (HR = 0.69; 95% CI: 0.47-0.98; P = 0.041). Independent prognostic factors for TACE were tumor size >5 cm, poor differentiation, and major resection, whereas poor differentiation, hepatitis B virus infection, cholelithiasis, and lymph node metastasis were identified for PMCT. Both predictive nomograms for TACE and PMCT were validated to be effective with area under curve values of 0.77 and 0.70, respectively. CONCLUSIONS: TACE provided better survival benefits compared to PMCT. However, there was a disparity in prognostic factors, suggesting evaluation of the two nomograms may be supportive in modality selection. Further prospective validation studies are required for the results to be applied in clinical medicine.

It is of great significance to investigate the spatio-temporal variation in nitrogen and phosphorus nutrients as a mechanism for controlling the sudden increase of algae in eutrophic water. Based on the geostatistical analysis, we studied the sources and occurring forms of nitrogen and phosphorus in different areas of Lake Taihu-a large shallow and eutrophic lake in China. We also examined the spatial distribution of the type of nutrient restriction and its reason by monitoring the sites seasonally from August 2014 to May 2015. The results showed that:① The concentrations of soluble nitrogen and phosphorus were higher in the winter than in other seasons, and they gradually decreased spatially from the northwest to southeast. The concentrations of particulate nitrogen, phosphorus, and chlorophyll a were highest in the summer, and the high-concentration regions in the winter and other seasons were located in the southern and northwestern parts of the lake, respectively. ② The composition of nitrogen and phosphorus in algae-and macrophyte-dominated regions changed substantially with the seasonal change. The algae-dominated regions were dominated by nitrate nitrogen and organic phosphorus in the winter and particulate nitrogen and phosphorus during the other seasons. However, the macrophyte-dominated region was dominated by particulate nitrogen and phosphorus in the winter and ammonia, organic nitrogen, and phosphorus during the other seasons. ③ The ratios of total nitrogen to total phosphorus in the algae-dominated region were greater than 16:1 in the autumn and winter but less than 16:1 in the summer and spring. Meanwhile, the ratios of total nitrogen to total phosphorus in the macrophyte-dominated region increased from less than 16:1 in the autumn and winter to more than 16:1 in the summer and spring. In the algae-dominated region, the spatial variation in the dissolved total nitrogen to total phosphorus ratio was consistent with the ratio of total nitrogen to total phosphorus. In the macrophyte-dominated region, the dissolved total nitrogen to total phosphorus ratio increased from less than 16:1 in the autumn to more than 16:1 in the summer, winter, and spring. The temporal and spatial variation in the ratio of particulate nitrogen to phosphorus was not significant (P>0.05), and this ratio was less than and greater than 16 in algae-and macrophyte-dominated regions, respectively.

The study of pathophysiological mechanisms in human liver disease has been constrained by the inability to expand primary hepatocytes in vitro while maintaining proliferative capacity and metabolic function. We and others have previously shown that mouse mature hepatocytes can be converted to liver progenitor-like cells in vitro with defined chemical factors. Here we describe a protocol achieving efficient conversion of human primary hepatocytes into liver progenitor-like cells (HepLPCs) through delivery of developmentally relevant cues, including NAD + -dependent deacetylase SIRT1 signaling. These HepLPCs could be expanded significantly during in vitro passage. The expanded cells can readily be converted back into metabolically functional hepatocytes in vitro and upon transplantation in vivo. Under three-dimensional culture conditions, differentiated cells generated from HepLPCs regained the ability to support infection or reactivation of hepatitis B virus (HBV). Our work demonstrates the utility of the conversion between hepatocyte and liver progenitor-like cells for studying HBV biology and antiviral therapies. These findings will facilitate the study of liver diseases and regenerative medicine.

BACKGROUND AND AIM: Mucosal traction as a "second hand" in endoscopic submucosal dissection (ESD) is very helpful for ESD operators. The efficacy of using a snare combined with endoclips to assist in ESD (SC-ESD) was evaluated whether it could achieve peroral external traction (PET) and peroral internal traction (PIT) and real-time adjustment of mucosal traction position. This study aimed to preliminarily evaluate its feasibility. METHODS: A snare was fixed using endoclips to the incised mucosal flap that was intended for dissection. Subsequently, the snare was adjusted to achieve mucosal traction. This study included 54 gastroesophageal neoplasias in 54 consecutive patients who underwent SC-ESD. The operative and clinical data were retrospectively collected. RESULTS: Peroral external tractions were performed for 22 esophageal neoplasias, and PETs and PITs were performed for 32 gastric neoplasias. The median (interquartile range) sizes of lesions were 4.3 (2.5) and 4.2 (2.1) cm, operation times were 73.4 (43.8) and 60.0 (45.2) min, and submucosal dissection times were 27.0 (32.0) and 31.0 (34.5) min in the esophagus and stomach, respectively. Mucosal traction position could be effectively adjusted by fixing the snare to the incised mucosal flap intended for dissection during SC-ESD. All lesions in the esophagus and stomach were completely resected, with R0 resection rates of 100%. Besides postoperative bleeding in a patient with gastric neoplasia, no intraoperative and postoperative perforations occurred. CONCLUSIONS: SC-ESD seems to be feasible for resecting gastroesophageal neoplasia, and two types of mucosal tractions (PET and PIT) could be effectively achieved during SC-ESD. Further prospective comparative study with a large sample is warranted.

Thirty-two samples were collected from eight typical areas in Lake Taihu. Three-dimensional excitation-emission matrix fluorescence spectra (EEMs) and a parallel factor analysis (PARAFAC) were applied to investigate the fluorescence properties, the sources, and environmental significance of particulate organic matter (POM) from the overlying water in Lake Taihu in summer. Differences in fluorescence characteristics between POM and DOM (dissolved organic matter), and that in POM between the grass lake and the algal lake were further examined. There are five kinds of fluorophores in Lake Taihu:tyrosine-like fluorophores (C1 and C2), humic-like fluorophores (C3 and C4), and tryptophan-like fluorophores (C5), among which significant correlations were found between C1 and C2, C3 and C4, and C5 and two humic-like fluorophores (C4, C3). By comparing with the fluorescence characteristics of DOM from earlier studies, it is found that there are differences in compositions, sources, and correlations with water quality between POM and DOM in Lake Taihu in summer. In summer, the contribution of endogenous inputs to POM is greater than that of exogenous inputs in Lake Taihu because the ranges in the fluorescence indices, FI, BIX, and HIX, are 1.78-2.35, 0.3-2.7, and 0.8-1.1, respectively. Significant correlations have been found between humic-like fluorescent components and TN, TP, Chla, COD, POC, and SS, suggesting that fluorescence analysis can be used as an important method for a semi-quantitative analysis of nutrients. The protein-like components of the algal lake mainly consist of tryptophan and tyrosine, while that of the grass lake mainly contain tyrosine and a few tryptophan. It is worth noting that, the grass lake contains more tyrosine relative to the algal lake (t test, P < 0.01). Significant positive correlations between the protein-like and the humic-like fluorescence are found in the algal lake, while it is not significant in the grass lake. Significant positive correlations are found between the fluorescent components (protein-like and humic-like) and chlorophyll-a in the algal lake, while the correlation was only found between humic-like components and chlorophyll-a in the grass lake.

Organic matter (OM), rich in carbon, nitrogen, phosphorus and other biogenic elements, has numerous geochemical and ecological functions in all kinds of water bodies, and is highly valued in ecological studies of both marine and inland environments. The formation, transformation, and fate of OM, and its accompanying nutrient regeneration process are key links in the biogeochemical cycling of nutrients in aquatic ecosystems. Furthermore OM, as a provider of nutrients, also plays a very significant role during the outbreak of algal blooms in inland lakes. Two extensive sampling campaigns were conducted at the same site (n=17) in Lake Taihu in February and May of 2015, and 34 (17×2) samples were collected. Excitation-emission matrix (EEM) fluorescence was combined with parallel factor analysis (PARAFAC) to characterize the spatial-temporal variations in the components and sources of particulate (POM) and dissolved (DOM) organic matter. Moreover, the correlations between fluorescence intensity and Chla were studied. Significant differences between the fluorescence peaks, fluorescence positions, and fluorescence intensities of POM and DOM indicated that the components of POM were more complex than those of DOM. Fluorescence intensities and fluorescence indices indicated that POM and DOM possessed endogenous characteristics and that estuaries possessed obvious exogenous characteristics. Five types of fluorophores were present in POM (C1p-C5p) and DOM (C1d-C5d):tyrosine-like fluorophores (C1 and C2), humic-like fluorophores (C3 and C4), and tryptophan-like fluorophores (C5). The fluorescent components of POM and DOM differed significantly. C3p was the dominant fluorophore in POM during spring and winter, and fluorescence intensities of C1p-C5p were higher in the Meiliang Bay and the central part of thelake than in other areas. C2d, C1d, and C5d were the dominant fluorophores in DOM during spring and winter, and fluorescence intensities of C1d-C5d were higher in the Zhushan Bay than in other areas. In spring, positive correlations were observed between C1d and C2d (P<0.01), C3d and C4d (P<0.01), C1p and C4p (P<0.05), and C2p and C5p (P<0.01). Remarkably, C3p and C3d (P<0.01) and C3p and C4d (P<0.05) were positively correlated, suggesting that these two forms of OM were correlated. Similarly, highly significant, positive correlations were observed between C1d and C2d (P<0.01), C3d and C4d (P<0.01), C1p and C2p (P<0.01), and C1d and C2p (P<0.01) during winter. Furthermore, Chla and C5d were significantly and positively correlated (P<0.01). However, both Chla/C2d and Chla/C3p were negatively correlated in spring. Furthermore, Chla and C4d were negatively correlated in winter.

The traditional, retrocolic/antegastric Roux-en-Y choledochojejunostomy is technically complicated, and the incidence of postoperative complications remains high. Here we report the outcome of 59 consecutively treated patients (study group, SG) that underwent a new choledochojejunostomy method in which the jejunal loop is passed behind the antrum pyloricum (retrogastric route). A retrospective comparison was made between this group of patients and 187 patients (control group, CG) that underwent conventional Roux-en-Y choledochojejunostomy (antegastric route). Baseline clinicopathological characteristics were similar in both groups, except for the BMI, which was significantly higher in the SG. The time spent on constructing the anastomosis, as well as overall postoperative complications, did not differ between groups. Compared with the CG, the incidence of postoperative delayed gastric emptying was decreased in the SG, and the time elapsed before the patients' first postoperative liquid food consumption was shorter. We ascribe these beneficial effects to the superiority of the modified, retropyloric choledochojejunostomy approach, and propose that this surgical technique is particularly suitable for obese patients, especially those with a short ascending bowel loop.
