Background: Peanut is an important source of dietary protein for human beings, but it is also recognized as one of the eight major food allergens. Binding of IgE antibodies to specific epitopes in peanut allergens plays important roles in initiating peanut-allergic reactions, and Ara h 2 is widely considered as the most potent peanut allergen and the best predictor of peanut allergy. Therefore, Ara h 2 IgE epitopes can serve as useful biomarkers for prediction of IgE-binding variations of Ara h 2 and peanut in foods. This study aimed to develop and validate an IgE epitope-specific antibodies (IgE-EsAbs)-based sandwich ELISA (sELISA) for detection of Ara h 2 and measurement of Ara h 2 IgE-immunoreactivity changes in foods. Methods: DEAE-Sepharose Fast Flow anion-exchange chromatography combining with SDS-PAGE gel extraction were applied to purify Ara h 2 from raw peanut. Hybridoma and epitope vaccine techniques were employed to generate a monoclonal antibody against a major IgE epitope of Ara h 2 and a polyclonal antibody against 12 IgE epitopes of Ara h 2, respectively. ELISA was carried out to evaluate the target binding and specificity of the generated IgE-EsAbs. Subsequently, IgE-EsAbs-based sELISA was developed to detect Ara h 2 and its allergenic residues in food samples. The IgE-binding capacity of Ara h 2 and peanut in foods was determined by competitive ELISA. The dose-effect relationship between the Ara h 2 IgE epitope content and Ara h 2 (or peanut) IgE-binding ability was further established to validate the reliability of the developed sELISA in measuring IgE-binding variations of Ara h 2 and peanut in foods. Results: The obtained Ara h 2 had a purity of 94.44%. Antibody characterization revealed that the IgE-EsAbs recognized the target IgE epitope(s) of Ara h 2 and exhibited high specificity. Accordingly, an IgE-EsAbs-based sELISA using these antibodies was able to detect Ara h 2 and its allergenic residues in food samples, with high sensitivity (a limit of detection of 0.98 ng/mL), accuracy (a mean bias of 0.88%), precision (relative standard deviation < 16.50%), specificity, and recovery (an average recovery of 98.28%). Moreover, the developed sELISA could predict IgE-binding variations of Ara h 2 and peanut in foods, as verified by using sera IgE derived from peanut-allergic individuals. Conclusion: This novel immunoassay could be a user-friendly method to monitor low level of Ara h 2 and to preliminary predict in vitro potential allergenicity of Ara h 2 and peanut in processed foods.

Hydrogels have entered the spotlight for applications in soft electronics. It is essential and challenging to obtain hydrogels that can function properly under varying environmental circumstances, that is, 30-90% relative humidity (RH) and -20 to 40 °C due to their intrinsic nature to lose and absorb water upon variations in humidity and temperature. In this work, a green solvent, solketal, is introduced into poly 3-dimethyl-2-(2-methylprop-2-enoyloxy)ethyl azaniumyl propane-1-sulfonate (poly(DMAPS)) zwitterionic hydrogels. Compared to glycerol, solketal endows hydrogels with greater possibility for further modification as well as improved water content and mechanical performance consistency over 30-90% RH. Encouragingly, the optimized hydrogel demonstrates its unique merits as a dielectric layer in iontronic sensors, featuring non-leaky ions, high sensitivity (1100 kPa-1 ), wide humidity, and temperature range applicability. A wide-humidity range healable and stretchable electrode is attained by combining the hydrogel substrate with Ag paste. A full-device healable and highly-sensitive sensor is developed. This study is a pioneering work that tackles the broad humidity range applicability issue of hydrogels, and demonstrates the ion-leakage-free ionic skins with zwitterionic dielectrics. The outcomes of the study will considerably promote advancements in the fields of hydrogel electronics and iontronic sensors.

This study sought to explore the effects and potential mechanisms of dietary supplementation with isoquinoline alkaloids (IA) from Macleaya cordata to alleviate lipopolysaccharide (LPS)-induced intestinal epithelium injury in broilers. A total of 486 1-day-old broilers were assigned at random to a control (CON) group, LPS group, and LPS+IA group in a 21-d study. The CON and LPS groups received a basal diet, while the LPS+IA group received a basal diet supplemented with 0.6 mg/kg IA. At 17, 19, and 21 days of age, the LPS and LPS+BP groups were injected intraperitoneally with LPS, and the CON group was intraperitoneally injected equivalent amount of saline solution. The results manifested that LPS injection caused intestinal inflammation and lipid peroxidation, disrupted intestinal barrier and function, and increased the abundance of harmful microorganisms. However, dietary IA supplementation alleviated LPS-induced adverse changes in intestinal morphology, apoptosis, mucosal barrier integrity, cecum microorganisms, and homeostasis disorder by decreasing inflammatory cytokines and enhancing antioxidant-related genes expressions; inhibited LPS-induced increases in TLR4 and NF-κB expressions and decreases in Nrf2 and GPX1 genes expressions. Our findings indicated that Macleaya cordata IA addition attenuated LPS-induced intestinal epithelium injury and disorder of intestinal homeostasis by enhancing the anti-inflammatory and antioxidant capacity of broiler chickens possibly via co-regulating TLR4/MyD88/NF-κB and Nrf2 signaling pathways.

The primary tools for developing pupils' creativity, capacity for verbal expression, and spiritual growth are literary reading and writing. Literature is a sort of art that elicits feelings and expresses the author's comprehension and outlook on social life via the use of language. Reading and writing literary works helps students develop their aesthetic sensibilities and capacity to create compelling images, as well as their spirituality and wisdom. This study suggests a data mining-based optimal design approach for analyzing association rules of influencing aspects of interest in literary courses. To increase the frequency and accuracy of data mining, association rules are used to obtain the association mapping relationship between data sets of influencing factors of interest in literature courses. Rough set theory is then used to distinguish between the feature sets of data sets in the same subspace and different subspaces. To identify the most prevalent factor that affects the interest of the curriculum's literary components and then to conduct simulation testing and analysis. The proposed arithmetic has a particular accuracy, which is 8.25% greater than the conventional arithmetic, according to simulation findings. This outcome demonstrates in full how the enhanced arithmetic decreases the amount of records in the scanning database by grouping and compressing the database, hence lowering the scanning time and pruning before the connection process of Liu arithmetic. Classical literary works are without a doubt the most valuable resources to feed, edify, forge, and grow the spirit, soul, and personality of contemporary individuals throughout history and in all nations. Education through literature develops one's character, spirit, emotions, and aesthetic sense. The importance of reaffirming the significant place of literature education in contemporary national basic education cannot be overstated.

Traditional alternating-current-driven electroluminescent (AC-EL) devices adopting a sandwich structure are commonly used in solid-state lighting and displays, while the emerging coplanar-electrode alternating-current-driven light-emitting variants manifest excellent application prospects in intelligent, multifunctional, and full-color displays, and sensing purposes. In this work, an asymmetrically enhanced coplanar-electrode AC-EL device with a universal and straightforward architecture is designed based on the impedance adjustment strategy. This newly devised asymmetric structure extends the functionalities of the coplanar-electrode AC-EL devices by overcoming the bottlenecks of complicated patterning procedures and high driving voltages of symmetric configuration. The developed device design enables a new type of information encryption and ultrahighly stretchable patterned displays. Notably, the novel encryption appliances demonstrate feasible encryption/decryption features, multiple encryptions, and practical applicability; the biaxially stretchable display devices achieve the highest tensile performance in the field of stretchable electroluminescent pattern displays, and outperform the ultrahighly stretchable sandwich devices in terms of simple patterning process, higher brightness, wider color gamut, and long-term stability. The proposed configuration opens up new avenues for AC-EL devices toward a plethora of smart applications in wearable electronics with intelligent displays, dynamic interaction of human-machine interface, and soft robotics.
