RESUMEN

The desert is a harsh habitat for flora and microbial life due to its aridness and strong radiation. In this study, we constructed the first complete and deeply annotated genome of the genus Pontibacter (Pontibacter korlensis X14-1(T) = CCTCC AB 206081(T), X14-1). Reconstruction of the sugar metabolism process indicated that strain X14-1 can utilize diverse sugars, including cellulose, starch and sucrose; this result is consistent with previous experiments. Strain X14-1 is also able to resist desiccation and radiation in the desert through well-armed systems related to DNA repair, radical oxygen species (ROS) detoxification and the OstAB and TreYZ pathways for trehalose synthesis. A comparative transcriptomic analysis under gamma radiation revealed that strain X14-1 presents high-efficacy operating responses to radiation, including the robust expression of catalase and the manganese transport protein. Evaluation of 73 novel genes that are differentially expressed showed that some of these genes may contribute to the strain's adaptation to radiation and desiccation through ferric transport and preservation.

Acinetobacter johnsonii A2 isolated from the natural community of Laguna Azul (Andean Mountains at 4,560 m above sea level), Serratia marcescens MF42, Pseudomonas sp. strain MF8 isolated from the planktonic community, and Cytophaga sp. strain MF7 isolated from the benthic community from Laguna Pozuelos (Andean Puna at 3,600 m above sea level) were subjected to UV-B (3,931 J m-2) irradiation. In addition, a marine Pseudomonas putida strain, 2IDINH, and a second Acinetobacter johnsonii strain, ATCC 17909, were used as external controls. Resistance to UV-B and kinetic rates of light-dependent (UV-A [315 to 400 nm] and cool white light [400 to 700 nm]) and -independent reactivation following exposure were determined by measuring the survival (expressed as CFU) and accumulation of cyclobutane pyrimidine dimers (CPD). Significant differences in survival after UV-B irradiation were observed: Acinetobacter johnsonii A2, 48%; Acinetobacter johnsonii ATCC 17909, 20%; Pseudomonas sp. strain MF8, 40%; marine Pseudomonas putida strain 2IDINH, 12%; Cytophaga sp. strain MF7, 20%; and Serratia marcescens, 21%. Most bacteria exhibited little DNA damage (between 40 and 80 CPD/Mb), except for the benthic isolate Cytophaga sp. strain MF7 (400 CPD/Mb) and Acinetobacter johnsonii ATCC 17909 (160 CPD/Mb). The recovery strategies through dark and light repair were different in all strains. The most efficient in recovering were both Acinetobacter johnsonii A2 and Cytophaga sp. strain MF7; Serratia marcescens MF42 showed intermediate recovery, and in both Pseudomonas strains, recovery was essentially zero. The UV-B responses and recovery abilities of the different bacteria were consistent with the irradiation levels in their native environment.

The ability of oysters to purge themselves of microbial contaminants was investigated by identifying the microorganisms retained by oysters after they have been subjected to ultraviolet (UV) light-treated seawater. A UV intensity of 960 muw per min per cm(2) reduced the microbial count of seawater from 263 to 13 per ml. The coliform multitube test (MPN) was reduced from a high of 17 to <0.18 per 100 ml. Over 75% of the microorganisms found in treated seawater were Acinetobacter/Moraxella, Vibrio/Pseudomonas type II, and Flavobacterium/Cytophaga. With the exception of coliforms, the microbial composition of oysters subjected to UV-treated seawater remained at levels comparable to the control oysters held in untreated seawater. Total counts ranged between 10(3) and 10(5)/g. The microorganism most frequently encountered were Flavobacterium/Cytophaga, Vibrio/Pseudomonas type II, Pseudomonas type III or IV, Acinetobacter/Moraxella, gram-positive cocci and Bacillus. Together they comprised over 90% of the flora. Coagulase-positive, deoxyribonuclease-positive, and beta-hemolytic cocci were found in some samples, as were V. parahaemolyticus, V. aliginolyticus, and Aeromonas species.
