Abstract Understanding morphological and physiological changes under different light conditions in native fruit species in juveniles' stage is important, as it indicate the appropriate environment to achieve vigorous saplings. We aimed to verify growth and morphophysiological changes under shade gradient in feijoa (Acca sellowiana (O. Berg) Burret) to achieve good quality saplings adequate to improve cultivation in orchards. The saplings were grown for twenty-one-month under four shading treatments (0%, 30%, 50%, and 80%). Growth, photosynthetic pigments, gas exchanges, chlorophyll fluorescence, and leaf anatomy parameters were evaluated. Saplings under full sun and 30% shade had higher height and diameter growth and dry mass accumulation due to higher photosynthesis rate. As main acclimatization mechanisms in feijoa saplings under 80% shade were developed larger leaf area, reduced leaf blade thickness, and enhanced quantum yield of photosystem II. Even so, the net CO2 assimilation and the electron transport rate was lower and, consequently, there was a restriction on the growth and dry mass in saplings under deep shade. Therefore, to obtain higher quality feijoa saplings, we recommend that it be carried out in full sun or up to 30% shade, to maximize the sapling vigor in nurseries and, later, this light environment can also be used in orchards for favor growth and fruit production.


Resumo A verificação de mudanças morfológicas e fisiológicas sob diferentes condições luminosas em espécies frutíferas nativas em estágio juvenil é importante, uma vez que indicam o ambiente adequado para a formação de mudas com alto vigor. Objetivou-se verificar o crescimento e as alterações morfofisiológicas sob gradiente de sombreamento em mudas de feijoa (Acca sellowiana (O. Berg) Burret) para obter mudas de boa qualidade, adequadas para fomentar os plantios da espécie em pomares. As mudas foram cultivadas por vinte e um meses sob quatro tratamentos de sombreamento (0%, 30%, 50% e 80%). Foram avaliados parâmetros de crescimento, pigmentos fotossintéticos, trocas gasosas, fluorescência da clorofila e anatomia foliar. Mudas a pleno sol e 30% de sombra apresentaram maior crescimento em altura, diâmetro e acúmulo de massa seca, devido à maior taxa de fotossíntese. Como principais mecanismos de aclimatação sob 80% de sombra, as mudas desenvolveram maior área foliar, redução da espessura do limbo foliar e aumento do rendimento quântico do fotossistema II. Mesmo assim, a assimilação líquida de CO2 e a taxa de transporte de elétrons foram menores e, consequentemente, houve restrição ao crescimento e acúmulo de massa seca das mudas no maior nível de sombreamento. Portanto, para a obtenção de mudas de feijoa de maior qualidade, recomendamos que seja realizada a pleno sol ou até 30% de sombra, para maximizar o vigor das mudas em viveiros e, posteriormente, este ambiente de luz também pode ser utilizado em pomares para favorecer o crescimento e a produção de frutos.

The aims of this work were to characterize the damage caused by Paraulaca dives in feijoa (Acca sellowiana) and develop a diagrammatic scale to evaluate the severity of herbivory. The evaluations were carried out in a feijoa progenies orchard with eight-year-old. The beetles caused damage mainly on leaves, from October to December (spring season). The distribution of beetles in the orchard occurred randomly, not following a defined pattern of incidence. The diagrammatic scale of herbivory severity was represented by seven levels (1%, 3%, 5%, 7%, 15%, 32%, and 55% of the leaf area consumed). The use of this diagrammatic scale significantly increased the precision and accuracy of severity estimates by inexperienced evaluators. This can help in strategies to control this pest to expand the cultivation of feijoa in Brazil.

Brazil is a large country with high biodiversity in its different regions. However, species of native fruits widely reported in the southern Brazil have not been properly explored so far, remaining underutilized by the food industry. This study evaluated the polyphenolic profile, as well as the composition of organic acids and sugars of the pulps of the feijoa, and the uvaia from southern brazilian highlands. The uvaia pulp showed the highest total polyphenol content and the highest antioxidant capacity by the methods used. The polyphenol (+)-catechin (6.54 mg 100g-1) was the major phenolic compound in uvaia pulp, which has not yet been reported in the literature for fruits of other regions. In addition, the feijoa pulp stood out for the presence of (-)-epicatechin (18.29 mg 100g-1). The pulps of native fruits in this study only quantified values for citric and malic acids. Malic acid (553.00 mg 100g-1) was the main acid in the uvaia pulp, and citric acid (455.60 mg 100g-1) was the main acid in the feijoa pulp. It is possible to note that the feijoa pulp showed the highest total sugar content (11.14 g 100g-1) and was the only pulp that contained sucrose. The uvaia pulp, conversely, showed fructose (3.10 g 100g-1) as the main sugar. The results obtained in this study contributed to the valuation and conservation of the species investigated, representing a promising alternative for the use of these native fruits in the development of new products.


O Brasil é um grande país e com elevada biodiversidade em suas diferentes regiões. Entretanto, espécies de frutas nativas amplamente encontradas no sul do Brasil ainda não foram devidamente exploradas, permanecendo desconhecidas e subutilizadas pela indústria de alimentos. Nesse sentido, o objetivo deste estudo foi avaliar o perfil fenólico, bem como a composição de ácidos orgânicos e de açúcares de polpas de feijoa (Acca sellowiana) e de uvaia (Eugenia pyriformis) provenientes do planalto sul brasileiro. A polpa de uvaia apresentou o maior teor de polifenóis totais e a maior capacidade antioxidante pelos métodos utilizados. A (+)-catequina (6.54 mg 100g-1) foi o principal composto fenólico da polpa da uvaia, o que ainda não foi relatado na literatura para frutas de outras regiões. Além disso, a polpa da feijoa se destacou pela presença de (-)-epicatequina (18.29 mg 100g-1). Dentre os ácidos orgânicos avaliados, as polpas de frutas nativas apresentaram valores para os ácidos cítrico e málico, sendo o ácido málico (553.00 mg 100g-1) o principal na polpa de uvaia, e o cítrico (455.60 mg 100g-1) na polpa de feijoa. Em relação aos açúcares, a polpa da feijoa apresentou o maior teor de açúcar total (11.14 g 100g-1) e foi a única polpa que continha sacarose. Já a polpa da uvaia apresentou a frutose (3.10 g 100g-1) como principal açúcar. Os resultados obtidos neste estudo contribuem para a valorização e conservação das espécies investigadas, representando uma alternativa promissora para a utilização dessas frutas nativas no desenvolvimento de novos produtos.

The present study aimed to evaluate the developmental phases of feijoa fruits associated with infestation by Anastrepha fraterculus (Wiedemann) (Diptera: Tephritidae) and the nonpreference of fruit flies for ripe fruits of feijoa. Two tests were carried out. In the first trial, we evaluated the association between the developmental stages of feijoa fruits and infestation by A. fraterculus. To accomplish this, fruits at five different stages of development, from green to ripe, were examined from the Alcântara cultivar. In the second trial, we evaluated the nonpreference of A. fraterculus for ripe fruits of Alcântara, Helena, Mattos and access 2316 cultivars. Physicochemical analyses were performed on the fruits of both trials. In the second test, analyses of polyphenol index and tannin concentration were also performed. Feijoa fruits in stage I are the likely targets of attack by A. fraterculus. The fruits of Alcântara cultivar from stage II, with 39 mm of transversal diameter and soluble solids, presented four puparium / fruit. The fruits of Alcântara and access 2316 cultivars showed greater and lesser infestation, respectively. The fruits of access 2316 also showed a higher concentration of tannins compared to the three commercial cultivars tested, which may have conferred the greatest protection against infestation by A. fraterculus. Feijoa fruits from Alcântara and Mattos cultivars showed high susceptibility to infestation by A. fraterculus, requiring management practices to guarantee commercial production.


O presente estudo teve como objetivos avaliar as fases de desenvolvimento de frutos de feijoa associadas à infestação por Anastrepha fraterculus (Wiedemann) (Diptera: Tephritidae) e a não preferência por frutos maduros. No primeiro ensaio, foi avaliado a influência dos cinco estágios de desenvolvimento dos frutos de feijoa na infestação por A. fraterculus na cultivar Alcântara. No segundo ensaio, foi avaliado a não preferência de A. fraterculus por frutos maduros das cultivares Alcântara, Helena, Mattos e acesso 2316. Foram realizadas análises físico-químicas nos frutos de ambos os ensaios. No segundo ensaio, também foram realizadas análises do índice de polifenóis e concentração de taninos. Frutos de feijoa no estágio I são os prováveis alvos de ataque por A fraterculus. Os frutos do cultivar Alcântara, a partir do estádio II, quando apresentavam 39 mm de diâmetro transversal e sólidos solúveis, apresentaram quatro pupários/fruto. Os frutos da cultivar Alcântara e do acesso 2316 apresentaram maior e menor infestação, respectivamente. Os frutos do acesso 2316 também apresentaram a maior concentração de taninos do que nas três cultivares comerciais testadas, o que pode ter conferido a maior proteção contra infestação por A. fraterculus. Frutos de feijoa das cultivares Alcântara e Mattos apresentaram alta suscetibilidade à infestação por A. fraterculus, o que requer práticas de manejo para garantir a produção comercial.

The aim of this work was to evaluate the effect of the essential oils of Syzygium aromaticum, Cymbopogon citratus, Eucalyptus citriodora and Rosmarinus officinalis on the mycelial development of the fungus Colletotrichum sp. in fragments of Feijoa sellowiana fruits. The essential oils were incorporated in the PDA (Potato-Dextrose-Agar) medium in the concentrations of 250, 500 and 1000 ppm, 0 ppm (PDA only) (negative control), and fungicide fluazinam 1% (positive control). The area under the mycelial growth curve (AUMGC) and percent inhibition of mycelial growth (PIMG) were calculated. In the second evaluation, fruits fragments bordering the disease symptom were immersed in essential oils aqueous solution of S. aromaticum, C. citratus, and E. citriodora, at the concentration of 5000 ppm, 0 ppm (water only - negative control) and fluazinam 1% (positive control). The immersion times in the treatments were: 2, 4, 8, 12 and 24 hours, with subsequent incubation in Agar-Agar medium at 25°C. This evaluation was performed daily for 15 days, observing the moment of fungal germination through the emission of the mycelium. It was verified from the obtained results that all treatments reduced the fungal growth, and the essential oils of C. citratus and S. aromaticum totally inhibited its growth from the dose 500 and 1000 ppm, respectively. Regarding the test on fruit fragments, the essential oil of S. aromaticum at the immersion times of 12 and 24 hours was effective in inhibiting the fungus until the 15th day of evaluation.

Acca sellowiana (feijoa) and Eugenia involucrata (cherry) are fruits species of Brazilian biodiversity (Myrtaceae family). In this study, a sampling process was used with three different harvesting sites. The composition of phenolic compounds of these fruits was determined by HPLC-DAD-MS/MS. Moreover, the antioxidant capacity of hydroethanolic extracts against hydrogen peroxide (H2O2), hydroxyl (OH), peroxyl (ROO-) and ABTS radicals was evaluated. Thirty and twenty-seven phenolic compounds were identified in feijoa and cherry, respectively. The major phenolic compounds found were pedunculagin isomer (5040.87, 3443.66 and 1324.95 µg·g-1) in feijoa and procyanidin (1406.54, 1888.00 and 1380.64 µg·g-1) in cherry. Hydroethanolic extract of these fruits was a potent scavenger of free radicals and excellent source of phenolic compounds. In hydroethanolic extracts of feijoa, the phenolic content increased by around 50%, while in the cherry the content was similar to that found in the fruit. For ORAC method, sample 2 of feijoa and cherry showed values of 383 and 126 µM·TE·g-1, respectively, featuring the highest antioxidant capacity. This study is the first to report the identification of castalagin, catechin and epicatechin in feijoa, and rutin in cherry. Besides, the health benefits, these fruits can contribute to biodiversity conservation.

BACKGROUND: Feijoa [Acca sellowiana (Berg) Burret] is a Brazilian native fruit with few commercial-level plantations and high agroindustrial potential. A genotype evaluation experiment was conducted from 1996 onward, aiming to obtain fruits based on the agronomical parameters; however, the selection based on chemical composition had not been evaluated with respect to developing a new cultivar. Accordingly, the present study aimed to discriminate seven accessions of feijoa in terms of nutritional composition, phenolic compounds and antioxidant activity using multivariate analysis (principal component analysis and multivariate contrast), targeting the potential production of a new cultivar with better nutritional value and high antioxidant capacity. RESULTS: Feijoa husk presented high content of ashes, lipids, proteins, carbohydrates, phenolic compounds and antioxidant activity compared to feijoa pulp. However, only feijoa pulp was selected to multivariate analysis because it is the fruit edible part. Data variability was explained in 78% and the feijoa pulp accessions were discriminated into four groups related to their characteristics. The accession 5 discrimination can be explained by the high content of ashes, carbohydrates, soluble solids, phenolic compounds and antioxidant activity. Accession 6 was also discriminated by the high content of total acidity, pH and proteins, as well as a low content of soluble solids. CONCLUSION: Feijoa accessions may be indicated for increasing plant selection via hybridization with the other accessions, aiming to produce new cultivars with better nutritional composition and antioxidant capacity. For example, accession 5 is the most suited fruit for human consumption and is a potential plant with respect to becoming a new cultivar. © 2020 Society of Chemical Industry.

Feijoa is a subtropical bush of the Myrtaceae family. It has unique fruit with organoleptic properties that make it an exotic fruit. Head space solid phase microextraction (HS-SPME) with a 65 µm divinylbenzene/polydimethylsiloxane (DVB/PDMS) fiber and gas chromatography coupled to mass spectrophotometry (GC-MS) was used to study the volatile fraction of feijoa fruit cultured in Caldas, Colombia. The profile analyzed included 134 volatile organic compounds (VOCs). 127 VOCs were classified based on the functional group using the spectral and structural networks correlation analysis. Methyl, ethyl and (Z)-3-hexenyl benzoate with 50% of the volatile composition, were the main compounds. Biosynthesis of the volatilome of feijoa fruit was associated with five main metabolic pathways. This study represents the first analysis of feijoa fruit commercialized in the region. This is an innovator view in elucidation of metabolic pathways that represent the biochemistry of the aroma of this fruit.

Past and contemporary human actions are causing numerous changes in patterns and processes at various ecosystem scales and trophic levels, including unintended downstream changes, such as species interactions. In its native range Acca sellowiana (Feijoa) combines some characteristics of human interactions: incipient domestication, restricted to subtropical Atlantic Forest highlands, associated with the threatened conifer Araucaria angustifolia (Araucaria), within a domesticated landscape with anthropogenic forest patches, and provides fruit at a time of resource shortage (Araucaria seeds-pinhão). We quantify the trophic relationships between Feijoa and vertebrates, and evaluate the influences on interactions caused by environmental variations, Feijoa domestication evidences, spatial distance and fruit availability. In four sites within protected areas, we selected 28 focal individuals of Feijoa (seven/site) and collected three temporal replicas between 2015 and 2016, when we measured productivity and frugivory via 45-second videos taken with camera traps. Using ecological network, rarefaction curves and variation partitioning analyses, we evaluate the frugivory network topology, the spatiotemporal structure of communities in relation to fruit availability and the influence of predictive variables on frugivory. We found a large spatiotemporal variation in productivity of Feijoa and that 20 species consumed Feijoa fruits, with a species degree of 2.8 (±5.7) and average Feijoa degree of 14.4 (±10.1), in a modular network with intermediary connectance. Rarefaction curves showed that richness and the independent records are congruent with the fruit amount. Variation partitioning showed that, for the focal individuals, canopy area, green coverage, patch size and distance to water influenced frugivory, and the Feijoa domestication influenced significantly the mammalian frugivory. Feijoa is an important resource that provides food during the time of year when Pinhão is absent, and attracts frugivores, maintain the residual diversity of vertebrates contributing to the structure of communities in highlands. Our insights allowed us to evaluate the magnitude of the interactions between vertebrates and an incipient domesticated tree, in a cultural landscape and highly threatened environment, under a basal foodweb approach with implications for bottom-up and top-down forces. The results contribute to understanding animal-plant relationships, including concepts that can be replicated for other sessile prey and mobile predators in any region or habitat under different gradients of management. Thus, this work shows how human actions can change not only patterns of distribution and abundance but also the diversity and direction of interspecific interactions among species.

The objective of this paper was to identify the direct and indirect effects of feijoa fruits (Acca sellowiana) traitson pulp weight, in order to use these traits in indirect genotypes selection. Fruits of five feijoa plants were collected in Rio Grande do Sul, in the years of 2009, 2010 and 2011. Six traits were evaluated: diameter, length, total weight, pulp weight, peel thickness and number of seeds per fruit. In the path analysis, with or without ridge regression, pulp weight was considered as the basic variable, and the other traits were considered as explanatory variables. Total weight and fruit diameter had high direct effect, and are the main traits associated with pulp weight. These traits may serve as criteria for indirect selection to increase feijoa pulp weight, since they are easy to be measured.


O objetivo deste trabalho foi identificar os efeitos diretos e indiretos de caracteres dos frutos de goiabeira-serrana (Accasellowiana) sobre o peso de polpa, visando à utilização desses caracteres para a seleção indireta de genótipos. Foram coletados frutos de cinco grupo de plantas de goiabeira-serrana no Rio Grande do Sul, nos anos de 2009, 2010 e 2011. Seis caracteres foram avaliados: diâmetro, comprimento, peso total, peso de polpa, espessura da casca e número de sementes por fruto. Na análise de trilha com e sem regressão em crista para controle da multicolinearidade, o peso de polpa foi considerado variável básica e os demais caracteres considerados explicativos. O peso total, seguido do diâmetro de fruto, tiveram efeito direto elevado e são os principais caracteres associados ao peso de polpa. Estes caracteres podem servir de critérios de seleção indireta para aumentar o peso de polpa de goiabeira-serrana, pois são de fácil mensuração.

RESUMEN

The objective of this paper was to identify the direct and indirect effects of feijoa fruits (Acca sellowiana) traitson pulp weight, in order to use these traits in indirect genotypes selection. Fruits of five feijoa plants were collected in Rio Grande do Sul, in the years of 2009, 2010 and 2011. Six traits were evaluated: diameter, length, total weight, pulp weight, peel thickness and number of seeds per fruit. In the path analysis, with or without ridge regression, pulp weight was considered as the basic variable, and the other traits were considered as explanatory variables. Total weight and fruit diameter had high direct effect, and are the main traits associated with pulp weight. These traits may serve as criteria for indirect selection to increase feijoa pulp weight, since they are easy to be measured.(AU)


O objetivo deste trabalho foi identificar os efeitos diretos e indiretos de caracteres dos frutos de goiabeira-serrana (Accasellowiana) sobre o peso de polpa, visando à utilização desses caracteres para a seleção indireta de genótipos. Foram coletados frutos de cinco grupo de plantas de goiabeira-serrana no Rio Grande do Sul, nos anos de 2009, 2010 e 2011. Seis caracteres foram avaliados: diâmetro, comprimento, peso total, peso de polpa, espessura da casca e número de sementes por fruto. Na análise de trilha com e sem regressão em crista para controle da multicolinearidade, o peso de polpa foi considerado variável básica e os demais caracteres considerados explicativos. O peso total, seguido do diâmetro de fruto, tiveram efeito direto elevado e são os principais caracteres associados ao peso de polpa. Estes caracteres podem servir de critérios de seleção indireta para aumentar o peso de polpa de goiabeira-serrana, pois são de fácil mensuração.(AU)

The Anastrepha fraterculus (Wiedemann) (Diptera: Tephritidae) is the main pest of fruit trees grown in temperate climates in the southern region of Brazil. The objective of this work was to evaluate the efficiency of the major commercial food lures used in Brazil for trapping and monitoring of A. fraterculus in plum, pear, and feijoa orchards. The assessed lures were hydrolyzed proteins of animal origin (CeraTrap) and plant origin (BioAnastrepha), torula yeast + borax (Torula), and grape juice. Response variables included the rate of adult capture (flies per trap per day, FTD) and the percentage of females captured. We also evaluated the number of times the weekly capture rate exceeded the traditional threshold of 0.5 FTD for each lure. Traps baited with grape juice, currently used for monitoring A. fraterculus in Southern Brazil, captured fewer adults and a lower percentage of females compared with the other lures. CeraTrap trapped a greater number of A. fraterculus adults and, in some cases, a lower percentage of females compared with the other lures in pears. Traps baited with CeraTrap had greater capture rates (FTD), particularly during the stages of fruit maturation and harvest, and even in years with low population density of A. fraterculus, thus demonstrating greater sensitivity in the detection of this pest. These results show that, in order to detect and monitor the presence of A. fraterculus in plum, feijoa, and pear crops, protein-based lures are superior to grape juice, especially the animal protein CeraTrap.

Given their distribution, importance, and richness, Myrtaceae species comprise a model system for studying the evolution of tropical plant diversity. In addition, chloroplast (cp) genome sequencing is an efficient tool for phylogenetic relationship studies. Feijoa [Acca sellowiana (O. Berg) Burret; CN: pineapple-guava] is a Myrtaceae species that occurs naturally in southern Brazil and northern Uruguay. Feijoa is known for its exquisite perfume and flavorful fruits, pharmacological properties, ornamental value and increasing economic relevance. In the present work, we reported the complete cp genome of feijoa. The feijoa cp genome is a circular molecule of 159,370 bp with a quadripartite structure containing two single copy regions, a Large Single Copy region (LSC 88,028 bp) and a Small Single Copy region (SSC 18,598 bp) separated by Inverted Repeat regions (IRs 26,372 bp). The genome structure, gene order, GC content and codon usage are similar to those of typical angiosperm cp genomes. When compared to other cp genome sequences of Myrtaceae, feijoa showed closest relationship with pitanga (Eugenia uniflora L.). Furthermore, a comparison of pitanga synonymous (Ks) and nonsynonymous (Ka) substitution rates revealed extremely low values. Maximum Likelihood and Bayesian Inference analyses produced phylogenomic trees identical in topology. These trees supported monophyly of three Myrtoideae clades.

Morphological disorders in a relevant portion of emerged somatic embryos have been a limiting factor in the true-to-type plantlet formation in Acca sellowiana. In this sense, the present study undertook a comparison between normal phenotype and off-type somatic plantlets protein profiles by means of the 2-D DIGE proteomics approach. Off-type and normal phenotype somatic plantlets obtained at 10 and 20 days conversion were evaluated. Results indicated 12 exclusive spots between normal and off-type plantlets at 10 days conversion, and 17 exclusive spots at 20 days conversion. Also at 20 days conversion, 4 spots were differentially expressed, up- or down-regulated. Two proteins related to carbohydrate metabolism were only expressed in off-types at 10 days conversion, suggesting a more active respiratory pathway. A vicilin-like storage protein was only found in off-types at 20 days conversion, indicating that plantlets may present an abnormality in the mobilization of storage compounds, causing reduced vigor in the development of derived plantlets. The presence of heat shock proteins were only observed during formation of normal phenotype somatic plantlets, indicating that these proteins may be involved in normal morphogenesis of plantlets formed. These new findings shed light on possible genetic or epigenetic mechanisms governing A. sellowiana morphogenesis.

Acca sellowiana (O. Berg) Burret sin. Feijoa sellowiana (Myrtaceae) is a semiwoody fruit species native to South Brazil, Uruguay, and Argentina; edible fruits are tasty. The naturally occurring populations in Santa Catarina State show high variability in fruit size, color, and other features. A breeding program launched in 1990 resulted in the release of four Brazilian commercial varieties. The conventional clonal propagation methods of this species, such as cutting and grafting, have shown low efficiency. Therefore, tissue culture techniques were developed for mass propagation. This chapter describes several protocols based on organogenesis and somatic embryogenesis. Additional techniques including synthetic seed technology and temporary immersion system are also described.

O objetivo deste trabalho foi desenvolver e testar uma técnica de enxertia para a clonagem da goiabeira serrana (Acca sellowiana). Utilizou-se porta enxertos e copas jovens com tecido vegetal formado na estação, ambos apresentando espessura entre 0,3 e 0,5 cm. Os porta enxertos foram produzidos a partir de sementes de frutos oriundos da coleção da Estação Experimental de Lages (EEL), Epagri. O material de copa foi obtido de plantas pré-selecionadas da mesma coleção. Os enxertos foram feitos em condições controladas e em período de alta atividade metabólica (primavera). A técnica descrita mostrou-se eficiente para a multiplicação clonal da goiabeira serrana, com índice médio de pega dos enxertos de 92%.


This study aimed to develop and test a grafting technique for cloning feijoa plants (Acca sellowiana). The technique consisted of grafting young vegetative shoots of the scion on equally young rootstock, both with a diameter ranging from 0.3 to 0.5 cm. The rootstocks were selected from seedlings of the feijoa collection of Lages Experimental Station, Epagri. The scion wood was selected from feijoa plants of the same mentioned collection. The grafts were done under controlled conditions during the growing season (spring). The results suggest that the technique is efficient for cloning feijoa plants, presenting an average grafting success index of 92%.

UNLABELLED: DNA methylation is an epigenetic regulatory mechanism of gene expression which can be associated with developmental phases and in vitro morphogenetic competence in plants. The present work evaluated the effects of 5-azacytidine (AzaC) and 2,4-dichlorophenoxyacetic acid (2,4-D) on Acca sellowiana somatic embryogenesis (SE) and global DNA methylation levels by high-performance liquid chromatography mass spectrometry (HPLC/MS/MS). 2,4-D-free treatments revealed no somatic embryo formation in both accessions tested. Treatments supplemented with 2,4-D pulse plus AzaC in the culture medium resulted in increased embryo formation. In AzaC-free treatment, HPLC/MS/MS analysis showed a gradual increase in methylation levels in cultures of both accessions tested during SE induction. Treatment with AzaC and 2,4-D-free resulted in a marked decrease in methylation for both accessions, ranging from 37.6 to 20.8 %. In treatment with 2,4-D and AzaC combined, the 85 accession showed increasing global methylation levels. Otherwise, the 101X458 accession, in the same treatment, showed a decrease between 10 and 20 days, followed by an increase after 30 days (39.5, 36.2 and 41.6 %). These results indicate that 2,4-D pulse combined with AzaC improves SE induction. However, the conversion phase showed that although positively influencing SE induction, AzaC had a dysregulatory effect on the stage of autotrophic plant formation, resulting in significantly lower conversion rates. The results suggest that DNA methylation dramatically influences SE in Acca sellowiana, and global DNA methylation dynamics are related to morphogenetic response. KEY MESSAGE: 5-Azacytidine combined with 2,4-D increases the number of Acca sellowiana somatic embryos. Global DNA methylation is directly affected by these compounds.
