RESUMEN

Canine leproid granuloma (CLG) is a chronic form of dermatitis that has been associated with nontuberculous mycobacterial infections in Africa, Oceania, the Americas, and Europe. We report here a case of CLG associated with a member of the Mycobacterium tuberculosis complex (MTBC), which could be of public health concern. An 8-y-old pet dog developed 0.5-1-cm diameter, raised, firm, nonpruritic, alopecic, painless skin nodules on the external aspects of both pinnae. Histologic examination revealed severe pyogranulomatous dermatitis with intracellular Ziehl-Neelsen-positive bacilli that were immunoreactive by immunohistochemistry using a polyclonal primary antibody that recognizes tuberculous and nontuberculous Mycobacterium species. DNA extracted from formalin-fixed, paraffin-embedded skin sections was tested by a Mycobacterium genus-specific nested PCR assay targeting the 16S rRNA gene. BLAST sequence analysis of 214-bp and 178-bp amplicons showed 99.5% identity with members of the MTBC; however, the agent could not be identified at the species level. Although CLG has been associated traditionally with nontuberculous mycobacterial infections, the role of Mycobacterium spp. within the MTBC as a cause of this condition, and the role of dogs with CLG as possible sources of MTBC to other animals and humans, should not be disregarded given its zoonotic potential.

Environmental mycobacteria such as those from the Mycobacterium avium-intacellulare complex may cause disseminated and severe disease in dogs with genetic predisposition. A series of cases of 4 miniature schnauzers with nonspecific clinical signs and the diagnostic tests are described. Complementary means of diagnosis including complete blood count, biochemical serum analyses and fine needle aspiration cytology staining were performed. The bacteriological culture followed by PCR amplification of 1245 and 901 insertion sequences, allowed the identification of Mycobacterium avium subsp. hominissuis. This environmental Mycobacteria normally do not cause severe disease in dogs or other species, but when CARD-9 gene presents mutations, dogs may become extremely susceptible and disease is fast, disseminated, and fatal. Antibiotic therapy can be applied under veterinary consideration in specific situations, as treatment is usually applied for a long period of time. Although zoonotic risk is low as the Mycobacterium is environmental, contamination of the location may be high, and immunosuppressed animals and humans can develop infection as well. This report may aid clinical veterinarians in the diagnosis, treatment, and prognosis in similar cases of this breed and others with the genetic predisposition.

This report describes the clinical features and molecular diagnosis of a case of canine leproid granuloma (CLG) caused by mycobacterial strains of the Mycobacterium simiae complex in Brazil. A 12-year-old non-neutered male Labrador Retriever dog was presented with a 2-week history of progressive painless cutaneous lesions. Ulcerated nodules with hematic crusts were observed on the dorsal surface of the right and left pinna and on the metacarpal, metatarsal, and digits. Complete blood count, serum biochemistry, aspiration cytology of cutaneous lesions, biopsy for histopathological evaluation, culture for aerobic and anaerobic bacteria, polymerase chain reaction and DNA sequencing to identify mycobacterial species were performed. According to the clinical and histopathological findings, a diagnosis of CLG was established. Despite the negative result of the bacterial culture, mycobacterial identification was made by sequencing the hsp65 gene. Our findings highlight that mycobacterial species closely related to members of the M simiae clade can be causative agents of CLG.

Background: Mycobacteriosis is caused by bacteria belonging to the genus Mycobacterium, with considerable zoonoticpotential and risk to public health. Infection in dogs is rare and is usually associated with immunosuppression, resultingfrom eating meat or contact with contaminated soil or fomites. Dogs are also known as potential sources for the spread ofatypical tuberculosis in humans and other animals. This paper aims to describe the clinical, cytological, histopathological,and molecular findings of a male canine seen at University Veterinary Hospital of Cuiabá, Mato Grosso, with generalizedlymphadenomegaly associated Mycobacterium intracellulare infection.Case: A 2-year-old male Lhasa Apso dog was referred to the University Veterinary Hospital in Cuiabá city, located in theMidwest region of Brazil. The patient had a history of intermittent claudication of the left pelvic limb for approximately6 months and lymphadenomegaly with progression for approximately 2 months. The dog had wheezing and generalizedlymphadenopathy (submandibular, axillary, and popliteal lymph nodes); cryptorchidism was also observed. The freshwater ornamental fish trade represents a major contributor to the livelihoods of many producers in Trinidad and Tobago, with stocks destined for local, regional and international markets. A review of clinical cases presented to the Aquatic Animal Health Unit at the University of the West Indies, School of Veterinary Medicine for the period September 2010 to December 2012 suggested that piscine mycobacteriosis may be widespread throughout the local ornamental fish industry. Thus, to determine the prevalence of mycobacteriosis in ornamental fish sold in pet stores, a total of 122 specimens were sourced from 24 retail suppliers across Trinidad. Fish were killed and internal organs were examined for lesions suggestive of granulomas. All wet-mount slides were acid-fast stained, regardless of the presence or absence of observed granuloma-like lesions. Histological analysis was performed on one randomly selected whole specimen from each facility. Mycobacterium sp. was identified using real-time PCR detecting the 16S rRNA gene in tissue samples. Associations between parasitism, facility biosecurity and presence of positive animals were determined. The prevalence of Mycobacterium sp. infection was 61 ± 7% (74/122), with positive specimens being acquired from 54.2% (13/24) of facilities examined. Further, 100% of facilities did not employ optimum biosecurity measures.

Mycobacteriosis was detected in seven out of one Atlantic mackerel (Scomber scombrus) that was purchased for human consumption from a fish market. The fish was apparently healthy but during cleaning, several granulomatous foci were noticed in the visceral organs. Histopathological examination of the lesions revealed numerous foci characterized by caseous necrosis in the center of the lesion surrounded by epithelioid giant cells. Ziehl-Neelsen staining revealed the presence of rod-shaped, acid-fast bacteria. Furthermore, immunohistochemical examination revealed the presence of a protein of mycobacterial origin in giant cells and macrophages. Based on gross and microscopic findings, mycobacteriosis was diagnosed. This report showed that due to its zoonotic potential, mycobacteriosis should be considered even in healthy-appearing fishes for human consumption.


A micobacteriose foi detectada em uma de sete cavalas do Atlântico (Scomber scombrus) que foi comprada para consumo humano em um mercado de peixe. O peixe era aparentemente saudável, mas durante a limpeza, vários focos granulomatosos foram notados nos órgãos viscerais. O exame histopatológico das lesões revelou numerosos focos caracterizados por necrose caseosa no centro da lesão circundada por células gigantes epitelioides. A coloração de Ziehl-Neelsen revelou a presença de bactérias em forma de bastonete, com rápida acidez. Além disso, o exame imunohistoquímico revelou a presença de uma proteína de origem micobacteriana em células gigantes e macrófagos. Com base em achados grosseiros e microscópicos, a micobacteriose foi diagnosticada. Este relatório mostrou que devido a seu potencial zoonótico, a micobacteriose deve ser considerada mesmo em peixes saudáveis para consumo humano.

Between November 2017 and August 2018, in order to determine the occurrence of tuberculosis in cattle slaughtered in the semi-arid region of Rio Grande do Norte, 11,610 bovine carcasses underwent a routine post-mortem inspection. One animal presented suggestive lesions and samples from lung, spleen and heart were submitted to microbiological culture in Stonebrink medium for 90 days and molecular analysis by nested-PCR. For histopathological examination and Ziehl Neelsen staining, the omentum was used. In the cultured samples, two of them (heart and spleen) showed growth, but were not confirmed as M. bovis by conventional PCR. At nested-PCR, the samples showed amplification for the TbD1 region. The presence of numerous granulomas was detected in the histopathological examination characterized by anecrotic center and areas of mineralization, as well as the presence of acid-fast bacilli (AFB) in the Ziehl Neelsen stain. Microbiological culture can show false negative results, despite being considered a gold standard technique, although it takes time. Nested-PCR and histopathology show fast and effective results for the diagnosis of the disease. The presence of positive animals represents a public health risk in the studied region. Therefore, one of the essential systems applied to the control of bovine tuberculosis is the epidemiological surveillance of animals in slaughterhouses.


Entre novembro de 2017 e agosto de 2018, com objetivo de determinar a ocorrência de tuberculose em bovinos abatidos no semiárido do Rio Grande do Norte, 11.610 carcaças de bovinos passaram por inspeção post-mortem de rotina. Um animal apresentou lesões sugestivas e amostras de pulmão, baço e coração foram submetidas ao cultivo microbiológico em meio Stonebrink por 90 dias e análise molecular pela nested-PCR. O omento foi utilizado para exame histopatológico e coloração de Ziehl Neelsen. Nas amostras cultivadas, duas (coração e baço) evidenciaram crescimento, mas M. bovis não foi confirmado na PCR convencional. Já na nested-PCR, as amostras exibiram amplificação para a região TbD1. Foi detectada a presença de inúmeros granulomas no exame histopatológico caracterizados por possuírem um centro necrótico e áreas de mineralização, bem como a presença de bacilos Álcool-ácido resistentes (BAAR)na coloração de Ziehl-Neelsen. A cultura microbiológica pode apresentar resultados falso-negativos, apesar de ser considerada uma técnica padrão-ouro, embora demorada. A nested-PCR e a histopatologia apresentam resultados rápidos e eficazes para o diagnóstico da enfermidade. A presença de animais positivos representa um risco a saúde pública na região estudada. Portanto, um dos sistemas essenciais aplicados ao controle da tuberculose bovina é a vigilância epidemiológica dos animais em abatedouros.

Background: Among the bacterial dermopathy the canine leproid granuloma (CLG) is a nodular pyogranulomatous disorder that affects the skin or subcutaneous tissue mainly in the dorsal face of ear pinna, head, and extremity of memberscaused by Mycobacterium spp. The pathogenicity is still not well clarified regarding the causative agent, which has notyet been completely typified, but phylogenetically, it is related to Mycobacterium tilburgii, M. simiae, and M. genavense,in Brazil, by the species M. murphy. The objective of this study is to report a case of canine leproid granuloma, throughcytology and histopathology, and present the therapeutic procedures until the regression of cutaneous lesion.Case: A 5-year-old Boxer breed, intac male weighing 32 kg, was assisted at the Veterinary Clinic of UNIFIMES, in MineirosCity, Mid-West Region of Brazil, GO, Brazil. The animal had 4 nodules in the ears with evolution of 30 days, with nopruritus and without previous treatment. During the physical exam, the animal had normal physiological parameters. Thecutaneous lesions were characterised by papules and alopecic nodules of firm to fibroelastic consistency, with progressiveincrease, located in the convex face of the ears. The fine needle aspiration puncture technique (FNAP) and histopathologyfor a definitive diagnosis was used, allowing the differentiation between inflammatory processes, infectious and neoplastic. Furthermore, blood was collected for hemogram and biochemical analysis for the assessment of renal and hepaticfunctions. In cytology, the stained blades by the Diff-quick stain in the microscopic exam had elevated cellularity, withseveral macrophages, and bacilliform structures in the negative image. Mycobacterium bovis is the main causative agent of bovine tuberculosis (bTB) being among the animal-adapted Mycobacterium tuberculosis complex. Herds can also be infected with non-tuberculous mycobacteria (NTM) causing a negative effect on the economy and on animal and human health through zoonotic infections. Molecular tools are required for mycobacteria identification; thus, it is laborious to determine the epidemiological information of mycobacteria among herds. We aimed to describe the mycobacterial pathogens associated with cases of suspected bTB lesions in cattle/buffaloes slaughtered for consumption and to investigate bTB transmission. We evaluated 74 lesion samples from 48 animals (27 bovine/21 buffaloes) from 16 mapped farms. Positives samples from nested-PCR were cultured in Lowenstein-Jensen (LJ), 2% pyruvate (LJ + P), and 2% glycerol (LJ + G) media, followed by Ziehl-Neelsen (ZN) staining technique and partial gene sequencing (hsp65, rpoB, and 16S-rRNA). Spoligotyping and 24-MIRU-VNTR were performed. The LJ + P increased the chance of obtaining bacilli. The respiratory tract and the oral cavity were the most important infection route. In addition, the calcified part of the lesions suggested chronic bTB. Spoligotypes of M. bovis (SIT986/SB0885) differed from others found in South America, and the MIRU-VNTR 24 loci suggested that bTB was associated to highly related strains. The NTM species found are of clinical importance in humans.

Background: The Harpy eagle (Harpia harpyia) is the largest raptor in the Neotropical region, distributed in low densities within primary forest habitats from southern Mexico to east-central Brazil, including Central America and Panama.Although locally extinct due to habitat degradation, human poaching and reduction of tropical forests in recent decades,some remnant populations are known to be small and isolated. Thus, information on its ecology, including infectiousdiseases, is critical for conservation efforts and management of populations of this emblematic species in the wild. AvianMycobacteriosis (AM) is a chronic disease affecting a wide range of birds and mammals and clinical cases have beensporadically reported, although in harpy eagles, has not been documented. In addition, scant information exists on pathologies affecting raptors, therefore, this report expands the knowledge on infectious diseases affecting wild birds. This studydescribes the first case of AM in an immature harpy eagle rescued from Darien gap and presents details of the clinicalmanifestation of the disease and its histological findings.Case: An immature harpy eagle was presented to a rescue center after being confiscated by Ministry of Environmentfrom a local farm in a remote region of Eastern Panama, in the Darien gap. The raptor had a history of having been keptin captivity for approximately five weeks, cohabiting with backyard poultry and fed corn, raw duck and chicken providedby farm owners. Stagnant water was offered for drinking. The harpy eagle arrived to the facility very lethargic and emaciated. Physical examination revealed damaged plumage, poor body condition and dyspnea. Complete blood count revealedanemia and leukocytosis based on lymphocytosis and eosinophilia. As micobacterioses são bacilos resistentes a ácidos, causando doença crônica e progressiva em animais. Os felinos podem ser suscetíveis a infecções por micobactérias, especialmente quando há imunossupressão causada por retrovírus e / ou lesões por combate. O objetivo deste artigo é relatar um caso de micobacteriose felina atípica. Foi tratado em uma clínica veterinária particular, um felino, feminino, com histórico de ovariosalpingohisterectomia por um mês e com lesões fistulares na região da cirurgia. Foi realizada biópsia incisional e avaliação histopatológica, onde a análise foi compatível com micobacteriose atípica. A ressecção cirúrgica da maioria dos tecidos afetados foi realizada e a antibioticoterapia foi iniciada com marbofloxacina 2,2 mg / kg, SID, por 7 dias. Em vista do diagnóstico definitivo de micobacteriose atípica, doxiciclina 5 mg / kg, BID, foi adicionada à terapia por 90 dias, com regressão total da doença. A combinação de medicamentos com exérese cirúrgica é uma boa combinação terapêutica para uma remissão mais rápida da condição clínica.


Mycobacteriosis are acid-resistant bacilli causing chronic and progressive disease in animals. Felines may be susceptible to mycobacterial infections, especially when there is immunosuppression caused by retroviruses and / or fight injuries. The aim of this paper is to report a case of atypical feline mycobacteriosis. Was treated at a private veterinary clinic, a feline, female with a history of ovariosalpingohisterectomy for a month and with fistular lesions in the region of surgery. An incisional biopsy and histopathological evaluation were performed, where the analysis was compatible with atypical mycobacteriosis. Surgical resection of most of the affected tissue was performed and antibiotic therapy was started using marbofloxacin 2.2 mg/kg, SID, for 7 days. In view of the definitive diagnosis of atypical mycobacteriosis, doxycycline 5 mg/kg, BID, was added to the therapy for 90 days, with total regression of the condition. Combination of drugs with surgical exeresis is a good therapeutic combination for a faster remission of the clinical condition.

We conducted a study of the two main populations of free-living Scarlet Macaws ( Ara macao) in Costa Rica to detect the causal agents of avian tuberculosis using noninvasive techniques. We analyzed 83 fecal samples collected between February and May 2016 from the central and southern Pacific areas in the country. Using PCR, we first amplified the 16S region of the ribosomal RNA, common to all Mycobacterium species. Then, products from the insertion sequence IS901 and from a 155-base pair DNA fragment evidenced the presence of the avian pathogenic Mycobacterium avium subsp. avium strain and a Mycobacterium genavense strain, respectively. Seven of 38 (18%) samples collected in the central Pacific area were positive for Mycobacterium spp. and 3 of 38 (8%) were positive for M. genavense, with one sample amplifying regions for both. Two of the 45 (4%) samples collected in the south Pacific area of Costa Rica were positive to M. a. avium. Our detection of avian tuberculosis pathogens in free-living Scarlet Macaws suggests that free-living macaws could excrete in their feces M. genavense, bird-pathogenic M. a. avium, and possibly other Mycobacteria (not detected in our study).

O granuloma leproide canino (GLC) é uma doença micobacteriana que cursa com lesão nodular, cutânea ou subcutânea, tipicamente auto limitante, decorrente de infecção pelo Mycobacterium. É uma doença dermatológica rara, usualmente relatada em países de clima tropical. O objetivo deste trabalho é caracterizar as alterações microscópicas e epidemiológicas de casos de GLC. Além disso, utilizar exames complementares para demonstração do Mycobacterium sp. na lesão analisada utilizando coloração de Ziehl-Neelsen, imuno-histoquímica e PCR para classificação e sequenciamento da espécie de Mycobacterium. Foram analisados 27 casos de GLC diagnosticados no período compreendido de 2005 a 2014 (21 exames histológicos e seis citológicos). A raça mais acometida foi Boxer (33,3%), seguida da Dachshund (11,1%). Foi frequente o acometimento de caninos de grande porte (64%) e de pelame curto (96%). Não foi possível estabelecer qualquer padrão de sazonalidade do GLC. As lesões se localizaram anatomicamente no pavilhão auricular na maioria dos casos (93%), constituindo-se de nódulos únicos ou múltiplos, ulcerados ou não. Em nenhum dos casos foi observado acometimento sistêmico. Lesões histológicas seguiram três padrões que variaram de nodulares a difusos, com infiltrado granulomatoso/piogranulomatoso com envolvimento de derme superficial e profunda. A observação de bacilos pela coloração de Ziehl-Neelsen confirmou o diagnóstico. Este teste demonstrou grande variação na quantidade de bacilos nas lesões, não relacionado estatisticamente ao tipo da lesão nem ao período evolutivo. A imuno-histoquímica foi positiva em 90% dos casos, e em alguns casos facilitou a identificação de bacilos, entretanto, em dois casos ocorreram resultados negativos mesmo havendo marcação pela coloração de Ziehl-Neelsen. Na técnica de PCR realizada em 18 amostras, 44% dos casos foram positivos, duas sequências inespecíficas, e demonstrou ainda, que Mycobacterium murphy foi responsável por 2 casos e Mycobacterium hodleri em um caso. Em outros três casos não se obteve 100% de semelhança entre o produto sequenciado e os demais Mycobacterium.(AU)

There was no evidence predilection for occurrence in any season. The lesions were localized, in most cases, at the pinna (93%), comprise one or multiple nodules, ulcerated or not. None of the cases was observed with systemic involvement. Histopathological findings included three patterns of nodular to diffuse, with inflammatory infiltration granulomatous/pyogranulomatous that involved superficial and deep dermis. Definitive diagnosis was made by the presence of acid-fast bacilli by Ziehl-Neelsen stain, showing wide variation in the number of bacilli in the lesions, not being statistically correlated to the type of injury or the evolution period. Immunohistochemistry was effective, and in some cases facilitated the identification of bacilli, but in two cases were negative even with marking by Ziehl-Neelsen staining. Mycobacterioses can produce nonspecific clinical signs in dogs and cats that make diagnosis difficult. Furthermore, the full characterization of mycobacterial agents is not always possible or practical. We characterized mycobacteria detected through cytology in 12 dogs and 7 cats with generalized clinical signs from the province of Buenos Aires in Argentina. In dogs, molecular testing confirmed the presence of Mycobacterium avium subsp. hominissuis (MAH) in 8 cases and M. fortuitum in 1 case. All dogs were Miniature Schnauzers, suggesting that this breed may be more susceptible to M. avium than other dog breeds. The cat isolates were 2 M. bovis, 1 M. fortuitum, and 1 MAH. Mycobacterial interspersed repetitive unit-variable-number tandem repeat patterns suggested possible links with cattle, swine, and humans studied previously in Argentina. The results show that pets may act as susceptible hosts with the potential risk of transmitting the infection to humans and other animals.
