ABSTRACT
The recalcitrance exhibited by many maize (Zea mays) genotypes to traditional genetic transformation protocols poses a significant challenge to the large-scale application of genome editing (GE) in this major crop species. Although a few maize genotypes are widely used for genetic transformation, they prove unsuitable for agronomic tests in field trials or commercial applications. This challenge is exacerbated by the predominance of transformable maize lines adapted to temperate geographies, despite a considerable proportion of maize production occurring in the tropics. Ectopic expression of morphogenic regulators (MRs) stands out as a promising approach to overcome low efficiency and genotype dependency, aiming to achieve 'universal' transformation and GE capabilities in maize. Here, we report the successful GE of agronomically relevant tropical maize lines using a MR-based, Agrobacterium-mediated transformation protocol previously optimized for the B104 temperate inbred line. To this end, we used a CRISPR/Cas9-based construct aiming at the knockout of the VIRESCENT YELLOW-LIKE (VYL) gene, which results in an easily recognizable phenotype. Mutations at VYL were verified in protoplasts prepared from B104 and three tropical lines, regardless of the presence of a single nucleotide polymorphism (SNP) at the seed region of the VYL target site in two of the tropical lines. Three out of five tropical lines were amenable to transformation, with efficiencies reaching up to 6.63%. Remarkably, 97% of the recovered events presented indels at the target site, which were inherited by the next generation. We observed off-target activity of the CRISPR/Cas9-based construct towards the VYL paralog VYL-MODIFIER, which could be partly due to the expression of the WUSCHEL (WUS) MR. Our results demonstrate efficient GE of relevant tropical maize lines, expanding the current availability of GE-amenable genotypes of this major crop.
ABSTRACT
The timing of Tiwanaku's collapse remains contested. Here we present a generational-scale chronology of Tiwanaku using Bayesian models of 102 radiocarbon dates, including 45 unpublished dates. This chronology tracks four community practices: residing short- vs. long-term, constructing monuments, discarding decorated ceramics, and leaving human burials. Tiwanaku was founded around AD 100 and around AD 600, it became the region's principal destination for migrants. It grew into one of the Andes' first cities and became famous for its decorated ceramics, carved monoliths, and large monuments. Our Bayesian models show that monument building ended ~AD 720 (the median of the ending boundary). Around ~AD 910, burials in tombs ceased as violent deaths began, which we document for the first time in this paper. Ritualized murders are limited to the century leading up to ~AD 1020. Our clearest proxy for social networks breaking down is a precise estimate for the end of permanent residence, ~AD 1010 (970-1050, 95%). This major inflection point was followed by visitors who used the same ceramics until ~AD 1040. Temporary camps lasted until roughly ~AD 1050. These four events suggest a rapid, city-wide collapse at ~AD 1010-1050, lasting just ~20 years (0-70 years, 95%). These results suggest a cascading breakdown of community practices and social networks that were physically anchored at Tiwanaku, though visitors continued to leave informal burials for centuries. This generation-scale chronology suggests that collapse 1) took place well before reduced precipitation, hence this was not a drought-induced societal change and 2) a few resilient communities sustained some traditions at other sites, hence the chronology for the site of Tiwanaku cannot be transposed to all sites with similar material culture.

Background: The changes in endothelial function, arterial stiffness, and heart rate variability (HRV) produced in the first trimester of pregnancy in women who develop gestational hypertension (GH) are still being investigated. Objective: to evaluate the HVR, endothelial function, and arterial stiffness changes during the first trimester of pregnancy and their relationship with the development of GH. Methods: A group of women normotensive during the first trimester (n = 43), who later did (GH; n = 11) or did not (no-GH; n = 32) develop GH in that pregnancy, were enrolled. In the first trimester, endothelial function and arterial stiffness were evaluated through photoplethysmography. HRV, parasympathetic (PNS), and sympathetic (SNS) indexes were measured in a 5-minute continuous electrocardiogram record at rest sitting. The Griess reaction measured urinary nitrite excretion (NOx). Results: Systolic blood pressure (SBP) values were higher in GH (no-GH: 105.8 ± 2.0 vs. GH: 112.7 ± 3.0 mmHg; p < 0.05). Endothelial function was decreased, and arterial stiffness was increased in GH. Only in GH the arterial stiffness was correlated with SBP (Pearson's r: 0.5594; 95%CI: 0.06106-0.8681; p < 0.05). In HRV, GH decreased low-frequency power and the ratio SD2/SD1. The inhibition of PNS was lower in GH. The NOx was reduced in GH (no-GH: 3.4 ± 0.4 vs. GH: 0.3 ± 0.1 µM/L; p < 0.001). NOx was correlated negatively with the SNS index only in GH. Conclusions: Developed GH is preceded early in pregnancy by endothelial dysfunction and increased arterial stiffness. In this context, there are SNS-PNS interrelation modifications with less inhibition of PNS.

Abstract Introduction: During the COVID-19 pandemic, there was a notable increase in the use of telemedicine as an alternative to provide healthcare services. In this context, the Virtual Cardiology Service of the Sistema Provincial de Salud (SIPROSA for its acronym in Spanish) of Tucumán, Argentina, was created. Objective: To describe the characteristics of patients with or without a prior diagnosis of high blood pressure (HBP and non-HBP, respectively) who were assessed through teleconsultation at the Virtual Cardiology Service of the SIPROSA during the COVID-19 pandemic due to high blood pressure (BP) levels. Materials and methods: Descriptive cross-sectional study conducted in 275 patients (HBP=167; non-HBP=108) assessed at the Virtual Cardiology Service between July and September 2021. Bivariate analyses (ANOVA, Student's t-test and chi square test) were performed to determine differences between groups (HBP vs. non-HBP) according to sex. In addition, Pearson's correlation coefficient was used to establish the correlation between age and body mass index; systolic blood pressure (SBP), diastolic blood pressure and pulse pressure values; and the number of antihypertensive drugs used. Results: Patient's mean age was 48±1 years, 69.45% were women, 61.81% had some degree of obesity, 22.91% were on a low-sodium diet, and 48.36% presented with at least one of the considered risk factors (RF) for heart disease (diabetes, dyslipidemia, smoking). In the HBP group, the average number of antihypertensive drugs used was 1.2, and age was correlated with SBP values in both women (r: 0.2631; p<0.01) and men (r: -0.3440; p<0.05). Conclusions: A high prevalence of obesity was found and almost half of participants presented with at least one RF. In the HBP group, most patients used only one antihypertensive drug and were not or did not know if they were on a low-sodium diet. Finally, a high percentage of patients without HBP used the Virtual Cardiology Service.
Resumen Introducción. Durante la pandemia por COVID-19 el uso de la telemedicina aumentó considerablemente como una alternativa para prestar atención en salud. En este contexto surgió el Consultorio Virtual de Cardiología del Sistema Provincial de Salud de Tucumán, Argentina (SIPROSA). Objetivo. Describir las características de pacientes con o sin diagnóstico previo de hipertensión arterial (HTA y no-HTA, respectivamente) que fueron valorados mediante teleconsulta en el Consultorio Virtual de Cardiología del SIPROSA durante la pandemia por COVID-19 debido a valores elevados de presión arterial (PA). Materiales y métodos. Estudio transversal descriptivo realizado en 275 pacientes (HTA=167; no-HTA=108) atendidos en el Consultorio Virtual de Cardiología del SIPROSA entre julio y septiembre de 2021. Se realizaron análisis bivariados (ANOVA, prueba t-Student y prueba X2) para determinar diferencias entre grupos (HTA vs. No-HTA) según el sexo. Además, se utilizó el coeficiente de correlación de Pearson para establecer la correlación entre, por un lado, la edad y el índice de masa corporal, y, por otro, valores de presión arterial sistólica (PAS), presión arterial diastólica y presión diferencial, y el número de medicamentos antihipertensivos usados. Resultados. El promedio de edad fue 48±1 años, 69.45% de los pacientes eran mujeres, 61.81% tenía algún grado de obesidad, 22.91% seguía una dieta hiposódica y 48.36% tenía al menos uno de los factores de riesgo (FR) de enfermedad cardiaca considerados (diabetes, dislipemia, tabaquismo). En el grupo HTA, el promedio de medicamentos antihipertensivos usados fue de 1.2 y la edad se correlacionó con los valores de PAS en mujeres (r: 0.2631; p<0.01) y hombres (r: -0.3440; p<0.05). Conclusiones. Se encontró una alta prevalencia de obesidad y que casi la mitad de los participantes tenía al menos un FR. En el grupo HTA, la mayoría de pacientes solo usaba un fármaco antihipertensivo y no realizaba o no sabía si realizaba dieta hiposódica. ABSTRACT
INTRODUCTION: Photobiomodulation has been widely used as a complementary treatment for dentin hypersensitivity, with reports of clinical success. However, the literature offers only one study, in which photobiomodulation was used for the treatment of sensitivity in molars with molar incisor hypomineralisation (MIH). The aim of the proposed study is to determine whether photobiomodulation enhances the results of treatment with glass ionomer sealant on molars with MIH that present sensitivity. METHODS AND ANALYSIS: The study involves 50 patients from 6 to 12 years of age that will be randomly allocated in two groups. Group 1 (control group) (n=25): toothpaste with a concentration of fluoride ≤1000 ppm two times a day, glass ionomer sealant and sham low-level laser (LLL) and group 2 (n=25): toothpaste with a concentration of fluoride ≤1000 ppm two times a day, glass ionomer sealant and active LLL. The evaluations will involve MIH record, Simplified Oral Hygiene Index (OHI), Schiff Cold Air Sensitivity Scale (SCASS) and visual analogue scale (VAS) before the procedure. Immediately after the procedure, the hypersensitivity index (SCASS/VAS) will be registered. Records of OHI and SCASS/VAS will be registered after 48 hours as well as 1 month after the procedure. The persistence of the sealant will also be registered. It is expected that by the second consultation, a decrease in sensitivity will be observed due to the treatments received in the two groups. ETHICS AND DISSEMINATION: This protocol has been approved by the local medical ethical committee (certificate: CEUCU 220516). The findings will be published in a peer-reviewed journal. Cysteine S-nitrosation is a redox-based post-translational modification that mediates nitric oxide (NO) regulation of various aspects of plant growth, development and stress responses. Despite its importance, studies exploring protein signaling pathways that are regulated by S-nitrosation during somatic embryogenesis have not been performed. In the present study, endogenous cysteine S-nitrosation site and S-nitrosated proteins were identified by iodo-TMT labeling during somatic embryogenesis in Brazilian pine, an endangered native conifer of South America. In addition, endogenous -S-nitrosothiol (SNO) levels and S-nitrosoglutathione reductase (GSNOR) activity were determined in cell lines with contrasting embryogenic potential. Overall, we identified an array of proteins associated with a large variety of biological processes and molecular functions with some of them already described as important for somatic embryogenesis (Class IV chitinase, pyruvate dehydrogenase E1 and dehydroascorbate reductase). In total, our S-nitrosoproteome analyses identified 18 endogenously S-nitrosated proteins and 50 in vitro S-nitrosated proteins (after GSNO treatment) during cell culture proliferation and embryo development. Furthermore, SNO levels and GSNOR activity were increased during embryo formation. These findings expand our understanding of the Brazilian pine proteome and shed novel insights into the potential use of pharmacological manipulation of NO levels by using NO inhibitors and donors during somatic embryogenesis.

Insulin vasorelaxant effect in metabolic syndrome has been shown on precontracted vessels. However, the insulin effects on basal vascular tone and its interrelationship with nitric oxide (NO) and K-channels are unknown. To test the effect of insulin on the basal vascular tone in isolated aortic rings from the cafeteria diet-induced hypertensive rats and to determine the role of NO and K-channels on this insulin effect. Male Wistar rats were randomized into two groups: one group fed with a cafeteria diet (CafR) and another fed with a standard chow diet (control rats: CR). Then, in isolated aortic rings, the insulin effect on the basal tone and the role of K-channels were evaluated. Also, the endothelial function, NO levels, and resting membrane potential were measured. CafR increased blood pressure (138 ± 6.2 mmHg; n = 9 vs. CR: 109 ± 1.4 mmHg; n = 9; p < 0.001) and vascular basal tone. Insulin 400 mU/ml reduced basal tone in aortic rings (-284 ± 47 mg; n = 9). This effect was unaffected by endothelium removal or NG-nitro-l-arginine methyl ester (L-NAME) treatment. Likewise, CafR showed low NO levels and a hyperpolarized resting membrane potential. Insulin decreased the resting membrane potential and the KCa and Kv channels blockers abolished this effect. In CafR, endothelial dysfunction is accompanied by an increased basal tone. Insulin reduced it by Kv and KCa channels dependent mechanisms, using an endothelium-independent pathway. These results highlight a novel insulin effect on basal tone of aortic rings from animals with metabolic syndrome and endothelial dysfunction, pathophysiological conditions associated with human hypertension.

INTRODUCCIÓN: A través de la Red Ministerial de Investigación en Salud (REMINSA) se actualizó el estado de situación de las áreas provinciales de investigación en salud pública. MÉTODOS: Se realizaron reuniones virtuales de debate con los referentes REMINSA y una encuesta en Google Forms® sobre funcionamiento normativo, organizacional, presupuestario, alcance de tareas y necesidades de mejoras. RESULTADOS: Se recibieron 21 respuestas de un total de 21 provincias con referentes. Se verificó la existencia de un amplio marco de normativas regulatorias de la investigación en salud humana y, en menor medida, para impulsar la investigación en el marco de la salud pública. El 71% tiene cierta estructura institucional, el 20% ofrece becas de investigación, y el 20% posee residencias con perfil en investigación. La principal actividad es la regulación o participación en comités de ética. La promoción, difusión de resultados y elaboración de proyectos de investigación basada en evidencia quedaron relegadas (20%). Las áreas de investigación están consolidadas en la mayoría de las provincias. DISCUSIÓN: El desafío consiste en promover el desarrollo de estos sistemas en las provincias que no cuentan con áreas ministeriales y fortalecer las provincias que sí las poseen. REMINSA ha identificado oportunidades de mejorar la comunicación entre jurisdicciones, las normativas de promoción de la investigación y el trabajo en red para la investigación, la gestión y la toma de decisiones.

This study aimed to evaluate vascular function changes and autonomic balance during the first trimester of pregnancy and its relationship with the new-born weight. This prospective study performed in pregnant (PG) women and after delivery (not pregnant: NPG) evaluated the endothelial function (EF) and arterial stiffness (AS) by a non-invasive method. We evaluated the heart rate variability (HRV), parasympathetic nervous system (PNS), sympathetic nervous system (SNS) indexes by electrocardiogram (5 min) and the urinary nitrite excretion (NOx). PG increased EF and NOx and decreased AS and HRV. PG decreased the PNS index and augmented the SNS index. The new-born weight positively correlated with the PNS index (Pearson's r: 0.4291; p<.05), NOx, HRV and negatively correlated with AS. In summary, in pregnancy, although haemodynamically, the SNS activation plays a compensatory role, the low rates of PNS inhibition are essential to ensure normal foetal growth.Impact StatementWhat is already known on this subject? In pregnancy, there are adaptive physiological changes in the cardiovascular system that include increases of EF and decreases AS with an SNS activation. The study of HRV lets to predict the SNS and PNS balance and how they affect blood pressure and vascular function.What the results of this study add? Although it is known that SNS activation plays a compensatory role in healthy pregnancy, this study adds the critical role of PNS. Early in pregnancy, the low rates of PNS inhibition are essential to ensure normal foetal growth.What the implications are of these findings for clinical practice and/or further research? The present results show a potential predictive value of SNS and PNS activity early in pregnancy. It will provide valuable information not only on the pregnant woman's vascular function but also on the new-born weight.

El cepillado dental es el método tradicional reconocido para eliminar la biopelícula. Objetivo: Evaluar la relación entre edad y cepillado dental, así como la asociación entre hábitos de cepillado dental y presencia de caries en un grupo de escolares de Montevideo, Uruguay. Material y métodos: Se realizó un estudio observacional, transversal, analítico, en 127 escolares de 4 a 11 años de edad, en Montevideo, Uruguay. Se realizó una encuesta a los padres en relación a los hábitos de cepillado dental de sus hijos y se estudiaron las variables tiempo de cepillado, Índice de Higiene Oral simplificado y se registró el índice para caries dental ICDAS. Resultados: La edad media de los participantes fue 7,93 años (DS ± 1,95). El tiempo promedio de cepillado fue de 47 segundos (DS ± 27 segundos). Los tiempos de cepillado que reportaron los padres difirieron significativamente de los observados en los niños. Al aumentar la edad, el tiempo de cepillado también aumentaba. Se ajustó un modelo de regresión múltiple para analizar la relación entre la disminución en la magnitud del IHOS debido al cepillado y las variables edad, género, tiempo de cepillado y ayuda en el cepillado. Se verificó asociación entre las variables ayuda y/o supervisión en el cepillado y presencia o no de caries dental. Conclusiones: Se encontró una relación significativa entre cepillado dental, edad y tiempo de cepillado en la población estudiada. A mayor edad, mayor tiempo de cepillado y mejor resultado en el índice de higiene oral. Los niños que recibían supervisión o ayuda de los padres fueron los que presentaron menos lesiones de caries.
A escovação dentária é o método tradicional reconhecido para a remoção do biofilme. Objetivo: Avaliar a relação entre idade e escovação dentária, bem como a associação entre hábitos de escovação dentária e a presença de cáries em um grupo de escolares em Montevidéu, Uruguai. Material e métodos: Um estudo observacional, transversal, analítico foi realizado em 127 escolares de 4 a 11 anos, em Montevidéu, Uruguai. Foi realizada um levantamento dos pais sobre os hábitos de escovação dentária de seus filhos e as variáveis foram estudadas para o tempo de escovação, índice simplificado de higiene bucal e o índice de cárie dentária do ICDAS. Resultados: A idade média dos participantes foi de 7,93 anos (DS s 1,95). O tempo médio de escovação foi de 47 segundos (DS x 27 segundos). Os tempos de escovação relatados pelos pais diferiram significativamente daqueles observados nas crianças. À medida que a idade aumentava, o tempo de escovação também aumentava. Um modelo de regressão múltipla foi ajustado para analisar a relação entre a diminuição da magnitude do IHOS devido à escovação e as variáveis de idade, sexo, tempo de escovação e auxílio de escovação. Houve associação entre as variáveis ajuda e/ou supervisão na escovação e presença ou não de cárie dentária Conclusões: Verificou-se uma relação significativa entre escovação dentária, idade e tempo de escovação na população estudada. Quanto mais velho, maior o tempo de escovação e melhor resultado no índice de higiene bucal. As crianças que receberam a supervisão ou a ajuda dos pais eram essas com menos os ferimentos da cavidade
Dental brushing is the recognized traditional method for removing the biofilm. Objective: To evaluate the relationship between age and toothbrushing, as well as the association between toothbrushing and caries lesions in schoolchildren in Montevideo, Uruguay. Material and methods: An observational, cross sectional and analytic study was carried on 127 schoolchildren aged 4 to 11 years old, in Montevideo, Uruguay. The parents were asked about the oral health habits of their children through a questionnaire, and the variables toothbrushing time, Simplified Oral Hygiene Index and ICDAS index were registered. Results: Mean age of the children was 7,93 years (SD ± 1,95). Mean time of toothbrushing was 47 seconds (DS ± 27 seconds). The toothbrushing times reported by the parents differed significantly from those observed in the children. As age increased, brushing time also increased. A multiple regression model was adjusted to analyze the relationship between the decrease in the magnitude of IHOS due to toothbrushing and the variables age, toothbrushing time and brushing aid. There was an association between the variables help and / or supervision in brushing and presence or not of dental caries. Conclusions: A significant relationship was found between toothbrushing, age, gender and brushing time in this group of children. When the age was higher, they spent more time brushing their teeth and they had better results in oral hygiene index. Children who had parents help or supervision had lower caries lesions

INTRODUCCIÓN: Una Agenda Nacional de Investigación en Salud Pública (ANISP) participativa y con priorización temática constituye un elemento estratégico para generar recomendaciones y políticas públicas basadas en evidencia, que impacten positivamente en la salud de las poblaciones y permitan lograr los objetivos sanitarios. En la actualización de la ANISP participaron la Dirección de Investigación en Salud (DIS) del Ministerio de Salud de la Nación (MSAL), a través de la Red Ministerial de Investigación en Salud (REMINSA), y actores de los niveles gubernamentales provinciales y nacionales pertenecientes a los sectores público, privado, de la salud, académico y de investigación. Se adaptó la herramienta original propuesta por la Organización Panamericana de la Salud, utilizada en el proceso en 2019. La actualización abarcó diferentes etapas. La selección de los temas contó con la legitimidad, reconocimiento y participación de los actores vinculados a la salud, a la gestión gubernamental y privada y a la investigación científica; se trabajó de manera federal y transversal, por consenso con las redes provinciales y un Comité Central Asesor en el MSAL. A partir de los lineamientos preliminares obtenidos, se elaboró una encuesta en línea semiestructurada, que fue distribuida a todos los actores federales y recibió 431 respuestas. El proceso resultó en 55 lineamientos priorizados, divididos en 6 áreas temáticas y 33 subtemas, seleccionados por votación según importancia, impacto y factibilidad.

The Lake Titicaca basin was one of the major centers for cultural development in the ancient world. This lacustrine environment is unique in the high, dry Andean altiplano, and its aquatic and terrestrial resources are thought to have contributed to the florescence of complex societies in this region. Nevertheless, it remains unclear to what extent local aquatic resources, particularly fish, and the introduced crop, maize, which can be grown in regions along the lakeshores, contributed to facilitating sustained food production and population growth, which underpinned increasing social political complexity starting in the Formative Period (1400 BCE to 500 CE) and culminating with the Tiwanaku state (500 to 1100 CE). Here, we present direct dietary evidence from stable isotope analysis of human skeletal remains spanning over two millennia, together with faunal and floral reference materials, to reconstruct foodways and ecological interactions in southern Lake Titicaca over time. Bulk stable isotope analysis, coupled with compound-specific amino acid stable isotope analysis, allows better discrimination between resources consumed across aquatic and terrestrial environments. Together, this evidence demonstrates that human diets predominantly relied on C3 plants, particularly quinoa and tubers, along with terrestrial animals, notably domestic camelids. Surprisingly, fish were not a significant source of animal protein, but a slight increase in C4 plant consumption verifies the increasing importance of maize in the Middle Horizon. These results underscore the primary role of local terrestrial food resources in securing a nutritious diet that allowed for sustained population growth, even in the face of documented climate and political change across these periods.

We propose and describe a new genus of interstitial Paramesochridae, Maliithipon gen. nov., to accommodate three species. Maliithipon wellsi sp. nov., type species of the genus, is described based on specimens collected in two sandy beaches in Isla Verde, the Philippines. Apodopsyllus aberrans Mielke, 1984a described from Panama, is allocated to the herein erected genus as Maliithipon aberrans (Mielke, 1984a) comb. nov. Maliithipon cf. aberrans is described, based on specimens collected in the Azores and previously identified as Apodopsyllus aberrans. The new genus is characterized by several distinct morphological features: mandible with uniramous, 2-segmented palp and gnathobase with thin and pointed teeth; maxillule with reduced armature, lacking endopod and exopod; maxilla large, with three syncoxal endites and endopod with reduced setal number; P5 very reduced in both sexes; caudal rami with six setae, two of which (setae III and VI) transformed; pseudoperculum well-developed, with four-lobed distal margin. The new genus is included in the subfamily Paramesochrinae Lang, 1944 and in the genus-group Scottopsyllus (sensu Huys, 1987). It has strong affinities with Leptopsyllus (Leptopsyllus) platyspinosus Mielke, 1984b and Wellsopsyllus (Intermediopsyllus) smirnovi (Kunz, 1992), which are considered as species inquirendae in this paper. Some remarks on the ecology and geonemy of the three studied species are provided. We also list the accompanying Paramesochridae obtained from the two new collecting sites of the genus, and all the genera of Paramesochridae recorded from other sites in the Philippines during the same survey.

RESUMEN INTRODUCCIÓN : Una Agenda Nacional de Investigación en Salud Pública (ANISP) participativa y con priorización temática constituye un elemento estratégico para generar recomendaciones y políticas públicas basadas en evidencia, que imparten positivamente en la salud de las poblaciones y permitan lograr los objetivos sanitarios. En la actualización de la ANISP participaron la Dirección de Investigación en Salud (DIS) del Ministerio de Salud de la Nación (MSAL), a través de la Red Ministerial de Investigación en Salud (REMINSA), y actores de los niveles gubernamentales provinciales y nacionales pertenecientes a los sectores público, privado, de la salud, académico y de investigación. Se adaptó la herramienta original propuesta por la Organización Panamericana de la Salud, utilizada en el proceso en 2019. La actualización abarcó diferentes etapas. La selección de los temas contó con la legitimidad, reconocimiento y participación de los actores vinculados a la salud, a la gestión gubernamental y privada y a la investigación científica; se trabajó de manera federal y transversal, por consenso con las redes provinciales y un Comité Central Asesor en el MSAL. A partir de los lineamientos preliminares obtenidos, se elaboró una encuesta en línea semiestructurada, que fue distribuida a todos los actores federales y recibió 431 respuestas. El proceso resultó en 55 lineamientos priorizados, divididos en 6 áreas temáticas y 33 subtemas, seleccionados por votación según importancia, impacto y factibilidad.
ABSTRACT INTRODUCTION : A participatory National Public Health Research Agenda (ANISP) with thematic prioritization is a strategic element to generóte evidence-based recommendations and public policies that have a positive impact on the health of populations and enable to achieve health objectives. The Directorate of Health Research (DIS) ofthe Argentine Ministry of Health (MSAL), through the Ministerial NetWork of Health Research (REMINSA), along with adors from the provincial and national government levels belonging to public, privóte, health, academic and research sectors participated in the update of the ANISP. They adapted the original tooI proposed by the Pan American Health Organizatlon and used in the process in 2019. The update included different stages. The selection ofthe topics had the legitimacy, recognition and participation ofthe actors involved, related to health, to government and privóte management and to scientific research; the work was conducted in a federal and transversal manner by consensus with the provincial networks and a Central Advisory Committee in the MSAL. Based on the preliminary guidelines obtained, a semi-structured online survey was developed and distributed to all federal actors, receiving 431 responses. The process resulted in 55 priorilized guidelines, divided into 6 thematic oreas and 33 sub-themes, selected by voting according to importance, impact and feasibility.

BACKGROUND: Oxidative stress plays an essential role in the vascular tone in hypertension; however, the mechanisms remain unclear. AIM: This study aimed to determine the antioxidant effect of tempol and vitamin C (Vit-C) on the basal tone and vascular remodeling of the aorta in nitric oxide (NO) deficiency-induced hypertensive rats. METHOD: Male Sprague-Dawley rats were induced to hypertension by Nω-nitro-L-arginine methyl ester (L-NAME). Animals were randomized as follows: vehicle (Control: CR), CR-tempol, CR-Vit-C, L-NAME, L-NAME-tempol, and L-NAME-Vit-C. After 6 weeks of treatment, the basal aortic tone was evaluated by sodium nitroprusside (SNP) and calcium-free medium. Endothelial function, NO, reduced-to-oxidized glutathione (GSH/GSSG) ratio, resting membrane potential (mP), and vascular remodeling were also measured. RESULTS: L-NAME rats showed an increased basal tone that was blunted by both SNP (-547 ± 69; n = 7 vs. CR: -7.5 ± 6.7 mg; n = 7; p < 0.001) and calcium-free medium. Tempol or Vit-C did not reverse hypertension, and the high basal tone was decreased only with tempol. In L-NAME rats, only tempol partially improved endothelial function, GSH-to-GSSG ratio, mP values, and vascular remodeling. CONCLUSIONS: Tempol decreased calcium-dependent basal aortic tone and improved vascular homeostasis in L-NAME rats. Vit-C did not lead to a similar effect, suggesting that alterations in the superoxide dismutase pathway may play a role in the basal aortic tone.

Objetivo: analizar los datos epidemiológicos y las medidas generales en la práctica odontológica tomadas por Colombia y Uruguay durante la pandemia por la COVID-19. Métodos: este trabajo es una revisión de la literatura. Se consultaron las bases de datos "PubMed", "Scielo" y el motor de búsqueda "Google Scholar". También se consultaron bases de datos de libre acceso como google, our world in data y las páginas oficiales de la Organización Mundial de la Salud, el Ministerio de Salud de Colombia, el Instituto Nacional de Salud y el Ministerio de Salud Pública de Uruguay. Se realizó un análisis descriptivo del comportamiento epidemiológico y de las medidas tomadas en la práctica odontológica en los dos países. Resultados: en Colombia, a diario, se presentan más de 3.000 casos nuevos y más de 100 muertes. Lo contrario ocurre en Uruguay, que ha logrado disminuir las tasas de contagio y de muertes, presentando menos de 10 casos y ninguna muerte diaria. Los profesionales de la salud han incrementado su lucha; los odontólogos se encuentran expuestos por su proximidad al momento de la atención, en el contacto con saliva, sangre y la generación de aerosoles, por lo que han modificado los protocolos de bioseguridad buscando prevenir los contagios asociados a la prestación de servicio. Conclusiones: Uruguay ha tenido un comportamiento ejemplar ante la pandemia; en Colombia los resultados son preocupantes dado el crecimiento exponencial. En ambos países la práctica odontológica se ha visto afectada y se han implementado nuevos protocolos para proteger a profesionales y pacientes.
Objective: Analyze the epidemiological data, general and dental practice measures taken by Colombia and Uruguay in the face of the COVID-19 pandemic. Methods: This study is a review. We search in the databases "PubMed", "Scielo" and in the search engine "Google Scholar". Also, were consulted the free databases like google, our world in data and the official pages of the World Health Organization, Ministry of Health of Colombia, The National institute of health and the Ministry of public Health of Uruguay. A descriptive analysis of the epidemiological behavior and the measures taken of dental practice in both countries was done. Results: Colombia had more than 3,000 new cases and more than 100 deaths daily. The opposite occurs in Uruguay, which has managed to decrease the infection and death rates, presenting less than 10 cases and no daily deaths. The struggles experienced by health professionals has increased, dentists are exposed, by their proximity at the time of care, to contact with saliva, blood and the generation of aerosols, therefore, they have modified the biosafety protocols, seeking to prevent infections associated with the practice. Conclusions: Uruguay has an exemplary behavior; in Colombia these results are worrying with an exponential growth. In both countries, dental practice has been affected and new protocols have been implemented that seek to protect the professionals and patients.

Peptidic messengers constitute a highly diversified group of intercellular messengers widely distributed in nature that regulate a great number of physiological processes in Metazoa. Being crucial for life, it seem that they have appeared in the ancestral group from which Metazoa evolved, and were highly conserved along the evolutionary process. Peptides act mainly through G-protein coupled receptors (GPCRs), a family of transmembrane molecules. GPCRs are also widely distributed in nature being present in metazoan, but also in Choanoflagellata and Fungi. Among GPCRs, the Allatotropin/Orexin (AT/Ox) family is particularly characterized by the presence of the DRW motif in the second intracellular loop (IC Loop 2), and seems to be present in Cnidaria, Placozoa and in Bilateria, suggesting that it was present in the common ancestor of Metazoa. Looking for the evolutionary history of this GPCRs we searched for corresponding sequences in public databases. Our results suggest that AT/Ox receptors were highly conserved along evolutionary process, and that they are characterized by the presence of the E/DRWYAI motif at the IC Loop 2. Phylogenetic analyses show that AT/Ox family of receptors reflects evolutionary relationships that agree with current phylogenetic understanding in Actinopterygii and Sauropsida, including also the largely discussed position of Testudines.
