ABSTRACT
Sacral nerve stimulation (SNS) is an alternative surgical approach to alleviate fecal incontinence and constipation. This study aimed to explore the effects and underlying mechanisms of SNS with acupuncture on gut transit time and colon c-kit protein expression in rats with slow transit constipation (STC). Fifty Sprague-Dawley rats were randomly divided into five groups: blank control, SNS, Mosapride, sham SNS, and STC model control group. The STC model was established by subcutaneous injection of morphine. Each group was treated over a 15-day period. Gut transit time was measured 1 day before the treatment started and after 5, 10, and 15 days of treatment. After the 15-day treatment, animals were sacrificed and colonic tissues were collected for analysis of c-kit protein expression, using western blot analysis. We found significant differences in gut transit time in the SNS group compared with the Mosapride group after 5 (P = 0.001) and 10 (P = 0.004) days of treatment. After 15 days of treatment, there were no differences in gut transit time among the SNS, Mosapride, and blank control groups. However, significant differences were observed when comparing the SNS and Mosapride groups with the STC model and sham SNS groups. A decreased c-kit protein expression was observed in the STC model control, sham SNS, and Mosapride groups, compared with the SNS group (P = 0.001). Our data indicate that SNS can decrease gut transit time and increase the expression of c-kit protein in rats with STC to improve colon transit function.

Our study aimed to investigate the co-localization and protein-protein interactions between ezrin and p65 in human breast cancer cells. Liquid chromatography-mass spectrometry (LCMS) was used to uncover novel protein interactions with ezrin in MDA-MB-231 cells. Endogenous co-immunoprecipitation was used to validate protein-protein interactions between ezrin and p65 in MDA-MB-231. Exogenous interactions between ezrin and p65 were validated in MDA-MB-231 cells via Flag-ezrin and HA-p65 co-transfection and followed by co-immunoprecipitation. Immunofluorescence staining was used to visualize ezrin and p65 co-localization in MDA-MB-231. LCMS results showed that there were 1000 proteins interacting with ezrin in MDA-MB-231 cells. Ezrin and p65 interactions were confirmed with both endogenous and exogenous methods. We were also able to visualize ezrin and p65 co-localization in MDA-MB-231. In summary, we found protein-protein interactions between Ezrin and p65 in human breast cancer cells.

Twelve polymorphic microsatellite loci were isolated in the Japanese gecko, Gekko japonicus. We genotyped one population from Wenzhou, Zhejiang Province, China (N = 36). The mean number of observed alleles per locus was 7.3 (range 4 to 13). Observed and expected heterozygosity values ranged from 0.200 to 0.944 and from 0.395 to 0.797, respectively. One locus (GJ20) showed significant departure from Hardy-Weinberg equilibrium; no linkage disequilibrium was found between any two loci. These informative microsatellite markers will be useful for population genetic analyses of G. japonicus and other species in the genus Gekko.

Association mapping based on linkage disequilibrium (LD) provides a promising tool to identify quantitative trait loci (QTLs) in plant resources. A total of 141 eggplant (Solanum melongena L.) accessions were selected to detect simple sequence repeat (SSR) markers associated with nine fruit traits. Population structure analysis was performed with 105 SSR markers, which revealed that two subgroups were present in this population. LD analysis exhibited an extensive long-range LD of approximately 11 cM. A total of 49 marker associations related to eight phenotypic traits were identified to involve 24 different markers, although no association was found with the trait of fruit glossiness. To our knowledge, this is the 1st approach to use a genome-wide association study in eggplant with SSR markers. These results suggest that the association analysis approach could be a useful alternative to traditional linkage mapping to detect putative QTLs in eggplant.

The genetic diversity of 10 Machilus thunbergii populations in eastern China was analyzed using inter-simple sequence repeat markers. The populations showed high genetic diversity, with an overall population genetic diversity of 0.2343. Genetic diversity varied largely among populations, and populations with the highest genetic diversity were mainly from the eastern and western parts of the natural distribution area. Small populations, lack of effective gene flow, and fragmentation of habitats have led to greater genetic differentiation among populations, with 41.18% of genetic variation existing among populations. Unweighted pair-group method with arithmetic mean cluster analysis indicated that populations distributed between latitudes 25° and 31°N were clustered together and should be prioritized for in situ conservation. Northern, eastern, and southern populations were located in peripheral areas of the distribution range and were clustered separately. Collection of distinctive germplasm from peripheral populations should be promoted and ex situ conservation of elite germplasm should be implemented.
