ABSTRACT

Yeast extracts (YEs) are used in foods because of their flavour properties and ability to reduce bitterness. The adenosine 5'-monophosphate (AMP) found in YEs is known to decrease the bitterness of some compounds. This study aimed to investigate the ability of YEs to inhibit bitter taste receptors (TAS2Rs) using in vitro cell-based assays. A screen of TAS2Rs activated by AMP and YEs revealed that AMP and the AMP-rich YE activated more TAS2Rs. The inhibitory effect of the AMP-rich YE on seven TAS2Rs activated by bitter agonists was studied. YE reduced TAS2R activation, increased the EC50 value and decreased the maximum amplitude, demonstrating competitive and non-competitive inhibitions. Amongst the nineteen TAS2Rs tested, seven showed 40 % or greater inhibition after treatment of AMP-rich YE. Our data provide a better understanding of the TAS2R inhibition mechanism of AMP-rich YEs and promote their use as a strategy to reduce bitterness in foods and medicines.

ABSTRACT

Polymer flooding is a well-established technique aimed at improved recovery factors from oilfields. Among the important parameters affecting the feasibility of a large deployment, polymer retention is one of the most critical since it directly impacts the oil bank delay and therefore the final economics of the project. This paper describes the work performed for the East-Messoyakhskoe oilfield located in Northern Siberia (Russia). A literature review was first performed to select the most appropriate methodology to assess polymer retention in unconsolidated cores at residual oil saturation. 4 polyacrylamide polymers were selected with molecular weights between 7 and 18 M Da and sulfonated monomer (ATBS) content between 0 and 5% molar. An improved 2-fronts dynamic retention method along with total organic carbon-total nitrogen analyzers were used for concentration measurement. Retention values vary between 93 and 444 The sentence could be rephrased µg/g, with the lowest given by the polymers containing ATBS, corroborating other publications on the topic. This paper also summarizes the main learnings gathered during the adaptation of laboratory procedures and paves the way for a faster and more efficient retention estimation for unconsolidated reservoirs.

ABSTRACT

BACKGROUND: Segmental duplications in genomes have been studied for many years. Recently, several studies have highlighted a biological phenomenon called breakpoint-duplication that apparently associates a significant proportion of segmental duplications in Mammals, and the Drosophila species group, to breakpoints in rearrangement events. RESULTS: In this paper, we introduce and study a combinatorial problem, inspired from the breakpoint-duplication phenomenon, called the Genome Dedoubling Problem. It consists of finding a minimum length rearrangement scenario required to transform a genome with duplicated segments into a non-duplicated genome such that duplications are caused by rearrangement breakpoints. We show that the problem, in the Double-Cut-and-Join (DCJ) and the reversal rearrangement models, can be reduced to an APX-complete problem, and we provide algorithms for the Genome Dedoubling Problem with 2-approximable parts. We apply the methods for the reconstruction of a non-duplicated ancestor of Drosophila yakuba. CONCLUSIONS: We present the Genome Dedoubling Problem, and describe two algorithms solving the problem in the DCJ model, and the reversal model. The usefulness of the problems and the methods are showed through an application to real Drosophila data.
