ABSTRACT

Bacillus spp. are probiotics and can secrete a variety of natural antimicrobiol active substances, of which lipopeptides are an important class. Up to now, about 90 lipopeptides have been identified, and most of them are cyclic lipopeptides. surfactin, iturin, fengycin, bacillomycin and polymyxins are widely studied, and the first three have huge potential for application due to their properties of surfactants and anti-fungal, anti-bacterial, anti-viral, anti-tumor and anti-inflammatory functions. In this paper, the research progress in the structure, function, synthesis regulation, separation, purification and production of surfactin, iturin and fengycin was reviewed. Synthetic biology is a vital means to increase the yield of lipopeptides, and in the future, lipopeptides can be used in crop cultivation, animal farming, food, medicine and petroleum industries as well as environmental protection. Future research should be strengthened on the discovery of new lipopeptides, synthesis of high-activity lipopeptides, economical production of lipopeptides on a large scale and their safety evaluation.

ABSTRACT

In the present study, we focused on designing a species-specific chloroplast vector for Capsicum annuum L. and finding out its transformation efficiency compared to a heterologous vector. The plastid transformation vector (CaIA) was designed to target homologous regions trnA and trnI of IR region. A selectable marker gene aadA, whose expression is controlled by psbA promoter and terminator, was cloned between two flanking regions. A heterologous vector pRB95, which targets trnfM and trnG of LSC region along with aadA driven by rrn promoter and psbA terminator, was also used for developing plastid transformation in Capsicum. Cotyledonary explants were bombarded with stabilized biolistic parameters: 900 psi pressure and 9 cm flight distance, and optimized regeneration protocol (0.7 mg/L TDZ + 0.2 mg/L IAA) was used to obtain transplastomic lines on selection medium (300 mg/L spectinomycin). The aadA integration and homoplasmy were confirmed by obtaining 1.2 and 3.7 kb amplicons in CaIA transformants and subsequently verified by Southern blotting, whereas in pRB95 transformants, integration was confirmed by PCR with 1.45 kb and 255 bp amplicons corresponding to aadA integration and flanks, respectively. The transformation efficiencies attained with two plastid vectors were found to be 20%, i.e., 10 transplastomic lines in 50 bombarded plates, with CaIA and 2%, i.e., 1 transplastomic line in 50 bombarded plates, with heterologous pRB95, respectively.

ABSTRACT

Haplotype is the combination of a series of genetic mutations that coexist on a single chromosome, each of which has its own unique haplotypes. As a common data analysis method, the analysis of haplotype is effective for the localization of heterozygosis SNPs on single chromosome, the excavation of disease genes and the search of maladies treatments. It mainly includes indirect computational inferential method and direct experimental method. In this review we introduced various haplotype analysis methods and applications, especially two important ones: single-molecule dilution and contiguity-preserving transposition sequencing common technology. Meanwhile, further research prospects on haplotype sequencing were proposed.

ABSTRACT

Drought is a worldwide problem, seriously influencing plant (crop) productivity. Wheat is a stable food for 35% of the world population, and moreover, about 60% of land area on the globe belongs to arid and semiarid zone. Wheat drought resistance is a multi-gene controlling quantitative character and wheat final production in field is realized mainly by physiological regulation under the condition of multi-environmental factor interaction. Exploring drought resistance physiological mechanisms for different wheat genotypes is of importance to finding new drought resistance gene resources and conventional breeding, and the basis for wheat drought resistance biotechnological breeding and platform. Photosynthesis is the main component for physiological machinery of wheat assimilates conversion and wheat production. Investigation on photosynthetic characteristics of different wheat genotypes at soil water deficits also has other implications for refine physiological regulation of photosynthesis in fields and field management of crops in arid and semiarid areas. By pot-cultivating experiments, investigation of photosynthesis for 10 wheat genotypes at seedling stage and tillering stage at soil water deficits (75%FC, 55%FC and 45%FC, respectively) was conducted. The main results were as followed: developmental stages influenced wheat photosynthesis greatly and tillering stage played more roles; there were significant difference in the main photosynthetic parameters, photosynthesis rate (Photo), stomatal conductance (Cond) and transpiration rate (Tr), among 10 wheat genotypes; general photosynthesis and drought resistance in different wheat genotypes was related much to their domesticated origin soil water environment and selected generations and there was a photosynthetic threshold effect in terms of different wheat genotypes at soil water deficits.
