ABSTRACT

Some endocrine-disrupting chemicals (EDCs) can affect the endocrine system through covalent interactions with specific sites, leading to deregulation of physiological homeostasis. The acrylamide (AA) present in some fried or baked foods is an example of an electrophile molecule that is able to form adducts with nucleophilic regions of nervous system proteins leading to neurological defects. A positive correlation between increased urinary AA metabolite concentration and reduced levels of thyroid hormones (TH) was described in adolescents and young adults. Thus, this study aimed to evaluate whether AA affects the physiology of the hypothalamus-pituitary-thyroid (HPT) axis and the possible repercussions in peripheral TH-target systems. For this, male Wistar rats were exposed to doses of 2.5 or 5.0 mg AA/Kg/day, based on the LOAEL (Lowest Observed Adverse Effect Level) during prepubertal development. The expression of molecular markers of HPT functionality was investigated in the hypothalamus, pituitary, thyroid, heart and liver, as well as the hormonal and lipid profiles in blood samples. Herein, we showed that AA acts as EDCs for thyroid gland function, increasing the transcript expression of several proteins related to TH synthesis and altering hypothalamus-pituitary-thyroid axis homeostasis, an effect evidenced by the higher levels of THs in the serum. Compensatory mechanisms were observed in TH-target tissues, such as an increase in Dio3 mRNA expression in the liver and a reduction in Mct8 transcript content in the hearts of AA-treated rats. Together, these results pointed out an allostatic regulation of the HPT axis induced by AA and suggest that chronic exposure to it, mainly associated with food consumption, might be related to the higher prevalence of thyroid dysfunctions.

ABSTRACT

Reduced plasma vitamin D (VD) levels may contribute to excessive white adipose tissue, insulin resistance (IR) and dyslipidaemia. We evaluated the effect of chronic oral VD supplementation on adiposity and insulin secretion in monosodium glutamate (MSG)-treated rats. During their first 5 d of life, male neonate rats received subcutaneous injections of MSG (4 g/kg), while the control (CON) group received saline solution. After weaning, groups were randomly distributed into VD supplemented (12 µg/kg; three times/week) and non-supplemented (NS) rats, forming four experimental groups (n 15 rats/group): CON-NS, CON-VD, MSG-NS and MSG-VD. At 76 d of life, rats were submitted to an oral glucose tolerance test (OGTT; 2 g/kg), and at 86 d, obesity, IR and plasma metabolic parameters were evaluated. Pancreatic islets were isolated for glucose-induced insulin secretion (GIIS), cholinergic insulinotropic response and muscarinic 3 receptor (M3R), protein kinase C (PKC) and protein kinase A (PKA) expressions. Pancreas was submitted to histological analyses. VD supplementation decreased hyperinsulinaemia (86 %), hypertriacylglycerolaemia (50 %) and restored insulin sensibility (89 %) in MSG-VD rats, without modifying adiposity, OGTT or GIIS, compared with the MSG-NS group. The cholinergic action was reduced (57 %) in islets from MSG-VD rats, without any change in M3R, PKA or PKC expression. In conclusion, chronic oral VD supplementation of MSG-obese rats was able to prevent hyperinsulinaemia and IR, improving triacylglycerolaemia without modifying adiposity. A reduced cholinergic pancreatic effect, in response to VD, could be involved in the normalisation of plasma insulin levels, an event that appears to be independent of M3R and its downstream pathways.

ABSTRACT

The present study evaluated recipes of home-prepared diets for dogs and cats published in Portuguese. A total of 106 diets were evaluated: eighty for dogs, twenty-four for cats and two intended for both species. A commercial software package was used to analyse the diets, and an ingredient chemical composition database was built based on the Brazilian Tables of Food Composition and United States Department of Agriculture Nutrient Database. The estimated chemical composition of each recipe was compared with the Nutritional Guidelines for Complete and Complementary Pet Food for Cats and Dogs (Fédération Européenne de L'industrie des Aliments Pour Animaux Familiers; FEDIAF, 2014) recommendations for maintenance (as units/MJ). Most recipes (48 %) had no precise determination of ingredients and quantities. All diets had at least one nutrient below the recommendations, and all investigated nutrients were deficient in at least one diet. The most frequent nutrients below recommendation were: Fe (68·3 % of the recipes for dogs; 100 % of the recipes for cats); vitamin E (82·9 % of the dog recipes; 84·6 % of the cat recipes); Zn (75·6 % for dogs; 88·4 % for cats); Ca (73·2 % for dogs; 73 % for cats); Cu (85·4 % for dogs; 69·2 % for cats); choline (85·4 % for dogs; 69·2 % for cats); riboflavin (65·8 % for dogs; 11·5 % for cats); thiamine (39 % for dogs; 80·7 % for cats); and vitamin B12 (61 % for dogs; 34·6 % for cats). These recipes may potentially expose animals to nutritional deficiencies, and it is important to inform the owners of the risks of providing home-prepared diets. Better training of professionals that intend to prescribe home-prepared diets is advisable.

ABSTRACT

The objective was to evaluate the effects of dietary fish oil (FO) and vitamin E (VE) supplementation on sperm sensitivity to lipid peroxidation (LP) in dogs. Using an incomplete replicate 3 × 3 Latin square design, five dogs were allocated into three groups. One of the squares was incomplete and had two dogs that were used with three treatments. The dogs were assigned to three different treatments, fed a control diet of balanced commercial food (control group; CG), control diet supplemented with 54 mg FO/kg body weight0·75 per d (FO group; FG) and FO plus 400 mg VE per d (FO and VE group; FEG) for 60 d. Semen samples were collected on days 0 and 60 and divided into two halves, peroxidised and control, with or without ascorbate-Fe2+, respectively. LP was measured in both halves by chemiluminescence as counts per min/mg protein. Fatty acid profile was determined by GC. Data were analysed using the mixed procedure (SAS). On day 0, LP increased in all groups in the peroxidised samples (P < 0·05). However, on day 60 LP decreased in peroxidised samples of both the FG and FEG (P < 0·05), but there were no differences between the FG and FEG (P > 0·1). Additionally, on day 60 total n-3 was higher in the FG and FEG compared with the CG (P < 0·05). Supplementation with FO alone or together with VE decreased LP in peroxidised samples. These results could indicate a protective effect of n-3 on sperm. More studies are needed to understand the mechanism whereby FO and/or FO plus VE decrease LP in dogs' sperm.

ABSTRACT

Surveys report that 25-57 % of cats are overweight or obese. The most evinced cause is neutering. Weight loss often fails; thus, new strategies are needed. Obesity has been associated with altered gut bacterial populations and increases in microbial dietary energy extraction, body weight and adiposity. This study aimed to determine whether alterations in intestinal bacteria were associated with obesity, energy restriction and neutering by characterising faecal microbiota using 16S rRNA gene sequencing in eight lean intact, eight lean neutered and eight obese neutered cats before and after 6 weeks of energy restriction. Lean neutered cats had a bacterial profile similar to obese rodents and humans, with a greater abundance (P<0·05) of Firmicutes and lower abundance (P<0·05) of Bacteroidetes compared with the other groups. The greater abundance of Firmicutes in lean neutered cats was due to a bloom in Peptostreptococcaceae. Obese cats had an 18 % reduction in fat mass after energy restriction (P<0·05). Energy reduction was concurrent with significant shifts in two low-abundance bacterial genera and trends in four additional genera. The greatest change was a reduction in the Firmicutes genus, Sarcina, from 4·54 to 0·65 % abundance after energy restriction. The short duration of energy restriction may explain why few bacterial changes were observed in the obese cats. Additional work is needed to understand how neutering, obesity and weight loss are related to changes in feline microbiota and how these microbial shifts affect host physiology.

ABSTRACT

High dietary energy density (ED) has been associated with weight gain. However, little is known about the long-term effects of ED on weight changes among free-living subjects, particularly in Japanese and other Asian populations. In this study, we assessed dietary habits and weight changes in participants (5778 males and 7440 females, 35-69 years old) of the Takayama study. ED was estimated using a validated FFQ at baseline only. Information on body weight (BW) was obtained by self-administered questionnaires at baseline and follow-up. Mean BW difference in 9·8 years was 17 (se 4221) g for men and -210 (se 3889) g for women. In men, ED was positively associated with BW at follow-up after controlling for age, BW, height, physical activity score, alcohol consumption, energy intake, years of education at the baseline and change of smoking status during the follow-up. On average, men in the highest quartile of ED (>5·322 kJ/g (>1·272 kcal/g)) gained 138 (se 111) g, whereas men in the lowest ED (<1·057) lost 22 (se 111) g (P for trend=0·01). The association between ED and BW gain was stronger in men with normal weight. In women, the association between ED and weight change was not statistically significant. In conclusion, contrary to some studies that report an association between ED and weight gain in the overweight only, our data suggest that high-ED diets may be associated with weight gain in the lean population as well, at least in male subjects.

ABSTRACT

The association between dietary patterns and metabolic cardiovascular risk factors has long been addressed but there is a lack of evidence towards the effects of the overall diet on the complex net of biological inter-relationships between risk factors. This study aimed to derive dietary patterns and examine their associations with metabolic cardiovascular risk factors following a theoretic model for the relationship between them. Participants included 417 adults of both sexes, enrolled to the cross-sectional population-based study performed in Brazil. Body weight, waist circumference, high-sensitivity C-reactive protein, blood pressure, total cholesterol:HDL-cholesterol ratio, TAG:HDL-cholesterol ratio, fasting plasma glucose and serum leptin were evaluated. Food consumption was assessed by two non-consecutive 24-h dietary recalls adjusted for the within-person variation of intake. A total of three dietary patterns were derived by exploratory structural equation modelling: 'Traditional', 'Prudent' and 'Modern'. The 'Traditional' pattern had a negative and direct effect on obesity indicators (serum LEP, body weight and waist circumference) and negative indirect effects on total cholesterol:HDL-cholesterol ratio, TAG:HDL-cholesterol ratio and fasting plasma glucose. The 'Prudent' pattern had a negative and direct effect on systolic blood pressure. No association was observed for the 'Modern' pattern and metabolic risk factors. In conclusion, the 'Traditional' and 'Prudent' dietary patterns were negatively associated with metabolic cardiovascular risk factors among Brazilian adults. Their apparent protective effects against obesity and high blood pressure may be important non-pharmacological strategies for the prevention and control of obesity-related metabolic disorders and CVD.

ABSTRACT

The digestive tract of ferrets is anatomically simple, with no caecum, ileocolic valve or external differentiation between the transition of ileum and colon. The species has a short large intestine that provides minor contributions to the digestive process. Aiming to better understand the digestibility efficiency of ferrets, the present study compared the digestibility of extruded diets with different amounts of macronutrients fed to dogs, cats and ferrets. Three formulations for cat maintenance were used (values in % of DM basis): high carbohydrate (HC; nitrogen-free extract (NFE) = 54 %, protein = 31 % and fat = 8 %); moderate carbohydrate (MC; NFE = 37 %, protein = 41 % and fat = 10 %); and low carbohydrate (LC; NFE = 19 %, protein = 46 % and fat = 23 %). Apparent total tract macronutrient digestibility was determined by the method of total collection of faeces. Results were compared by ANOVA, considering the diet and species effects and their interactions. Means were compared by the Tukey's test (P < 0·05). Dogs and cats presented similar food intakes, but ferrets consumed almost two times more food (g/kg body weight). Species × diet interactions were verified for apparent total tract digestibility (ATTD; P < 0·05). Ferrets presented lower DM digestibility than dogs and cats for all three diets (P < 0·05), lower NFE digestibility than dogs for the three diets and lower NFE digestibility than cats for the HC and LC diets (P < 0·05). For crude protein (CP), ferrets presented lower ATTD than dogs and cats (P < 0·05), whereas for fat, dogs and ferrets presented similar ATTD, and higher values than those presented by cats (P < 0·05). Kibble diets had a lower DM, CP and NFE digestibility when fed to ferrets compared with dogs and cats. Fat digestibility was similar between dogs and ferrets and higher than that for cats.

Obesity is a growing problem in dogs. Therefore, there is an increasing need of foods for obese dogs with high-fibre content to dilute energies and to reduce energy absorption. Soya hulls are cheap and are widely available as a fibre source. We aimed at evaluating the body condition of dogs fed diets containing 0 % soya hulls (0SH) or 16 % soya hulls (16SH) in replacement of maize. Twelve adult dogs, with 11·3 (se 1·6) kg average body weight (BW), 4·1 (se 0·1) years old and body condition score (BCS) between 4 and 7, were completely randomised assigned (six per treatment) and were fed the 0SH diet according to their maintenance energy requirements or the same amount in grams (g/kg BW(0·75)) of the 16SH diet once daily for 56 d. The animals were evaluated on days 0 and 57 for BW, BCS (1, very thin to 9, obese), subcutaneous fat thickness in the L7 vertebra using ultrasound (L7), canine BMI (CBMI) and body fat (BF). Data were analysed by the Student's t test and Kruskal-Wallis test (P < 0·05). The change (final - initial) in BW (-0·58 v. -0·49 kg), BCS (-1 v. -1), L7 (-2 v. 0·35 mm), CBMI (-0·85 v. -0·63 kg/m(2)) and BF (-5·0 v. -5·4 %) of dogs fed the 0SH and 16SH diets, respectively, were not different (P > 0·05). The 16SH diet, with 11·4 % restriction in metabolisable energy, did not change the BCS of adult dogs. Further studies evaluating the supply of soya hulls only to overweight/obese dogs should to be carried out, because these dogs may respond differently than the group evaluated, which had a BCS between 4 and 7 (ideal to overweight).

Dietary fibre may promote satiety and reduce energy consumption per gram of food. Associated with feeding management, dietary fibre may contribute to reduce anxiety in dogs submitted to food restriction to lose weight. The objective of the present study was to evaluate the food intake and the behaviour of dogs fed diets without soya hulls (0SH) or with soya hulls (16SH) once (1×) or twice (2×) daily. Eight adult Beagle dogs, with 11·3 (se 1·6) kg body weight (BW), 4·1 (se 0·1) years of age and body condition score between 4 and 7, were distributed in 4 × 4 Latin square design (n 8/treatment, 5 d/period) in a 2 × 2 factorial arrangement (0SH v. 16SH; 1× v. 2×). Food allowance was 50 % in excess of the daily metabolisable energy (ME) requirements; food residues were weighed. The behavioural test consisted in continuously observations for 24 h, using the scanning sampling technique (observations every 10 min). General behaviours, such as sleeping, barking, standing and others were recorded. Food intake in g/kg BW(0·75) per d of 0SH and 16SH was not different (P > 0·05). However, dogs fed the 0SH diet presented higher (P > 0·05) energy intake (kJ/kg BW(0·75) per d; P < 0·05) compared with those fed the 16SH diet. Dogs fed 2× daily had higher intake, both in g and in kJ, compared with those fed 1 × , independently of diet. There was no interaction between diets and feeding frequency (P > 0·05). No behavioural differences were observed (P > 0·05). The diet containing 16SH, despite reducing ME intake, did not restrict food intake (in g). Dogs fed 1× per d had lower food intake, possibly due to gastric capacity limitation.

Insulin acts in the hypothalamus, decreasing food intake (FI) by the IR/PI3K/Akt pathway. This pathway is impaired in obese animals and endoplasmic reticulum (ER) stress and low-grade inflammation are possible mechanisms involved in this impairment. Here, we highlighted the amygdala as an important brain region for FI regulation in response to insulin. This regulation was dependent on PI3K/AKT pathway similar to the hypothalamus. Insulin was able to decrease neuropeptide Y (NPY) and increase oxytocin mRNA levels in the amygdala via PI3K, which may contribute to hypophagia. Additionally, obese rats did not reduce FI in response to insulin and AKT phosphorylation was decreased in the amygdala, suggesting insulin resistance. Insulin resistance was associated with ER stress and low-grade inflammation in this brain region. The inhibition of ER stress with PBA reverses insulin action/signaling, decreases NPY and increases oxytocin mRNA levels in the amygdala from obese rats, suggesting that ER stress is probably one of the mechanisms that induce insulin resistance in the amygdala.
