ABSTRACT

Seven seawater polychlorinated biphenyls (PCBs) were measured in water (DAP), suspended particulate matter (SPM), and blue mussels (Mytilus edulis) collected from four beaches in northwest Portugal. PCBs were extracted using solid-phase-extraction, ultrasound-extraction and QuEChERS before GC-MS analysis. The two-year annual average concentrations of PCBs in DAP, SPM and the four-year analysis in mussels were âˆ¼ 4.4 ng/L, ∼15.9 µg/kg, and âˆ¼ 56.0 µg/kg. The results suggest higher concentrations of PCBs in summer for SPM and in spring for mussels, mainly those collected close to the Ave River estuary. The origins of PCBs remain uncertain. Risk assessment shows that PCBs in water are unlikely to harm local biota due to their low thyroid hormone toxicity equivalents (TEQ-TH; ∼1.4E-04 ng/L and âˆ¼ 4.1E-04 µg/kg) and on WHO toxicity equivalents (TEQ-WHO; ∼2.1E-05 ng/L and âˆ¼ 4.9E-05 µg/kg). However, the lifetime carcinogenic risk (LCR) for humans consuming local bivalves is concerning, as it exceeds 1.0E-06.

ABSTRACT

This review elucidates the physiological and endocrinological processes intrinsic to puberty and ovulation induction protocols in Bos indicus and Bos taurus beef heifers. Puberty is a complex physiological event involving gonadotropic and metabolic changes that lead to sexual maturity, first ovulation, and regular reproductive cycles, enabling females to reproduce. Exposure to progesterone-based hormonal protocols, with or without additional hormones, can reduce the age at first ovulation and improve sexual maturity through stimuli in the hypothalamus-pituitary-gonadal (HPG) axis and uterine development. However, inducing puberty differs from inducing ovulation, as it does not ensure the heifer will continue cycling or be ready to establish and maintain pregnancy after hormonal exposure. Regardless of the pharmacological basis, studies consistently report that beef heifers that had a corpus luteum (CL) prior to the timed-artificial insemination (TAI) protocol, have greater expression of estrus in response to synchronization and greater pregnancy per AI compared to heifers without a CL. The combination of P4 and E2 significantly impacts uterine development, increasing reproductive efficiency. Exposure to P4 causes a positive effect on inducing ovulation. However, studies indicate that the addition of E2 esters at the time of P4 device removal increases the ovulation rate. In general, the studies showed that fertility varied according to the type of the ovulation induction protocol used, but with inconsistent results. Although ovulation induction protocols are strategic tools to accelerate sexual maturity, a holistic view of the entire system is extremely important, combining integration with genetics and nutrition to enhance the reproductive outcomes of beef heifers. Future research is needed to understand and refine these protocols, driving the efficiency of beef cattle production systems.

ABSTRACT

Reproductive outcomes were evaluated in Nelore (Bos indicus) heifers submitted to one, two or no ovulation induction protocols based on progesterone (P4) and estradiol (E2) prior to a timed-artificial insemination (TAI) protocol. A total of 1,437 heifers (13.0 ± 0.8 mo old; 3.1 ± 0.1 of body condition score [BCS] and 279.9 ± 25.8 kg of body weight [BW]) were randomly assigned to 1 of 3 treatments: 0IND (n = 486): no ovulation induction protocol; 1IND (n = 481): one ovulation induction protocol; or 2IND (n = 470): two ovulation induction protocols. On Day -47, heifers from 2IND received a disinfected intravaginal P4 device (2 g, previously used for 21 d), kept until Day -40, when 0.5 mg of E2 cypionate (EC) was given. On Day -19, heifers from 2IND and 1IND underwent the same protocol. On Day 0, all heifers were submitted to the same TAI protocol, starting with a P4 device (0.5 g), 0.5 mg of cloprostenol sodium (PGF), and 1.5 mg of E2 benzoate. On Day 7, P4 device was removed, 0.5 mg of PGF, 0.5 mg of EC, and 200 IU of equine chorionic gonadotropin (eCG) were administered. The TAI was performed 2 d later (Day 9). Blood samples were collected on Days -47 and 0, to determine the presence of CL (circulating P4 concentrations ≥ 1.0 ng/mL). Ultrasound was performed on Days 40, 75 and between Day 150 and parturition to assess pregnancy per AI (P/AI) and pregnancy loss (PL). Statistical analyses were performed using SAS 9.4 (a-cP ≤ 0.05; A,B0.05 < P ≤ 0.10). The proportion of heifers with CL on Day -47 was similar among groups (3.4%). A greater proportion of heifers from 1IND had CL on Day 0, followed by 2IND, then 0IND (87.9a; 80.4b; 28.8c%). There was an effect of treatment on expression of estrus (2IND: 66.6a; 1IND: 67.2a; 0IND: 57.4b%), P/AI on Day 40 (2IND: 53.4a; 1IND: 43.9b; 0IND: 46.5b%), P/AI on Day 75 (2IND: 49.8a; 1IND: 40.5b; 0IND: 44.4ab%) and final P/AI (2IND: 45.5a; 1IND: 35.8b; 0IND: 40.5ab%). No differences were observed in PL (40-75 = 6.3%; 75-final = 9.6%; Total = 15.3%). Particularly within lighter heifers, there was an effect of treatment on P/AI on Day 40 (0IND: 39.2b; 1IND: 43.3ab; 2IND: 53.9a%) and on Day 75 (0IND: 36.6B; 1IND: 39.0AB; 2IND: 48.5A%). At the first pregnancy diagnosis, more nonpregnant heifers from 2IND had CL on Day 40 than 0IND, but 1IND did not differ from the other groups (85.4a; 74.8b; 80.8ab%). In conclusion, ovulation induction protocols performed prior to the TAI protocol increased the proportion of heifers with CL on Day 0. The use of two induction protocols resulted in greater fertility, particularly in lighter heifers, and increased cyclicity among nonpregnant heifers. These results indicate that this strategy may be an optimized method for inducing cyclicity and enhancing fertility of prepubertal Nelore heifers raised in pasture-based feeding systems.

ABSTRACT

Graphyne and two-dimensional porous carbon-based materials have garnered significant attention due to their interesting structural characteristics and essential properties for new technological applications. Within this scope, this work investigates the structural, thermal, electronic, optical, and mechanical properties of a novel two-dimensional allotrope that combines triangular (T) and hexagonal (H) rings, connected by acetylenic linkages (graphyne-like), thus named TH-graphyne (TH-GY). This study comprehensively characterizes the proposed system's behavior using density functional theory, ab initio molecular dynamics, and classical reactive molecular dynamics simulations. Our results confirm the structural stability of TH-GY. AIMD simulations demonstrate the material's thermal stability at elevated temperatures, while phonon dispersions indicate its dynamical stability. Electronic band structure calculations show that the system is metallic. The analysis of optical properties reveals intense activity in the visible and UV regions, with pronounced anisotropy. A machine learning interatomic potentials model was developed for TH-GY and used to determine the mechanical behavior of the system, which exhibits Young's modulus ranging from 263 to 356 GPa, highlighting its flexibility. Classical reactive MD simulations elucidate the fracture behavior of TH-GY, revealing distinct fracture patterns and mechanical anisotropy.

Three-dimensional (3D) fish hepatocyte cultures are promising alternative models for replicating in vivo data. Few studies have attempted to characterise the structure and function of fish 3D liver models and illustrate their applicability. This study aimed to further characterise a previously established spheroid model obtained from juvenile brown trout (Salmo trutta) primary hepatocytes under estrogenic stimulation. The spheroids were exposed for six days to environmentally relevant concentrations of 17α-ethinylestradiol-EE2 (1-100 ng/L). The mRNA levels of peroxisome (catalase-Cat and urate oxidase-Uox), lipid metabolism (acyl-CoA long chain synthetase 1-Acsl1, apolipoprotein AI-ApoAI, and fatty acid binding protein 1-Fabp1), and estrogen-related (estrogen receptor α-ERα, estrogen receptor ß-ERß, vitellogenin A-VtgA, zona pellucida glycoprotein 2.5-ZP2.5, and zona pellucida glycoprotein 3a.2-ZP3a.2) target genes were evaluated by quantitative real-time polymerase chain reaction. Immunohistochemistry was used to assess Vtg and ZP protein expressions. At the highest EE2 concentration, VtgA and ZP2.5 genes were significantly upregulated. The remaining target genes were not significantly altered by EE2. Vtg and ZP immunostaining was consistently increased in spheroids exposed to 50 and 100 ng/L of EE2, whereas lower EE2 levels resulted in a weaker signal. EE2 did not induce significant changes in the spheroids' viability and morphological parameters. This study identified EE2 effects at environmentally relevant doses in trout liver spheroids, indicating its usefulness as a proxy for in vivo impacts of xenoestrogens.

ABSTRACT

The aim was to compare reproductive outcomes of Nelore heifers submitted to timed AI (TAI) protocols, with 7 or 9 d of permanence of the intravaginal progesterone (P4) device and different times of prostaglandin F2α (PGF) administration, for first (n = 935) and second (n = 530) services. On Day -24, heifers without corpus luteum (CL) underwent a protocol for induction of ovulation. On Day 0, heifers received a P4 device (0.5 g) and 1.5 mg estradiol (E2) benzoate. In order for the TAI to be carried out on the same day, these treatments were performed 2 d later on the heifers treated with the 7-d protocol. Additionally, heifers received 0.5 mg PGF at different times, resulting in four experimental groups: 9dP4-PGFd9 (n = 365); 9dP4-PGFd7 (n = 369); 9dP4-PGFd0&9 (n = 364); 7dP4-PGFd0&7 (n = 367). These nomenclatures indicate for how many d the P4 device was kept and the specific day on which PGF was given. At P4 removal, all heifers received 0.5 mg E2 cypionate and 200 IU eCG, and TAI was performed 2 d later. Effects were considered significant when P ≤ 0.05 (superscript letters a,b) whereas a tendency was assumed when 0.05 < P ≤ 0.10. Groups 9dP4-PGFd0&9 and 7dP4-PGFd0&7 had lower percentage of heifers with CL at P4 removal. The diameter (mm) of the dominant follicle (DF) was affected by treatment at P4 removal (9dP4-PGFd9: 11.3 ± 0.3b; 9dP4-PGFd7: 11.8 ± 0.2ab; 9dP4-PGFd0&9: 12.6 ± 0.2a; 7dP4-PGFd0&7: 10.8 ± 0.2c) and at TAI (9dP4-PGFd9: 12.7 ± 0.3ab; 9dP4-PGFd7: 13.2 ± 0.2a; 9dP4-PGFd0&9: 13.4 ± 0.2a; 7dP4-PGFd0&7: 12.4 ± 0.3b). Expression of estrus (%) was affected by treatment (9dP4-PGFd9: 89.6a; 9dP4-PGFd7: 93.5a; 9dP4-PGFd0&9: 88.2ab; 7dP4-PGFd0&7: 85.6b). There were no differences among treatments for P/AI on Day 40 (30-35 d post AI), final P/AI (between Day 70 and parturition) and pregnancy loss (between Day 40 and final P/AI). When the permanence of the P4 device was compared, regardless of PGF treatments, 9-d protocols resulted in greater DF diameter at P4 removal and at TAI, and greater expression of estrus (90.4 vs. 85.6%) than the 7-d protocol. Despite that, the 7-d protocol resulted in greater P/AI on Day 40 (55.3 vs. 49.1%). In addition, there was an interaction between protocol duration and body weight, in which heavier heifers (≥ 307 kg) had greater P/AI when treated with the 7-d protocol, in comparison to 9-d. In conclusion, longer TAI protocols (9 d of P4 device duration) resulted in greater DF diameter and expression of estrus. However, the shorter TAI protocol (7 d of P4 device duration) produced greater P/AI on Day 40, particularly in heavier heifers. Within 9-d protocols, the additional dose of PGF on Day 0 or the anticipation of the PGF to Day 7 did not influence fertility.

ABSTRACT

In areas of machine learning such as cognitive modeling or recommendation, user feedback is usually context-dependent. For instance, a website might provide a user with a set of recommendations and observe which (if any) of the links were clicked by the user. Similarly, there is growing interest in the so-called "odd-one-out" learning setting, where human participants are provided with a basket of items and asked which is the most dissimilar to the others. In both of those cases, the presence of all the items in the basket can influence the final decision. In this article, we consider a classification task where each input consists of three items (a triplet), and the task is to predict which of the three will be selected. Our aim is not only to return accurate predictions for the selection task, but also to additionally provide interpretable feature representations for both the context and for each individual item. To achieve this, we introduce CARE, a specialized neural network architecture that yields Context-Aware REpresentations of items based on observations of triplets of items alone. We demonstrate that, in addition to achieving state-of-the-art performance at the selection task, our model can produce meaningful representations both for each item, as well for each context (triplet of items). This is done using only triplet responses: CARE has no access to supervised item-level information. In addition, we prove parameter counting generalization bounds for our model in the i.i.d. setting, demonstrating that the apparent sample sparsity arising from the combinatorially large number of possible triplets is no obstacle to efficient learning.

OBJECTIVE: The main objective of this study was to compare stress and anxiety levels in children undergoing surgical procedures with or without parental presence at induction of anesthesia by measuring salivary cortisol levels and applying the mYPAS. METHOD: Quasi-randomized trial with children aged 5-12 year, with ASA physical status I, II, or III, undergoing elective surgery. According to parents' willingness, the pair were defined as accompanied or unaccompanied group. Chi-square, Fisher's exact tests, Student's t test, Mann-Whitney, Hodges-Lehman and Spearman's tests were used for statistical analyzes. RESULTS: We included 46 children; 63% were preschool children mostly accompanied by their mothers (80%). The median mYPAS score was 37.5 (quartile range, 23.4-51.6) in unaccompanied children, and 55.0 (quartile range, 27.9-65.0) in accompanied children, with an estimated median difference of +11.8 (95% CI of 0 to 23.4; p = 0.044). There were no significant differences in the mean salivary cortisol levels. CONCLUSION: The level of anxiety was higher in accompanied children. ABSTRACT

KEY MESSAGE: We propose an "enviromics" prediction model for recommending cultivars based on thematic maps aimed at decision-makers. Parsimonious methods that capture genotype-by-environment interaction (GEI) in multi-environment trials (MET) are important in breeding programs. Understanding the causes and factors of GEI allows the utilization of genotype adaptations in the target population of environments through environmental features and factor-analytic (FA) models. Here, we present a novel predictive breeding approach called GIS-FA, which integrates geographic information systems (GIS) techniques, FA models, partial least squares (PLS) regression, and enviromics to predict phenotypic performance in untested environments. The GIS-FA approach enables: (i) the prediction of the phenotypic performance of tested genotypes in untested environments, (ii) the selection of the best-ranking genotypes based on their overall performance and stability using the FA selection tools, and (iii) the creation of thematic maps showing overall or pairwise performance and stability for decision-making. We exemplify the usage of the GIS-FA approach using two datasets of rice [Oryza sativa (L.)] and soybean [Glycine max (L.) Merr.] in MET spread over tropical areas. In summary, our novel predictive method allows the identification of new breeding scenarios by pinpointing groups of environments where genotypes demonstrate superior predicted performance. It also facilitates and optimizes cultivar recommendations by utilizing thematic maps.

ABSTRACT

The present study compared 2 strategies to initiate a progesterone (P4)-based timed artificial insemination (TAI) protocol for lactating dairy cows: only GnRH or estradiol benzoate (EB) plus GnRH (EB+GnRH). Lactating Holstein cows (n = 487; 184 primiparous and 303 multiparous) from 2 commercial dairy herds were used for their second or greater services postpartum. Each week, cows that were nonpregnant at the pregnancy diagnosis 32 d after a previous AI were randomly assigned to 1 of 2 experimental groups that differed only in the strategy to initiate (d 0) the TAI protocol. On d 0, every cow received a 2.0-g P4 implant; in the EB+GnRH group, cows were treated with 2.0 mg i.m. of EB and 16.8 µg i.m. of the GnRH analog buserelin acetate, whereas in the GnRH group, cows received only 16.8 µg i.m. of GnRH. On d 7 after the initial treatment, 0.530 mg i.m. of cloprostenol sodium (PGF) was administered in all cows, followed by a second dose on d 8, concomitant with 1.0 mg i.m. of estradiol cypionate and P4 implant withdrawal. The TAI was performed on d 10 (48 h after P4 device withdrawal) in both experimental groups. Only conventional Holstein semen was used throughout the study. The percentage of cows with corpus luteum (CL) on d 0 (73%) and overall ovulation rate after d 0 (54%) did not differ between groups. The CL regression between d 0 and the first PGF treatment was greater in the EB+GnRH group than the GnRH group (42% vs. 31%). Consequently, the proportion of cows with CL at PGF was greater when only GnRH was used on d 0 compared with EB+GnRH (86% vs. 82%), and the mean number of CL at PGF was greater (1.23 vs. 1.11). The expression of estrus near TAI was greater in GnRH group (84% vs. 77%), and cows showing estrus had greater (44% vs. 10%) pregnancy per AI (P/AI) on d 32 for both treatments. We found no effect of the presence of CL on d 0 or at PGF, nor of ovulation after d 0 or CL regression between d 0 and d 7 on fertility. However, fertility was critically impaired when cows did not have CL at both times, d 0 and at PGF treatment. We did not observe any interaction between treatment and other variables, and the P/AI was similar in cows receiving EB+GnRH or only GnRH on d 0 (37.8% vs. 36.6%). In summary, although there was no detectable difference in P/AI between treatments, this study demonstrated potential negative physiological outcomes caused by EB treatment on d 0 (greater incidence of luteolysis after d 0 and fewer cows with CL at PGF treatment). Overall, we found no benefit of adding EB at the initiation of a P4-based TAI protocol on fertility compared with using GnRH alone, despite differences in ovarian dynamics and expression of estrus.

ABSTRACT

From August 2019 to January 2020 the Brazilian Coast was impacted by the largest oil spill in the Tropical oceans ever since. Paiva Beach, one of the most preserved sandy beaches in the northeast tropical Brazilian coast, was among the most affected regions by the oil spill in October 2019. This area has important reef systems that harbor abundant macroalgal assemblages with very diverse epifaunal communities. The present study aims to evaluate the impacts of the 2019 oil spill on epifauna associated with the seaweed species Jania capillacea and Penicillus capitatus collected in Paiva Beach from July 2019 to October 2022, and use the interaction macroalgae/epifauna/timescale as a proxy to quantify the effects of oil spills on communities structure and recovery. The epifauna of both algae did not suffer severe changes in abundance immediately after the spill, however, the abundance of taxa like echinoderms, sea spiders (Pycnogonida) and peracarid crustaceans dropped during or soon after the event, whereas others, like sabellid worms, had strong increase in abundance, possibly related to the spill. Both phytal assemblages also had distinct temporal patterns in species diversity and abundance, the last being strongly correlated to amphipod abundance for some taxa. The epifauna of J. capillacea had a short-term tendency of decrease in taxonomic diversity, whereas the epifaunal communities of P. capitatus gained a few taxa in the months immediately after the disaster. The diversity profiles of both communities returned to pre-spill conditions a few months later, with a slower abundance recovery in J. capillacea. Local environmental characteristics like landscape heterogeneity, connectivity with other environments, and critical-species (mainly amphipods) were possibly the main buffering factors for the phytal communities after the oil spill.

RESUMO Objetivo: O principal objetivo deste estudo foi comparar os níveis de estresse e ansiedade em crianças submetidas a procedimentos cirúrgicos com ou sem presença dos pais na indução da anestesia, medindo os níveis de cortisol salivar e aplicando o mYPAS. Método: Ensaio quaserandomizado com crianças de 5 a 12 anos, com estado físico ASA I, II ou III, submetidas a cirurgia eletiva. De acordo com a disposição dos pais, o par foi definido como grupo acompanhado ou não acompanhado. Foram utilizados testes de qui-quadrado, exato de Fisher, t de Student, Mann-Whitney, Hodges-Lehman e Spearman para as análises estatísticas. Resultados: Foram incluídas 46 crianças; 63% delas em idade préescolar, principalmente acompanhadas por suas mães (80%). A pontuação mYPAS mediana foi de 37,5 (intervalo interquartil, 23,4-51,6) em crianças não acompanhadas e de 55,0 (intervalo interquartil, 27,9-65,0) em crianças acompanhadas, com uma diferença mediana estimada de +11,8 (IC de 95% de 0 a 23,4; p = 0,044). Não houve diferenças significativas nos níveis médios de cortisol salivar. Conclusão: O nível de ansiedade foi maior em crianças acompanhadas. RESUMEN Objetivo: El objetivo principal de este estudio fue comparar los niveles de estrés y ansiedad en niños sometidos a procedimientos quirúrgicos con o sin presencia de los padres en la inducción de la anestesia mediante la medición de los niveles de cortisol salival y la aplicación del mYPAS. Método: Ensayo cuasi-aleatorio con niños de 5 a 12 años, con estado físico ASA I, II o III, sometidos a cirugía electiva. Según la disposición de los padres, se definieron como grupo acompañado o no acompañado. Se utilizaron pruebas de chi-cuadrado, exacta de Fisher, t de Student, Mann-Whitney, Hodges-Lehman y Spearman para los análisis estadísticos. Resultados: Se incluyeron 46 niños; el 63% eran niños en edad preescolar, en su mayoría acompañados por sus madres (80%). La puntuación mYPAS mediana fue de 37,5 (rango intercuartílico, 23,4-51,6) en niños no acompañados y de 55,0 (rango intercuartílico, 27,9-65,0) en niños acompañados, con una diferencia mediana estimada de +11,8 (IC del 95% de 0 a 23,4; p = 0,044). No hubo diferencias significativas en los niveles medios de cortisol salival. Conclusión: El nivel de ansiedad fue mayor en los niños acompañados. ABSTRACT

A case of hepatic arteriovenous malformation is described in a calf that presented with a marked increase in abdominal volume and died 12 h after birth. At necropsy, the calf had marked abdominal distension and haemoperitoneum. The left hepatic lobe was reduced in size and covered by slightly whitish and intensely vascularized membranous tissue. At the edge of the left lobe there were cystic structures filled with translucent fluid. On cut surface, there were multiple cavitations well delimited by whitish walls and multiple dilated blood vessels that communicated with the cavitations. Histopathology revealed an increase in the number of vascular structures of variable calibre within loose fibrous connective tissue. The diagnosis of hepatic arteriovenous malformation was based on the lesions and confirmed by immunohistochemistry. Hepatic arteriovenous malformations are rare in all species, have not been described in cattle and should be included as a cause of mortality in neonates with post-partum haemoperitoneum.

ABSTRACT

BACKGROUND: There are few studies related to Coronavirus Disease 2019 (COVID-19) on the prevalence and nature of pain symptoms after hospital discharge, especially in individuals who develop moderate to severe disease forms. Therefore, this study aimed to evaluate the presence of chronic pain in patients discharged after hospitalization for COVID-19, and the relationship between the presence of chronic pain and intensive care stay, demographics, and risk factors for the worst Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) outcome. METHODS: A cross-sectional observational study was carried out on patients with COVID-19 who recovered after hospitalization. Patients were recruited at the least 3 months after discharge and their hospital's health files were prospected. The variables evaluated were demographics, the severity of SARS-CoV-2 infection (considering the need for intensive care), and the presence of chronic pain. The results were shown in a descriptive manner, and multivariate analysis expressed as Odds Ratios (ORs) and respective Confidence Intervals (CIs) for the outcomes studied. Statistical significance was set at p < 0.05. RESULTS: Of 242 individuals included, 77 (31.8%) reported chronic pain related to COVID-19, with no correlation with the severity of infection. Female sex and obesity were associated with a higher risk for chronic pain with ORs of 2.69 (Confidence Interval [95% CI 1.4 to 5.0]) and 3.02 (95% CI 1.5 to 5.9). The limbs were the most affected areas of the body. CONCLUSION: Chronic pain is common among COVID-19 survivors treated in hospital environments. Female sex and obesity are risk factors for its occurrence.

In a recommender systems (RSs) dataset, observed ratings are subject to unequal amounts of noise. Some users might be consistently more conscientious in choosing the ratings they provide for the content they consume. Some items may be very divisive and elicit highly noisy reviews. In this article, we perform a nuclear-norm-based matrix factorization method which relies on side information in the form of an estimate of the uncertainty of each rating. A rating with a higher uncertainty is considered more likely to be erroneous or subject to large amounts of noise, and therefore more likely to mislead the model. Our uncertainty estimate is used as a weighting factor in the loss we optimize. To maintain the favorable scaling and theoretical guarantees coming with nuclear norm regularization even in this weighted context, we introduce an adjusted version of the trace norm regularizer which takes the weights into account. This regularization strategy is inspired from the weighted trace norm which was introduced to tackle nonuniform sampling regimes in matrix completion. Our method exhibits state-of-the-art performance on both synthetic and real life datasets in terms of various performance measures, confirming that we have successfully used the auxiliary information extracted.

Mammal hepatocyte spheroids have been investigated as alternative experimental models in several contexts, since three-dimensional (3D) systems have shown the potential to mimic in vivo scenarios. The description of fish hepatocyte 3D models is still minimal. This study intends to further characterize brown trout primary hepatocyte spheroids at distinct time points up to 25 days in culture. Viability, biometry, histomorphology, and basal expression of a selection of genes (metabolism and detoxification, efflux transport, and estrogenic signalling) were considered. The gene expression of whole liver samples from the same fish donor were evaluated concurrently. After 12 days in culture, the hepatocyte spheroids exhibited biometric and morphological stability. From the 12th to the 20th day in culture, the basal expression levels for most of the selected genes did not vary. The targeted mRNA levels were higher in brown trout liver samples compared to hepatocyte spheroids. Despite that, data supported that this model resembles some in vivo features. As an experimental alternative model, it showed potential to be used in a stable time window that can be exploited for exposure tests to different xenobiotics, namely, estrogenic compounds.

ABSTRACT

Composted sewage sludge (CSS) is an organic fertilizer that can be used as a source of micronutrients in agriculture. However, there are few studies with CSS to supply micronutrients for the bean crop. We aimed to evaluate micronutrient concentrations in the soil and their effects on nutrition, extraction, export, and grain yield in response to CSS residual application. The experiment was carried out in the field at Selvíria-MS, Brazil. The common bean cv. BRS Estilo was cultivated in two agricultural years (2017/18 and 2018/19). The experiment was designed in randomized blocks with four replications. Six different treatments were compared: (i) four increasing CSS rates, i.e., CSS5.0 (5.0 t ha-1 of applied CSS, wet basis), CSS7.5, CSS10.0, CSS12.5; (ii) a conventional mineral fertilizer (CF); (iii) a control (CT) without CSS and CF application. The available levels of B, Cu, Fe, Mn, and Zn were evaluated in soil samples collected in the 0-0.2 and 0.2-0.4 m soil surface horizons. The concentration, extraction, and export of micronutrients in the leaf and productivity of common beans were evaluated. The concentration of Cu, Fe, and Mn ranged from medium to high in soil. The available levels of B and Zn in the soil increased with the residual rates of CSS, which were statistically not different from the treatments with CF. The nutritional status of the common bean remained adequate. The common bean showed a higher requirement for micronutrients in the second year. The leaf concentration of B and Zn increased in the CSS7.5 and CSS10.0 treatments. There was a greater extraction of micronutrients in the second year. Productivity was not influenced by the treatments; however, it was higher than the Brazilian national average. Micronutrients exported to grains varied between growing years but were not influenced by treatments. We conclude that CSS can be used as an alternative source of micronutrients for common beans grown in winter.

Objetivo ­ Verificar a suscetibilidade de criação de fômites de Sars-Cov-2 de acordo com temperatura, umidade e período de exposição. Métodos ­ Foi realizada uma revisão sistemática da literatura, entre março de 2021 e junho de 2022, utilizando-se a Biblioteca Virtual em Saúde (BVS), PubMed e Medline, através do cruzamento dos termos SARS-COV-2 AND desinfecção AND higienização AND fômites AND superfícies. Resultados ­ Dos 295 artigos encontrados, 24 foram selecionados e divididos em 3 categorias temáticas, "A influência das condições ambientais", "detecção por período de exposição" e "amostragens não experimental". Conclusões ­ Foi observado que existe a possibilidade da formação de fômites de Sars-Cov-2 e as condições ambientais influenciam diretamente, exigindo assim cuidado com superfícies possivelmente contaminadas. Abstract Background: Conventional dental care is often impossible in patients with Autism Spectrum Disorder (ASD). Non-collaborative behaviors, sometimes associated with aggressiveness, are usual justifications for premedication in this population. Thereby, this research focuses on the effects of oral midazolam versus oral ketamine plus midazolam as preanesthetic medication in ASD. Methods: The sample included 64 persons with ASD, aged 2-59 years, scheduled for dental care under general anesthesia. The primary objective of this study was to compare degrees of sedation between two parallel, double-blinded, equally proportional groups randomized to receive oral midazolam (0.5 mg.kg−1, maximum 15 mg) or oral midazolam (0.5 mg.kg−1) associated with oral S(+)-ketamine (3 mg.kg−1, maximum 300 mg). The secondary outcomes were the need of physical stabilization to obtain intravenous line, awakening time, and occurrence of adverse events. Results: According to the dichotomous analysis of sedation level (Ramsay score 1 and 2 versus Ramsay ≥ 3), oral association of S(+)-ketamine and midazolam improved sedation, with increased probability of Ramsay ≥ 3, Relative Risk (RR) = 3.2 (95% Confidence Interval [95% CI] = 1.32 to 7.76) compared to midazolam alone. Combined treatment also made it easier to obtain venous access without physical stabilization, RR = 2.05 (95% CI = 1.14 to 3.68). There were no differences between groups regarding awakening time and the occurrence of adverse events. Conclusion: The association of oral S(+)-ketamine with midazolam provides better preanesthetic sedation rates than midazolam alone and facilitates intravenous line access in patients with autism.
