ABSTRACT

Manganese (Mn) is a metal and important micronutrient. However, exposure to supraphysiological levels of Mn, which occur through fungicides, atmospheric emissions, drainages, and spills, has been related to health risks, including morphometric changes in the male reproductive organs and impairment on gametogenesis and sperm quality, impacting the fertile ability of adult animals. Despite the relevance of the fetal/perinatal period for toxicological studies on Mn, previous data only deal with the physical and neurological development of the offspring, without mentioning their reproductive development. The present study investigated whether exposure to Mn during fetal/perinatal phase, specifically during the period of formation and proliferation of Sertoli cells, impairs the reproductive development of male offspring in early postnatal life. Therefore, pregnant Wistar rats were randomly distributed into 3 experimental groups: Ctl (received saline solution), Mn-9 (received 9 mg/kg of MnCl2), and Mn-90 (received 90 mg/kg of MnCl2). The female rats received the experimental treatment by gavage from gestational day 13 to lactational day 15, i.e., postnatal day (PND) 15 of the pups. Oxidative damage to the genetic material of germ and Sertoli cells, together with a decrease in connexin 43 immunolabeling were observed in the testis of male pups evaluated at PND 15. In addition, an increase in the seminiferous tubules presenting slight epithelium vacuolization and cells with eosinophilic cytoplasm were observed, without apparent epididymal changes. In conclusion, it was demonstrated that Mn perturbed the initial testicular development by altering Sertoli cell integrity through oxidative insult, which may compromise the spermatogenesis in the long-term.

ABSTRACT

This study evaluated the effect of different extraction technologies and conditions in order to obtain jaboticaba skin extracts. Firstly, the skins were extracted by conventional extraction, according to a rotatable central composite design, varying ethanol concentration, solid:liquid ratio, and temperature. Next, ultrasound-assisted extraction was performed using different power densities and times. Finally, high-pressure extractions were performed with varying pressures and times. For agitated bed extraction, the highest anthocyanin content was observed for ethanol concentrations varying between 60% and 80%. Thus, the independent variables which more influenced anthocyanin content were ethanol concentration and solid:liquid ratio. Folin-Ciocalteu reducing capacity was linearly affected by the increase in temperature. Ethanol concentration was the variable that most influenced ABTS+. On the other hand, the increase in ethanol concentration decreased the antioxidant capacity by ABTS+. Considering the ultrasound extraction, increasing its power did not affect total monomeric anthocyanins content, while the increase in process time had better yields. The highest antioxidant capacity and total monomeric anthocyanins were found for the highest extraction time. Similarly, with ultrasound, the increase in high hydrostatic-assisted extraction time positively influenced anthocyanin content and antioxidant capacity. As a result, the ultrasound-assisted method was found to be the best extraction technology for anthocyanins recovery.

ABSTRACT

Objetivamos refletir como os censos do INEP podem possibilitar um critério para analisar a formação do psicólogo no Brasil. Estabelecemos uma estratégia metodológica baseada na pesquisa bibliográfica e análise documental. Contextualizamos o surgimento do INEP e a sua lógica de estabelecer o censo do ensino superior pelas figuras de Lourenço Filho e Anísio Teixeira, suas estadas na Universidade de Columbia e apropriações de ideias psicológicas, educacionais e administrativas. Adentramos tais censos, entre 1940-2010, para entender o expansionismo educacional e a formação do psicólogo. No censo de 2020,observamos: predominância de cursos privados sobre os públicos; expansão formativa em cursos interioranos; expressão da mercantilização da Psicologia; discrepância entre os cursos de Bacharelado e Licenciatura; equivalência e diminuição da oferta de cursos de Licenciatura no ensino público e privado. Concluímos que a análise empreendida ajuda a entender o campo da formação do psicólogo em suas nuances históricas, políticas, estatísticas, sociais e educacionais.


We aim to reflect on how the INEP's censuses mayprovide a criterion to analyze the professional education of psychologists in Brazil.We established a methodological strategy based on bibliographic-research and document analysis. We contextualized the INEP's emergence and its logic of establishing higher education census through the figures of Lourenço Filho and Anísio Teixeira, their stays at Columbia University and appropriations of psychological, educational and administrative ideas. Weexamedsuch censuses, between1940-2010, to understand the educational expansionism and professional education of psychologists.In the 2020 census, we observe: predominance of private courses over public ones; educationalexpansion in inland courses; expression of a commodified Psychology; discrepancy between the Bachelor's and the Teaching License Degree in the education of psychologists; equivalence and reduced offers of Psychology Degreecourses in public and private education. We conclude that the analysis undertakenhelps to understand the psychologist professional educationfield in its historical, political, statistical, social and educational nuances.

ABSTRACT

Rubber bushings and mounts are vastly used in automotive applications as support and interface elements. In suspension systems, they are commonly employed to interconnect the damping structure to the chassis. Therein, the viscoelastic nature of the material introduces a desirable filtering effect to reduce mechanical vibrations. When designing a suspension system, available literature often deals with viscoelastic mounts by introducing a linear or nonlinear stiffness behavior. In this context, the present paper aims at representing the rubber material using a proper viscoelastic model with the selection of different in-wheels motors. Thus, the mount dynamic behavior's influence in a suspension is studied and discussed thoroughly through numerical simulations and sensitivity analyses. Furthermore, guidelines are proposed to orient the designer when selecting these elements.

ABSTRACT

Eugenia copacabanensis and Myrciaria tenella are present in restingas of the Atlantic Forest, but little information is available about their chemical and biological potential. In this context, the hexane, dichloromethane, ethyl acetate and butanol fractions from the leaves of methanolic extract were analyzed by GC/MS and HPLC-DAD and the antioxidant potential was determined by DPPH and ABTS assays and using a Saccharomyces cerevisiae model. Dereplication allowed the identification of 68 compounds, 42 and 41 of which, respectively, are first reported here for E. copacabanensis and M. tenella. In vivo results revealed that the ethyl acetate and butanol fractions showed expressive antioxidant protection in the BY4741 and Δgsh1 strains, with greater impact on glutathione-deficient cells. With a high diversity of phenolic compounds, these polar fractions of E. copacabanensis and M. tenella leaves are potential protectors against intracellular oxidative stress.

ABSTRACT

The management of agrochemicals in Brazilian agriculture impacts global environmental sustainability and food security, since this country is one of the major agro-food exporters in the world. Acephate, carbendazim, and dithiocarbamates (DTCs) such as mancozeb, are among the most detected agrochemicals in Brazilian agro-food products, occurring in combination in several crops, especially in fruit cultures. The present study evaluated the impact of the exposure to isolated agrochemicals and all the combined possible mixtures (binary and ternary forms) on the reproductive parameters of male juvenile rats, known to be a vulnerable biological system and developmental window. Data were analyzed using Generalized Linear Models (GzLM), considering each agrochemical as an independent factor. The study revealed higher reproductive toxicity exerted by isolated agrochemicals when compared to the combined treatments, which exhibited mostly an antagonistic effect. Results suggest endocrine disruptive effects of each one separately on the weight of reproductive organs and testicular histomorphometry, besides changes in testicular SOD activity. The full factorial experimental design employed here allowed us to conclude that it is not possible to scale-up the effects of the isolated treatments to the mixtures, showing how difficult it is to know beforehand the response and cross-talk among the multiple physiological mechanisms disturbed by complex mixtures. Considering that food products are shared on a global scale and that some of these three agrochemicals have already been prohibited in EU countries, the consumption of some Brazilian products puts global human health at risk, that of children.

ABSTRACT

For the first time, juvenile toxicity of inorganic arsenic (As) was investigated in male rats, focusing on reproductive effects. As is a metalloid naturally occurring in the environment, being the inorganic forms the most toxics. Contaminated drinking water and agricultural products are the main prospectors of intoxication for general population. In the present study, Wistar male rats (21 days old) were distributed into three groups (n = 10/group): control (received vehicle-filtered drinking water), As1 (received AsNaO2 at 0.01 mg L-1 ) and As2 (received AsNaO2 at 10 mg L-1 ). The animals were euthanized on PND 53. Testicular damages increased in As1 and As2 compared to control (ie, presence of vacuolization, acidophilic cells, and epithelium degeneration). Testicular interstitium of As1 and As2 presented fluid's increase and intense inflammatory infiltration. In the epididymis there was reduction of sperm amount in the lumen, besides epithelium areas presenting cribriform aspect in As1 and As2, exfoliation of cells in the light (in As1) and vacuoles (in As2). In epididymis interstitium, inflammatory infiltrates were observed in initial segment of As1 and As2. AsNaO2 changed immunolabeling pattern for androgen receptor in epididymis of As2, although serum testosterone levels was statistically comparable to control. Mass spectrometry revealed higher As concentrations in testis and epididymis of As2 compared to As1 and Control. These results indicate compromise of spermatogenesis and epididymal histophysiology in AsNaO2 -treated animals, possibly impairing sperm quality and fertility in long-term, even at low levels of exposure. Investigations about the reversibility of reproductive damages are necessary to better understand the mechanisms of As reproductive toxicity.

ABSTRACT

M. floribunda fruit was studied to characterize its chemical composition. The chemical composition, bioactive compounds, antioxidant activity and volatiles of the fruit were determined. The chemical composition was determined according to AOAC and AOCS, the bioactive compounds by HPLC, the volatiles by GCMS and the antioxidant activity by ABTS+ and DPPH methods. The chemical composition of the freeze-dried fruit was 1.89 g·100 g-1, 2.43 g·100 g-1, 4.78 g·100 g-1 and 90.89 g·100 g-1 of ashes, lipids, proteins and total carbohydrates on a dry base, respectively. The concentration of the carotenoids was 52.22 mg·100 g-1 and for the flavonoid rutin was 78.56 mg·100 g-1. The gallic and ellagic acid contents were 5.45 mg·g-1 and 2.21 mg·g-1, respectively. The cis-ß-ocimene corresponded to 50.90% of the volatiles. The antioxidant activity by ABTS+ method was 550.14 µmol Trolox·g-1 and by the DPPH method the EC50 was 85.68 g·g-1. The fruits presented relevant antioxidant activity, a high concentration of carotenoids and of rutin.

Introducción: el formaldehído es un importante químico con muchos derivados y usos comerciales. Estudios epidemiológicos en poblaciones ocupacionalmente expuestas a formaldehído lo tiende a asociar con un número de efectos biológicos en humanos. Materiales y métodos: para determinar los posibles daños ocasionados en población expuesta, se realizó una evaluación genotóxica en 40 Médicos Legistas y Disectores hombres y mujeres expuestos a formaldehído; un grupo similar de 40 empleados administrativos hombres y mujeres de la misma institución pero no expuestos a formaldehído sirvieron como no expuestos; para determinar la frecuencia de diferentes tipos de aberraciones cromosómicas estructurales y numéricas. Resultados: en el grupo ocupacionalmente expuesto a formaldehído un significativo incremento en la incidencia de: fracturas cromatídicas, fracturas cromosómicas, cromosomas en anillo y poliploídias fue observado. La exposición a formaldehído fue tres veces mayor en el grupo de expuestos a formaldehído. El nivel de exposición a formaldehído más alto fue encontrado cuando los cadáveres eran cerrados, seguidos del embalsamamiento (formolización) de los cuerpos. Conclusión: los resultados demostraron que la exposición a formaldehído induce aberraciones cromosómicas en linfocitos de sangre periférica del personal que labora en las morgues de los Departamentos Médico Legales.


Introduction: formaldehyde is an important chemical with many derivatives and commercial uses. Epidemiological studies of occupational population exposed to formaldehyde prove to be associated to biological effects in humans. Materials and methods: chromosome analyses were carried out in lymphocytes of 40 forensic doctors and dissectors male and female exposed to formaldehyde (FA) for 3 -20 years. A similar group of 40 males and females from the same police Institution but without FA exposure served as a control group with non exposure in order to establish frequency and different types of numerical and structural chromosomal aberrations. The exposed group to formaldehyde showed significantly increased incidence of breaks of the chromatid type, breaks of the chromosomes type, ring chromosomes and polyploidies was observed. Results: total mean exposure time to FA was about 3 times longer than non exposure group. The highest exposures to formaldehyde were encountered when autopsied cases were closed, followed by when autopsies cases were embalmed. Conclusions: the results demonstrate that exposure to formaldehyde induces chromosomal aberrations in peripheral blood lymphocytes of personnel working in murders of legal medicine departments.

ABSTRACT

Objetivou-se averiguar em publicações científicas a influência dos distúrbios do sono no desenvolvimento infantil. Revisão integrativa da literatura, realizada no mês de dezembro de 2012 e março de 2014 nas bases de dados: LILACS, Pubmed, CINAHL, Scopus e Cochrane, abrangendo o periodo de publicação de 2003 a março de 2014, com os descritores desenvolvimento infantil e distúrbios do sono. Como amostra da pesquisa, foram selecionados oito artigos constantes na base de dados SCOPUS e três na PubMed. Dois desses artigos foram indexados repetidamente nas duas bases. Dessa forma, resultou em uma amostra final de nove artigos publicados entre 2007 e 2013; três são do tipo longitudinal e seis são revisões de literatura. A síntese dos resultados aponta evidências do distúrbio de sono como possível influenciador em alterações no desenvolvimento, principalmente, no aspecto cognitivo e comportamental.


This study aimed to verify, in scientific publications, the influence of sleep disorders on child development. It is an integrative literature review, undertaken in December 2012 and March 2014 in the LILACS, Pubmed, CINAHL, Scopus and Cochrane databases, in the period of publication covering 2003 - March 2014, with the descriptors 'child development' and 'sleep disorders'. As the research sample, eight articles were selected from the SCOPUS database and three from PubMed. Two of these articles were indexed repeatedly in these two databases. As a result, the final sample had nine articles, published between 2007 and 2013; three are of the longitudinal type and six are literature reviews. The summary of the results indicates evidence of sleep disorders as possible influences on alterations in development, mainly, in the cognitive and behavioral aspects.


El propósito del estudio fue analizar la influencia de los disturbios del sueño en el desarrollo infantil en publicaciones científicas. Revisión integrativa de la literatura, realizada en diciembre de 2012 y marzo de 2014 en las bases de datos: LILACS, Pubmed, CINAHL, Scopus y Cochrane, abarcando el periodo de publicación de 2003 a marzo de 2014, con los descriptores desarrollo infantil y disturbios del sueño. Como muestra de la investigación, fueron seleccionados ocho artículos constantes en la base de datos SCOPUS y tres en la PubMed. Dos de eses artículos fueron indexados repetidamente en las dos bases mencionadas. De esa forma, la muestra final resultó en nueve artículos publicados entre 2007 y 2013; tres son del tipo longitudinal y seis son revisiones de literatura. La síntesis de los resultados apunta evidencias del disturbio de sueño como posible influenciador en alteraciones en el desarrollo, principalmente, en el aspecto cognitivo y de comportamiento.

ABSTRACT

Medical genetics is a field marked by fast progress. Even though it was at one point confined to a group of relatively rare diseases, today it has become a central component in the understanding of disorders and it is the subject of interest for all medical specialties. This paper, shares data on the chromosomal alterations and variations that have been diagnosed in Ecuadorian patients since 1998. A total of 2,636 individual cases have been analyzed by G-banding technique until February 2012. The present work shows this collection of data and the important findings that have appeared throughout these years in hopes that it can contribute to have a deeper understanding of the incidence of chromosomal aberrations and alterations in the Ecuadorian population.

ABSTRACT

Shiga toxin-producing Escherichia coli (STEC) is an important foodborne pathogen, and ruminants are recognized as the main natural reservoir. The purposes of this study were to detect E. coli O157 in bovine feces and surface water in a beef cattle farm of Gualeguaychú, Argentina; to characterize the isolates; and to establish the clonal relatedness by pulsed-field gel electrophoresis. Between September 2005 and November 2006, 288 samples of bovine feces and 79 samples of water troughs were studied. E. coli O157 was detected by immunomagnetic separation and polymerase chain reaction as screening techniques. The rfb(O157) gene was detected in 3.8% of the 288 fecal samples and in 17.7% of the 79 water samples. The stx gene was detected in all rfb(O157)-positive fecal samples and in 5.1% of water samples. Eleven E. coli O157 strains isolated from bovine fecal samples and eight from water samples were characterized. The most frequent stx genotype identified was stx(1) and stx(2c(vh-a)). Twelve (63.2%) strains harbored fliC(H7), eae, and ehxA genes. Using pulsed-field gel electrophoresis with the enzyme XbaI, a total of eight patterns with at least 72.1% similarity were identified among the 19 strains. The patterns of 15 strains were grouped into four clusters: two of them included only bovine strains and the other two only aquatic strains. No genetic correlation was established between the bovine and water STEC strains detected. The prevalence of STEC O157:H7 established in the herd studied was higher than that previously reported for Argentine grazed cattle.
