BACKGROUND: There is increasing evidence that platelet-derived extracellular vesicles (PEVs) may be involved in the mechanisms of inflammatory storm and organ damage in sepsis. However, there are no available studies on PEVs and renal injury in patients with urosepsis. METHODS: We analyzed the concentration and ratio of PEVs in plasma by flow cytometry and measured plasma IL-1ß/IL-6/TNF-α/NGAL levels by ELISA. Correlation analysis was also used to examine the concentration of PEVs in relation to levels of inflammatory factors and indicators of kidney damage, as well as the severity of the disease. Finally, the receiver operating characteristic curves were produced for PEVs concentrations as a diagnosis of S-AKI/AKI. RESULTS: We found significantly higher levels of IL-1ß/IL-6/TNF-α/NGAL in patients with urogenital sepsis. Furthermore, the concentrations of PEVs in plasma were significantly elevated in patients with urosepsis, especially in patients with Gram-negative bacterial infections, which were significantly and positively correlated with IL-1ß/IL-6/TNF-α/NGAL levels. The area under the curve for PEVs diagnosing S-AKI and AKI was 0.746 [0.484, 1.000] and 0.943 [0.874, 1.000] respectively. CONCLUSION: Overall, the present study suggested that PEVs may mediate the release of inflammatory mediators in patients with urosepsis and participate in the mechanism of acute kidney injury, as well as having potential as diagnostic indicators of S-AKI and AKI and as early warning indicators of the severity of patients with urosepsis.

Introduction: Erectile dysfunction (ED) is a common disease among elderly men, and novel therapy methods are needed for drug-refractory ED. As an extracellular vesicle, stem cell-derived exosomes displayed erectile function improvement in rat ED models in some preclinical studies. However, the therapeutic efficacy has not been comprehensively evaluated. Aim: To study the therapeutic effects of stem cell-derived exosomes on ED in preclinical studies and to investigate the potential mechanisms responsible for the efficacy. Methods: The systematic literature search was conducted in Web of Science, PubMed, and Embase to retrieve studies utilizing stem cell-derived exosomes for ED treatment. We extracted data of intracavernous pressure/mean artery pressure (ICP/MAP), and cavernosum structural changes in rat ED models before and after stem cell-derived exosome therapy. RevMan 5.3 was used to perform meta-analyses of ICP/MAP and cavernosum microstructural changes. Publication bias was assessed with the Egger test and funnel plot by Stata 15.0 (StataCorp). Main Outcome Measures: Outcomes included ICP/MAP, smooth muscle, and endothelial markers-such as the ratio of smooth muscle to collagen and the expression of α-SMA (alpha smooth muscle actin), CD31 (cluster of differentiation 31), nNOS and eNOS (neuronal and endothelial nitric oxide synthase), TGF-ß1 (transforming growth factor ß1), and caspase 3 protein-to evaluate erectile function and microstructural changes. Forest plots of effect sizes were performed. Results: Of 146 studies retrieved, 11 studies were eligible. Pooled analysis showed that stem cell-derived exosomes ameliorated damaged ICP/MAP (standardized mean difference, 3.68; 95% CI, 2.64-4.72; P < .001) and structural changes, including the ratio of smooth muscle to collagen and the expression of α-SMA, CD31, nNOS, eNOS, TGF-ß1, and caspase 3 protein. Subgroup analysis indicated that exosome type and ED model type made no difference to curative effects. Conclusion: This meta-analysis suggests the therapeutic efficacy of stem cell-derived exosomes for ED. Exosomes may restore erectile function by optimizing cavernosum microstructures.

Extracellular vesicles (EVs) are vesicular bodies with a double-layered membrane structure that are detached from the cell membrane or secreted by the cells. EVs secreted by platelets account for the main part in the blood circulation, which account for about 30% or even more. Many types of cells are regulated by PEVs, including endothelial cells, leukocytes, smooth muscle cells, etc. Nevertheless, despite the growing interest in the study of extracellular vesicles, there are still only a few studies on the role of PEVs. Therefore, this overview mainly focuses on one method of isolation and the functions of PEVs in tissues found so far, including promoting tissue repair and mediating tissue damage, which can be used for researchers to continue to explore the role of PEVs in other fields.

The generation of construction and demolition waste1 (CDW) in China has increased dramatically in recent decades due to the rapid urbanization. Yet there is a very limited utilization of this waste, meaning that there is an urgent need to address this issue in order to reduce the reliance on virgin materials and improve the lived environment in China. This problem contrasts with the EU experience where many member states already exceed an 80% utilization rate of CDW due to extensive policy measures. We argue that the supervision of CDW is an important and underestimated element in addressing this waste stream, and that China and other rapidly developing states can build on the EU experience to address this issue rapidly and efficiently. This paper took a comparative policy analytical approach to summarize advanced experiences promoting CDW utilization, highlighting 14 advanced policy measures. We then conducted a case study approach applied to a Chinese metropolitan city, Wuhan, to identify the key measures to promote CDW utilization in the local area by questionnaire. On this basis, we then proposed a whole process supervision framework for the six most important policy measures, to cover the whole process of the generation, transport and recycling of this waste. This approach provides a mode for policymakers to prioritize the most important policy measures to address CDW problem in China through a timely and data-driven process.

Natural materials, especially natural colorants, have achieved global prominence and might be regarded as an environmentally beneficial alternative to hazardous synthetic dyes. The color limitation of natural dyes hinders their application in textiles. The present work aims to prepare more color shades of wool yarns via dyeing with ternary natural dye mixtures without adding mordants. In this study, a sustainable dyeing approach for wool yarn was evaluated with three natural dyes, madder red (MR), gardenia blue (GB), and gardenia yellow (GY), by following an industrial dyeing procedure in the absence of a mordant. In the beginning, a preliminary assessment of dye stabilities was carried out, and it was found that the three natural dyes were sensitive to temperature and acid (degradation tendency). Then, the dyeing behavior was systematically evaluated, including a single natural dye, a binary natural dye mixture, and a ternary natural dye mixture. The results of wool yarn dyeing with a single natural dye show that the dye exhaustion percentage (E%) of MR, GY, and GB was in the ranges of 78.7-94.1%, 13.4-44.1%, and 54.8-68.5%, respectively. The dyeing results of wool yarns dyed with binary and ternary natural dye mixtures (a color triangle framework of dyed wool yarn) were characterized by colorimetric values (L*, a*, b*, C*, h, and K/S), and are presented to enlighten various colorful shades. Finally, color uniformity and colorfastness tests confirmed the vital contribution of natural dyes toward wool yarn coloration. Particularly, colorfastness to washing confirmed the stability of natural dyes with reference to the lower amount of dyes released into the effluent, which is beneficial for the environment. Overall, this study provides a good background for enhancing the industrialization trend of natural dyes by modulating their dyeing scheme.

OBJECTIVES: Coagulation factors participates in the inflammatory cascade, known to play a crucial role in the development of acute kidney injury (AKI). Thus, it's likely that some factors may be associated with AKI. Among them, low levels of fibrinogen and antithrombin III (ATIII) activity have been proved to increase mortality in patients with sepsis. Moreover, they are also reported to be associated with higher incidence of AKI. However, the association between coagulation parameters, especially fibrinogen and ATIII, and prognosis of AKI has not been examined. METHODS: Data were acquired from Multiparameter Intelligent Monitoring in Intensive Care Database IV (MIMIC-IV) version 1.0. Cox proportional hazards regression model was used to estimate the relationship between coagulation parameters and in-hospital mortality in critically ill patients with AKI. Subgroup analysis was also conducted to assess the robustness of the association. Restricted cubic spline (RCS) curve was utilized to examine the nonlinear relationships between fibrinogen or ATIII and in-hospital mortality. Kaplan-Meier method was used to estimate cumulative incidence of mortality by fibrinogen or ATIII levels. Receiver-operating characteristic (ROC) curve was plotted and area under curve was calculated to evaluate predictive ability of fibrinogen or ATIII. RESULTS: A total of 5914 eligible patients were enrolled in fibrinogen cohort study and 115 patients were eligible for ATIII cohort study. The baseline of low fibrinogen (<150 mg/dL) or ATIII (<80%) activity was associated with significantly higher in-hospital mortality (fibrinogen HR [95% CIs] 2.01 [1.79, 2.27]; ATIII 3.73 [1.11, 12.54]). The HR [95% CIs] of low fibrinogen remained significant 1.29 (1.13, 1.48) in multivariate analysis. The RCS curve showed nearly linear relationship. Subgroup analysis also proved the robustness of the association between fibrinogen and in-hospital mortality. Kaplan-Meier survival curve and ROC demonstrated the predictive capability of fibrinogen and ATIII. CONCLUSION: Low fibrinogen is an independent predictor of in-hospital mortality in critically ill patients with AKI. Low ATIII activity is also likely to impact the risk of in-hospital death.

Background: Renal sarcoma (RS) is rarely seen in clinical practice. The purpose of this study was to develop a prognostic nomogram model, which could predict the probability of overall survival (OS) and cancer-specific survival (CSS) in adult patients with RS. Methods: Patients diagnosed with RS were recruited from the SEER database between 2004 and 2015, and randomized to two cohorts: the training cohort and the validation cohort. Uni- and multivariate Cox regression analyses in the training cohort were used to screen independent prognostic factors for OS and CSS. Prognostic nomograms for OS and CSS were created separately for adult RS patients based on independent risk factors. The area under the receiver operating characteristic (ROC) curves, calibration curves, and decision curve analysis (DCA) were used to validate the nomograms. Results: A total of 232 eligible patients were recruited, including 162 in the training cohort and 70 in the validation cohort. Sex, histological type, SEER stage, and surgery were independent prognostic factors for OS, while histological type, SEER stage, surgery, chemotherapy were independent prognostic factors for CSS. Based on the above independent prognostic factors, prognostic nomograms for OS and CSS were created respectively. In the training cohort, the AUCs of the nomograms for OS and CSS were 0.742 and 0.733, respectively. In the validation cohort, the AUCs of the nomograms for OS and CSS were 0.837 and 0.758, respectively. The calibration curves of the nomograms showed high consistencies between the predicted and actual survival rates. Finally, the DCA demonstrated that the nomograms in the wide high-risk threshold had a higher net benefit than the SEER stage. Conclusion: A prognostic nomogram for renal sarcoma was created and validated for reliability and usefulness in our study, which assisted urologists in accurately assessing the prognosis of adult RS patients.

For many centuries, traditional grass cloth has been used as an important raw material for home textiles in China, but its market can be expanded by incorporating color. Reactive Red 2 (R2), Reactive Blue 194 (B194), and Reactive Orange 5 (O5) were used in this work to explore the dyeing behavior of sustainable traditional grass fiber using industrial dyeing methods. Initially, an L16 (4^4) orthogonal design was schematically applied to carry out the dyeing process and it was determined that the total dye fixation rate (T%) of B194 dye was the best among the three dyes. Accordingly, a statistical Taguchi technique was analyzed on a larger scale to optimize the dyeing process parameters (salt concentration, fixation time, fixation temperature, and solution pH) of B194, in which solution pH was found to be the most influential factor in achieving the highest T%. This phenomenon was also verified using analysis of variance (ANOVA), where the solution pH was found to be the biggest contributor (50%) and statistically significant (p < 0.05). Finally, confirmation tests were conducted under optimized conditions and a higher T% (53.18%) was determined compared to initial conditions (48.40%). Later, Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM) were used to analyze the structural characteristics and found that grass cloth was chemically stable, yet gummy materials were still observed on their surface, which was also confirmed from digital photographs. Generally, the color coordinates and fastness properties were also satisfactory.

Liquid ammonia (LA) dyeing is a zero-effluent and sustainable dyeing technology investigated for textiles. In the present work, three bi-functional reactive dyes, Reactive Red 195 (R195), Reactive Yellow 145 (Y145), and Reactive Blue 194 (B194), were used to dye ramie fiber in liquid ammonia, and the dye exhaustion (%) and fixation (%) were compared with ramie fibers dyed with the same dyes in an aqueous dyeing method. Dyeing with a single reactive dye, a binary dye mixture, and a ternary dye mixture in liquid ammonia showed that all the dyes are highly compatible as they showed similar uptake. The total dye exhaustion percentage of dyeing with the ternary dye mixture was 22.6%. After dyeing, a cationic fixing agent (CFA)/decamethylcyclopentasiloxane (D5) micro-emulsion was applied and the dye fixation rate was 96.7% accompanied by high colorfastness to washing (Grade 4-5) and produced uniform shades. Finally, a color triangle of dyed ramie fibers was prepared to exhibit many colorful shades. This work demonstrates the viability of dyeing of textile fibers in liquid ammonia.

A two-stage sequential pretreatment including caustic mercerization (CM) and liquid ammonia (LA) treatment was applied to investigate the influence on dyeing performance and handle of knit cotton fabric, and the relationship between dye size and dyeing properties. Various techniques were applied to characterize all the treated fabrics. X-ray diffraction (XRD) and Fourier-transform infrared (FTIR) analyses of the treated fabrics confirmed that both sequential treatments decreased the crystallinity of cotton fabric more than only the CM or LA treatment. The pattern of cellulose I was transferred to a mixed configuration of cellulose II and cellulose III after the CM/LA or LA/CM treatment. Thermal performances measured by thermogravimetric analysis (TGA) and differential thermogravimetry (DTG) techniques showed that the thermal stability of the treated cotton only marginally decreased. The wicking height increased after the sequential CM/LA treatment, indicating that the hydrophilicity of the fabric increased. The dye absorption and color uniformity were better for the reactive dye with a smaller molecular weight (Reactive Red 2) compared with the one with a larger molecular weight (Reactive Red 195). The total dye fixation efficiency (T%) increased to 72.93% and 73.24% for Reactive Red 2 dyeings of CM/LA- and LA/CM-cotton fabric from 46.75% of the untreated fabric, respectively; the T% increased to 65.33% and 72.27% for Reactive Red 195 dyeings of CM/LA- and LA/CM-cotton fabric from 35.17% of the untreated fabric, respectively. The colorfastness and dye exhaustion and fixation percentages of the samples were enhanced after the treatments. Furthermore, compared to the single CM or LA treatment, the softness handle properties were further improved after the fabrics were sequentially treated by CM/LA. The developed pre-treatment of CM/LA can be used in the textile industry to promote the dyeability, handle, and mechanical properties of knit cotton fabrics.

The dyeing process commonly deteriorates the luster of pre-mercerized cotton fabric, so post-mercerization processes are regularly applied to compensate for this. Herein, the influence of combining pre-mercerization with CS (caustic solution) or LA (liquid ammonia) and post-mercerization with CS or LA on the morphological structure, dyeing performance, tensile strength, and stiffness of woven cotton fabric was investigated. The crystallinity index values greatly decreased from 73.12 to 51.25, 58.73, 38.42, and 40.90% after the combined mercerization processes of LA-LA, CS-CS, LA-CS, and CS-LA, respectively. Additionally, the CS-LA- and LA-CS-treated samples exhibited a mixture of cellulose II and cellulose III allomorphs. The combined mercerization processing of cotton fabric resulted in slightly worse thermal stability. The LA and CS pre-mercerization processes increased the dye exhaustion, although the former decreased the dye fixation rate while the latter increased it by 4% for both dyes. The color strength of the dyed cotton fabric increased after both post-mercerization processes. Moreover, the fabric stiffness and mechanical properties showed an increasing trend due to the combined mercerization efforts.

Renal cancer is one of the most extensively studied human tumors today, with clear cell renal cell carcinoma accounting for approximately 80% of all cases. Despite recent advances in research on clear cell renal cell carcinoma, advanced distant metastasis of the disease, delay in diagnosis, as well as drug resistance remain major problems. In recent years, as an important mediator of material and information exchange between cells in the tumor microenvironment, exosomes have attracted widespread attention for their role in tumor development. It has been reported that tumor-derived exosomes may act as regulators and have an important effect on the metastasis, drug resistance formation, and providing targets for early diagnosis of clear cell renal cell carcinoma. Therefore, the extensive study of tumour-derived exosomes will provide a meaningful reference for the development of the diagnostic and therapeutic field of clear cell renal cell carcinoma. This article reviews the biological role and research progress of tumor-derived exosomes in different aspects of premetastatic niche formation, tumor angiogenesis, and epithelial-mesenchymal transition during the progression of clear cell renal cell carcinoma. In addition, the role of tumor-derived exosomes in the development of drug resistance in clear cell renal cell carcinoma is also addressed in this review. Furthermore, recent studies have found that cargoes of exosomes in serum and urine, for example, a series of miRNAs, have the potential to be biological markers of clear cell renal cell carcinoma and provide meaningful targets for early diagnosis and monitoring of tumors, which is also covered in this article.

Combretastatin-4 (CA-4) as a tubulin polymerization inhibitor draws extensive attentions. However, due to its weak stability of cis-olefin and poor metabolic stability, structure modifications on cis-configuration are being performed. In this work, we constructed a series of novel CA-4 analogues with linkers on olefin containing diphenylethanone, cis-locked dihydrofuran, α-substituted diphenylethanone, cyclobutane and cyclohexane on its cis-olefin. Cytotoxic activity of all analogues was measured by an SRB assay. Among them, compound 6b, a by-product in the preparation of diphenylethanone analogues, was found to be the most potent cytotoxic agents against HepG2 cells with IC50 values of less than 0.5 µM. The two isomers of 6b induced cellular apoptosis tested by Annexin V-FITC and propidium iodide (PI) double staining, arrested cells in the G2/M phase by PI staining analysis, and disrupted microtubule network by immunohistochemistry study in HepG2 cells. Moreover, 6b-(E) displayed a dose-dependent inhibition effect for tubulin assembly in in vitro tubulin polymerization assay. In addition, molecular docking studies showed that two isomers of 6b could bind efficiently at colchicine binding site of tubulin similar to CA-4.
