ABSTRACT

In the Amazon and Orinoco basins, mercury has been released from artisanal and industrial gold mining since the Colonial time, as well as a result of deforestation and burning of primary forest, that release natural deposits of methyl mercury, affecting the local aquatic vertebrate fauna. This study reports the presence of mercury in river dolphins' genera Inia and Sotalia. Mercury concentrations were analysed in muscle tissue samples collected from 46 individuals at the Arauca and Orinoco Rivers (Colombia), the Amazon River (Colombia), a tributary of the Itenez River (Bolivia) and from the Tapajos River (Brazil). Ranges of total mercury (Hg) concentration in muscle tissue of the four different taxa sampled were: I. geoffrensis humboldtiana 0.003-3.99 mg kg-1 ww (n = 21, Me = 0.4), I. g. geoffrensis 0.1-2.6 mg kg-1 ww (n = 15, Me = 0.55), I. boliviensis 0.03-0.4 mg kg-1 ww (n = 8, Me = 0.1) and S. fluviatilis 0.1-0.87 mg kg-1 ww (n = 2, Me = 0.5). The highest Hg concentration in our study was obtained at the Orinoco basin, recorded from a juvenile male of I. g. humboldtiana (3.99 mg kg-1 ww). At the Amazon basin, higher concentrations of mercury were recorded in the Tapajos River (Brazil) from an adult male of I. g. geoffrensis (2.6 mg kg-1 ww) and the Amazon River from an adult female of S. fluviatilis (0.87 mg kg-1 ww). Our data support the presence of total Hg in river dolphins distributed across the evaluated basins, evidencing the role of these cetaceans as sentinel species and bioindicators of the presence of this heavy metal in natural aquatic environments.

The giant otter (Pteronura brasiliensis) is an aquatic mammal of the Mustelidae family, endemic to South America. Its original distribution corresponds to the region from the Guyanas to Central-North Argentina, but it is extinct or on the verge of extinction in most of its historical range. Currently, the species is considered endangered by the World Conservation Union (IUCN). Based on its geographic distribution in the South American continent and on some morphological characters, two subspecies were suggested: P. brasiliensis brasiliensis, occurring in the Amazon and Orinoco River Basins, and P. brasiliensis paranensis, in the Paraná and Paraguai River Basins. However, there is no consensus on assuming this subspecies division and no detailed studies have been carried out to elucidate this question. This study aims to evaluate the genetic diversity and population structure of Pteronura brasiliensis along its range in Brazil to check the possibility of the existence of two distinct subspecies using also a reciprocal monophyly criterion. We analyzed the control region, and the Cytochrome b and Cytochrome c Oxidase subunit I genes of the mitochondrial DNA in several giant otter populations from the Amazon and Paraguai River Basins. Analyses have indicated some degree of geographic correlation and a high level of inter-population divergence, although the subspecies division is not highly supported. As we observed strong population structure, we cannot rule out the existence of further divisions shaping the species distribution. The results suggest that a more complex population structure occurs in P. brasiliensis, and the conservation practice should concentrate on preserving all remaining local populations.


A ariranha (Pteronura brasiliensis) é um mamífero aquático da família Mustelidae, endêmico da América do Sul. Sua distribuição original se estendia desde as Guianas até o centro-norte da Argentina, mas está extinta ou à beira da extinção na maior parte de sua distribuição histórica. Atualmente a espécie é considerada como ameaçada de extinção pela World Conservation Union (IUCN). Em função de sua distribuição no continente sul-americano e de algumas características morfológicas, duas subespécies foram sugeridas: P. brasiliensis brasiliensis, com ocorrência nas bacias do Amazonas e Orinoco, e P. brasiliensis paranensis, ocorrendo nas bacias dos Rios Paraná e Paraguai. Inexiste, contudo, um consenso sobre a validade da divisão em subespécies e nenhum estudo detalhado foi realizado para elucidar esta questão. Este trabalho tem o objetivo de avaliar a diversidade genética e a estrutura populacional de P. brasiliensis ao longo de sua distribuição no Brasil para verificar a existência de duas subespécies baseando-se também em um critério de monofilia recíproca. A região controle e os genes do Citocromo b e da Subunidade I da Citocromo c Oxidase do DNA mitocondrial foram analisados em diversas populações de ariranha que ocorrem nas bacias dos rios Amazonas e Paraguai. As análises indicaram um grau moderado de correlação geográfica e um alto nível de divergência inter-populacional, embora a divisão em subespécies não seja bem sustentada. Como uma forte estruturação populacional foi observada, não é possível descartar a existência de outras subdivisões nesta espécie. Os resultados indicam a presença de uma estrutura populacional mais complexa em P. brasiliensis, o que implica que medidas de conservação deveriam concentrar seus esforços preservando todas as populações locais remanescentes.

ABSTRACT

The Amazon river dolphin, genus Inia, is endemic to the major river basins of northern South America. No previous studies have focused on the genetic structure of this genus. In this work, 96 DNA samples from specimens of this genus were collected in the Orinoco basin (four rivers), the Putumayo River, a tributary of the Colombian Amazon and the Mamoré, and the Tijamuchí and Ipurupuru rivers in theBolivian Amazon. These samples were used to amplify a fragment of 400 bp of the mitochondrial DNA (mtDNA) control region. In addition, 38 of these samples were also used to sequence 600 bp of the mitochondrial cytochrome b gene. The analysis of the population structure subdivision with an analysis of molecular variance (AMOVA) revealed important aspects about the genetic structure of Inia groups fromthese three geographically separate regions. By comparing the control region DNA and cytochrome b sequences, distinct types of nonshared haplotypes were observed. The net genetic divergence of control region sequences was 6.53% between the Orinoco and Bolivian rivers, 5.32% between the Putumayo and Bolivian rivers, and 2.50% between the Orinoco and Putumayo rivers. For the cytochrome b gene, these values were 2.48%, 2.98%, and 0.06%, respectively. The nucleotide sequences were analyzed phylogenetically using several genetic distance matrices and applying neighbor-joining, maximum likelihood, and maximum parsimony procedures. The results support the proposal to subdivide the Inia genus into at least two evolutionarily significant units: one confined to the Bolivian river basin and the other widely distributed across the Amazon and Orinoco basins.

ABSTRACT

To resolve the population genetic structure and phylogeography of the West Indian manatee (Trichechus manatus), mitochondrial (mt) DNA control region sequences were compared among eight locations across the western Atlantic region. Fifteen haplotypes were identified among 86 individuals from Florida, Puerto Rico, the Dominican Republic, Mexico, Columbia, Venezuela, Guyana and Brazil. Despite the manatee's ability to move thousands of kilometers along continental margins, strong population separations between most locations were demonstrated with significant haplotype frequency shifts. These findings are consistent with tagging studies which indicate that stretches of open water and unsuitable coastal habitats constitute substantial barriers to gene flow and colonization. Low levels of genetic diversity within Florida and Brazilian samples might be explained by recent colonization into high latitudes or bottleneck effects. Three distinctive mtDNA lineages were observed in an intraspecific phylogeny of T. manatus, corresponding approximately to: (i) Florida and the West Indies; (ii) the Gulf of Mexico to the Caribbean rivers of South America; and (iii) the northeast Atlantic coast of South America. These lineages, which are not concordant with previous subspecies designations, are separated by sequence divergence estimates of d = 0.04-0.07, approximately the same level of divergence observed between T. manatus and the Amazonian manatee (T. inunguis, n = 16). Three individuals from Guyana, identified as T. manatus, had mtDNA haplotypes which are affiliated with the endemic Amazon form T. inunguis. The three primary T. manatus lineages and the T. inunguis lineage may represent relatively deep phylogeographic partitions which have been bridged recently due to changes in habitat availability (after the Wisconsin glacial period, 10 000 B P), natural colonization, and human-mediated transplantation.

Os autores mostram quais os topicos que chamaram a atencao da populacao estudada em uma palestra contra o fumo, e sugerem que a estes seja dada especial atencao em proximas iniciativas neste sentido. Sugerem ainda, que futuras campanhas sejam baseadas em um programa continuado de esclarecimento quanto aos prejuizos do fumo a saude da populacao e nao apenas em uma iniciativa isolada. Mostram, ainda que esta populacao e suscetivel a isto, visto que 69% dela gostaria de parar de fumar e 74% ja fizeram tentativas com este fim
