ABSTRACT

Bladder cancer (BCa) is the most common malignant tumor of the urinary system, and its incidence is increasing year by year. At present, for all patients with resectable non-metastatic muscle-invasive BCa, radical cystectomy + bilateral pelvic lymph node dissection is strongly recommended, but they still face the risk of recurrence, metastasis and death. In recent years, the proportion of patients with advanced and metastatic BCa is increasing among patients with newly diagnosed BCa. Although current treatment models are diverse, they often struggle to achieve significant efficacy due to their low effectiveness and adverse effects, resulting in low survival rates for patients with advanced and metastatic BCa. Therefore, the treatment of BCa still faces great challenges, and there is an urgent need to discover an effective new antitumor drug. With the improvement of medical standards, traditional Chinese medicine has shown great advantages in the treatment of BCa. Traditional Chinese medicine is mild and easy to accept, and can inhibit tumor progression through a multi-pathway, multi-way and multi-target manner, so as to exert its anticancer effect. Taraxaci Herba is a medicinal and food homologous plant, which has many biological activities, such as antibacterial, anti-inflammatory, anti-oxidation, anti-tumor, protecting liver and gallbladder, reducing blood sugar and enhancing immunity, and it has shown a clear anticancer effect in breast cancer, liver cancer, gastric cancer, tongue cancer and lung cancer. By reviewing previous studies worldwide, this article summarizes the mechanism of Taraxaci Herba extract in inducing autophagy and apoptosis, inhibiting cell migration and invasion, regulating cell cycle and proliferation, regulating cell metabolism, inhibiting tumor angiogenesis, combining the effects of chemotherapeutic drugs, and regulating the transduction of related signal pathways. On this basis, this study systematically elaborates on the potential mechanism of Taraxaci Herba against BCa, in order to provide a theoretical basis for the research and treatment of BCa.

ABSTRACT

Metastatic pheochromocytoma and paraganglioma(MPPGL)is a rare neuroendocrine tumour in which genetic factors play an important role.In recent years,with the continuous progress of genetic testing technol-ogy,more and more susceptibility genes have been proved to be associated with MPPGL,making early identifica-tion of MPPGL possible.Recent studies have shown that genes associated with the development of MPPGL include SDHA,SDHB,SDHC,SDHD,SDHAF2,FH,MDH2,VHL,IDH1,PDH1/2,SLC25A11,GOT2,DLST,CSDE1,MAML3,H3F3A,MERTK,PCDHGC3,and KIF1B,with SDHA,SDHB,SDHC,SDHD,and SDHAF2 being the common pathogenic genes.Potential mutations affect the clinical manifestations of MPPGL,such as malignant potential and genetic prediction,which can help to better understand the clinical course and treat accordingly.Genetic testing for pheochromocytomas and paragangliomas allows for early detection of genetic syndromes and facilitates close follow-up of high-risk patients.This article provides a review of the progress of research on susceptibility genes identified in MPPGL in recent years,with a view to providing a certain theoretical basis for further related research.

ABSTRACT

Solitary kidney, renal duplication and malrotation are rare congenital renal malformations in urology department, and probably contributed to some complications such as obstruction, hydronephrosis, infection, stones. In this case report, we firstly presented a male patient with rarely multiple renal malformations, including solitary kidney, renal duplication, misaligned malrotation of upper and lower moieties, and accompanied by complete staghorn stones and hydronephrosis, who was treated with open pyelolithotomy under general anesthesia. After the operation, obstruction, hydronephrosis, and infection were relieved.

ABSTRACT

Pasteurella multocida can infect a wide range of host, including humans and animals of economic importance. Genomics studies on the pathogen have produced a large amount of omics data, which are deposited in GenBank but lacks a dedicated and comprehensive resource for further analysis and integration so that need to be brought together centrally in a coherent and systematic manner. Here we have collected the genomic data for 176 P. multocida strains that are categorized into 11 host groups and 9 serotype groups, and developed the open-access P. multocida Database (PamulDB) to make this resource readily available. The PamulDB implements and integrates Chado for genome data management, Drupal for web content management, and bioinformatics tools like NCBI BLAST, HMMER, PSORTb and OrthoMCL for data analysis. All the P. multocida genomes have been further annotated for search and analysis of homologous sequence, phylogeny, gene ontology, transposon, protein subcellular localization and secreted protein. Transcriptomic data of P. multocida are also selectively adopted for gene expression analysis. The PamulDB has been developing and improving to better aid researchers with identifying and classifying of pathogens, dissecting mechanisms of the pathogen infection and host response.

ABSTRACT

Objective: To explore the early diagnostic value of dynamic changes for high-sensitive troponin I (hs-cTnI) in patients with acute coronary syndrome (ACS). Methods: A total of 186 ACS patients treated in our hospital from 2014-02 to 2015-04 were studied. The patients were divided into 2 groups:AMI group, n=169 and UA group, n=17, in addition, there was a Control group, n=13 healthy subjects. Blood levels of hs-cTnI were measured at admission and 2h, 4h after admission. Dynamic changes of hs-cTnI was calculated by slop coefifcient of hs-cTnI (Δhs-cTnI) at 3 time points and relative changes of hs-cTnI values. ROC curve was made by SPSS 16.0 statistical software and the best diagnostic cutoff point was determined by Youden index. Results: In clinical practice, hs-cTnI=1 ng/ml was critical condition, hs-cTnI=0.15 ng/ml was recommended for AMI diagnosis. When hs-cTnI Conclusion: AMI could not be effectively diagnosed by hs-cTnI alone at admission, using combined relative changes of hs-cTnI andΔhs-cTnI was superior to 0 h hs-cTnI for AMI diagnosis.

ABSTRACT

<p><b>BACKGROUND</b>Several studies have evaluated the association between polymorphisms of thymidylate synthase (TS) and cancer risk in diverse populations but with conflicting results. By pooling the relatively small samples in each study, it is possible to evaluate the association using a meta-analysis.</p><p><b>METHODS</b>A comprehensive search was conducted to identify all case-control studies on TS on a 28-bp tandem repeats in 5'untranslated region (5'UTR) and a 6-bp insertion (ins) and deletion (del) mutation in 3'UTR of the gene and cancer risk. Meta-analysis was conducted using a fixed and random effect model.</p><p><b>RESULTS</b>Our meta-analysis on a total of 13 307 cancer cases and 18 226 control subjects from 37 published case-control studies showed no significant association between the risk of cancer and the 5'UTR 28-bp tandem repeats polymorphism (3R/3R vs. 2R/2R: OR = 1.06, 95%CI, 0.93 - 1.20) or the 3'UTR 6-bp ins/del polymorphism (del6/del6 vs. ins6/ins6: OR = 0.93, 95%CI, 0.81 - 1.08) with significant between-study heterogeneity. In the cancer type- and ethnic subgroup-stratification analyses, we did not find any association between TS polymorphisms and cancer risk either.</p><p><b>CONCLUSION</b>TS 5'UTR 28-bp tandem repeats and 3'UTR 6-bp ins/del polymorphisms may not be associated with cancer risk.</p>

ABSTRACT

<p><b>AIM</b>To investigate the protective role of HO/CO systems in IL-1beta induced islest apoptosis and to explore the mechanisms of the protective effect of fructose-1, 6-disphosphate (FDP).</p><p><b>METHODS</b>The pancreases of the rats were removed to collect islets cells. The cells were incubated with IL-1beta with/or FDP. Cell activity, insulin secretion, HO-1 activity, CO content and apoptotic percentage were detected.</p><p><b>RESULTS</b>HO-1 activity and CO content of the normal control group were low. IL-1beta induced a significant decrease of cell activity and insulin release, flow cytometry analysis showed that apoptotic percentage of islet cells remarkably increased following the addition of IL-1beta, FDP obviously improved the islets cellular activity damaged by IL-1beta, and basic amount of insulin secretion and stimulated by high glucose were improved (P < 0.01). Content of CO and activity of HO-1 were higher in the IL-1beta group than the normal control group (P < 0.05), and there were significant differences between the FDP groups and IL-1beta group. FDP decreased cell apoptotic percentage. Activities of HO-1 and content of CO were higher than that in the IL-1beta group (P < 0.01).</p><p><b>CONCLUSION</b>FDP can attenuate the IL-1beta induced apoptosis of cultured beta cells, the mechanism of which may be improved HO-1 activity and CO content.</p>

ABSTRACT

Objective: To observe the influence of silencing MAT1 gene by short interfering RNA (siRNA) on the proliferation and invasion of human pancreatic cancer cells and discuss the feasibility of using MAT1 gene as therapeutic target for treatment of pancreatic cancer. Methods: BxPC-3 cells were transfected with sequence-specific siRNA targeting MAT1 gene with liposome mediation. The mRNA and protein expression of MAT1 were confirmed by RT-PCR and Western blot, respectively. The cell proliferation was measured by Trypan blue exclusion staining. The invasion ability was determined by Boyden chamber model in vitro. Results: The mRNA and protein expression of MAT1 were significantly down-regulated by siRNA in BxPC-3 cells compared with the control groups. The expression of MAT1 mRNA was reduced by 55.2% and 64.3% at 24 h and 48 h, respectively (P <0.01). The cell proliferation and invasion ability of BxPC-3 cells were significantly inhibited in vitro (P < 0.01). Conclusion: The results suggest that siRNA targeting MAT1 gene inhibits the cell proliferation and invasion of BxPC-3 cells in vitro. MAT1 may be a potential target for gene therapy of human pancreatic cancer.
