ABSTRACT

This study aimed to assess the cortisol, body and reproductive development of prepubertal Holstein and Holstein-Gir ¾ heifers at 27 months of age maintained in an integrated livestock-forest (ILF) system for 60 summer days compared to the monoculture system in full sun (FS). The ILF system promoted changes (P=0.02) in the cortisol levels of Holstein-Gir ¾ heifers and did not affect weight gain in any of the breed groups studied. Animals in ILF system presented a lower (P=0.006) vulvar development for the rima height parameter and similar for the vulva width parameter. The ovarian follicular population of Holstein-Gir ¾ heifers in the ILF system was lower (P=0.004); however, for the Holstein heifers, no statistical difference was found, and numbers were higher (P=0.08) in the ILF system. None of the other ovarian parameters studied had any changes, and we also found important racial differences. Weight gain (P=0.003), vulvar development (P<0.001), and mean follicular size (P=0.008) were higher in the Holstein-Gir ¾ animals. Based on such results, the effect of the ILF system at 27 months of age on stress and reproductive parameters in the Holstein breed is considered positive, although negative effects have been detected on reproductive parameters in the Holstein-Gir ¾ breed.

ABSTRACT

This study evaluated the effect of crude protein (CP) reduction in four diets (156, 139, 132, and 127 g Kg-1 DM) maintaining constant metabolizable protein (188 g/day) on the follicular fluid and cumulus-oocyte complexes of mid-lactating Girolando cows. Twenty-two Girolando cows with average of 21.55 ±3.19 L daily milk yield, 105.30 ±22.62 days in lactation and 3.22 ±0.03 body condition score were selected. To reduce CP in diets and maintain constant metabolizable protein, urea and soybean meal were gradually replaced by lignosulfonate-treated soybean meal (SoyPass®, Cargill), resulting in an increase in rumen-undegradable protein and a reduction in rumen degradable protein. A linear and quadratic reduction was observed in the plasma and follicular fluid urea nitrogen concentration following CP reduction, with the most intense reduction occurring in the 127 g Kg-1 DM group (p<0.001). As CP reduced, there was a tendency for a linear increase in the follicular growth rate (P=0.0696), on the number and proportion of viable oocytes (P<0.09), and also a linear increase for the number (P=0.0397) and proportion (P<0.09) of grade I viable oocytes. Plus, there was a linear effect for the number of cumulus oophorus cells. Cows fed with the lowest amount of CP had cumulus-oocyte complexes with higher numbers of cumulus oophorus cells (P=0.0238). Also, the reduction of diet crude protein was followed by a decrease in the probability of oocytes' DNA degradation. In conclusion, the reduction of CP in the diet of mid-lactating Girolando cows, reduces urea nitrogen concentration in both blood plasma and follicular fluid, and, as a consequence, increases the viability of oocytes and the number of cumulus oophorus cells while reducing oocytes' DNA degradation of follicular included cumulus-oocyte complex. The reduction on dietary CP may improve in vivo oocytes' embryo development impacting fertility of lactating dairy cows.

ABSTRACT

Background: Lacaune is an important sheep breed and shows reproductive seasonality which affects dairy product supply. The most common hormonal protocols are based on progesterone-releasing devices in order to reach reproductive activity. Due to international consumer demand for natural, green, and clean methods, such issues encourage the minimal or absent use of synthetic hormones in livestock. In this sense, the present study tested the efficiency of induction and estrus synchronization protocols in lacaune ewes, considering: application interval of synthetic prostaglandin at the breeding season; length of synthetic progesterone exposure; and eCG dose at the outbreeding season. Materials, Methods & Results: In study 1: 52 ewes were submitted to estrus synchronization protocol using two doses of d-cloprostenol 7 or 9 days apart. In study 2: 47 ewes were submitted to estrus induction protocol using an intravaginal device containing 60 mg of medroxyprogesterone acetate maintained for 6, 9, or 12 days. In study 3: 46 ewes were submitted to synchronized estrus induction protocol using intravaginal progesterone device for six days. On the fifth day, d-cloprostenol and 300 IU or 400 IU of eCG were administered. For all studies, signs of estrus beginning/ending were recorded and natural mating was performed. After 30 days, transrectal ultrasound was performed for pregnancy diagnosis. In study 1, groups presented similar rate of estrus behavior (78.8%) and pregnancy (average 75.6%). Meanwhile, compared to G9days (37.8 ± 7.2 h), interval between the second dose of prostaglandin and beginning of estrus was smaller in G7days (31.5 ± 7.8 h). No difference was found among variables studied in study 2 (P > 0.05), nevertheless the SD for the estrus duration was shorter (P < 0.05) in G6days and G12 days when compared with the G9days . A eficiência reprodutiva de doadoras de embriões Bos taurus é afetada pelas estações devido ao estresse calórico em países de clima tropical e temperado. O objetivo do estudo foi avaliar o efeito das condições climáticas de verão e inverno na produção de oócitos e embriões in vitro na raça Gir Leiteiro (Bos indicus) em fazendas da região Sudeste do Brasil. Os parâmetros avaliados foram: total de oócitos recuperados (OT), oócitos viáveis (OV) e percentual de oócitos viáveis sobre o total (POV); embriões produzidos (EP) e percentual de embriões produzidos (PEP) sobre o total de estruturas recuperadas pela aspiração folicular guiada por ultrassonografia (OPU). Os oócitos foram obtidos de 153 doadoras pluríparas de diferentes idades e ordens de parição de 10 rebanhos. As médias obtidas em 380 aspirações foliculares (OPU) nos meses de verão foram: OT=14,8; OV=12,6; EP= 3,4; PEP=22,3%, e no inverno de OT=13,4; OV=10,8; EP=3,5; PEP=26,1%, apresentando coeficiente de variação de até 84%, porém sem diferença estatísticas (p<0,05) entre estes períodos, utilizando-se o teste de Kruscal Wallis s (p<0,05). Apenas a percentagem de oócitos viáveis foi maior (p<0,05) no período de verão (POV=84,4 vs. 80,0%), contudo, o total de embriões PIVE foi semelhante nos dois períodos. Conclui-se que a produção de embriões da raça Gir Leiteiro, criadas na região Sudeste do Brasil, não foi afetada pela sazonalidade climática.(AU)


The reproductive performance of B.taurus embryo donors is affected in many tropical and temperate countries. The objective of this study was to evaluate the effects of the climatic conditions of summer and winter climatic conditions on the production of oocytes and in vitro embryos in dairy cows of the breed Gir Leiteiro (Bos indicus) at farms in Southeast of the Brazil. The variable answers evaluated were the total oocytes recovered (TO) and percentage of viable oocytes over the total (PVO); embryo production (EP) and percentage of embryos in viable oocytes (PE) over the total of structures recovered by follicular aspiration guided by ultrasonography (OPU). The oocytes analyzed were obtained from 10 herds and 153 pubertal donors of different ages and orders of parturition. The average from 380 ovum pick up (OPU) during summer were: TO=14.8; PVO=12.6; EP=3.4; PE=22.3%, and TO=13.4; PVO=10.8; EP=3.5; PE=26.1%, with variation coefficient of up to 84%, and no differences evaluated through the Kruscal Wallis (p<0.05). Only the percentage of viable oocytes was higher (p<0.05) in the summer period (PVO=84.4 vs. 80.0%). However, the embryo production (IVEP) was similar in both seasons (summer and winter). Therefore, we could conclude that the embryo production of Dairy Gir cows in Southeast of the Brazil was not affected by weather seasonality.(AU)

ABSTRACT

This study investigated the influence of heat shock during in vitro maturation on embryo development following in vitro fertilization (IVF) or parthenogenesis (Part). Immature bovine cumulus-oocyte complexes were exposed to heat shock (41.0°C) during the first 12 hr of in vitro maturation (IVM), followed by 12 hr at 38.5°C. Control group consisted of in vitro maturation for 24 hr at 38.5°C. Oocytes were in vitro-fertilized or activated with ionomycin and cultured in vitro for 192 hr post-in vitro insemination or parthenogenetic activation (hpia). There was an interaction (p < .01) between temperature of IVM and method of oocyte activation (IVF or Part) for cleavage at 48 hpia. Heat shock had a negative impact (p < .01) on cleavage of IVF embryos, whereas no (p > .05) effect was found in the Part embryos. Embryo development towards blastocyst stage at 168 and 192 hpia decreased in both IVF and Part embryos derived from heat-shocked oocytes. Heat shock increased (p < .05) the apoptotic index in Part blastocysts, but no effect (p > .05) was found in IVF counterparts. Heat shock also down-regulated the expression of AQP3 (p < .01) and up-regulated the expression of HSP70.1 (p < .01) in Part blastocysts, whereas it down-regulated the expression of ATP1A1 (p < .05) in IVF blastocysts. In conclusion, the effects of heat shock during IVM on early embryo cleavage and blastocyst apoptosis are influenced by the method of oocyte activation and expression of some genes can be disturbed in embryos derived from heat-shocked oocytes.

ABSTRACT

O objetivo do trabalho foi avaliar o perfil cromatográfico por gel filtração de proteínas do plasma seminal de 16 touros jovens da raça Nelore e suas associações com parâmetros andrológicos, congelação do sêmen e reação acrossômica induzida. As concentrações de proteínas totais no plasma seminal variaram de 3,18 a 52,7 mg/mL, com média de 26,8_+20,5 mg/mL. O perfil por gel filtração das proteínas do plasma seminal apresentou oito diferentes frações. Os picos com maior quantidade de proteínas foram os picos com peso molecular variando entre 640-240 kDa, 187-70 kDa e 12,4-4,7 kDa, além de estarem presentes em mais de 80% dos touros avaliados. O pico com variação entre 54,7-20,4 kDa também apresentou boa proporção de proteínas e frequência de ocorrência entre os touros (81%), sendo segundo a literatura a faixa de maior interesse para exploração quanto a proteínas associadas a fertilidade e/ou sucesso na criopreservação do sêmen. A concentração espermática esteve mais associada ao pico de maior peso molecular (entre 187-70 kDa) e a motilidade e padrão de recuperação espermática encontraram-se mais próximos aos grupos de proteínas com peso molecular menor do que 10 kDa. Já o resultado de reação acrossômica induzida (RAI) não demostrou associação com nenhum grupo proteico. Conclui-se separação dos grupos de proteínas do plasma seminal em função do seu peso molecular através de gel de superose 12 foi uma importante ferramenta para análise inicial do perfil proteico de touros Nelore, sendo recomendado seu uso como procedimento inicial de quaisquer pesquisas científicas que visem explorar o perfil proteico do plasma seminal de touros.(AU)

Total seminal plasma protein concentrations ranged from 3.18 to 52.7 mg/mL, average of 26.8±20.5 mg/mL. The seminal plasma protein profile by gel filtration showed eight different fractions. The highest protein peaks were those with molecular weight ranging from 640-240 kDa, 187-70 kDa to 12.4-4.7 kDa, and were present in more than 80% of the bulls. The peak with a variation between 54.7-20.4 kDa also presented a good protein proportion and frequency of occurrence among the bulls (81%). According to the literature, this is the peak with most interest in exploration of proteins associated with fertility and semen cryopreservation success. Sperm concentration was more associated with the highest molecular weight peak (between 187-70 kDa) and sperm motility and freezability were closer to protein groups with molecular weight less than 10 kDa. The result of induced acrosome reaction (RAI) showed no association with any protein groups. The separation of seminal plasma protein groups according to their molecular weight by superose gel 12 was an important tool for the initial analysis of the protein profile of Nellore bulls, and its use as an initial procedure for any scientific research aiming at explore the protein profile of seminal plasma of bulls.(AU)

Holstein-Gyr crossbred cattle are strategic for dairy systems in tropical countries, since they combine milk yield genetics with adaptability to tropical climate. However, Holstein (Bos taurus) and Gyr (Bos indicus) breeds present remarkable differences regarding reproductive physiology. Brazil stands out as the world’s largest user of embryo in vitro production (IVP) in bovine, and the use of this technique is increasing in dairy systems. As Holstein-Gyr crossbreds are important oocyte donors for IVP, the present work aimed at investigating whether increased Gyr or Holstein breed composition influences donor’s performance. Sixteen Holstein-Gyr crossbred females presenting increased (HG, 71.4 to 87.5% Holstein; n = 9) or decreased (GH, 40.2 to 46.6% Holstein; n = 7) Holstein composition were submitted to three ovum pick up (OPU) sessions. We observed similar (P = 0.2946) antral follicle count between HG and GH donors (24.8 ± 3.2 vs 29.4 ± 2.8 respectively; mean ± SEM). Groups also display similar morphological oocyte grading (Grade I: 0.1 ± 0.1 vs 0.1 ± 0.1 – P = 0.9680; Grade II: 0.9 ± 0.5 vs 1.9 ± 0.5 – P = 0.1942; Grade III, 4.0 ± 1.2 vs 7.2 ± 1.4 – P = 0.1047, HG vs GH respectively; mean ± SEM). Additionally, the proportion of viable oocyte was similar between HG and GH groups (27.8% vs 31.9%, respectively, P = 0.3500) and oocyte lipid area fraction (6.8% vs 9.5%, respectively; P = 0.1539). Our results indicate that the individual variation has more influence than breed composition of crossbred oocyte donors.

Sixteen Holstein-Gyr crossbred females presenting increased (HG, 71.4 to 87.5% Holstein; n = 9) or decreased (GH, 40.2 to 46.6% Holstein; n = 7) Holstein composition were submitted to three ovum pick up (OPU) sessions. Holstein-Gyr crossbred cattle are strategic for dairy systems in tropical countries, since they combine milk yield genetics with adaptability to tropical climate. However, Holstein (Bos taurus) and Gyr (Bos indicus) breeds present remarkable differences regarding reproductive physiology. Brazil stands out as the world's largest user of embryo in vitro production (IVP) in bovine, and the use of this technique is increasing in dairy systems. As Holstein-Gyr crossbreds are important oocyte donors for IVP, the present work aimed at investigating whether increased Gyr or Holstein breed composition influences donor's performance. ABSTRACT

The aim of this study was to evaluate the effect of culture media and stage of development in the osmotic ability of in vitro-fertilized bovine embryos and the expression of aquaporin 3 (Aqp3) and Na/K ATPase isoform 1 (ATPAse1) genes in embryos (i) with different ability to undergo rehydration and (ii) following vitrification. In experiment 1, in vitro fertilized presumptive zygotes were co-cultured in SOFaac or modified CR2aa medium and embryos at blastocyst and expanded blastocyst stages at day 7 post-insemination were exposed to NaCl hypertonic medium (900 mOsm) for 5 min following 120 min of culture in isotonic medium in order to evaluate dehydration and rehydration, respectively. No difference (P>0.05) on blastocyst rate was found between CR2aa and SOFaac medium but embryos co-cultured in SOFaac medium underwent greater (P<0.05) dehydration. Embryos at expanded blastocyst stage underwent greater dehydration but slower rehydration than embryos at blastocysts stage (P<0.05). In the experiment 2, the amount of Aqp3 and ATPase1 transcripts were quantified in blastocysts with high or low rehydration after exposure to hypertonic medium. No difference (P>0.05) on relative amount of transcripts was found in either genes. In the experiment 3, expanded blastocysts produced in a co-culture system were vitrified, warmed and then cultured for 72 h for analysis of embryo survival and amount of Aqp3 and ATPase1 transcripts. Lower (P<0.05) embryo survival rate was found for vitrified-warmed embryos (57.9%) than for their fresh counterparts (84.6%). There was no difference on expression of ATPase1 gene but lower (P<0.01) amount of Aqp3 transcripts was found in the vitrified-warmed embryos. In conclusion, embryo ability to undergo shrinkage and swelling is influenced by medium used in a co-culture system and by embryo stage. Rehydrating ability of embryos after exposure to NaCl hypertonic medium is not associated with variations on expression of Aqp3 and ATPase1 genes, but the vitrification can alter gene expression of in vitro-fertilized bovine embryos produced in a co-culture system.

A importância do (F1) para o Brasil é notória, associando rusticidade e produtividade, porém é grande a dificuldade do produtor em manter seu rebanho nesse grau de sangue. No laboratório da Embrapa Gado de Leite em Juiz de Fora, MG foram produzidos in vitro 266 embriões (F1), a partir de complexos cumulus-oócitos (CCO's) de vacas Gir puncionadas e fecundadas com sêmen de touro Holandês. Destes, 53 embriões foram transferidos a fresco para receptoras de rebanhos comerciais de uma Cooperativa da Zona da Mata, MG, enquanto 72 embriões foram inovulados em rebanho controlado da Embrapa Campo Experimental de Coronel Pacheco. A produção de embriões F1 por fecundação in vitro apresentou taxa de produção de blastocisto de 15,58%. A taxa de gestação obtida foi de 26,4% no rebanho comercial e 40,2% no rebanho da Embrapa. Considerando-se o nascimento de 50% de machos e fêmeas, conclui-se que em rebanhos de 100 vacas seriam obtidas 13 fêmeas (13% de reposição) nos rebanhos comerciais e 20 fêmeas (20% de reposição) no rebanho da Embrapa. Foram muitas as dificuldades de operacionalização para o uso de dessa biotecnologia que requer um mínimo de escrituração zootécnica, controle sanitário, observação de cios e manejo reprodutivo.

The pregnance rate obtained was 26.4% in the cows from the commercial herds and 40.2% in the herds of Embrapa Gado de Leite. Considering the birth of 50% female cows, we can estimate a replacement annual rate of 13% (26.2% for gestation) for commercial herds and 20% (40% for pregnancy) for the controlled herd. There were many difficulties encountered in the operation, such as (i) the low level of adoption of technology; (ii) little investment in feeding, handling and sanity conditions of the herds; (iii) conditions capable to compromising the utilization of embryos, which requires a minimum of structure in terms of zoo-technical documentation, observation of rutting and reproduction management.

Um total de 38 touros adultos, aprovados andrologicamente, foi submetido ao teste de libido em curral, sendo avaliados oseventos: reflexo de Flehmen (1), perseguição ativa, com posicionamento (2), impulso de monta (3), tentativa de monta (4),monta abortada (5), monta abortada com perseguição (6) e monta completa (7). Após esta avaliação, os touros foram definidosem três categorias: alta libido – eventos 6 e 7; média libido – eventos 3 a 5 e baixa libido – eventos 1 e 2. Registrou-se altapercentagem (68,42 por cento) de animais com alta ou média libido. Durante os testes de libido, as freqüências médias dos eventosde 1 a 7 foram, respectivamente, 3,02 ± 1,6; 0,44 ± 079; 0,92 ± 1,17; 0,68 ± 0,98; 0,10 ± 0,38; 0,10 ± 0,31 e 0,23 ± 0,48. Obse rvou-se que, após dois minutos e 30 segundos de teste, os touros de baixa libido efetuaram o dobro do evento 1 (reflexo deFlehmen), quando comparados aos touros de alta libido (p>0,05). Observaram-se correlações negativas de média magnitudeentre o evento 1 e os eventos 3, 4, respectivamente, -0,40 e -0,42. Já os eventos 3, 4, 6 e 7 apresentaram correlações de médiapara alta magnitude, com a classificação final da libido, respectivamente, 0,47; 0,37; 0,44 e 0,67. Para os touros que reagiramao teste a média do tempo para a primeira atitude de interesse por uma fêmea (TIS) foi de 40,88 ± 53,02 segundos, comvariações de 10 a 210 segundos. Já o tempo para a primeira atitude de monta (TAM) foi de 101,20 ± 74,03 segundos, variandode 10 a 250 segundos. O TR, para os touros que efetuaram monta completa foi de 200,18 ± 107,77 segundos, variando de 10a 310 segundos.. Registraram-se correlações significativas entre TIS e TAM (0,48) e de TR e TAM (0,65). Conclui-se que oestudo detalhado dos eventos sexuais podem indicar importantes marcadores do comportamento sexual de touros da raçaNelore, direcionando as adaptações necessárias nos testes de libido em curral para touros zebuínos.


A total of 38 Nelore adult bulls, andrologicaly evaluated, were pen tested for libido, being observed the following Sexual reaction(SR): Flehmen reflex (1), active riding with mounting positioning (2), impulse of mounting (3), attempts to mount (4), abortemounting (5), mounting attempts with new attempts (6), and full mounting (7). Bulls were classified as high libido (scores 6 an7), medium libido (scores 3 to 5) and low libido (scores 1 and 2). It was registered high rate (68.42 percent) of bulls with medium anhigh libido. Mean frequency for SR 1 to 7 were, respectively, 3.02 ± 1.6; 0.44 ± 0.79; 0.92 ± 1.17; 0.68 ± 0.98; 0.10 ± 0.38; 0 .100.31 e 0.23 ± 0.48. After 2 minutes and 30 seconds of libido test, bulls classified as low libido realized two times more the S1, when compared to bulls classified as high libido (p>0,05). Negative correlations among SR 1 and SR 3, 4 of -0.40 e -0.4respectively, were recorded. SR 3, 4, 6 and 7 showed correlations varying from medium to high with final libido classificatiorespectively, 0,47; 0.37; 0.44 e 0.67. Among bulls that react to the libido test, the mean reaction time for the first reaction tofemale in heat (RF) was 40.88 ± 53.02 seconds (ranging from 10 to 210 seconds) and the mean mounting reaction time (MTwas 101.20 ± 74.03 seconds (ranging from 10 to 250 seconds). RT for bulls that realized full mounting was 200.18 ± 107.7second, with variations from 10 to 310 seconds. Correlations of 0.48 between RF and MT and 0.65 between RT and MT werecorded (p<0,05). It was concluded that the sexual events are important for make adaptations in pen libido test for zebu bullFurthermore, the pen libido test was efficient in selecting bulls that realized higher number of full mountings.Keywords: sexual behavior, Nelore bulls, Zebu.

ABSTRACT

O presente estudo reporta observações sobre idade, peso e concentração plasmática de progesterona em novilhas mestiças (zebu x holandês) durante o período peripuberal. As médias de idade e peso na puberdade foram 29,1-2,0 meses e 326-19,2 kg respectivamente. Dos cinco animais acompanhados, dois apresentaram elevações transitórias de progesterona (entre 0,5 e 1,0 ng/ml) anteriores à puberdade. Dentre os que não apresentaram elevação transitória, dois tiveram corpo lúteo de baixa produção de progesterona (1,2 e 1,6 ng/ml) após a primeira ovulação e um apresentou corpo lúteo de longa duração (acima de 28 dias). Os dados observados indicam que as elevações transitórias de progesterona anteriores à puberdade podem ocorrer ocasionalmente, no entanto parece ser necessária a instalação de uma função luteal competente após o primeiro cio.


The present study reports on age, body weight and pre-liminary observations on the plasmatic progesterone leveis in cross-breeding heifers (zebu x holstein) during peripuberal period. The mean age and body weight of tive heifers at puberty were 29.1-2.0 months and 326.8-19.2 kg respectively. Two of tive heifers exhibited after the first estrous low progesterone productive corpus luteum (1.2 and 1.6 ng/ml) and the third heifer exhibited one long-lived corpus luteum (>28 days long). The data observed in this study indicated that transiente increases of plasmatic progesterone prior puberty can not occur but should be necessary for the competent luteal funtioh, after the first estrous.
