ABSTRACT

Clathrin-mediated endocytosis (CME) is one of the main pathways for plant cells to internalize the membrane proteins in response to changing environmental conditions. The Epsin-like Clathrin adaptor proteins (ECAs) play important roles in the assembly of clathrin coat; however, their involvement in plant response to heat stress remains unclear. Here we report that SlECA4 responded to heat stress, and the silencing and knockout of SlECA4 increased tomato sensitivity to heat stress, while the overexpression of SlECA4 enhanced tomato tolerance to heat stress. Meanwhile, the treatment with a CME inhibitor, ES9-17, reduced tomato heat tolerance. SlECA4 localized to the plasma membrane (PM), the trans-Golgi network/early endosomes (TGN/EE), and the prevacuolar compartment (PVC)/late endosomes. In SlECA4-KO line, both CME and recycling from the TGN/EE to the PM were inhibited. These data suggest that SlECA4 involved in CME. After heat treatment, more punctate structures of SlECA4:GFP accumualted in tobacco leaf epidermal cells by transient expression. Furthermore, compared to WT, the rate of CME was inhibited under heat stress in SlECA4-KO line. Taken together, the Epsin-like Clathrin adaptor protein SlECA4 plays a positive role in tomato tolerance to heat stress via the CME pathway.

ABSTRACT

Autophagy, a highly conserved protein degradation system, plays an important role in protecting cells from adverse environmental conditions. ATG8-INTERACTING PROTEIN1 (ATI1) acts as an autophagy receptor, but its functional mechanisms in plants' heat stress tolerance remain unclear. In this study, using LC-MS/MS, we identified malate dehydrogenase (SlMDH3) as a SlATI1-interacting protein. Further studies showed that heat stress induced the expression of SlMDH3 and SlMDH3 co-localized with SlATI1 under both 22 °C and 42 °C heat treatment conditions. Moreover, silencing of SlMDH3 increased the sensitivity of tomato to heat stress, as evidenced by exacerbated degradation of chlorophyll; accumulation of MDA, H2O2, and dead cells; increased relative conductivity; and inhibition of stress-related gene expression. Conversely, overexpression of SlMDH3 improved tomato's heat tolerance, leading to opposite effects on physiological indicators and gene expression compared to SlMDH3 silencing. Taken together, our study suggests that SlMDH3 interacts with SlATI1 and positively regulates tomato heat tolerance.

ABSTRACT

MnNb2 O6 anode has attracted much attention owing to its unique properties for holding Li ions. Unluckily, its application as a Li-ion battery anode is restricted by low capacity because of the inferior electronic conductivity and limited electron transfer. Previous studies suggest that structure and component optimization could improve its reversible capacity. This improvement is always companied by capacity increments, however, the reasons have rarely been identified. Herein, MnNb2 O6 -C nanofibers (NFs) with MnNb2 O6 nanoparticles (~15 nm) confined in carbon NFs, and the counterpart MnNb2 O6 NFs consisting of larger nanoparticles (40-100 nm) are prepared by electrospinning for clarifying this phenomenon. The electrochemical evaluations indicate that the capacity achieved by the MnNb2 O6 NF electrode presents an activation process and a degradation in subsequence. Meanwhile, the MnNb2 O6 -C NF electrode delivers high reversible capacity and ultra-stable cycling performance. Further analysis based on electrochemical behaviors and microstructure changes reveals that the partial structure rearrangement should be in charge of the capacity increment, mainly including pseudocapacitance increment. This work suggests that diminishing the dimensions of MnNb2 O6 nanoparticles and further confining them in a matrix could increase the pseudocapacitance-dominated capacity, providing a novel way to improve the reversible capacity of MnNb2 O6 and other intercalation reaction anodes.

ABSTRACT

A designed method for the preparation of 3-aminomethylated maleimides via Morita-Baylis-Hillman (MBH) reaction was developed. This phosphine-catalyzed coupling adopted maleimides and 1,3,5-triazinanes as the substrate, giving a series of 3-aminomethylated maleimide derivatives with a double bond retained on the maleimide ring in 41-90% yield. Acylation, isomerization, and Michael addition of the obtained products demonstrated the synthetic application of the present protocol. The results of control experiments indicated that phosphorus ylide formation and elimination take place during the reaction pathway.

ABSTRACT

A total of 480 one-day-old AA broiler chicks were randomly allocated to one of four treatments in a 2 × 2 factorial to investigate the effects of tannic acid (TA) on growth performance, relative organ weight, antioxidant capacity, and intestinal health in broilers dietary exposed to aflatoxin B1 (AFB1). Treatments were as follows: (1) CON, control diet; (2) TA, CON + 250 mg/kg TA; (3) AFB1, CON + 500 µg/kg AFB1; and (4) TA+AFB1, CON + 250 mg/kg TA + 500 µg/kg AFB1. There were 10 replicate pens with 12 broilers per replicate. Dietary AFB1 challenge increased the feed conversion ratio during days 1 to 21 (P < 0.05). The TA in the diet did not show significant effects on the growth performance of broilers during the whole experiment period (P > 0.05). The liver and kidney relative weight was increased in the AF challenge groups compared with the CON (P < 0.05). The addition of TA could alleviate the relative weight increase of liver and kidney caused by AFB1 (P < 0.05). Broilers fed the AFB1 diets had lower activity of glutathione peroxidase, catalase, total superoxide dismutase, S-transferase, and total antioxidant capacity in plasma, liver and jejunum, and greater malondialdehyde content (P < 0.05). Dietary supplemented with 250 mg/kg TA increased the activities of antioxidative enzymes, and decreased malondialdehyde content (P < 0.05). In addition, AFB1 significantly reduced the villus height and crypt depth ratio in the ileum on day 42 (P < 0.05). In conclusion, supplementation with 250 mg/kg TA could partially protect the antioxidant capacity and prevent the enlargement of liver in broilers dietary challenged with 500 µg/kg AFB1.

ABSTRACT

Porcine epidemic diarrhea virus (PEDV) has become a challenging problem in pig industry all over the world, causing significant profit losses. Tannins and organic zinc have been shown to exert protective effects on the intestinal dysfunction caused by endotoxins. However, there is little information on tannic acid-chelated zinc (TAZ) supplementation in the diet of newborn piglets. This study was conducted to determine the effects of TAZ on the intestinal function of piglets infected with PEDV. Thirty-two 7-day-old piglets were randomly allocated to 1 of 4 treatments in a 2 × 2 factorial design consisting of 2 diets (0 or 50 mg/kg BW TAZ) and challenge (saline or PEDV). On day 9 of the trial, 8 pigs per treatment received either sterile saline or PEDV solution at 106 TCID50 (50% tissue culture infectious dose) per pig. Pigs infected with PEDV had greater diarrhea rate and lower average daily gain (ADG) (P < 0.05). PEDV infection decreased plasma D-xylose concentration, most antioxidative enzyme activities in plasma and intestine, as well as the small intestinal villus height (P < 0.05). Plasma diamine oxidase and blood parameters were also affected by PEDV infection. Dietary supplementation with TAZ could ameliorate the PEDV-induced changes in all measured variables (P < 0.05). Moreover, TAZ decreased the concentration of malondialdehyde in plasma, duodenum, jejunum, and colon (P < 0.05). Collectively, our results indicated that dietary TAZ could alleviate PEDV induced damage on intestinal mucosa and antioxidative capacity, and improve the absorptive function and growth in piglets. Therefore, our novel findings also suggest that TAZ, as a new feed additive for neonatal and weaning piglets, has the potential to be an alternative to ZnO.

ABSTRACT

Multicolor imaging, which maps the distribution of different targets, is important for in vivo molecular imaging and clinical diagnosis. Fluorine 19 magnetic resonance imaging (19 F MRI) is a promising technique because of unique insights without endogenous background or tissue penetration limit. Thus multicolor 19 F MRI probes, which can sense a wide variety of molecular species, are expected to help elucidate the biomolecular networks in complex biological systems. Here, a versatile model of activatable probes based on fluorinated ionic liquids (ILs) for multicolor 19 F MRI is reported. Three types of ILs at different chemical shifts are loaded in nanocarriers and sealed by three stimuli-sensitive copolymers, leading to "off" 19 F signals. The coating polymers specifically respond to their environmental stimuli, then degrade to release the loaded ILs, causing 19 F signals recovery. The nanoprobes are utilized for non-invasive detection of tumor hallmarks, which are distinguished by their individual colors in one living mouse, without interference between each other. This multicolor imaging strategy, which adopts modular construction of various ILs and stimuli-responsive polymers, will allow more comprehensive sensing of multiple biological targets, thus, opening a new realm in mechanistic understanding of complex pathophysiologic processes in vivo.
