ABSTRACT

This case report is one of the rare cases of bilateral pheochromocytoma associated with neurofibromatosis type 1. The interest lies in the clinical form in which the diagnosis was revealed. We report the case of a 38-year-old woman admitted for severe hypertension resistant to triple therapy. Clinical examination revealed Cafe-au-lait spots, which are pigmented birthmarks that appear as patches on the skin with a light to dark brown colour. More than six spots are present in an estimated 95% of people diagnosed with neurofibromatosis type 1 (NF1). Abdominal computed tomography (CT) showed bilateral adrenal tumor involvement. The diagnosis of pheochromocytoma was made by measuring urinary Vanillylmandelic acid (VMA). The evolution was favorable after the excision of the tumor, with normalization of blood pressure. In conclusion: resistant hypertension with café au lait spots may indicate pheochromocytoma, especially bilateral, suggesting an underlying genetic condition like NF1, warranting systematic screening.


En este reporte se documenta un caso raro de feocromocitoma bilateral asociado a neurofibromatosis tipo 1. El interés radica en la forma clínica en la que se reveló el diagnóstico. Presentamos el caso de una mujer de 38 años que ingresa por hipertensión arterial severa resistente a triple terapia. El examen clínico reveló manchas café con leche, que son marcas de nacimiento pigmentadas que aparecen como manchas en la piel y que son de un color marrón claro a oscuro. Se estima que el 95% de las personas diagnosticadas con neurofibromatosis tipo 1 (NF1) presentan más de seis manchas. La tomografía computarizada (TC) abdominal mostró afectación tumoral suprarrenal bilateral. El diagnóstico de feocromocitoma se realizó mediante la medición del ácido vanilmandélico (VMA) urinario. La evolución fue favorable tras la extirpación del tumor, con normalización de la presión arterial. En conclusión, la hipertensión resistente con manchas café con leche puede indicar feocromocitoma, especialmente si es bilateral, lo que sugiere una afección genética subyacente como la NF1, que justifica un tamizaje sistemático.

ABSTRACT

The first step in comprehending the properties of Au10 clusters is understanding the lowest energy structure at low and high temperatures. Functional materials operate at finite temperatures; however, energy computations employing density functional theory (DFT) methodology are typically carried out at zero temperature, leaving many properties unexplored. This study explored the potential and free energy surface of the neutral Au10 nanocluster at a finite temperature, employing a genetic algorithm coupled with DFT and nanothermodynamics. Furthermore, we computed the thermal population and infrared Boltzmann spectrum at a finite temperature and compared it with the validated experimental data. Moreover, we performed the chemical bonding analysis using the quantum theory of atoms in molecules (QTAIM) approach and the adaptive natural density partitioning method (AdNDP) to shed light on the bonding of Au atoms in the low-energy structures. In the calculations, we take into consideration the relativistic effects through the zero-order regular approximation (ZORA), the dispersion through Grimme's dispersion with Becke-Johnson damping (D3BJ), and we employed nanothermodynamics to consider temperature contributions. Small Au clusters prefer the planar shape, and the transition from 2D to 3D could take place at atomic clusters consisting of ten atoms, which could be affected by temperature, relativistic effects, and dispersion. We analyzed the energetic ordering of structures calculated using DFT with ZORA and single-point energy calculation employing the DLPNO-CCSD(T) methodology. Our findings indicate that the planar lowest energy structure computed with DFT is not the lowest energy structure computed at the DLPN0-CCSD(T) level of theory. The computed thermal population indicates that the 2D elongated hexagon configuration strongly dominates at a temperature range of 50-800 K. Based on the thermal population, at a temperature of 100 K, the computed IR Boltzmann spectrum agrees with the experimental IR spectrum. The chemical bonding analysis on the lowest energy structure indicates that the cluster bond is due only to the electrons of the 6 s orbital, and the Au d orbitals do not participate in the bonding of this system.

ABSTRACT

Resumen Objetivos : Monitorear la actividad de oviposición del mosquito Aedes aegypti y de casos de dengue y chikun gunya en cuatro localidades de Argentina templada, durante la epidemia del 2023. Métodos : Durante el verano y otoño del 2023, se mo nitoreó semanalmente mediante ovitrampas la actividad de oviposición del mosquito vector, y se registró el arribo de casos con dengue o chikungunya a Tandil, Olavarría, Bahía Blanca y Laprida. Resultados : La variación mensual del porcentaje de trampas positivas fue similar en las tres primeras localidades; en Laprida no se detectó el mosquito. Por el contrario, se observó una diferencia significativa del porcentaje de trampas que alguna vez resultó positiva en cada localidad, siendo mayor en Olavarría (83%), que en Bahía Blanca (67%) y Tandil (49%). Respecto a las enfermedades, se registraron 18 casos importados de dengue y 3 de chikungunya. Además, se registró el primer caso autóctono de dengue en la región, siendo el más austral hasta el momento. Conclusión : Es imprescindible sensibilizar y capacitar a los integrantes de los sistemas de salud de las nue vas regiones expuestas al Ae. aegypti para la detección temprana de casos, y a la población en general para potenciar las acciones de prevención.


Abstract Objectives : To monitor the oviposition activity of the mosquito Aedes aegypti and of dengue and chikungunya cases in four localities of temperate Argentina, during the 2023 epidemic. Methods : During the summer and autumn of 2023, the oviposition activity of the mosquito vector was monitored weekly using ovitraps, and the arrival of cases with dengue or chikungunya in Tandil, Olavarría, Bahía Blanca and Laprida were registered. Results : Monthly variations of the percentage of positive traps were similar in the first three locations; in Laprida the mosquito was not detected. On the con trary, a significant difference was observed in the per centage of total traps that ever tested positive in each locality, being higher in Olavarría (83.3%) than in Bahía Blanca (68.6%) and Tandil (48.7%). Regarding diseases, 18 imported cases of dengue and 3 of chikungunya were registered. In addition, the first autochthonous case of dengue in the region was recorded, being the southern most until known. Conclusion : It is essential to raise awareness and train the members of the health systems of the new regions exposed to Ae. aegypti for early detection of cases, and to the general population to enhance pre vention actions.

ABSTRACT

Despite the remarkable theoretical applications of silicene, its synthesis remains a complex task, with epitaxial growth being one of the main routes involving depositing evaporated Si atoms onto a suitable substrate. Additionally, the requirement for a substrate to maintain the silicene stability poses several difficulties in accurately determining the growth mechanisms and the resulting structures, leading to conflicting results in the literature. In this study, large-scale molecular dynamics simulations are performed to uncover the growth mechanisms and characteristics of epitaxially grown silicene sheets on Au(111) and Au(110) substrates, considering different temperatures and Si deposition rates. The growth process has been found to initiate with the nucleation of several independent islands homogeneously distributed on the substrate surface, which gradually merge to form a complete silicene sheet. The results consistently demonstrate the presence of a buckled silicene structure, although this characteristic is notably reduced when using an Au(111) substrate. Furthermore, the analysis also focuses on the quality and growth mode of the silicene sheets, considering the influence of temperature and deposition rate. The findings reveal a prevalence of the Frank-van der Merwe growth mode, along with diverse forms of defects throughout the sheets.

RESUMEN La displasia fibrosa es una enfermedad ósea benigna y progresiva, de base genética que puede afectar a uno o varios huesos. Su alta incidencia en la edad pediátrica sirvió de base para que anteriormente fuera considerada exclusiva de esta etapa; sin embargo, a la fecha, existen reportes que evidencian su desarrollo y crecimiento posterior en la segunda década de vida. Cuando está asociada a desequilibrios endocrinos y manchas café con leche, forma parte del síndrome de McCune-Albright. El presente reporte de caso muestra las características clínicas y radiológicas de la región bucofacial de un paciente adulto portador del síndrome de McCune-Albright. Las lesiones en los maxilares afectan la morfología y causan disfunción. A nivel dentario, los cambios producidos se traducen en maloclusión y defectos estructurales. En los estudios radiológicos se observaron cambios con densidad mixta y el patrón de vidrio esmerilado.


ABSTRACT Fibrous dysplasia is a benign and progressive bone disease of genetic basis that can affect one or several bones. Its high incidence in the pediatric age served as a basis for it to be previously considered exclusive to this stage; however, there are reports to date that show its development and later growth in the second decade of life. When associated with endocrine imbalances and café-au-lait spots, it is part of the McCune-Albright syndrome. The present case report shows the clinical and radiologic features of the oral-facial region of an adult patient with McCune-Albright syndrome. Lesions in the jaws affect the morphology and cause dysfunction. At the dental level, the changes produced result in malocclusion and structural defects. Radiological studies showed changes with mixed density and ground glass pattern.


RESUMO A displasia fibrosa é uma doença óssea benigna, progressiva e de base genética que pode afetar um ou mais ossos. A sua elevada incidência no grupo etário pediátrico foi a base para que anteriormente fosse considerada exclusiva desta fase; no entanto, existem relatos até à data do seu desenvolvimento e crescimento mais tardio na segunda década de vida. Quando associada a desequilíbrios endócrinos e manchas café-com-leite, faz parte da síndrome de McCune-Albright. Este relato de caso mostra as características clínicas e radiológicas da região bucofacial de um paciente adulto com síndrome de McCune-Albright. As lesões nos maxilares afetam a morfologia e provocam disfunções. A nível dentário, as alterações produzidas resultam em má oclusão e defeitos estruturais. Nos estudos radiológicos, foram observadas alterações com densidade mista e padrão de vidro despolido.

ABSTRACT

Surface water pollution has become relevant because growing population and intense industrial activities. Thus, to protect the environment from contamination, recently the electroanalytical sensors that require small sample volume and easy preparation have shown a prominent performance for pharmaceuticals monitoring. For this purpose, a miniaturized electrochemical platform was developed based on recycling obsolete computer integrated circuits (microchips), fitting with the ideals of green chemistry and circular economy. The gold microelectrodes array (Au-µEA) was easily exposed by polishing the device surface and then characterized by optical microscopy, scanning electron microscopy and cyclic voltammetry. To enhance the analytical performance for isoniazid detection, the Au-µEA was modified with electrochemically reduced graphene oxide (ERGO). The developed sensor presented a linear range between 5 and 100 µmol L-1 and a limit of detection of 1.38 µmol L-1 demonstrating a reliable performance. Looking to its environmental application, the ERGO/Au-µEA sensor was used for isoniazid quantification in lagoon, river, tap water and synthetic effluent spiked samples with recovery values between 92.5 and 108.4%. Thus, this research field opens up new possibilities in global water-related issues contributing with innovative sustainable solutions.

ABSTRACT

The current research describes the synthesis and characterization of 2-acetylpyridine N(4)-cyclohexyl-thiosemicarbazone ligand (HL) and their two metal complexes, [Au(L)Cl][AuCl2] (1) and [Pd(L)Cl]·DMF (2). The molecular structures of the compounds were determined by physicochemical and spectroscopic methods. Single crystal X-ray diffraction was employed in the structural elucidation of the new complexes. The complexes showed a square planar geometry to the metal center Au(III) and Pd(II), coordinated with a thiosemicarbazone molecule by the NNS-donor system and a chloride ion. Complex (1) also shows the [AuCl2]- counter-ion in the asymmetric unit, and complex (2) has one DMF solvent molecule. These molecules play a key role in the formation of supramolecular structures due to different interactions. Noncovalent interactions were investigated through the 3D Hirshfeld surface by the dnorm function and the 2D fingerprint plots. The biological activity of the compounds was evaluated in vitro against the human glioma U251 cells. The cytotoxicity results revealed great antitumor activity in complex (1) compared with complex (2) and the free ligand. Molecular docking simulations were used to predict interactions and properties with selected proteins and DNA of the synthesized compounds.

ABSTRACT

Aneurysmal coronary artery disease (ACAD) has been reported rarely in patients with neurofibromatosis type 1 (NF1), mostly in adults. We report on a female newborn affected by NF1 with ACAD disclosed during investigation for an abnormal prenatal ultrasound along with a review of the previously reported cases. The proposita had multiple café-au-lait spots and had no cardiac symptoms. Echocardiography, and cardiac computed tomography angiography confirmed aneurysms on the left coronary artery, left anterior descending coronary artery, and of the sinus of Valsalva. Molecular analysis detected the pathogenic variant NM_001042492.3(NF1):c.3943C>T (p.Gln1315*). Literature findings on ACAD in NF1 indicated that this mostly occurs in males, showing predilection for the development of aneurysms at the left anterior descending coronary artery, and manifesting predominantly as acute myocardial infarction, inclusively in teenagers, though it may be also asymptomatic as in our case. This report documents the first case of ACAD in a patient with NF1 diagnosed at birth, emphasizing that its early diagnosis is essential to prevent potential life-threatening events attributable directly to coronary lesions.

ABSTRACT

A series of neutral acridine-based gold(III)-NHC complexes containing the pentafluorophenyl (-C6F5) group were synthesized. All of the complexes were fully characterized by analytical techniques. The square planar geometry around the gold center was confirmed by X-ray diffraction analysis for complexes 1 (Trichloro [1-methyl-3-(9-acridine)imidazol-2-ylidene]gold(III)) and 2 (Chloro-bis(pentafluorophenyl)[1-methyl-3-(9-acridine)imidazol-2-ylidene]gold(III)). In both cases, the acridine rings play a key role in the crystal packing of the solid structures by mean of π-π stacking interactions, with centroid-centroid and interplanar distances being similar to those found in other previously reported acridine-based Au(I)-NHC complexes. A different reactivity when using a bulkier N-heterocyclic carbene ligand such as 1,3-bis-(2,6-diisopropylphenyl)-2-imidazolidinylidene (SIPr) was observed. While the use of the acridine-based NHC ligand led to the expected organometallic gold(III) species, the steric hindrance of the bulky SIPr ligand led to the formation of the corresponding imidazolinium cation stabilized by the tetrakis(pentafluorophenyl)aurate(III) [Au(C6F5)4]- anion. Computational experiments were carried out in order to figure out the ground state electronic structure and the binding formation energy of the complexes and, therefore, to explain the observed reactivity.

ABSTRACT

Organic reactions carried out in water under mild conditions are state-of-the-art in terms of environmentally benign chemical processes. In this direction, plasmonic catalysis can aid in accomplishing such tasks. In the present work, cyclodextrin-mediated AuPd bimetallic nanoparticles (NPs) were applied in room-temperature aqueous Suzuki-Miyaura reactions aiming at preparing biaryl products based on fluorene, isatin, benzimidazole and resorcinol, with yields of 77 % up to 95 %. AuPd NPs were revealed to be a physical mixture of Au and Pd particles circa 20 and 2 nm, respectively, through X-ray diffraction, dynamic light scattering, UV-Vis spectroscopy and transmission electron microscopy analyses.

ABSTRACT

Neurofibromatosis type 1 is one of the most common genetic autosomal dominant disorders described, with a prevalence of 1 in 2000 to 1 in 3000 individuals. It is characterized by skin, nerves, and bone abnormalities. Non-related to NF1, hypospadias is a displacement in the urethral opening which in the majority of patients has an idiopathic cause. Here, we describe a patient with neurofibromatosis type 1, hypospadias, and unilateral cryptorchidism. The heterozygous variants c.6789_6792delTTAC, p.(Tyr2264Thrfs*5) and c.140A>G, p.(Tyr47Cys) were found in the NF1 and NR5A1 genes, respectively. This case contributes to the phenotypical characterization of patients with NF1 but also with hypospadias caused by a mutation in the NR5A1 gene, which usually leads to severe sex disorders.

ABSTRACT

Herein, we report the synthesis of Au nanoparticles (AuNPs) in chitosan (CTS) solution by chemically reducing HAuCl4. CTS was further functionalized with glycidyl methacrylate (chitosan-g-glycidyl methacrylate/AuNP, CTS-g-GMA/AuNP) to improve the mechanical properties for cellular regeneration requirements of CTS-g-GMA/AuNP. Our nanocomposites promote excellent cellular viability and have a positive effect on cytokine regulation in the inflammatory and anti-inflammatory response of skin cells. After 40 days of nanocomposite exposure to a skin wound, we showed that our films have a greater skin wound healing capacity than a commercial film (TheraForm®), and the presence of the collagen allows better cosmetic ave aspects in skin regeneration in comparison with a nanocomposite with an absence of this protein. Electrical percolation phenomena in such nanocomposites were used as guiding tools for the best nanocomposite performance. Our results suggest that chitosan-based Au nanocomposites show great potential for skin wound repair.

ABSTRACT

Graphene-TiO2 composites have been investigated in various photocatalytic reactions showing successful synergy compared to pristine TiO2. In the present work, graphene oxide (GO) was synthesized by the Hummers method and then reduced graphene oxide-TiO2 composites (rGO/TiO2) were obtained by an in situ GO photoreduction route. X-ray diffraction, FTIR, Raman, UV-vis DRS, and photoluminescence were the main characterization techniques. The obtained composites containing 1 and 3 wt.% rGO were evaluated in the cyanide (50 mg/L) oxidation and Au-cyanide complex (300 mg/L) degradation under UV-A light. The composites showed higher photocatalytic activity than TiO2, mainly with the 1% rGO content. Cyanate and gold nanoparticles, deposited on the photocatalyst's surface, were the main byproducts during the photocatalyst assessment. The improved photocatalytic activity of the composites was attributed to a higher rate of electron transfer and a lower rate of charge recombination due to the chemical interaction of rGO with TiO2.

ABSTRACT

Cytogenetic data showed the enrichment of repetitive DNAs in chromosomal rearrangement points between closely related species in armored catfishes. Still, few studies integrated cytogenetic and genomic data aiming to identify their prone-to-break DNA sites. Here, we aimed to obtain the repetitive fraction in Rineloricaria latirostris to recognize the microsatellite and homopolymers flanking the regions previously described as chromosomal fusion points. The results indicated that repetitive DNAs in R. latirostris are predominantly DNA transposons, and considering the microsatellite and homopolymers, A/T-rich expansions were the most abundant. The in situ localization demonstrated the A/T-rich repetitive sequences were scattered on the chromosomes, while A/G-rich microsatellite units were accumulated in some regions. The DNA transposon hAT, the 5S rDNA, and 45S rDNA (previously identified in Robertsonian fusion points in R. latirostris) were clusterized with some microsatellites, especially (CA)n, (GA)n, and poly-A, which were also enriched in regions of chromosomal fusions. Our findings demonstrated that repetitive sequences such as rDNAs, hAT transposons, and microsatellite units flank probable evolutionary breakpoint regions in R. latirostris. However, due to the sequence unit homologies in different chromosomal sites, these repeat DNAs only may facilitate chromosome fusion events in R. latirostris rather than working as a double-strand breakpoint site.

PURPOSE: Brow ptosis can interfere with blepharoplasty results and can be corrected using several techniques. The present study was performed to compare two techniques for brow suspension, both with concomitant upper eyelid blepharoplasty. METHOD: A prospective, interventional, randomized study involving 27 female patients was performed to compare the effects of two different techniques of brow suspension: eyebrow suspension with nylon thread (ESN) and internal browpexy fixation (IBF), both combined with upper lid blepharoplasty. Qualitative assessment was carried out using a questionnaire, and quantitative angular measurement analysis of the brow position was performed using digital photographs taken with a Vectra H1 camera before, 60, and 120 days after surgery. The data were transferred to an Excel table and statistically analyzed. RESULTS: ESN was performed in 14 patients and IBF in 13 patients. Both groups were homogeneous in terms of age. The main preoperative complaints were excess skin on the upper eyelid (81.4%) and reduced visual field (59.2%) in both groups. Most of the patients expected aesthetic improvement, and the surgical results were considered satisfactory for all of them. There was no significant difference between ESN and IBF in the quantitative evaluation of eyebrow position. CONCLUSION: Both ESN and IBF resulted in a high degree of patient satisfaction. Both techniques provided similar brow suspension, emphasizing that IBF is technically easier to perform and produces less scarring.

OBJECTIVE: To evaluate sociodemographic and medical predictors of patient return to a neurofibromatosis subspecialty clinic. STUDY DESIGN: Data were collected from the Washington University Neurofibromatosis Clinical Program electronic medical records. A total of 713 subjects with initial visits to the Washington University Neurofibromatosis Clinical Program between July 1, 2005 and December 18, 2020 were included. Variables collected included sex, race, ethnicity, age, date of first visit, place of residence, diagnosis, insurance payer, physician recommendation for return, and subject return. Return rates for each demographic group were calculated. Bivariate analyses were performed to inform variable inclusion in the model, and a binary logistic regression model was calculated to predict subject return. RESULTS: The overall return rate was 76%. The binary logistic regression model was statistically significant (χ29 = 131.094; P < .001) and showed that subjects who self-identified as Black and/or African American, presented with or received a diagnosis of café-au-lait macules at their initial visit, were from a rural area, were older, or who lived farther from the Washington University Neurofibromatosis Clinical Program were less likely to return to clinic. CONCLUSIONS: These findings support the implementation of tailored communication and monitoring interventions to improve the care for children with neurofibromatosis type 1.

ABSTRACT

This study is about the intervention carried out by a group of professionals from the Interdisciplinary Mental Health Team of the Emergencies Department in a public hospital in Buenos Aires City between 2017 and 2019. It arose from the need to respond to a public health problem related to the increasing number of patients living in situations involving homelessness, drug abuse, violent behaviour and criminal background. The multiple facets of this problem made us think about the impossibility of addressing it only in the individual sense of each patient. We were forced to admit that we were facing a problem of a complex, collective, social and institutional order, which the predominant medical model had neither been able to handle nor understand. We sought to include a Jungian approach to this contemporary problem found in large Latin American cities. Amplifying a symbolic base allowed us to understand creatively the complexity of the phenomenon, which could not be read in any manner other than the social and collective.


Cette étude porte sur l'intervention menée dans un hôpital public de Buenos Aires entre 2017 et 2019 par un groupe de professionnels de l'équipe interdisciplinaire de Santé Psychique. Elle fut mise en place du fait du besoin de répondre au problème de santé publique lié au nombre croissant de patients vivant sans domicile fixe, avec des problèmes de drogue, de comportement violent et de criminalité. Les aspects multiples de ce problème nous amenèrent à penser qu'il était impossible d'y faire face seulement sur la base de chaque patient en tant qu'individu. Nous avons été forcés d'admettre que nous étions confrontés à un problème complexe, collectif, d'ordre social et institutionnel, que le modèle médical prédominant n'avait pu ni résoudre ni comprendre. Nous avons cherché à inclure une approche Jungienne à ce problème contemporain que l'on retrouve dans les grandes villes d'Amérique Latine. L'amplification d'un fondement symbolique nous a permis de comprendre de manière créative la complexité du phénomène, dont la lecture ne pouvait qu'être sociale et collective.


Este trabajo trata sobre una intervención realizada por un grupo de profesionales del Equipo Interdisciplinario de Salud Mental del Departamento de Emergencias de un Hospital Público de la Ciudad de Buenos Aires entre 2017-2019. Surgió ante la necesidad de dar respuesta a una problemática de Salud Pública relacionada con el incremento de pacientes que viven en situación de calle con consumo de drogas, conductas violentas y antecedentes delictivos. Las múltiples facetas de este problema nos llevó a reflexionar sobre la imposibilidad de darle solución si solamente lo enfocábamos desde lo individual de cada paciente. Tuvimos que admitir que nos encontrábamos ante un problema complejo, de orden colectivo, social e institucional que el modelo médico predominante era incapaz de comprender o resolver. Entonces pensamos observar desde un enfoque Junguiano este problema contemporáneo que encontramos en las grandes urbes de América Latina. Amplificar una base simbólica nos posibilitó comprender creativamente la complejidad del fenómeno, que no podía ser abordado sino desde lo social y colectivo.

Surgiu da necessidade de responder a um problema de saúde pública relacionado ao número crescente de pacientes que vivem em situações envolvendo situação de rua, abuso de drogas, comportamento violento e antecedentes criminais. As múltiplas facetas desse problema nos fizeram pensar na impossibilidade de abordá-lo apenas no sentido individual de cada paciente. Fomos forçados a admitir que estávamos diante de um problema de ordem complexa, coletiva, social e institucional, que o modelo médico predominante não era capaz de lidar nem entender. Procuramos incluir uma abordagem junguiana para esse problema contemporâneo encontrado nas grandes cidades latino-americanas. Ampliar uma base simbólica nos permitiu entender criativamente a complexidade do fenômeno, que não poderia ser lido de outra maneira além do social e coletivo.

ABSTRACT

Abstract McCune - Albright syndrome is a genetic disease with cutaneous mosaicism caused by post-zygotic activating mutations in GNAS locus, it has a triad of fibrous bone dysplasia, café-au-lait macules and precocious puberty. We examined a 22-year-old female patient with café au lait spot in right side of the abdomen, with a chessboard - like distribution, extending to right thigh with geographical contours, she has also an ovarian cyst, scoliosis and truncal obesity. Biopsies were taken from the hyperpigmented area and processed for light microscopy and for transmission electron microscopy. Light microscopy showed increased melanin pigment with HE staining. Immunohistochemistry with melanocytic markers (HMB-45 and Melan-A) revealed a normal number of melanocytes. Transmission electron microscopy demonstrated normal epidermal structures, such as desmosomes, cytokeratin filaments and hemidesmosomes. With high magnifications an irregular melanossomal contour was seen, with some indentations in their outline.

ABSTRACT

Detection of copper plays a prominent role in the environmental protection and human health. Herein, we firstly design and construct an "off-on" upconversion fluorescence resonance energy transfer (UFRET) probe with low toxicity for the Cu2+ determination by using NaYF4: Yb3+, Er3+ upconversion nanoparticles (UCNPs) and Au NPs. UCNPs with positive charge and Au NPs with negative charge are respectively employed as the donor and acceptor, and bound together to form UFRET probe. The upconversion fluorescence quenching of UCNPs occurs by Au NPs through FRET (defined as "off" state). When Cu2+ exists in samples, Cu2+ reacts with 4-mercaptobenzoic acid (4-MBA) capped on the surface of Au NPs to make Au NPs detach from UCNPs, leading to the termination of FRET and the recovery of upconversion fluorescence (defined as "on" state). "Off-on" typed UFRET probe has excellent sensing performances, including linear range of 0.02-1 µM Cu2+ concentration, the limit of detection of 18.2 nM, high selectivity to Cu2+ and good recovery. The probe has been successfully used to determine Cu2+ in spiked tap water with satisfactory results. The probe will provide theoretical and technical support for the design of new sensitive heavy metal ion detection probe.
