ABSTRACT

Yeast and fibrolytic enzymes serve as additives incorporated into the nutrition of ruminants to regulate rumen fermentation and increase the digestibility of fiber, thereby enhancing the efficiency of rumen fermentation. Two experiments were conducted to assess the impact of five diets: a control diet without additives, diets with yeast (Saccharomyces cerevisiae) or exogenous fibrolytic enzymes (EFE), and diets with a blend of 0.7yeast + 0.3EFE or 0.7EFE + 0.3Yeast (based on recommended levels in g/kg of total DM). In the first experiment, 40 five-month-old Santa Ines lambs (mean weight 25.0 ± 1.3 kg) were distributed in a completely randomized design (5 treatments and 8 lambs) for 81 days to evaluate performance, ingestive behaviour, and serum metabolites. In the second experiment, 25 Santa Ines male lambs weighing 25.7 ± 4.1 kg were housed in metabolic cages, in a randomized design with 5 treatments and 5 lambs, evaluating digestibility, nitrogen balance, and rumen pH. EFE supplementation increased intakes of dry matter (DM), total digestible nutrients (TDN), and apNDF (mean of 38.1, 5.26, and 27%, respectively) compared to yeast or the 0.7yeast-0.3EFE blend. Feed conversion was most efficient (mean of 27.1%) in lambs fed Yeast, 0.7EFE + 0.3yeast, and the control diet. Lambs fed 0.7yeast + 0.3EFE spent less time eating (mean of 16.5%) and more time idling (mean of 10.75%), whereas EFE-fed lambs spent more time eating (mean of 19.73%), and 0.7EFE + 0.3yeast-fed lambs spent more time ruminating (mean of 20.14%). Control group lambs chewed and ruminated less (means of 24.64 and 17.21%, respectively) compared to other treatments. Lambs on the 0.7yeast + 0.3EFE blend had higher eating and rumination efficiency rates for DM and apNDF (mean of 19.11 and 17.95%, respectively) compared to other additive treatments or individual additives. They also exhibited lower (means 7.59 g/d) urinary N excretion, with improved N retention (mean 3185 g/d) compared to the control group. There were significant effects on serum albumin and cholesterol concentrations, with the 0.7yeast + 0.3EFE blend showing higher albumin (mean 4.08 g/dL) levels, while diets without additives and yeast-EFE blends had higher cholesterol (mean of 62.51 g/dL) concentrations. Including Saccharomyces cerevisiae yeast along with 0.7 yeast + 0.3 EFE blend is recommended when feeding similar lamb diets to those used herein because it improves the efficiency of intake, rumination of DM and NDF, and nitrogen utilization without affecting the lamb performance.

ABSTRACT

Data and descriptive information were gathered from 226 peer-reviewed scientific publications from beef cattle experiments in which enteric methane and other animal response variables were measured. The dataset was based on the bibliography used by Arndt et al. (2022) but expanded to also include more recent studies published from 2019 to 2023. All articles were identified for inclusion in the dataset using the "Web of Science Core Collection", the "Commonwealth Agricultural Bureau International (CABI)", and the "EBSCO Discovery Service" databases with the search terms "methane" and "enteric" in combination with "beef", "cattle", "rumen", and "ruminant". Additionally, the search term "rumen" was used in combination with "energy balance", "energy metabolism", or "energy partitioning". For dataset inclusion, it was necessary for all studies to be written in English and at a minimum, quantify feed dry matter intake and enteric methane emissions as well as provide measures of variance for these estimates. Studies were primarily designed as completely randomized, randomized block, or crossover experiments. The dataset includes 895 records (rows) and 138 variables (columns). Reported variables include publication information, experimental design, animal description, methane measurement method, diet nutrient composition, and means and measures of variance for feed dry matter intake and enteric methane emissions. Additionally, depending on the study, data reported on rumen fermentation parameters, nutrient digestibility, nitrogen excretion, weight gain, and carbon dioxide and hydrogen emissions were included. This dataset can be used to explore the efficacy of enteric methane mitigation strategies and their impact on beef cattle nutrition and production. Furthermore, the dataset can potentially be used to investigate possible nutrient and feed additive interactions.

BACKGROUND: Visna-maedi is a notifiable disease in Norway, and eliminating the disease is a national goal. The import of sheep into Norway is very limited, and strict regulations apply to the movement of small ruminants between flocks and within defined geographical regions. Several outbreaks have occurred in the last 50 years, and the most recent before 2019 occurred in Trøndelag county in Central Norway in 2002. A national surveillance programme for small ruminant lentivirus infection exists since 2003. RESULTS: In 2019, the national surveillance programme detected seropositive animals for small ruminant lentivirus in a sheep flock in Trøndelag. Based on the result of polymerase chain reaction analysis and histopathological findings, the Norwegian Food Safety Authority concluded the diagnosis of maedi. Further investigations detected maedi in eight additional sheep flocks in the same county. The flocks were placed under restrictions, and the authorities also imposed restrictions on 82 contact flocks. Sequencing of partial gag genes indicated that the virus in the current outbreak was related to the small ruminant lentivirus detected in the same area between 2002 and 2005. CONCLUSIONS: The outbreak investigation shows the need for sensitive and specific diagnostic methods, and an improved and more targeted surveillance strategy. It also demonstrates the risk of disease spreading between flocks through animal movements, and highlights the importance of biosecurity and structured livestock trade. In addition to allowing livestock trade only from flocks documented free from maedi, it may be necessary to monitor sheep flocks over many years, when aiming to eliminate maedi from the Norwegian sheep population.

ABSTRACT

Livestock producers would benefit from more precise predictions of the growth response from nutrients consumed. Previously published models are often limited by the realities of data collection and are unable to account for alterations to body composition, due in part to the response of visceral organs to an alternate diet. The computerized tomography (CT) scanning of lambs enables the analysis of changes in body composition of individual animals over time, potentially supporting better model development and testing. The aim of this experiment was to develop a repeatable method for the analysis of live lamb body composition using CT scans. A secondary aim was to compare the data collected from CT scanning during a feeding trial to two predictive lamb growth models. Cross-bred lambs were fed two feeding levels at two stages of maturity, with CT scans at the beginning and end of each eight-week feeding period. The CT scan derived values for body composition taken at the beginning of feeding periods were used as inputs for two existing lamb growth models. Predictions of body composition were compared with CT scan derived values at the end of feeding periods. The CT scan analysis method used a proportion of images from each lamb to reduce manual image editing. The method was developed by comparing the estimated mass and volume of empty body components using all available CT scans to estimated values using a reduced number of scans from 12 lambs. The CT scan derived lean tissue mass aligned with model predictions at the end of each feeding period, however, CT scan derived fat mass was greater than predictions by both models especially for the high feeding level at the later stage of maturity. These results highlight that the analysis of body composition using CT scans requires further validation, particularly for the viscera, and that models likely require refinement to better predict the efficiency of energy utilisation by different tissues. The use of live animal CT scans can provide more accurate predictions of the growth of saleable products than measuring liveweight alone and will enable ruminant growth models to better adapt to different genetics and changing diets than comparative slaughter. To replicate the current data using comparative slaughter would require four times the animals, as individual lambs were CT scanned four times in this study, demonstrating the potential value of CT scanning in live animal research.

ABSTRACT

According to the Domestic Animal Diversity Information System (DAD-IS) of the FAO, Italy has one of the largest numbers of local small ruminant breeds among European countries. In Southern Italy, namely the Campania Region, Bagnolese and Laticauda sheep breeds and Cilentana goat breeds are considered endangered according to the DAD-IS. Conservation of endangered animal breeds is a goal of the European Union (EU). However, the role of infectious diseases as risk factors for endangered breeds has rarely been considered. Small ruminant lentiviruses (SRLV) infect sheep and goats, causing slow-progressive, persistent, and debilitating diseases that can lead to animal death and productivity loss. In this study, we investigated the presence of SRLV in Bagnolese, Laticauda, and Cilentana breeds using a commercial ELISA in parallel with an in-house ELISA. The results of the two tests were in good agreement (Cohen Kappa 0.84, 95 % CI = 0.76-0.93). Discrepancies between the two tests were resolved using western blotting. In total, 430 samples were tested (248 Bagnolese, 125 Laticauda, and 57 Cilentana). The apparent prevalence rates were 12.5 %, 6.4 %, and 1.7 % in Bagnolese, Laticauda, and Cilentana, respectively. In the molecular analysis of 11 proviral partial sequences, subtypes B2 and A24 were identified in two Bagnolese herds. Owing to the beneficial role of sheep and goat breeding in marginal areas, it is important to screen the entire population and implement control/eradication of SRLV infections in conjunction with each conservation program.

The objective of the current study was to evaluate the effects of soybean oil inclusion in diets on feeding behaviour, digestibility, performance, and blood metabolites of feedlot lambs. Forty non-castrated Santa Inês lambs with a mean age of 5 months and initial body weight of 34.88 ± 3.19 kg were used in a 40-day feeding trial. The lambs were distributed in five experimental diets with the inclusion of increasing soybean oil (SO) levels: 0, 30, 60, 90, and 120 g/kg DM. The SO inclusion promoted a linear reduction in DM intake (p < 0.001), crude protein (CP, p < 0.001), non-fibrous carbohydrates (NFC, p < 0.001), and total digestible nutrients (TDN, p = 0.004). There was an increasing quadratic effect on the intake of ether extract (EE; p = 0.002) and decreasing for neutral detergent fiber (p = 0.005). The soybean oil inclusion promoted the greater apparent digestibility of CP (p = 0.016), EE (p = 0.005), NDFom (p < 0.001), and TDN (p < 0.001); on the other hand, the apparent digestibility of NFC (p = 0.005) was decreased. The average daily gain decreased (p < 0.001) with SO inclusion. The SO inclusion increased feeding time (p = 0.004), reduced the efficiency of DM rumination (p = 0.001), and reduced the concentration of blood N-ureic (p < 0.001). Considering the productive parameters, SO can be included in diets and it is recommended that we include SO of up to 41 g/kg DM in diets for fattening lambs as the ideal maximum level. The strategy implemented to adapt lambs to increasing levels of high-fat diet mitigated the detrimental effects of lipids on the rumen, with high-density energy intake being the constraining factor on performance.

Given the significance of goat farming in Greece, our study aimed to explore biosecurity conditions on goat farms and refers to an investigation performed on 119 farms countrywide in Greece. The objective of the present work was to evaluate and report on potential associations between the socio-demographic characteristics of goat farmers and practices that play a role in biosecurity on farms. Data were obtained during a large cross-sectional study performed across Greece in 119 goat herds. The socio-demographic characteristics of farmers, as well as 16 variables related to biosecurity, were recorded; these were practices or events that could potentially enhance or compromise biosecurity on farms. A score based on all the biosecurity-related variables was devised by assigning a '1' or '-1' mark for each variable recorded on a farm that could enhance or compromise, respectively, biosecurity. Among variables potentially enhancing biosecurity on farms, the application of disinfections on the farm and the maintenance of isolation for sick animals were practiced more frequently; among variables potentially compromising biosecurity on farms, the presence of hunters in the area around the farm and grazing practices for animals were recorded more frequently. The median overall score for biosecurity-related practices for all farms in the study was 1 (interquartile range: 2.5; minimum: -4; maximum: 5). Among socio-demographic characteristics, the presence of working staff on a farm was associated with five individual biosecurity-related practices and the length of farming experience with three individual biosecurity-related practices. In the multivariable analysis, female farmers (p = 0.007) and the presence of working staff on the farm (p = 0.025) emerged as the two significant socio-demographic characteristics with an association with the overall biosecurity level on farms. This knowledge can be useful when developing biosecurity programs on goat farms. Recognition of locally applied farm-level practices enhancing biosecurity could form a basis for farmers to apply more rigorous and effective relevant plans.

ABSTRACT

The inclusion of spent hemp biomass (SHB), an extracted byproduct from industrial cannabidiol (CBD) production, in the diets of dairy cows and lambs appears to be safe with minor effects on the metabolism, including a decrease in circulating cholesterol and increase bilirubinemia, both associated with liver metabolism. Those effects could be consequence of the presence of cannabinoids, particularly Δ9-tetrahydrocannabinol (THC) and CBD in the SHB. This study aimed to study the transcriptional profile of the liver of dairy cows and lambs fed SHB. Dairy cows received SHB or alfalfa pellet for four weeks of intervention (IP) and four weeks of withdrawal periods (WP). Finishing lambs were fed a control diet (CON), 10% (LH2), or 20% (HH2) SHB for 2 months or 1 month followed by 1-month SHB withdrawal (LH1 and HH1, respectively). RNA sequencing was performed, and the mRNA was annotated using the latest reference genomes. The RNAseq data were filtered, normalized for library size and composition, and statistically analyzed by DESeq2. The bioinformatic analysis was performed by using DAVID, Gene Set Enrichment Analysis (GSEA), and the Dynamic Impact Approach. Using a 0.2 FDR cut-off, we identified only ≤24 differentially expressed genes (DEG) in the liver by feeding SHB in dairy cows and a larger number of DEGs in lambs (from 71 in HH1 vs. CON to 552 in LH1 vs. CON). The KEGG analysis demonstrated that feeding SHB in dairy cows and lambs had relatively minor to moderate metabolic alterations in dairy cows and lambs mainly associated with amino acids and lipid metabolism whereas cholesterol synthesis was overall activated in lambs. GSEA identified activation of the PPAR signaling pathway only in dairy cows. We found an opposite effect on activation of metabolism of drug and xenobiotics by cytochrome P450 enzymes in dairy cows and lambs receiving less SHB but an inhibition in HH2 lambs. Immune system-related pathways were inhibited by feeding SHB in lambs, but the impact was minor. Cumulatively, inclusion of SHB containing cannabinoids in dairy and lambs demonstrate very little effects on the alteration of transcriptomic profile of the liver.

ABSTRACT

Mycotoxins, unavoidable contaminants in feed and feed ingredients, have the potential to influence the incidence and severity of various diseases upon ingestion. Sheep coccidiosis is an enteric disease caused by protozoa of Eimeria spp. However, the extent to which the presence of aflatoxin b1 (AFB1) synergistically exacerbates damage to intestinal health in lambs with Eimeria remains unclear. 50-day-old female lambs were randomly assigned to a 2 × 2 factorial arrangement of treatments for 15 days to assess the impact of AFB1 exposure on lambs with or without Eimeria (E.) ovinoidalis infection. Our findings reveal that AFB1 synergistically intensifies damage to intestinal health in lambs challenged by E. ovinoidalis. This is evidenced by disruptions to the intestinal microbiota and reductions in the production of short-chain fatty acids. AFB1 further aggravates damage to the cecal mechanical barrier. Additionally, AFB1 contributes to the entry of lipopolysaccharide into the bloodstream, activating the inflammatory response. Interestingly, AFB1 exposure history results in an early peak of oocyst excretion and a decreased number of oocyst excretion in E. ovinoidalis infected lambs. This may be closely linked to the destruction of the intestinal epithelial cell structure and its apoptosis, as indicated by a decreased ratio of Bcl-2 to Bax and increased caspase-3 levels. Mechanistically, proteomics analysis identified mitochondrial dysfunction (inhibition of the oxidative phosphorylation pathway) as the primary factor intensifying intestinal epithelial cell destruction caused by coccidia, exacerbated by AFB1 through the inhibiting the conversion of NADH to NAD+ in the cecum of lambs via down-regulation of the PGC-1α/NRF1/TFAM pathway. Overall, these results offer novel insights into the AFB1 complicity in accelerating intestinal damage caused by E. ovinoidalis in lambs. Targeting the mitochondrial oxidative phosphorylation pathway of the intestine may represent a new therapeutic strategy against the detrimental effects of mycotoxin and coccidia.

This study investigates the dynamic changes in milk nutritional composition and microbial communities in Tibetan sheep and goats during the first 56 days of lactation. Milk samples were systematically collected at five time points (D0, D7, D14, D28, D56) post-delivery. In Tibetan sheep, milk fat, protein, and casein contents were highest on D0, gradually decreased, and stabilized after D14, while lactose and galactose levels showed the opposite trend. Goat milk exhibited similar initial peaks, with significant changes particularly between D0, D7, D14, and D56. 16S rRNA gene sequencing revealed increasing microbial diversity in both species over the lactation period. Principal coordinates analysis identified distinct microbial clusters corresponding to early (D0-D7), transitional (D14-D28), and mature (D56) stages. Core phyla, including Proteobacteria, Firmicutes, Bacteroidetes, and Actinobacteria, dominated the milk microbiota, with significant temporal shifts. Core microbes like Lactobacillus, Leuconostoc, and Streptococcus were common in both species, with species-specific taxa observed (e.g., Pediococcus in sheep, Shewanella in goats). Furthermore, we observed a highly shared core microbiota in sheep and goat milk, including Lactobacillus, Leuconostoc, and Streptococcus. Spearman correlation analysis highlighted significant relationships between specific microbial genera and milk nutrients. For instance, Lactobacillus positively correlated with total solids, non-fat milk solids, protein, and casein, while Mannheimia negatively correlated with protein content. This study underscores the complex interplay between milk composition and microbial dynamics in Tibetan sheep and goats, informing strategies for livestock management and nutritional enhancement. ABSTRACT

This study delves into the dynamic interaction between various fungal strains, substrates, and treatment durations to optimize the nutritional value of these by-products. Six fungi, including Penicillium chrysogenum, Fusarium sp., Fusarium oxysporum, Fusarium solani, Penicillium crustosum, and Cosmospora viridescens, were evaluated across three substrates: wheat straw (WS), cedar sawdust (CW), and olive pomace (OP) over treatment periods of 4, 8, and 12 weeks. The study discerned profound impacts of these fungi across multiple parameters, including cellulose variation (C.var), lignin variation (L.var), and in vitro true digestibility variation (IVTD.var). Our results demonstrated that the various fungi had a significant effect on all parameters (p < .001). Noteworthy, F. oxysporum and F. solani emerged as exemplars, displaying notable lignin degradation, cellulose liberation, and IVTD enhancement. Importantly, P. crustosum demonstrated substantial cellulose degradation, exhibiting optimal efficacy in just 4 weeks for all substrates. Notably, F. sp. excelled, yielding favorable results when treating WS. P. chrysogenum achieved optimal outcomes with 8-week treatment for WS. Both Fusarium sp. and P. chrysogenum exhibited slight cellulose release, with remarkable reduction of WS lignin compared to other substrates. Especially, WS and OP displayed superior digestibility enhancements relative to CW. It should be noted that the treatment duration further shaped these outcomes, as prolonged treatment (12 weeks) fostered greater benefits in lignin degradation and digestibility, albeit with concomitant cellulose degradation. These findings underscore the intricate balance between fungal strains, substrates, and treatment durations in optimizing the nutritional value of lignocellulosic agro-industrial by-products. The outcomes of this study lead to the enhancement in the overall value of by-products, promoting sustainable livestock feed and advancing agricultural sustainability.

Animals representing a wide range of taxonomic groups are known to select specific food combinations to achieve a nutritionally balanced diet. The nutrient balancing hypothesis suggests that, when given the opportunity, animals select foods to achieve a particular target nutrient balance, and that balancing occurs between meals and between days. For wild ruminants who inhabit landscapes dominated by human land use, nutritionally imbalanced diets can result from ingesting agricultural crops rich in starch and sugar (nonstructural carbohydrates [NCs]), which can be provided to them by people as supplementary feeds. Here, we test the nutrient balancing hypothesis by assessing potential effects that the ingestion of such crops by Alces alces (moose) may have on forage intake. We predicted that moose compensate for an imbalanced intake of excess NC by selecting tree forage with macro-nutritional content better suited for their rumen microbiome during wintertime. We applied DNA metabarcoding to identify plants in fecal and rumen content from the same moose during winter in Sweden. We found that the concentration of NC-rich crops in feces predicted the presence of Picea abies (Norway spruce) in rumen samples. The finding is consistent with the prediction that moose use tree forage as a nutritionally complementary resource to balance their intake of NC-rich foods, and that they ingested P. abies in particular (normally a forage rarely eaten by moose) because it was the most readily available tree. Our finding sheds new light on the foraging behavior of a model species in herbivore ecology, and on how habitat alterations by humans may change the behavior of wildlife.

ABSTRACT

This study evaluated the effects of supplementation of rumen-protected methionine (RPM) on body thermoregulation and conception rate of Nelore cows exposed to high temperature-humidity index (THI). On -31 days before the artificial insemination protocol, 562 lactating, multiparous cows were assigned to receive (MG) or not (CG) RPM supplementation (3 g/cow mixed into 100 g of mineral supplement). Both groups remained in tropical pastures and received supplementation for 77 days. A subset of cows (n = 142) remained with an intravaginal thermometer collecting intravaginal temperature (IT). The respective minimum, average, and maximum environmental THI were 72.8, 78.0, and 83.3. Effects of treatment × hour of the day were detected (P < 0.0001) for IT. From 1330 to 1730 h and 1830 to 1900 h, IT was higher (P < 0.05) for CG versus MG cows when exposed to moderate and high THI. The supplementation with RPM did not affect conception rate (CG = 64.4% vs. MG = 58.2%; P > 0.05). In conclusion, 3 g of RPM supplementation lowered internal body temperature and possibly altered critical THI threshold in Nelore cows with no impact on reproduction.

ABSTRACT

The research on automatic monitoring methods for greenhouse gases and hazardous gas emissions is currently a focal point in the fields of environmental science and climatology. Until 2023, the amount of greenhouse gases emitted by the livestock sector accounts for about 11-17% of total global emissions, with enteric fermentation in ruminants being the main source of the gases. With the escalating problem of global climate change, accurate and effective monitoring of gas emissions has become a top priority. Presently, the determination of gas emission indices relies on specialized instrumentation such as breathing chambers, greenfeed systems, methane laser detectors, etc., each characterized by distinct principles, applicability, and accuracy levels. This paper first explains the mechanisms and effects of gas production by ruminant production systems, focusing on the monitoring methods, principles, advantages, and disadvantages of monitoring gas concentrations, and a summary of existing methods reveals their shortcomings, such as limited applicability, low accuracy, and high cost. In response to the current challenges in the field of equipment for monitoring greenhouse and hazardous gas emissions from ruminant production systems, this paper outlines future perspectives with the aim of developing more efficient, user-friendly, and cost-effective monitoring instruments.

ABSTRACT

Behaviour is often the fundamental driver of disease transmission, where behaviours of individuals can be seen to scale up to epidemiological patterns seen at the population level. Here we focus on animal behaviour, and its role in parasite transmission to track its knock-on consequences for parasitism, production and pollution. Livestock face a nutrition versus parasitism trade-off in grazing environments where faeces creates both a nutritional benefit, fertilizing the surrounding sward, but also a parasite risk from infective nematode larvae contaminating the sward. The grazing decisions of ruminants depend on the perceived costs and benefits of the trade-off, which depend on the variations in both environmental (e.g., amounts of faeces) and animal factors (e.g., physiological state). Such grazing decisions determine the intake of both nutrients and parasites, affecting livestock growth rates and production efficiency. This impacts on the greenhouse gas costs of ruminant livestock production via two main mechanisms: (1) slower growth results in longer durations on-farm and (2) parasitised animals produce more methane per unit food intake. However, the sensitivity of behaviour to host parasite state offers opportunities for early detection of parasitism and control. Remote monitoring technology such as accelerometers can detect parasite-induced sickness behaviours soon after exposure, before impacts on growth, and thus may be used for targeting individuals for early treatment. We conclude that livestock host x parasite interactions are at the centre of the global challenges of food security and climate change, and that understanding livestock behaviour can contribute to solving both.

ABSTRACT

To better understand mechanisms of serotonin- (5-HT) mediated vasorelaxation, isolated lateral saphenous veins from cattle were assessed for vasoactivity using myography in response to increasing concentrations of 5-HT or selective 5-HT receptor agonists. Vessels were pre-contracted with 1 × 10-4 M phenylephrine and exposed to increasing concentrations of 5-HT or 5-HT receptor agonists that were selective for 5-HT1B, 5-HT2B, 5-HT4, and 5-HT7. Vasoactive response data were normalized as a percentage of the maximum contractile response induced by the phenylephrine pre-contraction. At 1 × 10-7 M 5-HT, a relaxation was observed with an 88.7% decrease (p < 0.01) from the phenylephrine maximum. At 1 × 10-4 M 5-HT, a contraction was observed with a 165% increase (p < 0.01) from the phenylephrine maximum. Increasing concentrations of agonists selective for 5-HT2B, 5-HT4, or 5-HT7 resulted in a 27%, 92%, or 44% (p < 0.01) decrease from the phenylephrine maximum, respectively. Of these 5-HT receptor agonists, the selective 5-HT4 receptor agonist resulted in the greatest potency (-log EC50) value (6.30) compared with 5-HT2B and 5-HT7 receptor agonists (4.21 and 4.66, respectively). To confirm the involvement of 5-HT4 in 5-HT-mediated vasorelaxation, blood vessels were exposed to either DMSO (solvent control) or a selective 5-HT4 antagonist (1 × 10-5 M) for 5-min prior to the phenylephrine pre-contraction and 5-HT additions. Antagonism of the 5-HT4 receptor attenuated the vasorelaxation caused by 5-HT. Approximately 94% of the vasorelaxation occurring in response to 5-HT could be accounted for through 5-HT4, providing strong evidence that 5-HT-mediated vasorelaxation occurs through 5-HT4 activation in bovine peripheral vasculature.

ABSTRACT

Understanding the transfer of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) in farm animals is essential for ensuring food safety, but such information for suckler ewes (Ovis aries) has been lacking. This work quantifies the accumulation, tissue distribution, and depuration kinetics of PCDD/Fs in these animals. Six suckler ewes (EXP group) were exposed to PCDD/Fs through contaminated hay (2.3-12.7 ng toxic-equivalent kg-1 dry matter) and then allowed to depurate by switching to noncontaminated hay from 29 days of lactation. Four control ewes were fed continuously with noncontaminated hay. At different time points covering depuration, weaning and slaughter, PCDD/F analysis of milk (three time points), blood and sternal adipose tissue (five time points), Longissimus thoracis muscle, liver, and empty body homogenate at slaughter (188 days of depuration) was performed. A relevant PCDD/F bioaccumulation was observed from oral intake in milk and adipose tissue (biotransfer factors of 1.24 and 1.06 day kg-1 lipids for the sum toxic-equivalent, respectively) in the EXP ewes, especially for penta- and hexa-chlorinated congeners. The EXP ewes' adipose tissue started at 10-fold the EU maximum level (ML) and showed depuration below the ML after 130 days. Specific PCDD/F accumulation in the ewe liver was observed, especially for dibenzofurans. These toxicokinetic data can inform recommendations to ensure the chemical safety of sheep food products.

ABSTRACT

Visna/Maedi virus (VMV) is lentiviral disease of sheep responsible for severe production losses. Multiple genomic regions associated with infection were reported indicating genetic complexity. In this study, a combined genome-wide approach using a high-density SNP array has been performed, comparing VMV-infected (n = 78) and non-infected (n = 66) individuals of the Valle del Belice breed. The serological tests showed a seroprevalence of 26%. The comparison among results from different approaches (GWAS, Fisher's exact test and the FST analysis) revealed two association signals: on OAR03 close to the GRIN2B gene and on OAR05 close to the TMEM232 gene. To the best of our knowledge, there has been no previous association between these genes and lentiviral infection in any species. The GRIN2B gene plays a role in pain response, synaptic transmission, and receptor clustering, while TMEM232 is involved in the development of immune-related disorders. The results highlighted new aspects of the genetic complexity related to the resistance/susceptibility to VMV in sheep, confirming that studies on different breeds can lead to different results. The ideal approach for validation of the markers identified in our study is to use samples from a population independent from the discovery population with the same phenotype used in the discovery stage.

ABSTRACT

Non-aureus staphylococci and mammaliicocci (NASM) are the microorganisms most frequently isolated from milk. Given their numerosity and complexity, MALDI-TOF MS is one of the preferred species identification approaches. Nevertheless, reference mass spectra for the novel species Staphylococcus borealis were included only recently in the Bruker Biotyper System (MBT) library, and other species of veterinary interest such as S. rostri are still absent. This work provides an updated picture of the NASM species found in milk, gained by retrospectively analyzing the data relating to 21,864 milk samples, of which 6,278 from clinical mastitis (CM), 4,039 from subclinical mastitis (SCM), and 11,547 from herd survey (HS), with a spectrum library including both species. As a result, S. borealis was the second most frequently isolated NASM (17.07%) after S. chromogenes (39.38%) in all sample types, with a slightly higher percentage in CM (21.84%), followed by SCM (17.65%), and HS (14.38%). S. rostri was also present in all sample types (3.34%), reaching 8.43% of all NASM in SCM and showing a significant association (p < 0.01) with this condition. Based on our findings, the presence of S. borealis and S. rostri in milk and their potential association with mastitis might have been overlooked, possibly due to the difficulties in differentiating these species from other closely related NASM. Our results indicate that S. borealis could be a more frequent contributor to bovine udder infections than previously thought and that S. rostri should also not be underestimated considering its significant association with SCM.
