ABSTRACT

Within a study focused on Sinapis pubescens subsp. pubescens wild from Sicily (Italy), an edible species still unexplored, our earlier published work has demonstrated good in vitro antioxidant properties for the flower and leaf hydroalcoholic extracts, exhibiting quite different qualitative-quantitative phenolic profiles. Herein, further research was designed to elucidate the role played by phenolic compounds in the different antioxidant mechanisms highlighted for the extracts. To achieve this goal, the crude extracts were subjected to liquid-liquid partitioning with solvents of increasing polarity; then, the fractions were investigated for their antioxidant properties using different in vitro assays. For both flowers and leaves, the ethyl acetate fractions exhibited the best activity in DPPH and reducing power assays, followed by n-butanol. The total phenolic content determination indicated these fractions as the phenolic-rich ones, which were characterized by HPLC-PDA/ESI-MS analysis. Conversely, the phenolic-rich fractions did not show any chelating activity, which was highlighted for the more hydrophobic ones.

ABSTRACT

Recently, our research team has started a study on Brassica fruticulosa subsp. fruticulosa, an edible plant traditionally used to treat various ailments, little investigated to date. Good in vitro antioxidant properties were highlighted for the leaf hydroalcoholic extract, with the secondary higher than the primary ones. In continuation of the ongoing research, this work was designed to elucidate the antioxidant properties of the phenolic compounds contained in the extract. For this purpose, a phenolic-rich ethyl acetate fraction (Bff-EAF) was obtained from the crude extract by liquid-liquid extraction. The phenolic composition was characterized by HPLC-PDA/ESI-MS analysis and the antioxidant potential was investigated by different in vitro methods. Furthermore, the cytotoxic properties were evaluated by MTT, LDH and ROS determinations on human colorectal epithelial adenocarcinoma cells (CaCo-2) and human normal fibroblasts (HFF-1). Twenty phenolic compounds (flavonoid and phenolic acid derivatives) were identified in Bff-EAF. The fraction exhibited good radical scavenging activity in the DPPH test (IC50 = 0.81 ± 0.02 mg/mL), and moderate reducing power (ASE/mL = 13.10 ± 0.94) and chelating properties (IC50 = 2.27 ± 0.18 mg/mL), contrary to what previously observed for the crude extract. Bff-EAF reduced in a dose-dependent manner CaCo-2 cell proliferation after 72 h of treatment. This effect was accompanied by the destabilization of the cellular redox state due to the antioxidant and pro-oxidant activities displayed by the fraction at lower and higher concentrations. No cytotoxic effect was observed on HFF-1 fibroblasts, used as control cell line.

ABSTRACT

In the past few years, evidence has supported the role of plants as a valuable tool for the development of promising therapeutic support options for many diseases, including cancer. We recently discovered that the edible wild plant Crithmum maritimum L. effectively inhibits the growth of hepatocellular carcinoma (HCC) cells and we provide insights into the biological mechanisms involved. Here, we aimed to characterize the effect of ethyl acetate extract of Crithmum maritimum on the bioenergetic phenotype of HCC cells and if this is associated with the anti-tumour effect we previously described. Results show that Crithmum maritimum significantly increases cellular respiration and reduces lactic fermentation in HCC cells, and that this reduction of the fermentative glycolytic phenotype is linked to inhibition of HCC growth. These data provide new preclinical evidence supporting the role of Crithmum maritimum L. as a nutraceutical option to expand the therapeutic opportunities in the management of HCC.

ABSTRACT

Recipes on the composition of the "salad of the monks" (Capuchin monks) have been reported in Italy since the 17th century. Different wild edible plants were highly regarded as an important ingredient of this mixed salad. Among these, some species played a key role for both their taste and nutritional properties: Plantago coronopus L. (PC), Rumex acetosa L., Cichorium intybus L., and Artemisia dracunculus L. In the present study, the micromorphological and phytochemical features as well as the antioxidant and anti-inflammatory properties of extracts of these fresh and blanched leaves, were investigated. The extracts obtained by blanched leaves, according to the traditionally used cooking method, showed the highest content of bioactive compounds (total phenols 1202.31-10,751.88 mg GAE/100 g DW; flavonoids 2921.38-61,141.83 mg QE/100 g DW; flavanols 17.47-685.52 mg CE/100 g DW; proanthocyanidins 2.83-16.33 mg CyE/100 g DW; total chlorophyll 0.84-1.09 mg/g FW; carbohydrates 0.14-1.92 g/100 g FW) and possess the most marked antioxidant (IC50 0.30-425.20 µg/mL) and anti-inflammatory activity (IC50 240.20-970.02 µg/mL). Considering this, our results indicate that increased consumption of the investigated plants, in particular of PC, raw or cooked briefly, could provide a healthy food source in the modern diet by the recovery and enhancement of ancient ingredients.

Nowadays, a growing body of evidence supports the view that plants offer an extraordinary opportunity to discover and develop new promising therapeutic strategies for many diseases, including cancer. Here we tested the anticancer action against Hepatocellular carcinoma (HCC) of extracts obtained from two plants harvested in Apulia, namely Brassica oleracea L. and Crithmum maritimum L. B. oleracea was grown in biodynamical agriculture without any agrochemical input, instead C. maritimum was collected on Apulian coasts and is still commonly eaten in Apulia. HCC, one of the most frequent tumors worldwide, is estimated to become the third leading cause of cancer-related deaths in Western Countries by 2030. The approved synthetic drugs for the treatment of HCC are currently inadequate in terms of therapeutic results and tolerability. Hence, aim of the present study was to test the anticancer action against HCC of extracts obtained from Brassica oleracea L. and Crithmum maritimum L. We preliminary prepared extracts from both plants using four solvents with different polarity: hexane, ethyl acetate, methanol and ethanol. Then, we tested the effect of the different fractions in inhibiting HCC cell growth. Finally, we characterized the mechanism of action of the most effective fraction. We found that ethyl acetate fractions from both plants were the most effective in inhibiting HCC growth. In particular, we demonstrated that these fractions effectively reduce HCC growth by exerting, on one hand, a cytostatic effect through their action on the cell cycle, and on the other hand by triggering apoptosis and necrosis. Our findings support the notion that ethyl acetate fractions from Apulian B. oleracea and C. maritimum can be in perspective considered as promising tools to expand the opportunities to identify new and not toxic anticancer therapeutic approaches for HCC. Further pharmacological investigations will shed light on how this could be effectively achieved. ABSTRACT

Edible wild plants (EWP) continue to be an important food source for indigenous communities. A survey was conducted to identify the consumption and management of EWP known as quelites in the Zongolica region of Mexico. 15 species of quelites are consumed mainly during the rainy season, whose local name is associated with the plant's shape, smell and flavor. Changes in food patterns and land use threaten the permanence and consumption of these species. Indigenous and local knowledge is crucial for the use, management and conservation of this group of plants, whose consumption can be leveraged to address malnutrition and unhealthy food use.

ABSTRACT

Edible and medicinal weeds being commercialised for the first time in an urban horticulturalists’ fair were studied with an ethnobotanical approach in Bariloche (Patagonia, Argentina). The effects of three community workshops carried out previously were analysed, paying particular attention to the cosmovision, attitudes and behaviour of the horticulturalists regarding these plants. The methodology also included 25 open interviews, participative observation and a field surveys carried out together with the informants. A total richness of 29 species of edible and medicinal use was registered, the majority being of European origin. Before the workshops, the ideas and appreciation of the use of wild plants as food were generally negative, being associated with poverty. In contrast, their medicinal uses were more readily accepted. Following the workshops, however, attitudes and behaviour reflected a more positive general assessment, leading to their commercialisation and a marked tendency towards innovation in knowledge that could be combined with the participants’ traditional knowledge. The importance of this kind of applied ethnobotany project should be noted, and its contribution to the development of local communities.


Desde una aproximación etnobotánica, se estudiaron las malezas comestibles y medicinales comercializadas por primera vez en una feria urbana de horticultores de Bariloche (Patagonia, Argentina), analizando los alcances de tres talleres comunitarios llevados a cabo previamente, y teniendo particular interés en las concepciones, actitudes y comportamientos acerca de dichas plantas entre los horticultores. La metodología incluyó también 25 entrevistas libres, observación participante y relevamientos a campo junto a informantes. Se registró una riqueza de total de 29 especies de uso medicinal y comestible, la mayoría de origen europeo. Antes de los talleres, las concepciones y valoraciones acerca de la utilización de las malezas como alimento eran en general negativas, asociándolas con la pobreza. En cambio, sus usos medicinales fueron mejor aceptados. Sin embargo, luego de los talleres, las actitudes y los comportamientos reflejaron valoraciones generales más positivas resultando en la comercialización de las mismas y una marcada tendencia hacia la innovación de saberes que podían hibridizarse con los saberes tradicionales de las personas. Se rescata la importancia de este tipo de proyectos de etnobotánica aplicada y su aporte al desarrollo de las comunidades locales.
