ABSTRACT

Passive acoustic monitoring (PAM) is a promising method for biodiversity assessment, which allows for longer and less intrusive sampling when compared to traditional methods (e.g., collecting specimens), by using sound recordings as the primary data source. Insects have great potential as models for the study and monitoring of acoustic assemblages due to their sensitivity to environmental changes. Nevertheless, ecoacoustic studies focused on insects are still scarce when compared to more charismatic groups. Insects' acoustic activity patterns respond to environmental factors, like temperature, moonlight, and precipitation, but community acoustic perspectives have been barely explored. Here, we provide an example of the usefulness of PAM to track temporal patterns of acoustic activity for a nocturnal assemblage of insects (Orthoptera). We integrate satellite remote sensing and astronomically measured environmental factors at a local scale in an Andean Forest of Colombia and evaluate the acoustic response of orthopterans through automated model detections of their songs for nine weeks (March and April of 2020). We describe the acoustic frequency range and diel period for the calling song of each representative species. Three species overlapped in frequency and diel acoustics but inhabit different strata: canopy, understory, and ground surface level. Based on the acoustic frequency and activity, we identified three trends: (i) both sampled cricket species call at lower frequency for shorter periods of time (dusk); (ii) all sampled katydid species call at higher frequency for longer time periods, including later hours at night; and (iii) the diel acoustic activity span window seems to increase proportionally with dominant acoustic frequency, but further research is required. We also identified a dusk chorus in which all the species sing at the same time. To quantify the acoustic response to environmental factors, we calculated a beta regression with the singing activity as a response variable and moon phase, surface temperature and daily precipitation as explanatory variables. The response to the moon phase was significant for the katydids but not for the crickets, possibly due to differences in diel activity periods. Crickets are active during dusk, thus the effects of moonlight on acoustic activity are negligible. The response to precipitation was significant for the two crickets and not for the katydids, possibly because of higher likelihood of rain interrupting crickets' shorter diel activity period. Our study shows how the local survey of orthopteran acoustic assemblages, with a species taxonomic resolution coupled with remote-sensing environmental measurements can reveal responses to environmental factors. In addition, we demonstrate how satellite data might prove to be a useful alternative source of environmental data for community studies with geographical, financial, or other constraints.

ABSTRACT

Cell penetrating peptides (CPPs) are molecules capable of passing through biological membranes. This capacity has been used to deliver impermeable molecules into cells, such as drugs and DNA probes, among others. However, the internalization of these peptides lacks specificity: CPPs internalize indistinctly on different cell types. Two major approaches have been described to address this problem: (i) targeting, in which a receptor-recognizing sequence is added to a CPP, and (ii) activation, where a non-active form of the CPP is activated once it interacts with cell target components. These strategies result in multifunctional peptides (i.e., penetrate and target recognition) that increase the CPP's length, the cost of synthesis and the likelihood to be degraded or become antigenic. In this work we describe the use of machine-learning methods to design short selective CPP; the reduction in size is accomplished by embedding two or more activities within a single CPP domain, hence we referred to these as moonlighting CPPs. We provide experimental evidence that these designed moonlighting peptides penetrate selectively in targeted cells and discuss areas of opportunity to improve in the design of these peptides.

ABSTRACT

Candida genus comprises several species that can be found in the oral cavity and the gastrointestinal and genitourinary tracts of healthy individuals. Under certain conditions, however, they behave as opportunistic pathogens that colonize these tissues, most frequently when the immune system is compromised by a disease or under certain medical treatments. To colonize the human host, these organisms require to express cell wall proteins (CWP) that allowed them to adhere and adapt to the reactive oxygen (ROS) and nitrogen (RNS) species produced in the macrophage during the respiratory burst. The aim of this study was to determine how four Candida species respond to the oxidative stress imposed by cumene hydroperoxide (CHP). To this purpose, C. albicans, C. glabrata, C. krusei and C. parapsilosis were exposed to this oxidant which is known to generate ROS in the membrane phospholipids. Accordingly, both mock and CHP-exposed cells were used to extract and analyze CWP and also to measure catalase activity and the levels of protein carbonylation. Results indicated that all four species express different CWP to neutralize ROS. Most relevant among these proteins were the glycolytic enzymes enolase and glyceraldehyde-3-phosphate dehydrogenase, known as moonlight proteins because in addition to participate in glycolysis they play an important role in the cell response to ROS. In addition, a thiol-specific antioxidant enzyme (Tsa) was also found to counteract ROS.

ABSTRACT

In order for Candida species to adhere and colonize human host cells they must express cell wall proteins (CWP) and adapt to reactive oxygen species (ROS) generated by phagocytic cells of the human host during the respiratory burst. However, how these pathogens change the expression of CWP in response to oxidative stress (OSR) is not known. Here, fifteen moonlight-like CWP were identified that expressed differentially in four species of Candida after they were exposed to H2O2 or menadione (O2(-)). These proteins included: (i) glycolytic enzymes, such as glyceraldehyde-3-phosphate dehydrogenase (Gapdh), fructose-bisphosphate aldolase (Fba1), phosphoglycerate mutase (Gpm1), phosphoglycerate kinase (Pgk), pyruvate kinase (Pk) and enolase (Eno1); (ii) the heat shock proteins Ssb1 and Ssa2; (iii) OSR proteins such as peroxyredoxin (Tsa1), the stress protein Ddr48 (Ddr48) and glutathione reductase (Glr1); (iv) other metabolic enzymes such as ketol-acid reductoisomerase (Ilv5) and pyruvate decarboxylase (Pdc1); and (v) other proteins such as elongation factor 1-beta (Efb1) and the 14-3-3 protein homolog. RT-PCR revealed that transcription of the genes coding for some of the identified CWP are differentially regulated. To our knowledge this is the first report showing that moonlight-like CWP are the first line of defense of Candida against ROS, and that they are differentially regulated in each of these pathogens.

The occurrence of coat colour polymorphisms in populations may promote the ecological success of species by permitting a wider spectrum of use of different subsets of available resources. We conducted an analysis of temporal segregation by comparing night brightness with nocturnal activity of spotted and melanistic oncillas (Leopardus tigrinus). Melanistic oncillas were more active during bright nights and spotted oncillas and other species were more active during dark nights. Each colour morph occupied a temporal niche outside the confidence interval of the other colour morph, demonstrating the ecological significance of polymorphic colour patterns in this felid species.


A ocorrência de polimorfismo no padrão de pelagem de populações pode promover o sucesso ecológico das espécies por permitir um amplo espectro de uso de diferentes parcelas de recursos disponíveis. Nós testamos a existência de diferença na segregação temporal do gato-do-mato-pequeno (Leopardus tigrinus), comparando a luminosidade durante períodos de atividade noturna de indivíduos pintados e melânicos. Indivíduos melânicos de gato-do-mato-pequeno foram mais ativos durante noites claras e indivíduos pintados de gato-do-mato-pequeno e outras espécies foram mais ativas durante noites escuras. Cada forma de coloração ocupou um nicho temporal fora do intervalo de confiança do outro, demonstrando a significância ecológica dos padrões de polimorfismo de colorações nesta espécie de felino.

ABSTRACT

The nightly activity patterns of Noctilio leporinus, a piscivorous neotropical bat, were documented by visual observation over one lunar cycle at a large dock in Golfito, Costa Rica. The activity patterns were compared to the intensity of the moonlight, and a strong correlation between darkness and activity was observed (n = 776, r2 = 0.96). This correlation has been observed in many species of mammals and may indicate adaptation by potential prey species to avoid predation by visually oriented nighttime predators and/or an adaptation by predators to maximize hunting success.


Documenté los patrones de actividad nocturna de Noctilio leporinus, murciélago piscívoro neotropical, mediante observaciones visuales durante un ciclo lunar en un muelle de Golfito, Costa Rica. Considerando la intensidad de la luz lunar, hubo una fuerte correlación entre la oscuridad y la actividad (n = 776, r2 = 0.96). Esta correlación ha sido observada en muchas especies de mamíferos y puede indicar una adaptación de presas potenciales para evadir los depredadores nocturnos y/o una adaptación de los depredadores para maximizar el éxito de cacería.
