ABSTRACT

Apresentamos uma explicação alternativa para a noção de drogadição, refletindo sobre os motivos que levam uma pessoa a perseverar no consumo de drogas, mesmo sabendo dos malefícios decorrentes. Referenciado pelas obras de Humberto Maturana e pela teoria dos afetos na Ética de Spinoza, assumimos o pressuposto de que os seres humanos são movidos por paixões e formulamos o circuito dos afetos da drogadição. Identificamos nesse circuito três complexos afetivos: complexo singular pessoal, complexo de alívio/oblívio e complexo psicossocial, que formam uma dinâmica espiral de afetos tristes. Tais afetos compelem o indivíduo a continuar nessa dinâmica, e o papel das drogas passa a ser acessório. Cada complexo afetivo é caracterizado por afetos de origens distintas e a compreensão da particularidade e da especificidade de cada um organiza a ação de cuidado correspondente, a fim de interromper a dinâmica espiral do circuito. Presentamos una explicación alternativa para la noción de drogadicción, reflexionando sobre los motivos que llevan a una persona a perseverar en el consumo de drogas, incluso sabiendo de los perjuicios que causan. Referenciado por las obras de Humberto Maturana y por la teoría de los afectos en la Ética de Spinoza, asumimos la presuposición de que los seres humanos son movidos por pasiones y formulamos el circuito de los afectos de la drogadicción. En ese circuito identificamos tres complejos afectivos: complejo singular personal, complejo de alivio/olvido y complejo psicosocial que forman una dinámica espiral de afectos tristes. Tales afectos compelen al individuo a continuar en esa dinámica y el papel de las drogas pasa a ser accesorio. Cada complejo afectivo se caracteriza por afectos de orígenes distintos y la comprensión de la particularidad y de la especificidad de cada uno organiza la acción de cuidado correspondiente, de forma que interrumpa la dinámica espiral del circuito. We present an alternative explanation for drug addiction, reflecting on the reasons that lead a person to persevere in drug consumption despite knowing the harm that this consumption brings. Based on the works by Humberto Maturana and on the theory of affects in Spinoza's Ethics, we assume that human beings are moved by passions and formulate the circuit of affects of drug addiction. In this circuit, we have identified three affective complexes: the singular personal complex, the relief/oblivion complex, and the psychosocial complex, which form a spiral dynamics of sad affects. Such affects impel the individual to continue in this dynamics, and drugs play only an accessory role. Affects from different origins characterize each affective complex and understanding their particular and specific features guides the corresponding care action, in order to interrupt the spiral dynamics of the circuit. ABSTRACT

PURPOSE: Temozolomide (TMZ) has anti-tumor activity in patients with malignant glioma. Hyperbaric oxygen (HBO) may enhance the efficacy of certain therapies that are limited because of the hypoxic tumor microenvironment. We examined the combined effects of TMZ-HBO in a rat glioma model. METHODS: After stereotactic injection of C6/LacZ rat glioma cells into the Wistar rats brain, the rats were randomly assigned to three treatment groups [group 1, control treatment; group 2, TMZ alone; group 3, a combination of TMZ and HBO]. Rats were sacrificed 18 days after treatment, and number of intra-/peri-tumoral vessels, microendothelial proliferations, immunohistochemistry and necrotic area were evaluated. RESULTS: Tumoral tissue was stained only sparsely with GFAP. Temozolomide treatment was significantly decreased in tumor tissue intratumoral vessel number / total tumor area level. The level of Ki67 was significantly decreased in the tumor tissue of the group 3. Additionally, the total necrotic area / total tumor volume (%) was decreased significantly in tumor tissue of the group 3 rats compared to group1 and 2. CONCLUSION: The combination of hyperbaric oxygen with temozolomide produced an important reduction in glioma growth and effective approach to the treatment of glioblastoma.(AU)


OBJETIVO: A temozolomida (TMZ) tem atividade anti-tumoral em pacientes com glioma maligno. Oxigênio hiperbárico (HBO) pode aumentar a eficácia de terapias que são limitadas devido a um microambiente do tumor hipóxico. Foram examinados os efeitos combinados de TMZ-HBO em um modelo de glioma em rato. MÉTODOS: Após a injeção estereotáxica de células de glioma de rato C6/LacZ no cérebro de ratos Wistar, os ratos foram distribuídos aleatoriamente em três grupos de tratamento: Grupo 1: tratamento de controle. Grupo 2: TMZ sozinho. Grupo 3: uma combinação de TMZ e HBO. Os ratos foram sacrificados 18 dias após o tratamento. Foram avaliados o número de vasos intra-/peri-tumoral, proliferação microendotelial, imunohistoquímica e área necrótica . RESULTADOS: O tecido tumoral foi marcado apenas esparsamente com GFAP. O tratamento com temozolomida diminuiu significativamente o tecido intratumoral e a área total do tumor. O nível de Ki67 foi significativamente diminuído no tecido do tumor do grupo 3. Além disso, a superfície necrótica total / volume total do tumor (%) diminuiu significativamente no tecido do tumor do grupo 3 em comparação com grupo 1 e 2. CONCLUSÃO: A combinação de oxigênio hiperbárico com temozolomida produziu uma redução importante no crescimento do glioma podendo ser abordagem eficaz para o tratamento do glioblastoma.(AU)

ABSTRACT

Genetic variation at the EF-hand domain containing 2 gene (EFHC2) locus has been associated with fear recognition in Turner syndrome. The aim of this study was to examine whether EFHC2 variants are associated with non-syndromic anxiety-related traits [harm avoidance (HA) and behavioral inhibition (BI)] and with panic disorder (PD). Our sample comprised 127 PD patients and 132 controls without psychiatric disorder. We genotyped nine SNPs within the EFHC2 locus and used PLINK to perform association analyses. An intronic SNP (rs1562875) was associated with HA (permuted p=0.031) accounting alone for over 3% of variance in this trait. This same SNP was nominally, but not empirically, associated with BI (r(2)=0.022; nominal p=0.022) and PD (OR=2.64; nominal p=0.009). The same association was found in a subsample of only females. In sum, we observed evidence of association between a variant in EFHC2, a gene previously associated with the processing of fear and social threat, and HA. Larger studies are warranted to confirm this association.
