ABSTRACT

Background: Lung adenocarcinoma (LUAD) is a pulmonary malignant disease that poses a high risk of mortality and morbidity. Previous study indicated that ORC1 plays an oncogenic function. However, the precise regulatory function that ORC1 serves in the progression of LUAD is still not clearly known. Methods: Bioinformatics analyses were performed using TCGA and GEO datasets. The human LUAD cell line NCIH1355, NCIH1568 as well as BEAS-2B cell line (human normal lung epithelial cell) were utilized for in vitro study. LUAD cell proliferation were determined via CCK-8 assays and RT-qPCR for ki-67. The relation of ORC1 and SLC7A11 was detected by Western blot and qPCR with or without sh-RNA. The expression level ACSL4, the biomarker of ferroptosis, were detected using RT-qPCR. Results: ORC1 and SLC7A11 exhibit high expression levels in both LUAD patients and cell lines, and are strongly associated with poor prognosis. In vitro experiments demonstrate that ORC1 and SLC7A11 promote proliferation of LUAD cell lines while inhibiting gefitinib-induced ferroptosis. Additionally, the function of ORC1 in LUAD cells is dependent on SLC7A11. Conclusion: ORC1 promotes LUAD cell proliferation and inhibits ferroptosis in a SLC7A11-dependent manner. This implies that ORC1 could potentially serve as a useful diagnosis biomarker and treatment target.

ABSTRACT

The effect of plugging the puncture channel with a mixture of hemocoagulase injection on the complications of CT-guided percutaneous transthoracic need biopsy (PTNB) was discussed. The medical records of PTNB were retrospectively studied from June 2017 to May 2022. In the study, the puncture channel of 626 patients were blocked, while remain 681 patients' were not. The Mantel Haenszel method performed layered analysis and evaluated the correlation of adjusted confounding factors. The Odds Ratio and its 95% confidence interval were calculated using the Woof method. The incidence of high-level pulmonary hemorrhage was significantly reduced in patients with lesions ≤ 2 cm and different needle lengths. Patients with different pleural-needle tip angle and perineedle emphysema were blocked, and the incidence of pneumothorax and thoracic implants was significantly reduced. Through puncture channel plugging, the incidence of pulmonary hemorrhage, pneumothorax and thoracic catheterization of PTNB under CT guidance was reduced.

ABSTRACT

Purpose To assess the expression of genes that are relevant to pyroptosis and the relationship between these genes and prognosis in uterine corpus endometrial carcinoma (UCEC). Methods The research identifies 16 pyroptosis regulators with different expressions in normal endometrium and UCEC. In accordance with the differentially expressed genes (DEGs), the various kinds of UCEC are classified into two sub-types. With the help of the Cancer Genome Atlas (TCGA), the prognostic value of all pyroptosis-related genes for survival was assessed, and a multigene model has constructed accordingly. Ten genes were modeled by applying the minimum criteria for determining risk score selection (LASSO) Cox regression method. Meanwhile, by referring to the TCGA atlas, UCEC patients were divided into the high- and low-risk subgroups. The effects of the gene with significant differences on the proliferation of two cancer cells were also verified. Results The survival rate of UCEC cases with higher risk was higher than that with lower risk (P < 0.001). Through the median risk score of TCGA atlas, UCEC cases were ranked as patients with higher risk and patients with lower risk. The low risk has a significant relationship with the prolongation of overall survival (OS) (p = 0.001) in the low-risk subgroup. Moreover, the KEGG and gene ontology (GO) enrichment models indicated that among the patients in the high-risk subgroup, their immune-related genes were concentrated but with decreased immune status. Conclusion The apoptosis-related genes are crucial for the immunity of tumors and may forecast the prognosis of UCEC (AU)

ABSTRACT

The overall incidence of rectal gastrointestinal stromal tumor (RGIST) has risen, but it remains a rare disease. Furthermore, tumor rupture is associated with poor prognosis. The present study reported a rare case of RGIST with indolent biological behavior. The biological behavior of this RGIST was analyzed and its malignant potential was evaluated using a guideline-based risk stratification assessment. The patient was diagnosed with a rectal tumor at the Third Affiliated Hospital of Qiqihar Medical University (Qiqihar, China) in April 2020 and a partial resection biopsy was then performed. This resection counts as a rupture. The biopsy confirmed RGIST and the patient refused further examination and treatment due to economic concerns. However, the patient survives with no tumor progression and metastasis until now, May 2022. In conclusion, based on the present case, tumor rupture in indolent RGIST is not necessarily associated with poor outcome.
