ABSTRACT

Biocompatible, biodegradable, and injectable hydrogels are a novel and promising approach for bone regeneration. In this study, poly(caprolactone)-poly(ethylene glycol)-poly(caprolactone) (PCL-PEG-PCL), PCL-PEG-PCL-gelatin (Gel), PCL-PEG-PCL-Gel/nano-hydroxyapatite (nHA) injectable hydrogels were synthesized and evaluated in a mouse model of subcutaneous transplantation after 14 days. PCL-PEG-PCL-Gel and PCL-PEG-PCL-Gel/nHA hydrogels were fabricated with in situ precipitation method. Structure, intermolecular interaction, and the reaction between the PCL-PEG-PCL, Gel, and nHA were evaluated using a scanning electron microscope (SEM), Fourier-transform infrared spectroscopy (FT-IR), proton nuclear magnetic resonance (H-NMR), and carbon nuclear magnetic resonance (C-NMR). Fourteen days after subcutaneous injection, the existence of an immune system reaction was investigated using Hematoxylin and Eosin (H&E) staining. Using immunofluorescence imaging, the number of CD68+ cells was determined in the periphery of the hydrogel. The CD8/CD4 lymphocyte ratio was also calculated in blood samples. We monitored the expression of CCL-2, BCL-2, IL-10, and CD31 using real-time PCR assay. The chemical evaluation revealed the successful integration of Gel and nHA to the PCL-PEG-PCL backbone. Histological examination showed the lack of inflammation at the site of injection. No toxicological effects were determined in hepatic and renal tissues. The addition of nHA to the PCL-PEG-PCL-Gel decreased biodegradation time. None of the hydrogels caused statistically significant differences in the number of CD68 cells (p > 0.05). The CD8/CD4 lymphocyte ratio remained unchanged in all groups (p > 0.05). Compared to the PCL-PEG-PCL group, the addition of nHA and Gel increased the expression of CCL-2, BCL-2, IL-10, and CD31 (p < 0.05). In conclusion, the current study showed that PCL-PEG-PCL-Gel/nHA hydrogels could be used in in vivo conditions without prominent toxic effects and inflammatory responses.

ABSTRACT

Injectable hydrogels attract more attention to hard tissue engineering for the fulfilment of the defects with irregular shapes. Therefore, the researchers investigated the biocompatibility and immune response to the injectable PCL-PEG-PCL-Col/nHA hydrogels in a mouse model. The histological examination was done via H&E. The activation of the immune cells was evaluated by using antibodies against the CD68, CD4, and CD8 markers. The expression of CCL-2, BCL-2, IL-10, and CD31 genes was measured. Moreover, serum levels of the ALT, ALP, AST, and Urea were detected. The results of the chemical analysis showed that the collagen and Nano-hydroxyapatite were successfully integrated into the PCL-PEG-PCL hydrogels. The histological examination revealed a delayed biodegradation rate after the addition of the collagen and Nano-hydroxyapatite. No prominent pro-inflammatory response was found at the site of the injection. There are no significant differences in the levels of the CD68 and CD8/CD4 lymphocyte ratio among groups (p > .05). The expression of the CD31, IL-10 was significantly increased in the PCL-PEG-PCL-Col/nHA hydrogel (p < .05). ALT, ALP, AST, and Urea levels were not altered pre- and post-transplantation of the hydrogels (p > .05). These in vivo results demonstrated that the injectable PCL-PEG-PCL-Col/nHA hydrogels are biocompatible and suitable for further research in hard tissue regeneration.

ABSTRACT

This is a case report of unusual lesions caused by Thelazia gulosa in cattle. After several ineffective treatments of suspected infectious keratoconjunctivitis in calves and cows, T. gulosa was found associated with retrobulbar granuloma-like masses. The masses with multiple digit-like protrusions on conjunctival surface ejected multiple worms on firm pressure on clinical examination. Samples of tissues that contained worms were surgically removed, fixed in ethanol and submitted to the parasitology and histopathology labs for morphological identification of worms and the nature of the tissue masses, respectively. The infestation was present only in young calves (<3months) and high-producing cows. Histopathology showed fibrovascular granulation tissue, containing a moderate to marked inflammatory infiltrate. Ivermectin treatment (200µg/kg, SC, once) with and without surgical excision resolved the infestation. To the best of our knowledge, this is the first report of tissue invasion by T. gulosa.

ABSTRACT

Hydatid Disease is the name given to the condition caused by the zoonotic tapeworm Echinococcus granulosus. The tapeworm spends most of its adult life in the intestine of its definitive host, namely canids and in particular the dog. The tapeworm eggs become voided in the canids' faeces and as a result of ingesting the eggs, infection passes to the intermediate host, commonly herbivores while grazing. However, humans can become accidentally infected and hydatid cysts may develop throughout the body. During April 2010-February 2014, a total 198 camels, which had been sent to the abattoir, the daily number of hydatid infected livers and lungs of camels slaughtered at Tabriz abattoir were recorded. To be sure about the validity of recorded data, observed data were collected daily. Approximately 29 (14.64 %) of camels were infected according to this survey. Age wise, the prevalence of infection in young animals (under the age of 5 years) was 4 (2.02 %), whereas in animals between 5 and 10 years and over, the prevalence of infection was 11 (5.55 %) and 14 (7.07 %) respectively. Sex wise, female animals had a higher prevalence with 17 (19.76 %) cases in camels, whereas in the males, there were 12 (10.71 %) cases in camels. There was a notable difference found in our study between male and female animals (P < 0.05). Infections were recorded in two visceral organs with the Lungs having the highest prevalence in camels, and the liver had low infected in camels. The results of this study suggest that infection of camels with hydatid cyst is common in Tabriz, Iran and that this may constitute economic and health problems in the meat industry.

OBJECTIVE: Bovine cysticercosis is a cattle infection caused by a tapeworm, Taenia saginata. While the condition is relatively innocuous, the parasite infects the small intestine of humans in its mature stage and causes a few specific symptoms such as abdominal pain and nausea. METHODS: Between February 2013 and February 2014, a total of 640 cattle were randomly selected from all the cattle sent to the abattoir, and some internal organs and skeletal muscles of these cattle were inspected. RESULTS: Overall, 11 (1.71%) cattle were infected with the larval form of the cestode parasite T. saginata. In addition, the infection was more prevalent in cattle aged above 12 months than in those aged below 12 months [10 (2.06%) vs. 1 (0.64%)]. The prevalence of infection was significantly higher in female animals [8 (3.72%)] than in male animals [3 (0.70%)] (p<0.05). However, no significant difference was found between the rates in the 2 age groups or in different seasons. While the infections were detected in several visceral organs, no significant difference was found between their infection rates. CONCLUSION: The comparatively high prevalence of Cysticercus bovis infection in the cattle in Tabriz, Iran, may contribute to economic and health problems in the country's meat industry. On the other hand, the role of public health education in C. bovis infection control cannot be neglected.

ABSTRACT

In February 2012, 100% mortality was reported in a herd with 79 local sheep that were kept around of Abhar, Northwest of Iran. The ration for adult sheep was daily mixed (40 kg straw, 25 kg wheat and 2 kg Vit-C premix) and accidentally 1 500 g of salinomycin (Salinomycin 12% Premix; Aras Bazar Laboratories, Iran) had been added to the ration (22388 mg/kg = 22388 ppm) and overnight was fed to herd. At the morning, 78 sheep were founded dead and one of them showed convulsive seizures. Postmortem examination revealed pulmonary congestion and edema, hemorrhages in abomasum, large pale kidney and white streak lines in myocardium. Main histopathologic lesions were extensive subepicardial and intercardiomyofibers hemorrhages, extensive cardiomyolysis and myocarditis in heart, severe hyperemia and extensive acute tubular necrosis (ATN) in kidneys and focal necrosis and retention of bile cholangitis in the liver. In this study, on the basis of the history, observation of the ionophore remnant in the ration, clinical signs, gross and histopathological findings, acute salinomycin intoxication is definitely diagnosed.

ABSTRACT

Arsenic contamination of groundwater has been previously reported in Ghopuz, a village located in the Northwest of Iran. Samples were taken from consuming and irrigation water and plants of the region for chemical analysis. A seven-year old ewe, which had lived in and fed a lifelong at the same place, with clinical signs such as weakness, wasting and inappropriate integument was necropsied. Grossly, buccal erosion, stomatitis, cutaneous ulcers and serous atrophy of fat deposits were observed. Rumen contents, wool and several tissue samples were obtained for toxicological and histopathological examinations. Mean arsenic concentration in the spring water, irrigation water and grass/algae were 70.11, 48.74 and 141.85 ppb (µg/kg), respectively. Arsenic levels were 486.73, 247.94, 127.92, 125.97 and 231.24 ppb in wool, skin, rumen contents, liver and kidney, respectively. Microscopic study revealed hyperemia and heavy parasitic infestation of the abomasal wall. Hyperemia and regeneration of renal tubule epithelia were observed in kidneys and hyperkeratosis, suppurative deep dermatitis and paniculitis were found in skin. Periacinar fibrosis and a poorly differentiated cholangiocarcinoma were seen in liver. In pancreas, reduced cell density of islands of Langerhans was noticeable. In the central nervous system, perineuronal and perivascular edema, ischemic changes in gray matter neurons, and microcavitation of white matter were present. Our findings confirmed chronic arsenic toxicosis in small ruminants in this region. It can be concluded that long-term consumption of arsenic contamined water and forage may be associated with chronic arsenic poisoning in domestic animals and human beings, with consequent neoplastic disease and induction of diabetes in this region.

Cottonseed meal is widely used as one of the protein supplement in poultry diets. Its mechanism of toxic action on chicken embryo is poorly understood. In this study, direct effects and abnormalities of cottonseed on chicken embryo were studied. Oxidative stress, cholinergic stress, mineral analysis and microscopic lesions were analyzed in chicken embryo which injected cottonseed extraction in 0.1, 1 and 10 mg concentration (with free gossypol 0.25 ppm, 2.5 ppm and 25 ppm respectively) at day 4 of incubation. Higher group had 100 percent mortality. Serum of alive chicken embryo at day 20 of incubation were measured for FRAP (ferric reducing ability of serum), total SH groups assay, cholinesterase assay and potassium concentration. The results expressed as mean±SD show to increase oxidative stress, cholinergic stress but significant difference (p<0.05) wasn't observed between groups. The significant difference was observed in potassium concentration in serum. Some evidence of hematotoxicity such as hemorrhage and higher number of puntate reticulocytes were detected. It is concluded, hematoxicity and hyperkalemia are toxicity mechanisms that could initiate in low concentrations of cottonseed in chick embryo.

In 2008, African swine fever was introduced into Georgia, after which it spread to neighboring Armenia, Azerbaijan, and the Russian Federation. That same year, PCR and sequence analysis identified African swine fever virus in samples from 3 dead female wild boars in northwestern Iran. Wild boars may serve as a reservoir.

ABSTRACT

There are no reports of toxicological studies of Pterocarya fraxinifolia. The leaves are used for fishing, which also an anesthetic agent. Currently, many drugs utilized in anesthesia practice are modified cholinergic transmission and acetylcholine esterase inhibitors; these are parts of anaesthetic pharmacy. Therefore, cholinergic assessment was surveyed in chicken embryo, which Pterocarya fraxinifolia extractes were injected in 0.1, 1 and 10 mg concentration at day 4 of incubation. Serum and brain cholinesterase were analyzed on day 20 of incubation. The signs were not due to the changes of cholinesterase activity. In histopathology examination, massive necrosis was observed in the spinal cord. Other tissues such as heart, kidneys, skeletal bones and muscles, trachea and lungs, digestive system and endocrine glands were completely developed. This data suggests that the spinal cord is a target organ of the bioactive component of this plant.
