ABSTRACT

Objective: Paraquat (PQ) is a highly toxic herbicide that causes pulmonary fibrosis (PF), and no specific antidote is available against it. Teucrium polium L. is a plant that exhibits antioxidant and anti-inflammatory activities. The present study evaluates the preventive and therapeutic effects of T. polium extract (TPE) against PQ-induced lung fibrosis in rats. Materials and Methods: We divided rats into five groups of eight. Groups one and two received saline and PQ (20 mg/kg, i.p.), respectively. Groups three to five were treated with TPE (50, 100, and 200 mg/kg, by gavage) started one week before PQ administration and lasted three weeks after PQ administration. Results: Our findings showed that PQ significantly increased lung malondialdehyde, nitric oxide, hydroxyproline, lung index, Ashcroft score, red blood cells accumulation, and inflammatory cell infiltration. Moreover, PQ decreased catalase and glutathione peroxidase activities and glutathione content. The results of hematoxylin-eosin and Masson's trichrome staining indicated that PQ destroyed lung parenchyma and developed PF (p<0.05 to p<0.001). Gavage with TPE significantly improved biochemical and histological abnormalities induced by PQ in rats (p<0.05 to p<0.001). Conclusion: The current survey indicated that treatment with TPE could reduce and reverse PQ-induced PF, which may be due to the phenolic compounds present in TPE.

BACKGROUND: Bed bugs as blood-sucking insects have become a public health problem in urban communities throughout the world. The objective of this study was to determine the effects of resident education and self-implementation of Integrated Pest Management (IPM) strategy for eliminating bed bug infestation in infected apartments in Ahvaz City, Iran. METHODS: This interventional study was conducted in seventy apartments infested with bed bug (Cimex lectularius) in Ahvaz City, southwest Iran, during a 6-month period. The bed bug infestations reported to Health centers were inspected visually and confirmed by medical entomology experts. Then, the heads of the households were received techniques/tools of the IPM program and trained by medical entomology experts before self-implementation of control methods. Finally, the infected apartments were inspected by the experts at 1, 3, and 6 months after intervention and data were recorded in a checklist. RESULTS: From the seventy infected apartments, 57%, 28% and 15% were considered as low, moderate, and high level infestation respectively. The bed bug infestation was eliminated from 53 apartments (76%) after one month and it reached to 62 apartments (88%) by the end of third month. Finally, after six months of applying IPM program, bed bugs infestation was eliminated from all infected apartments (100%). Residents expressed their 100% satisfaction with applying the bed bug IPM strategy. CONCLUSION: Training residents to implement the IPM program can reduce pest control costs, the volume of pesticides consumed, and human exposure to chemical pesticides, resulting in increased human and environmental health and safety.

BACKGROUND: The common bed bug (Cimex lectularius), a nocturnal blood-feeding ectoparasite, is considered an urban pest with public health importance in human environments. We aimed to determine the repellency effect of oregano essential oil, Origanum vulgare, against this pest under laboratory conditions. METHODS: The essential oil was prepared from dried leaves using hydro-distillation method. A gas chromatography-mass spectrometer (GC-MS) was used for analysis and identification of oregano essential oil compounds. Treated surface method in Petri dish was carried out to evaluate repellency potential of the oil using 4th and 5th instar nymphs and adults. The concentration-repellency response of oregano essential oil was calculated and compared with a commercial insect repellent stick containing 33% N, N-diethyl-meta-toluamide (DEET). RESULTS: The oregano essential oil consisted of 158 compounds with terpineol (22.85%) and α-terpinene (20.60%) being the major components by volume. The EC50 and EC99 of oregano oil (effective concentrations causing 50% and 90% repellency of bed bugs) were 1.61 and 6.57mg/cm2 at 9h after application, respectively. The 40% oregano essential oil exhibited 100% repellency against bed bug at 3, 5, 9 and 24h after application while the repellency index of DEET 33% was 100% at 3 and 5h and it decreased to 80% and 27% at 9 and 24 hours. CONCLUSION: Oregano oil 40% exhibited more repellency compared to commercial insect repellent stick containing 33% DEET. Further studies are warranted to confirm the effectiveness of oregano essential oil in personal protecting against bed bug biting.

BACKGROUND: The key enzyme in the process of melanin biosynthesis is tyrosinase. Skin hyperpigmentation and browning of foods are undesirable phenomena which tyrosinase represents. Therefore, tyrosinase inhibitors have been used increasingly for medicinal and cosmetic products. OBJECTIVES: In this study, inhibitory effects of four plants including: physalis alkekengi L., Alcea rosea L., Bunium persicum B. Fedtsch. and Marrubium vulgare L. on diphenolase activity of mushroom tyrosinase were evaluated. MATERIALS AND METHODS: The inhibitory activities of hydroalcoholic extracts of plants against oxidation of L-Dopa (as a substrate) by mushroom tyrosinase were investigated. RESULTS: The hydroalcoholic extract of P. alkekengi showed the most tyrosinase inhibitory effect with IC50 of 0.09 mg/mL vs. 0.38, 0.38 and 2.82 mg/mL of B. persicum, A. rosea and M. vulgare, respectively. M. vulgare exhibited uncompetitive inhibition and other plants showed mixed type inhibition on mushroom tyrosinase. CONCLUSIONS: All plants could inhibit mushroom tyrosinase, but more investigations on human tyrosinase and clinical studies are needed.

ABSTRACT

In the present study, protective effect of Teucrium polium L. (Labiatae) extract on acetaminophen-induced hepatotoxicity was investigated in mice. Animals were divided into six groups, each group consist of 8 mice. Group one as the negative control group received normal saline, while group two received only crude extract of T. polium L. (500 mg/Kg) for five days and group three as the positive group received acetaminophen (500 mg/Kg). Groups four, five and six received crude extract in doses of 125, 250 and 500 mg/Kg, respectively, and on the fifth day, one hour after the last administration, acetaminophen was given orally (500 mg/Kg). Then on the 6(th) day, animals were sacrificed, their blood was collected to determine serum enzyme activities of ALT, AST and ALP to measure the serum levels of directed and total bilirubin. The livers were removed for histological examination. The results of this study showed the protective effect in all doses but the most significant protection was observed in doses of 250 and 500 mg/Kg (p < 0.05). Also these findings were supported and confirmed by histological examination.

BACKGROUND: Degenerative Joint Disease (DJD) is the most common joint disease in human beings. Previous studies have explained that glucosamine is preferred as placebo and in efficacy compared with NSAID's in treatment of patients' knee osteoarthritis. Alendronate was used to treat osteoporotic patients and its efficacy was established. OBJECTIVES: The aim of this study was to compare the efficacy of administration of glucosamine alone and its combination with alendronate in osteoarthritis of the knee. PATIENTS AND METHODS: The study included 130 patients with osteoarthritis who randomly received glucosamine alone (group II) (500mg TDS), or combination of glucosamine (500mg TDS) and alendronate (70mg weekly) (group I) for 12 weeks. Patients were evaluated on 1, 3, 6 and 12 weeks after beginning the treatment to evaluate efficacy of each treatment. RESULTS: Statistically, there was no significant difference in pain index (P > 0.05) but in the two groups the mean of pain index decreased in a similar fashion. The stiffness index in combination treatment group (group I) decreased more than glucosamine group (group II) (P < 0.05). The function of joints in combination treatment group (group I) improved after 12weeks. The bone mineral density (BMD) at 12weeks in combination therapy group improved. CONCLUSIONS: Combination therapy of glucosamine and alendronate indicated significant improvement of stiffness, function, BMD of osteoarthritis compared with glucosamine alone but there was no statistically significant decrease in pain index. It can be concluded that the combination of glucosamine and alendronate provide better and more rapid improvement in patients with osteoarthritis.

BACKGROUND: Major depressive disorder (MDD) is the most common disorder in our community. Hypericum perforatumm, is a herb with a long application history in treating depression. A controlled study to evaluate the effects of this herbal medicine in Iran did not exist. OBJECTIVES: This study aimed to assess the effect of Hypericum perforatom (perforan), in combination with tricyclic antidepressants in MDD treatment. MATERIALS AND METHODS: The present study was a placebo-controlled double blind randomized clinical trial including 40 patients with major depressive disorder who referred to Psychiatric Clinic and Golestan Hospital in 2011. Patients were randomly distributed in 4 blocks with 10 patients in each. 5 out of 10 patients took medicine (group I) and the others received placebo (group II). RESULTS: After six weeks, both groups indicated some improvement, there was also a significant degree of recovery in the perforan group (P = 0.04). CONCLUSIONS: The results of the current study suggested that combinations of St. John's Wort and tricyclic antidepressants compared to tricyclic antidepressants alone had significant effect on mild to moderate depression improvement. According to the numerous side effects of antidepressants, their continuous use was not well tolerated. There is a strong global tendency to use herbal medicines. Hypericum perforatom (St. John's Wort) is one of the best types of unapproved MDD treatments. Also, the most common reason to discontinue antidepressant treatments is their sexual side effects. None of the patients in the two groups had complained about sexual side effects which was consistent with other studies and it was an advantage of using this herbal medicine. Perforan group had improved in quality of sleep and the increase of energy. This improvement had not been reported in previous studies. Sleep problems are common complications of specific serotonin reuptake inhibitors (SSRIs).

ABSTRACT

Several studies have demonstrated that herbal extracts possess various biological effects including anti-inflammatory and anti-cancer activities. The present study was aimed to investigate the protective effects of the Astragalus gypsicolus (AG) hydroalcoholic extract in early allergic sensitized mice induced by ovalbumin. Phytochemical assay was used to recognize the main active constituents in the AG hydroalcoholic extract. Mice were immunized with subcutaneous injection of ovalbumin and aluminum hydroxide. Efficiency of sensitization was assessed by serum IgE levels and eosinophil count. After sensitization, two doses of extract (250 mg/kg and 500 mg/kg) were injected intrapritoneally. On day 14, mice were challenged with intrapritoneal injection of ovalbumin. IL-4 and IFNγ levels in broncoalveolar lavage fluid, which had been collected on day 15, were assessed by Enzyme-Linked Immunosorbent Assay (ELISA) kit. Our results indicate two main active constituents including flavonoids and terpenoids are present in the AG hydroalcoholic extract. Intrapritoneal injection of the AG hydroalcoholic extract was able to decrease IL-4 and increase IFNγ. It seems the AG hydroalcoholic extract has the potential to modulate the balance of Th1/Th2 cytokines in allergy.
