ABSTRACT

Volatile components of Lonicerae Japonicae Flos in bud stage extended type Beihua 1 were determined by the headspace solid-phase micro-extraction, compared with traditional cultivar Damaohua. There are fifty-two volatile compounds were identified and the relative content of the volatiles was calculated by the area normalization method. Thirty-nine compounds were found in Beihua 1, whereas thirty-three components in Damaohua. Total twenty identical compounds existed in Beihua 1 and Damaohua. The contents of alcohols and hydrocarbons of Beihua 1 were higher significantly than that of Damaohua, while significantly lower than that of Damaohua in ketones content. Besides, twenty components were only detected in Beihua 1, such as methyl nicotinate, hexadecanoic acid, methyl ester,acetophenone, nonanoic acid.

ABSTRACT

OBJECTIVE: To evaluate the quality coherence of the prepared slices of Polygoni Cuspidati Rhizoma commercially available in China and provide a reference for their quality evaluation. METHODS: The fingerprints were obtained using HPLC method, and analyzed with Chromatographic Fingerprint Similarity Evaluation System (2004A Version) provided by Chinese Pharmacopoeia Commission. The experiment was carried out with an Agilent TC-C18 column (250 mm x 4.6 mm, 5 microm) using acetonitrile-0.05% aqueous solution of phosphoric acid as mobile phase at the flow rate of 1.0 mL/min, detection wavelength of 230 nm, and temperature of 25 degrees C. RESULTS: The fingerprint for Polygoni Cuspidati Rhizoma was established. 21 common characteristic peaks were identified and the similarities of 24 batches of prepared slices of Polygoni Cuspidati Rhizoma were between 0.882 - 0.988. CONCLUSION: The quality of the prepared slices of Polygoni Cuspidati Rhizoma commercially available in China is comparable.

ABSTRACT

OBJECTIVE: To analyze the volatile oil from Radix Euphorbia Pekinensis. METHODS: Volatile oil was extracted from Radix Euphorbia Pekinensis by steam distillation and GC-MS was employed for detecting the content of the constituents. The relative content of the chemical constituents were calculated using area normalization method. RESULTS: 35 peaks were separated and 29 constituents were identified. The yield rate of volatile oil extracted from the dried radix of Euphorbia Pekinensis was 0.3%. CONCLUSION: The main components of volatile oil were agarospirol and hedycargol and their contents were 49.23% and 20.66% respectively. This paper provides valuable experimental data for further research of Radix Euphorbia Pekinensis.

ABSTRACT

OBJECTIVE: To reveal the correlation between HQT gene and the biosynthesis of chlorogenic acid in Lonicera japonica. METHODS: RT-PCR was used to measure the relative expression of HQT gene and reference gene Actin, and agarose gel electrophoresis was used to analyse the PCR results. RESULTS: The brightness of Actin gene strips of different organs was properly similar to each other,but the brightness of HQT gene strips was significantly different. HQT gene strips of alabastrum were the brightest,the brightness of HQT gene strip of leaves took the second place, and the brightness of HQT gene strips of stems was very faint. This result was in accordance with the content of chlorogenic acid in different organs of Lonicera japonica. CONCLUSION: The expression of HQT gene with the biosynthesis of chlorogenic acid has necessary relation with Lonicera japonica.

ABSTRACT

OBJECTIVE: To explore the genetic basis of using three species of Asarum as Herba Asari to determine the taxonomic positions of Asarum heterotropoides and A. siebodii; and to apply DNA molecular analysis as a tool for identification of Herba Asari. METHOD: PCR, purification, sequence analysis were prerformed. RESULT: MS sequences of the three Asarum species were obtained. 3 botanical sources of Herba Asari are closely clustered together on the topology tree; one inner branch is composed of A. heterotropoides and A. sieboldii, whereas another branch contains A. sieboldii. Their ITS sequences are different. CONCLUSION: Three plant species of Herba Asari are closely related, and there are genetic reasons that they are used as the sources of the same medicine. The classification placement of A. sieboldii is not certain. The differences of ITS sequences of the botanical sources of Herba Asari can be used as a means of identification.
