ABSTRACT

This study aims to investigate the gene expression of sperm-borne phospholipase C zeta (PLCζ), WW domain-binding protein 2N-Terminal Like (WBP2NL), and Tumor necrosis factor (TNF-α), as a negative control, in spermatozoa and their relationship with fertility and seminal quality in stallions. Ejaculates from 40 Criollo stallions were used, whose fertility was assessed on the basis of their pregnancy rate per cycle in at least two breeding seasons. Pregnancy rates ranged from 20% to 90% and were used to divide the stallions into two groups: High rates (≥ 50%) (n = 25), and Low rates (< 50%) (n = 15). A computer-assisted sperm analysis system - (CASA) analyzed semen after collection. Also were evaluated the physical and functional integrity of the plasmatic membrane and sperm morphology alterations. All stallions expressed PLCζ, WBP2NL, and TNF-α. PLCζ positively correlates with conception rate, total motility (TM), progressive motility (PM), plasmatic membrane functionality, and integrity. A simple linear regression was detected between pregnancy rate and PLCζ expression (P = 0.003), TM (P < 0.001) and PM (P < 0.001). PLCζ gene expression was higher (P = 0,012) in the High rates group than in the Low group. WBP2NL and TNF-α did not correlate with seminal quality and stallion's fertility. It was concluded that PLCζ gene expression in the spermatozoa might be used as a biomarker of fertility and seminal quality in stallions. Parameters of sperm kinetics also showed, positive correlation between TM, PM and pregnancy rate.

ABSTRACT

The recovery of critical metals from spent lithium-ion batteries (LIBs) is rapidly growing. Current methods are energy-intensive and hazardous, while alternative solvent-based strategies require more studies on their 'green' character, metal dissolution mechanism and industrial applicability. Herein, we bridged this gap by studying the effect of dilute HCl solutions in hydroxylated solvents to dissolve Co, Ni and Mn oxides. Ethylene glycol emerged consistently as the most effective solvent, dissolving up to four times more Co and Ni oxides than using aqueous acidic media, attributed to improved chloro-complex formation and solvent effects. These effects had a significant contribution compared to acid type and concentration. The highest Co dissolution (0.27 M) was achieved in 0.5 M HCl in 25 % (v/v) glycerol in water, using less acid and a significant amount of water compared to other solvent systems, as well as mild temperatures (40 °C). This solvent was applied to dissolve battery cathode material, achieving 100 % dissolution of Co and Mn and 94 % dissolution of Ni, following what was concluded to be a mixed mechanism. These results offer a simple alternative to current leaching processes, reducing acid consumption, enhancing atomic efficiency, and paving the way for optimized industrial hydrometallurgical processes leaning to 'greener' strategies.

ABSTRACT

Solubilizing agents are widely used to extract poorly soluble compounds from biological matrices. Aqueous solutions of surfactants and hydrotropes are commonly used as solubilizers, however, the underlying mechanism that determines their action is still roughly understood. Among these, ionic liquids (IL) are often used not only for solubilization of a target compound but in liquid-liquid extraction processes. Molecular dynamics simulations can shed light into this issue by providing a microscopic insight of the interactions between solute and solubilising agents. In this work, a new coarse-grained (CG) model was developed under the MARTINI framework for gallic acid (GA) while the CG models of three quaternary ammonium ionic liquids and salts (QAILS) were obtained from literature. Three QAILS were selected bearing in mind their potential solubilising mechanisms: trimethyl-tetradecylammonium chloride ([N1,1,1,14]Cl) as a surfactant, tetrabutylammonium chloride ([N4,4,4,4]Cl) as a hydrotrope, and tributyl-tetradecylammonium chloride ([N4,4,4,14]Cl) as a system combining the characteristics of the other compounds. Throughout this hydrotrope-to-surfactant spectrum and considering the most prevalent GA species across the pH range, the solvation of GA at two concentration levels in aqueous QAILS solutions were studied and discussed. The results of this study indicate that dispersive interactions between the QAILS and GA are generally the driving force in the GA solubilization. However, electrostatic interactions play an increasingly significant role as the GA becomes deprotonated, affecting their placement within the micelle and ultimately the solvation mechanism. The hydrotropic mechanism seen in [N4,4,4,4]Cl corroborates recent models based on the formation of a hydrotrope-solute aggregates driven by dispersive forces. This work contributes to the application of a transferable approach to partition and solubilization studies using molecular dynamics, which could complement experimental assays and quickly screen molecular candidates for these processes.

ABSTRACT

The present case report aimed to determine the responsiveness of the endometrium and the ovaries of an X0 mare after hormonal treatment. On transrectal palpation, the uterus was flaccid and smaller than normal, and the ovaries were small and smooth. The endometrium had normal histological architecture, with an atrophic glandular epithelium. A karyotype evaluation was performed, and 70 cells presented 63 chromosomes, lacking one sex chromosome. Circulating hormonal levels of total estrogens were 43.93 pg/mL; progesterone 0.01 ng/mL; testosterone 48 pg/mL; FSH 30.3 ng/mL; and LH 1.71 ng/mL. Immunohistochemistry tests showed the presence of estrogens and progesterone receptors in the endometrial samples of the X0 mare. 17ß estradiol was administrated on three consecutive days and long-action progesterone on the fourth day. After hormonal stimulation, the mare showed changes in endometrial ultrasonography and histology. After treatment with estradiol, uterine edema was noted, and after progesterone, a reduction in edema was observed. At the request of the owner, no further treatment or follow-up occurred. This report showed that the endometrium is functional, but the ovaries did not change macroscopically under hormonal therapy.

ABSTRACT

Characterization of the uterine proteome before the entry of the conceptus to the uterus is essential to know the factors involved in the physiological events of gestation. The objective of the study was to compare proteomic profile of uterine fluid collected on day 5 post ovulation of cyclic and inseminated mares. Samples of endometrial secretion were recovered over 2 cycles during the fifth day post ovulation. The first cycle constituted the Cyclic group and in the following cycle, the same mares were inseminated and considered as the Inseminated group. All the samples were subjected to two-dimensional electrophoresis (2D-PAGE). A total of 107 spots were visualized by 2D-PAGE. Three spots with differences in abundance between the inseminated and cyclic mares and with presence in at least 80% in one of the groups were selected and identified. The selected spots were extracted, digested by trypsin and analyzed by matrix assisted laser desorption/ionization time-of-flight/time-of-flight (MALDI-TOF/TOF) mass spectrometry (MS) for protein identification. Three proteins were identified: ceruloplasmin (CP) serotransferrin (TF) and albumin (ALB). The identified proteins in this study were related to iron metabolism and immunological tolerance suggesting that changes in their abundance during the 5 days after ovulation are probably a signaling mechanism of the potential equine conceptus to the maternal immune system for its immunological recognition. Probably changes in abundance of CP, ALB and TF represent a mechanism of endometrial preparation for the maternal recognition, attachment and development of a potential equine embryo. There is also evidence to support an alternative hypothesis suggesting that protein changes are inflammatory events, resulting from a previous inflammation due to residual seminal effects.

ABSTRACT

Hydrogels have captivated the attention of several research and industry segments, including bioengineering, tissue engineering, implantable/wearable sensors and actuators, bioactive agent delivery, food processing, and industrial processes optimization. A common limitation of these systems is their fixed shape. The concept of hydrogel moldability is often assigned to the injectability potential of liquid precursors, and this feature is often lost right after hydrogel formation. Hydrogel modulation is a recent trend that advocates the importance of designing materials with shape fitting ability targeting on-demand responses or defect filling purposes. Here, we present a compliant and cell encapsulation-compatible hydrogel prepared from unmodified natural origin polymers with the ability to undergo extreme sequential shape alterations with high recovery of its mechanical properties. Different fragments of these hydrogels could be bonded together in spatiotemporally controlled shape- and formulation-morphing structures. This material is prepared with affordable off-the-shelf polysaccharides of natural origin using a mild and safe processing strategy based solely on polyelectrolyte complexation followed by an innovative partial coacervate compaction and dehydration step. These unique hydrogels hold potential for multifield industrial and healthcare applications. In particular, they may find application as defect filling agents or highly compliant wound healing patches for cargo release and/or cell delivery for tissue regeneration and cell-based therapies.

ABSTRACT

There are few microbiological surveys on reptiles in Brazil. The study described here focuses on a species of snake of great medical interest and serves as basis for other studies and comparisons. The samples were collected directly from the colon of healthy adult Jararacas. The material was seeded in MacConkey Agar (Difco®) and XLT4 Agar (Difco®). The isolates were identified through the API 20E Identification System (BioMériux®). The isolates of Salmonella sp. were submitted to serotyping. Finally, the colonies were submitted to the antimicrobial sensitivity test. Several genera of the Enterobacteriaceae family were obtained (Citrobacter, Enterobacter, Escherichia, Klebsiella, Kluyvera, Morganella, Proteus, Providencia and Salmonella), as well as a genus of morphologically similar Gram-negative bacteria (Aeromonas). Salmonella, Citrobacter and Escherichia were the most frequent isolates. Fourteen (14) serotypes of Salmonella were identified; 13 are of the subspecies IIIb and 1 of the subspecies IV. Aeromonas, Enterobacter, Escherichia and Klebsiella presented a higher resistance rate to antibiotics. Kluyvera and Salmonella were the genera most sensitive to the tested drugs. Aminoglycosides presented good antimicrobial action, but chloramphenicol was the only drug to which no isolate presented resistance.


Existem poucos levantamentos microbiológicos em répteis no Brasil. O estudo aqui descrito enfoca uma espécie de serpente de grande interesse médico e serve como base para outros estudos e comparações. As amostras foram colhidas diretamente do cólon de jararacas adultas e saudáveis. O material foi semeado em ágar MacConkey (Difco®) e ágar XLT4 (Difco®). Os isolados foram identificados por meio do Sistema API 20E de Identificação Bioquímica (Galerias API-BioMériux®). Os isolados de Salmonella sp. foram submetidos à sorotipagem. Por fim, as colônias foram submetidas ao teste de sensibilidade antimicrobiana. Foram obtidos vários gêneros da família Enterobacteriaceae (Citrobacter, Enterobacter, Escherichia, Klebsiella, Kluyvera, Morganella, Proteus, Providencia e Salmonella) e um gênero de bactéria Gram-negativa morfologicamente semelhante (Aeromonas). Salmonella, Citrobacter e Escherichia foram os isolados mais frequentes. 14 sorotipos de Salmonella foram identificados, sendo 13 da subespécie IIIb e 1 da subespécie IV. Aeromonas, Enterobacter, Escherichia e Klebsiella apresentaram maior taxa de resistência aos antibióticos. Kluyvera e Salmonella foram os gêneros mais sensíveis às drogas testadas. Aminoglicosídeos apresentaram boa ação antimicrobiana, mas cloranfenicol foi a única droga para qual nenhum isolado apresentou resistência.
