ABSTRACT

BACKGROUND: Supplementation with Angiotensin-(1-7) [(Ang-1-7)] has received considerable attention due to its possible ergogenic effects on physical performance. The effects of a single dose of Ang-(1-7) on the performance of mountain bike (MTB) athletes during progressive load tests performed until the onset of voluntary fatigue have previously been demonstrated. This study tested the effects of Ang-(1-7) in two different exercise protocols with different metabolic demands: aerobic (time trial) and anaerobic (repeated sprint). METHODS: Twenty one male recreational athletes were given capsules containing an oral formulation of HPßCD-Ang-(1-7) (0.8 mg) and HPßCD-placebo (only HPßCD) over a 7-day interval; a double-blind randomized crossover design was used. Physical performance was examined using two protocols: a 20-km cycling time trial or 4 × 30-s repeated all-out sprints on a leg cycle ergometer. Data were collected before and after physical tests to assess fatigue parameters, and included lactate levels, and muscle activation during the sprint protocol as evaluated by electromyography (EMG); cardiovascular parameters: diastolic and systolic blood pressure and heart rate; and performance parameters, time to complete (time trial), maximum power and mean power (repeated sprint). RESULTS: Supplementation with an oral formulation of HPßCD-Ang-(1-7) reduced basal plasma lactate levels and promoted the maintenance of plasma glucose levels after repeated sprints. Supplementation with HPßCD-Ang-(1-7) also increased baseline plasma nitrite levels and reduced resting diastolic blood pressure in a time trial protocol. HPßCD-Ang-(1-7) had no effect on the time trial or repeat sprint performance, or on the EMG recordings of the vastus lateralis and vastus medialis. CONCLUSIONS: Supplementation with HPßCD-Ang-(1-7) did not improve physical performance in time trial or in repeated sprints; however, it promoted the maintenance of plasma glucose and lactate levels after the sprint protocol and at rest, respectively. In addition, HPßCD-Ang-(1-7) also increased resting plasma nitrite levels and reduced diastolic blood pressure in the time trial protocol. ABSTRACT

PURPOSE: To investigate the relationship between the recovery of neuromuscular fatigue and the recovery of amount of work done above critical power (W´). METHODS: Ten healthy men performed, on different days, constant work rate exercises until task failure to determine critical power (CP) and W´. In the three following visits, participants performed two exhausting constant work rate exercises estimated to induce task failure within 6 min (P61 and P62), interspaced by 3, 6 or 15 min of recovery. Neuromuscular function was assessed before and periodically after the P61 using percutaneous electrical femoral nerve stimulation. The W´ recovery was measured from the total work performed above CP during the P62. RESULTS: The P61 induced a full use of W´ and a reduction in maximal voluntary contraction (MVC, - 19 ± 4%), voluntary activation (VA, - 6 ± 2%) and twitch force stimulated at 1 Hz (- 37 ± 11%), 10 Hz (- 50 ± 16%) and 100 Hz (- 32 ± 11%), when compared to baseline (P < 0.05). The time constant of VA recovery was significantly faster than the time constant of W´ recovery (P < 0.05), but there was no significant difference between the time constant of W´ recovery and the time constant of recovery of MVC or twitch force stimulated at 1, 10 and 100 Hz (P > 0.05). However, the time constant of W´ recovery was only associated to the time constant of MVC recovery (r = 0.73, P < 0.05). CONCLUSION: The W´ recovery is not associated to the recovery of peripheral or central fatigue alone. Rather, W´ seems to be associated to the recovery of the overall capacity to generate force.

ABSTRACT

This study investigated the effect of caffeine on neuromuscular function, power and sprint performance during the days following an eccentric-based exercise. Using a randomly counterbalanced, crossover and double-blinded design, eleven male jumpers and sprinters (age: 18.7 ± 2.7 years) performed a half-squat exercise (4 x 12 repetitions at 70% of 1 RM), with eccentric action emphasized by using a flexible strip attached to their knees (Tirante Musculador®). They ingested either a capsule of placebo or caffeine (5 mg.kg-1 body mass) 24, 48 and 72 h after. Neuromuscular function and muscle power (vertical countermovement-jump test) were assessed before and after the half-squat exercise and 50 min after the placebo or caffeine ingestion at each time-point post-exercise. Sprint performance was measured at pre-test and 75 min after the placebo or caffeine ingestion at each time-point post-exercise. Maximal voluntary contraction (overall fatigue) and twitch torque (peripheral fatigue) reduced after the half-squat exercise (-11 and -28%, respectively, P < 0.05) but returned to baseline 24 h post-exercise (P > 0.05) and were not affected by caffeine ingestion (P > 0.05). The voluntary activation (central fatigue) and sprint performance were not altered throughout the experiment and were not different between caffeine and placebo. However, caffeine increased height and power during the vertical countermovement-jump test at 48 and 72 h post half-squat exercise, when compared to the placebo (P < 0.05). In conclusion, caffeine improves muscle power 48 and 72 h after an eccentric-based exercise, but it has no effect on neuromuscular function and sprint performance.

ABSTRACT

Pacing during a high-intensity cycling time trial (TT) appears to prevent premature task failure, but the performance fatigability during a self-paced exercise is currently unknown. Therefore, the current study characterized the time course of performance fatigability during a 4-km TT. Eleven male cyclists performed three separated TTs in a crossover, counterbalanced design. The TTs lasted until the end of the fast-start (FS; 600 ± 205 m), even-pace (EP; 3,600 ± 190 m), and end-spurt (ES; 4,000 m) phases. Performance fatigability was characterized by using isometric maximal voluntary contractions (IMVCs), whereas the muscle activation [i.e., voluntary activation (VA)] and contractile function of knee extensors [e.g., peak torque of potentiated twitches (TwPt)] were evaluated using electrically evoked contractions performed before and 1 min after each specific part of the trial. Gas exchange, power output (PO), and electromyographic activity (EMG) were also recorded. EMG/PO showed an abrupt increase followed by a continuous decrease toward the end of FS, resulting in a drop in IMVC (-12%), VA (-8%), and TwPt (-23%). EMG/PO was stable during EP, with no additional drop on IMVC, VA, or TwPt (-12%, -6%, and -22%, respectively). EMG/PO increased abruptly during the ES, but there was no change in IMVCs, VA, or TwPt (-13%, -8%, and -26%, respectively). These findings demonstrate that the performance fatigability during a self-paced exercise is characterized by a large drop in contractile function and muscle activation at the beginning of the trial (i.e., FS), without additional change during the middle and end phases (i.e., EP and ES).NEW & NOTEWORTHY The time course of performance fatigability throughout a self-paced exercise is currently unknown. The results showed that a large amount of muscle activation and contractile function impairments are attained early on a self-paced exercise (first ∼15% of the total time trial distance) and maintained throughout the test. This novel finding characterizes the performance fatigability from a contractile function and muscle activation perspective, which brings new insights for future studies focused on real-world exercise training and competition.

ABSTRACT

The link between total work performed above critical power (CP) and peripheral muscle fatigue during self-paced exercise is unknown. We investigated the influence of caffeine on the total work done above CP during a 4-km cycling time trial (TT) and the subsequent consequence on the development of central and peripheral fatigue. Nine cyclists performed three constant-load exercise trials to determine CP and two 4-km TTs ~75 min after oral caffeine (5 mg/kg) or cellulose (placebo) ingestion. Neuromuscular functions were assessed before and 50 min after supplementation and 1 min after TT. Oral supplementation alone had no effect on neuromuscular function ( P > 0.05). Compared with placebo, caffeine increased mean power output (~4%, P = 0.01) and muscle recruitment (as inferred by EMG, ~17%, P = 0.01) and reduced the time to complete the TT (~2%, P = 0.01). Work performed above CP during the caffeine trial (16.7 ± 2.1 kJ) was significantly higher than during the placebo (14.7 ± 2.1 kJ, P = 0.01). End-exercise decline in quadriceps twitch force (pre- to postexercise decrease in twitch force at 1 and 10 Hz) was more pronounced after caffeine compared with placebo (121 ± 13 and 137 ± 14 N vs. 146 ± 13 and 156 ± 11 N; P < 0.05). There was no effect of caffeine on central fatigue. In conclusion, caffeine increases muscle recruitment, which enables greater work performed above CP and higher end-exercise peripheral locomotor muscle fatigue. NEW & NOTEWORTHY The link between total work done above critical power and peripheral fatigue during a self-paced, high-intensity exercise is unclear. This study revealed that caffeine ingestion increases muscle recruitment, which enables greater work done above critical power and a greater degree of end-exercise decline in quadriceps twitch force during a 4-km cycling time trial. These findings suggest that caffeine increases performance at the expense of greater locomotor muscle fatigue.
