ABSTRACT

The study of arthropods is still scarce in paleoparasitology, especially their molecular identification. In this experimental study, we amplify DNA using a polymerase chain reaction from 2 ectoparasite species, Rhipicephalus sanguineus and Pediculus humanus capitis, in experimentally desiccated feces. This study shows perspectives for the study of the identification of arthropods in coprolites when taxonomic identification is not possible.

ABSTRACT

This study aimed to assess anthropogenic impact of surrounding population in the Private Reserve of Natural Heritage at Pantanal, the world's largest freshwater wetland ecosystem located in the centre of South America. Viral aetiological agents of acute gastroenteritis as rotavirus A (RVA), noroviruses, human adenoviruses, klassevirus and of hepatitis, as hepatitis A virus, were investigated in different aquatic matrices. Annual collection campaigns were carried out from 2009 to 2012, alternating dry and rainy seasons. Viral particles present in the samples were concentrated by the adsorption-elution method, with negatively charged membranes, and detected by qualitative and quantitative PCR. From a total of 43 samples at least one virus was detected in 65% (28) of them. Viruses were detected in all matrices with concentrations ranging from 2 × 102 to 8·3 × 104 genome copies per litre. A significant higher RVA frequency was observed in the dry season. Our data revealing dissemination of human enteric viruses in water matrices both inside and outside the reserve could be useful to trace faecal contamination in the environment and to minimize the risk of infection by exposure of susceptible individuals. SIGNIFICANCE AND IMPACT OF THE STUDY: This study is part of a collaborative project designed to investigate the environmental and health conditions of the Private Reserve of Natural Heritage at Pantanal, the largest seasonally flooded wetland in the world. The project aimed to promote health and quality of human and wildlife extending technical-scientific knowledge about pathogens present in the region. By assessing the occurrence of human enteric viruses in different water matrices we demonstrated the anthropogenic impact of surrounding population and pointed out the potential risk of infection by exposure of susceptible individuals.

ABSTRACT

Oncicola venezuelensis Marteau, 1977 was found parasitizing adults of Leopardus pardalis (Linnaeus) found dead in Serra da Capivara National Park, Piauí state, Brazil, a new geographical locality reported for the species. The diversity of Oncicola Travassos, 1916 species is large, but genetic data are scarce. This article presents the results of genetic, morphological and ultrastructural studies carried out for taxonomic purposes. The first ultrastructural view showed a globular, short proboscis with 36 hooks, divided into six longitudinal rows of six hooks each. Hooks differ in size and shape: hooks I, II and III have a 'chisel-shaped' tip. The genetic profile included new sequences of ribosomal DNA ITS1, 5.8S and ITS2, and partial 28S rRNA regions. The results of maximum-likelihood tree analyses for each region showed Oligacanthorhynchidae Southwell et Macfie, 1925 close to Gigantorhynchidae Hamann, 1892 (supported >91%). Both use mammals and birds as definitive hosts. Morphological and ultrastructural studies combined with genetic analysis shed more light on the diversity of Oncicola species.

ABSTRACT

The first record of Echinostoma (Trematoda: Echinostomatidae) in coprolites was from a mummified human body in Minas Gerais State, Brazil. The finding raised questions on this parasite's incidence in prehistoric populations and the natural hosts of each species in remote times. Echinostomes occur worldwide and, despite the wide range of hosts, there is no record of Echinostomatidae in felines in Brazil. This study reports the finding of Echinostomatidae eggs in felid coprolites in the Furna do Estrago Archaeological Site, located in Pernambuco State in the Brazilian semiarid. Despite the possibility of false parasitism, the finding expands the distribution of this Digenea in remote times and raises the hypothesis of other cases of echinostomiasis in pre-Colombian populations.

ABSTRACT

The identification of parasites from ancient cultures expands our list of parasites infective to extant humans. A partially mummified human body from the archeological site of Lapa do Boquete, Minas Gerais State, Brazil, was recently discovered. It was interred between 600 and 1,200 yr ago. Dietary analysis showed that the mummified body was from a society that had a mixed subsistence of agriculture and gathering of wild foods. Coprolites from the body contained numerous helminth eggs. The eggs were identified as those of Echinostoma sp. and hookworm. Hookworm infection in pre-Columbian populations is already established, but this is the first evidence of Echinostoma sp. eggs found in human coprolites. The diagnosis of a true infection, as opposed to false parasitism, is discussed. The possibility of Echinostoma ilocanum infection is discussed, as this is a common species found in humans in the Asiatic region, which could have been introduced in South America in the pre-Columbian period. Alternative possibilities are also considered, including indigenous Brazilian Echinostoma species.

ABSTRACT

Trichuris eggs were found in Kerodon rupestris (Rodentia, Caviidae) coprolites collected in archaeological layers dated from 30,000 yr BP (before present) to 8,450 yr BP. Adult worms and eggs of this genus were not found in a search of living mammals of the region. Results indicate that K. rupestris was a host for an unknown Trichuris species not found in this rodent presently. Climate changes that occurred by 10,000 yr ago in the region could be the cause of its disappearance. The finding of parasites in archaeological material can show the antiquity of host-parasite relationships and parasite losses through time.

ABSTRACT

An important point in paleoparasitology is the correct diagnosis of the origin of coprolites found in archaeological sites. The identification of human and animal coprolites, through the study of the shape, size, characteristics after rehydration, alimentary contents, and the presence of parasites, has proved to be accurate for human coprolites. For non-human ones we compared coprolites with recent faeces of animals collected near the archaeological sites, following the methodology above mentioned. In this paper anteaters coprolites (Tamandua tetradactyla; Myrmecophaga tridactyla) with eggs of Gigantorhynchus echinodiscus (Archiancanthocephala; Gigantorynchidae) were identified.
