ABSTRACT

ETHNOPHARMACOLOGICAL RELEVANCE: Abrus cantoniensis Hance (AC), an abrus cantoniensis herb, is a Chinese medicinal herb used for the treatment of hepatitis. Total saponins extracted from AC (ACS) are a compound of triterpenoid saponins, which have protective properties against both chemical and immunological liver injuries. Nevertheless, ACS has not been proven to have an influence on drug-induced liver injury (DILI). AIM OF THE STUDY: This study used network pharmacology and experiments to investigate the effects of ACS on acetaminophen (APAP)-induced liver injury. MATERIALS AND METHODS: The targets associated with ACS and DILI were obtained from online databases. Cytoscape software was utilized to construct a "compound-target" network. In addition, Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were used to analyze the related signaling pathways impacted by ACS. AutoDock Vina was utilized to evaluate the binding affinity between bioactive compounds and the key targets. To validate the findings of network pharmacology, in vitro and in vivo experiments were conducted. Cell viability assay, transaminase activity detection, immunofluorescence assay, immunohistochemistry staining, RT-qPCR, and western blotting were utilized to explore the effects of ACS. RESULTS: 25 active compounds and 217 targets of ACS were screened, of which 94 common targets were considered as potential targets for ACS treating APAP-induced liver injury. GO and KEGG analyses showed that the effects of ACS exert their effects on liver injury through suppressing inflammatory response, oxidative stress, and apoptosis. Molecular docking results demonstrated that core active compounds of ACS were successfully docked to core targets such as CASP3, BCL2L1, MAPK8, MAPK14, PTGS2, and NOS2. In vitro experiments showed that ACS effectively attenuated APAP-induced damage through suppressing transaminase activity and attenuating apoptosis. Furthermore, in vivo studies demonstrated that ACS alleviated pathological changes in APAP-treated mice and attenuated inflammatory response. Additionally, ACS downregulated the expression of iNOS, COX2, and Caspase-3, and upregulated the expression of Bcl-2. ACS also suppressed the MAPK signaling pathway. CONCLUSIONS: This study demonstrated that ACS is a hepatoprotective drug through the combination of network pharmacology and in vitro and in vivo experiments. The findings reveal that ACS effectively attenuate APAP-induced oxidative stress, apoptosis, and inflammation through inhibiting the MAPK signaling pathway. Consequently, this research offers novel evidence supporting the potential preventive efficacy of ACS.

ABSTRACT

Solar heating is generally regarded as a clean and low-carbon heating method, while its high initial investment hinders its promotion in economically underdeveloped areas. With the implementation of the clean heating policy and the proposal of the carbon neutralization target, rural bulk coal heating in northern China is restricted. The Chinese government proposes to widely adopt solar heating to meet the heating demands of rural residents. In this research, the application of solar assisted heat pump systems in Beijing, Tianjin, Hebei and its surrounding areas in China is numerically simulated. A new evaluation method under the same initial investment constraint is proposed to verify its benefits throughout the entire life cycle. The results indicate that although solar thermal heating has the lowest environmental impact and carbon emissions among various heating methods, it is not the best solution to rural clean heating. The reason is that equal investment in other projects can bring much more benefits, such as roof solar photovoltaic. In contrast to the air source heat pump and photovoltaic panel scheme with the same initial investment, solar heating has obvious negative environmental impact, 53.3 % higher economic cost, 35.9 tons more carbon emissions, and 105.9 % higher roof area occupation. The sensitivity analysis of solar fraction, geographical coordinates, and energy price also supports the above findings. The recommendation is proposed to promote air source heat pumps or solar photovoltaic, rather than solar thermal collectors, so as to reduce the cost of rural clean heating and carbon emission reduction.

ABSTRACT

Nanozyme, with enzyme-mimicking activity and excellent stability, has attracted extensive attention. However, some inherent disadvantages, including poor dispersion, low selectivity, and insufficient peroxidase-like activity, still limit its further development. Therefore, an innovative bioconjugation of a nanozyme and natural enzyme was conducted. In the presence of graphene oxide (GO), histidine magnetic nanoparticles (H-Fe3O4) were first synthesized by a solvothermal method. The GO-supported H-Fe3O4 (GO@H-Fe3O4) exhibited superior dispersity and biocompatibility because GO was the carrier and possessed outstanding peroxidase-like activity because of the introduction of histidine. Furthermore, the mechanism of the peroxidase-like activity of GO@H-Fe3O4 was the generation of •OH. Uric acid oxidase (UAO) was selected as the model natural enzyme and covalently linked to GO@H-Fe3O4 with hydrophilic poly(ethylene glycol) as a linker. UAO could specifically catalyze the oxidation of uric acid (UA) to generate H2O2, and subsequently, the newly produced H2O2 oxidized the colorless 3,3',5,5'-tetramethylbenzidine (TMB) to blue ox-TMB under the catalysis of GO@H-Fe3O4. Based on the above cascade reaction, the GO@H-Fe3O4-linked UAO (GHFU) and GO@H-Fe3O4-linked ChOx (GHFC) were used for the detection of UA in serum samples and cholesterol (CS) in milk, respectively. The method based on GHFU exhibited a wide detection range (5-800 µM) and a low detection limit (1.5 µM) for UA, and the method based on GHFC exhibited a wide detection range (4-400 µM) and a low detection limit (1.13 µM) for CS. These results demonstrated that the proposed strategy had great potential in the field of clinical detection and food safety.

ABSTRACT

The COVID-19 pandemic created a significant economic decline and altered market behavior, forcing buyers and dealers online. The traditional local market merchants are not fully equipped with e-commerce business techniques and strategies, which is a barrier to their e-commerce behavior and success. The study aims to help small-medium firms adapt to an uncertain economic environment instead of reducing or shutting down business-like in Pakistan. From health to education, economy to domestic and social protection, various researches have been done since 2020. The researcher used primary data sources and did a Quantitative study after collecting the 240 samples size of data from the successful e-commerce players of Pakistan. The results confirm that customer satisfaction is essential for entrepreneurs to succeed, as customers were not satisfied with online shopping during COVID-19. Customer knowledge management (CKM) and job satisfaction (JS) are studied as potential and realized capacity variables. CKM act as a strategic asset to collect and assimilate the external customer knowledge. In contrast, satisfied employees act as a valuable asset that dynamically responds to changing customer needs and business environment by efficiently utilizing their knowledge and skills and reaching business success which is mirrored in customer satisfaction. Conclusive results enable practitioners to perceive the business success during economic crises in the organization's absorptive capacity.

ABSTRACT

The Yellow River is an important ecological shelter zone and economic belt in China. However, rapid urbanization and industrialization has produced a fragile ecological environment conditions and unbalanced economic development in the Yellow River Basin (YRB). Ecological protection and high-quality development of the YRB has been China's national strategy since 2019. As the only coastal province with the largest economy and population in the YRB, the sustainable development of Shandong Province is of great importance in the region. This study evaluated the dynamic trend of sustainability levels of the nine cities in Shandong Province in the YRB through emergy analysis. Emergy-based indicators were established and analyzed from 2010 to 2019, taking account of the ecological service emergy (ESE) needed to dilute pollutants and emergy equivalent loss (EEL) on ecosystem quality and human health damage. Results showed that emergy sustainable indicators (ESI) in Tai'an, Heze, Dezhou, and Liaocheng ranged from 1 to 10, which had the potential for sustainable development. The ESI value of Jinan, Jining, Zibo, Dongying, and Binzhou was less than 1, which indicated that these cities were under great ecological pressure. The value of emergy indicators for sustainable development (EISD) of the nine cities all declined from 2010 to 2016, but remained stable from 2017 to 2019. Based on findings from the emergy analysis regarding policy implications and local conditions, the study concludes by providing proposals to improve regional sustainability.

ABSTRACT

The ecological conservation and high-quality development of China's Yellow River Basin is a national strategy proposed in 2019. Under China's goal of achieving a carbon peak by 2030 and carbon neutrality by 2060, clarifying the carbon footprint of each province and the transfer paths of embodied carbon emissions is crucial to the carbon reduction strategy for this region. This paper uses input-output model and multi-regional input-output model to account for the carbon footprint of nine provinces in the Yellow River Basin, and to estimate the amount of embodied carbon transfer between provinces and industrial sectors. Social network analysis is applied to identify the critical industries in the inter-provincial embodied carbon emission transfers from the three major industries. We found that the per capita carbon footprint of the Yellow River Basin decreased by 23.4% in 2017 compared to 2012. Among the sectoral composition of the carbon footprint of each province, "Processing and manufacturing of petroleum, coking, nuclear fuel, and chemical products", "Construction", "Other services", and "Metal processing and metal, non-metallic products" are the four sectors with a higher proportion of emissions. The embodied carbon emission transfer between the provinces in middle and lower reaches of the Yellow River Basin is much higher than that between the upstream provinces. Among carbon emission transfer network of three major industries in nine provinces,the secondary industry in Shaanxi has the highest centrality and is the most critical industry. This study provides a theoretical basis and data support for formulating carbon emission reduction plans in the Yellow River Basin.

ABSTRACT

Hepatocellular carcinoma is one of the malignancies worldwide with a high mortality rate and an increasing incidence. Molecular Targeted agents are its common first-line treatment. Organoid technology, as a cutting-edge technology, is gradually being applied in the development of therapeutic oncology. Organoid models can be used to perform sensitivity screening of targeted drugs to facilitate the development of innovative therapeutic agents for the treatment of hepatocellular carcinoma. The purpose of this review is to provide an overview of the opportunities and challenges of hepatocellular carcinoma organoids in targeted drug sensitivity testing as well as a future outlook.

ABSTRACT

The subaperture processing is one of the essential strategies for low frequency ultrawideband synthetic aperture radar (LF UWB SAR) imaging, especially for the real-time LF UWB SAR imaging because it can improve the parallelization of the imaging algorithm. However, due to the longer synthetic aperture of LF UWB SAR, the traditional subaperture imaging encounters an azimuth ambiguities problem, which severely degrades the focused quality of the imaging results. In this paper, the reason for the presence of azimuth ambiguities in the LF UWB SAR subaperture imaging and its influence on image quality is first analyzed in theory. Then, an extended subaperture imaging method based on the extension of subaperture length before Range Cell Migration Correction (RCMC) was proposed. By lengthening the subaperture length, the azimuth ambiguities are effectively eliminated. Finally, the extended part of subaperture is wiped off before the azimuth compression (AC), and the LF UWB SAR image of high focused quality is obtained. The correctness of the theory analysis and the effectiveness of the proposed method have been validated through simulated and real LF UWB SAR data.
