ABSTRACT

Pesticides, which are associated with endocrine dysfunction, immunological dysregulation, and cancer, are widespread sources of drinking water contamination. The state of Paraná has a population of 11 million, is the second largest grain producer in Brazil and is a leading consumer of pesticides. In this study, we analyzed the extent of drinking water contamination from 11 proven, probable, or potentially carcinogenic pesticides (alachlor, aldrin-dieldrin, atrazine, chlordane, DDT-DDD-DDE, diuron, glyphosate-AMPA, lindane-γ-HCH, mancozeb-ETU, molinate, and trifluralin) in 127 grain-producing municipalities in the state of Paraná. Extensive contamination of drinking water was found, including legacy pesticides such as aldrin-dieldrin (mean 0.047 ppb), DDT-DDD-DDE (mean: 0.07), chlordane (mean: 0.181), and lindane-HCH (mean: 2.17). Most of the municipalities were significantly above the maximum limits for each one of the currently allowed pesticides (67% for alachlor, 9.44% for atrazine, 96.85% for diuron, 100% for glyphosate-AMPA, 80.31% for mancozeb-ETU, 91.33% for molinate, and 12.6% for trifluralin). Ninety-seven percent of municipalities presented a sum of all pesticides at levels significantly above (189.84 ppb) the European Union preconized limits (<0.5 ppb). Using the mean pesticide concentration in water (ppb), the exposed population for each municipality, and the benchmark cancer risk for pesticides, we estimated the minimum number of cancer cases attributable to pesticide-contaminated drinking water during the period (total of 542 cases). More than 80% were attributed to mancozeb-ETU and diuron. Glyphosate-AMPA and diuron-attributable cases strongly correlated with the total cancer cases in the same period (R = 0.8117 and 0.8138, respectively) as well as with breast cancer cases (R = 0.7695 and 0.7551, respectively). Water contamination was significantly correlated with the sum of the estimated cancer cases for all 11 pesticides detected in each city (R = 0.58 and p < 0.0001). These findings reveal extensive contamination of drinking water in the state of Paraná and suggest that contamination may increase the risk of cancer in this region.

ABSTRACT

Post-transcriptional (PtscM) and post-translational (PtrnM) modifications of nucleotides and amino acids are covalent modifications able to change physio-chemical properties of RNAs and proteins. In the ribosome, the adequate assembly of rRNAs and ribosomal protein subunits in the nucleolus ensures suitable translational activity, with protein synthesis tuned according to intracellular demands of energy production, replication, proliferation, and growth. Disruption in the regulatory control of PtscM and PtrnM can impair ribosome biogenesis and ribosome function. Ribosomal impairment may, in turn, impact the synthesis of proteins engaged in functions as varied as telomere maintenance, apoptosis, and DNA repair, as well as intersect with mitochondria and telomerase activity. These cellular processes often malfunction in carcinogenesis and senescence. Here we discuss regulatory mechanisms of PtscMs and PtrnMs on ribosomal function. We also address chemical modification in rRNAs and their impacts on cellular metabolism, replication control, and senescence. Further, we highlight similarities and differences of PtscMs and PtrnMs in ribosomal intermediates during aging and carcinogenesis. Understanding these regulatory mechanisms may uncover critical steps for the development of more efficient oncologic and anti-aging therapies.

ABSTRACT

Brazil has a herd of over 9 million goats, and the northeast of Brazil is home to over 93% of this herd. Caprine mycoplasmosis are widely disseminated worldwide, being highly contagious with high rates of morbidity and mortality, causing considerable economic loss to goat herders. In addition, there has been a lack of research using molecular testing to monitor the health and detect Mollicutes in this herd in Brazil. Therefore, the aim of this study is to associate animal management with the presence of the caprine origin Mollicutes in goats, in the southwest region of the state of Bahia, Brazil. A cross-sectional study was conducted on twelve farms, and statistical analyses were performed to identify associations between the presence of Mollicutes and the management of goats. Molecular testing identified Mollicutes class, Mycoplasma agalactiae (Ma) and M. conjunctivae (Mc), in the samples analyzed. Statistical associations were observed between animals from intensive livestock facilities and the presence of Mollicutes in nasal samples and dairy ranch animals and the presence of Mollicutes in ocular samples and animals from extensive ranching sites and positive results of Mollicutes in genital samples. We conclude that mycoplasmas are present in goat herds in the southwestern region of Bahia, which supports the need for more focused studies of mycoplasmas throughout the country. Our research also demonstrated the presence of two important opportunistic bacteria, Mc and Ma, and, to the best of our knowledge, this is the first time that M. conjunctivae was detected in Brazilian goats by molecular testing.

ABSTRACT

Cytokines act as protein mediators of the immune system and exert pleiotropic effects on the source cells and/or on target cells. Cytokines are formed in a cascade, bind to specific receptors, and influence the activity, differentiation, proliferation, and survival of immune cells of both T helper 1 (Th1) type (which has proinflammatory properties) and Th2 type (with an anti-inflammatory function). The female reproductive system is regulated by the immune system via cytokines at various physiological stages: during the ovarian cycle, maternal recognition, embryo implantation, gestation, and birth, participating in homeostasis and protection from pathogens. These processes interact under the hormonal influence of the hypothalamic­pituitary­gonadal axis. This review is aimed at addressing the involvement of some cytokines in female reproductive physiology, highlighting the maternal recognition of the embryo and implantation as immunologically important steps for fetal survival. The scientific knowledge on the role of cytokines in female reproduction processes, such as the Th1/Th2 balance and immune tolerance should advance the research in various fields of assisted reproduction in humans and animals, such as artificial insemination, embryo transfer, and in vitro fertilization. The same is true for the development of contraceptive methods and understanding of pathological processes such as uterine infections and autoimmune diseases.

The aim of this study was to identify and determine the prevalence of Eimeria species affecting sheep raised extensively in a semiarid region of Brazil. Fecal samples of native sheep were collected during the rainy and dry seasons. The degree of infection was determined by counting oocysts per gram (OPG) of feces, and the morphometric method was used for species identification. Oocysts were found in all the properties assessed, in which 68.3% of the animals were infected. The prevalence of oocysts was influenced by the season and animal category (P<0.05). It was higher during the rainy season than the dry season (80.2% vs. 55.8%) and highest in young animals than the adults animals (68.2% vs. 39.6%). The OPG was lower during the dry season (1,269 ± 312 vs. 4,400 ± 1,122). Ten species were found; of these, E. ovinoidalis, E. granulosa, E. faurei, and E. crandallis were the most frequent. E. ovinoidalis and E. crandallis were found in all properties, with their prevalences being 19.4% and 13.6% respectively. The high prevalence of pathogenic species shows that eimeriosis is a risk for animals raised extensively in the semiarid region.

Resumo Objetivou-se neste estudo identificar e determinar a prevalência de espécies de Eimeria que parasitam ovinos criados extensivamente em região semi-árida. Amostras de fezes de ovinos nativos foram coletados durante as estações chuvosa e seca. O grau de infecção foi determinado pela contagem de oocistos por grama de fezes (OoPG)e o método morfométrico foi utilizado para a identificação das espécies. Foram encontrados oocistos em todas os rebanhos avaliados e observou-se que 68,3% dos animais estavam infectados. A prevalência de oocistos foi influenciada pela estação climática e pela categoria dos animais (P<0,05), sendo mais alta durante a estação chuvosa em relação a estação seca (80,2% vs. 55,8%) e em animais jovens em relação aos animais adultos (68,2% vs. 39,6%). O OoPG foi menor durante a estação seca (1.269 ± 312 vs. 4.400 ± 1.122). Dez espécies foram encontradas sendo a E. ovinoidalis, E. granulosa, E. faurei, e E. crandallis as mais frequentes. E. ovinoidalis e E. crandallis foram encontrados em todas as propriedades, com prevalências de 19,4% e 13,6%, respectivamente. A alta prevalência de espécies patogênicas mostra que eimeriose é um risco para os animais criados extensivamente na região semiárida.

ABSTRACT

The aim of this brief review is to clarify the role of melatonin in the production and preservation of mammalian gametes and embryos. Melatonin is an indoleamine synthesized from tryptophan in the pineal gland and other organs that operates as a hypothalamic-pituitary-gonadal axis modulator and regulates the waxing and waning of seasonal reproductive competence in photoperiodic mammals. A major function of the melatonin rhythm is to transmit information about the length of the daily photoperiod to the circadian and circannual systems in order to provide time-of-day and time-of-year information, respectively, to the organism. Melatonin is also a powerful antioxidant and anti-apoptotic agent, which is due to its direct scavenging of toxic oxygen derivatives and its ability to reduce the formation of reactive species. Mammalian gametes and embryos are highly vulnerable to oxidative stress due to the presence of high lipid levels; during artificial breeding procedures, these structures are exposed to dramatic changes in the microenvironment, which have a direct bearing on their function and viability. Free radicals influence the balance between oxidation-reduction reactions, disturb the transbilayer-phospholipid asymmetry of the plasma membrane and enhance lipid peroxidation. Melatonin, due to its amphiphilic nature, is undoubtedly useful in tissues by protecting them from free radical-mediated oxidative damage and cellular death. The supplementation of melatonin to semen extender or culture medium significantly improves sperm viability, oocyte competence and blastocyst development in vitro.

ABSTRACT

The objective of this research was to study the population genetic structure of four herds of Mediterranean buffaloes in Brazil. It was used pedigree data from 6,588 buffaloes of Mediterranean breed born from 1980 to 2002. Of the total number of animals studied, 60.5, 15.3 and 2.1% had a pedigree in the first, second and third ascendancy, respectively. The effective number of herds that provided breeding males was 1.60 for parents, 1.16 for grandparents and 1.00 for great-grandparents. There were 923 founder animals and only 71 effective founders. The effective number of ancestors explaining the genetic variability of the population was 71 and only 30 ancestors accounted for 50% of the genetic variability of the population. The average relatedness coefficient (AR) between individuals and inbreeding (F) of the population were estimated at 0.37 and 0.34% respectively. The average estimate of generation interval was 8.71±2.85 years. The variability of the current population is the result of a few ancestors, who are mostly also founders showing that the population was developed from a narrow genetic base which characterizes the occurrence of founder effect.


O objetivo deste trabalho foi estudar a estrutura genética populacional de bubalinos da raça Mediterrâneo, em quatros rebanhos, no Brasil. Foram utilizados dados de pedigree de 6.588 bubalinos da raça Mediterrâneo nascidos no período de 1980 a 2002. Do total de animais estudados, 60,5; 15,3 e 2,1% possuíam pedigree na primeira, segunda e terceira ascendência, respectivamente. O número efetivo de rebanhos que forneceram machos reprodutores foi de 1,60 para pais; 1,16 para avôs e 1,00 para bisavôs. O número de animais fundadores foi 923 e o número efetivo de fundadores foi apenas 71. O número efetivo de ancestrais que explicaram a variabilidade genética da população foi de 71 e somente 30 ancestrais explicaram 50% da variabilidade genética da população. Os coeficientes médios de relação (CR) entre os indivíduos da população e de endogamia (F) foram estimados em 0,37 e 0,34%, respectivamente. A estimativa média do intervalo de gerações foi de 8,71±2,85 anos. A variabilidade da população atual é fruto da contribuição de poucos ancestrais, que são, na sua maioria, também fundadores, evidenciando que a população se desenvolveu a partir de uma base genética estreita, o que caracteriza a ocorrência do efeito fundador.

ABSTRACT

Com o objetivo de verificar a eficiência do efeito Doppler como método de diagnóstico precoce da gestação em caprinos, 13cabras da raça Saanen foram monitoradas aos 30, 45, 60, 75, 90, 105 e 120 dias de gestação, utilizando-se um equipamentobaseado no efeito Doppler e comparando-se à ultra-sonografia em tempo real. Quanto ao percentual de acerto para gestaçãopositiva, foram verificadas diferenças estatísticas (P 0,05) entre os métodos aos 30, 45, 60, 75 e 105 dias de gestação, sendoque a ultra-sonografia em tempo real sempre apresentou resultados superiores. Comparando-se a eficiência do efeitoDoppler, entre as idades gestacionais, verificou-se um baixo percentual de acerto aos 30 (0%) e 45 (22,2%) dias de gestação,aumentando este percentual a partir dos 60 dias pós-monta (66,7%). Para esse parâmetro, quando do uso do efeito Doppler,foram verificadas diferenças estatísticas (P 0,05) aos 90 (55,6% vs 22,2%) e aos 120 (55,6% vs 33,3%) dias de idadegestacional em cabras com gestação múltipla ou simples, respectivamente. Observou-se uma maior freqüência cardíacamaterna nas cabras com gestação múltipla aos 105 e 120 dias de idade gestacional, quando comparadas com as degestação simples. ABSTRACT

O objetivo deste trabalho foi o de determinar a concentraçäo hidrogeniônica (pH), acidez total titulável e tempo de reduçäo do azul de metileno no fluido ruminal de caprinos com ou sem raça definida, mantidos em pastagens artificiais, exclusivas de gramíneas, ou em caatinga, durante as épocas chuvosa e seca do ano. O delineamento experimental foi inteiramente casualizado, sendo analisado em esquema fatorial 23 (animais com e sem raça definida, pastagens artificiais e em caatinga, épocas chuvosa e seca do ano). O tipo de pastagem influenciou significativamente (p < 0,05) os valores encontrados para acidez total titulável. Em relaçäo ao pH, o tipo de pastagem e época do ano foram significativamente (p < 0,05) os fatores mais incisivos. A atividade microbiana, avaliada através dos parâmetros estudados, foi mais intensa na época das águas e na caatinga em relaçao à da seca e pastagens artificiais, respectivamente. Na avaliaçäo dos resultados desses testes, deve-se sempre levar em consideraçäo principalmente a época do ano e o tipo de pastagem, sem, no entanto, desprezar o fator raça
