ABSTRACT

Sex steroids are pervasive in mammals and evolutionarily conserved, but differences in the nuances of endocrine profiles characterize distinct species. Two sex steroids, testosterone and progesterone, feature prominently in the life history of mammalian taxa, but neither one has been analyzed from wild giraffes. Our study was designed to address this gap in knowledge by examining how these sex steroids are related to biological features and giraffe life history. We conducted the research at Rooipoort Nature Reserve a 44,000 ha private nature reserve in South Africa on a population of South African giraffes (Giraffa camelopardalis giraffa). Eleven adult giraffe cows and seven adult bulls were immobilized and various biological samples and morphological measurements were obtained. We analyzed both testosterone and progesterone using Ultra High Performance Liquid Chromotography - Tandem Mass Spectrometry (UHPLC-MS/MS). We found that non-pregnant females had lower progesterone concentrations than pregnant females and that those in the last trimester of pregnancy showed a slight drop in progesterone, as well as an increase in testosterone. Among males, chronological age was correlated with testicle size, testosterone concentrations, and ossicone volume. We propose that the progesterone decline functions partly to accelerate resumption of ovulation because giraffes become pregnant while lactating, and that the testosterone elevation provides an endocrine mileu for female defense of neonatal calves, given that lion predation is a major threat to calf survival. We suggest that male reproductive strategies are mediated by the age-related impacts of testosterone on growth in both body mass and ossicone volume as a consequence of sexual selection. We conclude by noting that the robust and solid ossicones of male giraffes function in mate competition as wedges that are used to topple opponents by raising their legs and placing them in an off-balanced position that can seriously injure them when falling to the ground.

ABSTRACT

Diet selection concerning browse availability of giraffes (Giraffa camelopardalis) was studied over 15 months in an arid environment in South Africa. A global positioning system collar was fitted to a giraffe individual to assess the specific areas, consisting of different vegetation types, that the population utilised during different seasons. Results are provided on diet selection in relation to browse availability between seasons and vegetation types, including tree densities and the amount of the total evapotranspiration tree equivalents. Diet selections of the giraffe population changed in response to the availability of browse material from July to October. The availability of important resource areas had a significant (p < 0.05) effect on the spatial ecology, and an increase in home range size was noted. Information that is important for the well-being of giraffes was identified. This included nutritional stress and the limited variety of the most utilised tree species available for browsing, especially during critical dry periods. The results demonstrate the importance of assessment of giraffes' diet selection in relation to browse availability, especially before introduction to a new area, to limit the lack of population growth and underperformance. This study provides valuable information towards understanding the resources and habitats required for successful giraffe management.

ABSTRACT

Elephant milk composition is unique, as are its changes over lactation. Presented here is the milk non-dedicated metabolite composition of three African elephants. Their lactation times are overlapping and span day one to thirty months. Metabolites were identified and quantified by 1H nuclear magnetic resonance spectroscopy. Lactose and short oligosaccharides are a large component of the metabolites, with lacto-N-difucohexaose I as the major oligosaccharide. These were followed by metabolites of lipids, amino acids, and the citric acid cycle. The content of lactose, lacto-N-difucohexaose I, 2'-fucosyllactose, and some unidentified oligosaccharides decrease over lactation, while that of difucosyllactose and other unidentified ones increase. The high content of glutamate, as a glucogenic amino acid, supported the uprated synthesis of saccharides by the milk gland cells. The content of succinate and choline increase over lactation, indicating higher energy expenditure and phospholipid synthesis during later lactation.

ABSTRACT

Adult giraffes reach heights of 4.5 m with a heart-to-head distance of over 2 m, making cranial blood supply challenging. Ultrasound confirmed that the giraffe jugular vein collapses during head movement from ground level to fully erect, negating the possibility of a siphon mechanism in the neck. We showed that a short-length siphon structure over a simulated head-to-heart distance for a giraffe significantly influences flow in a collapsible tube. The siphon structure is determined according to brain case measurements. The short-length siphon structure in a shorter-necked ostrich showed no significant increase in flow. The shorter head-to-heart distance might be the reason for the lack of effect in ostriches. A siphon mechanism situated in the cranium is certainly possible, with a significant effect exerted on the amount of pressure the heart must generate to allow adequate cranial blood perfusion in a long-necked giraffe. The study validated that a cranial-bound siphon structure can operate and will be of significant value for adequate cranial blood perfusion in long-necked species such as giraffes and might also have existed in extinct species of long-necked dinosaurs.

ABSTRACT

One of the highest occurrences of mortalities among giraffes (Giraffa camelopardalis) takes place during immobilisations, captures and translocations. Common mistakes, human error, unforeseen risks, the awkward anatomy and the sheer size of the animal are leading factors for giraffes' mortalities during these operations. Many risks can be circumvented but some risks are unpreventable, often due to terrain characteristics (rivers, deep ditches, holes and rocky terrain). From 2011 to 2021, seventy-five giraffes were successfully immobilised and captured to collect biological and physiological data from eight different study areas across South Africa. A 0% mortality and injury rate was achieved and, therefore, the techniques described in this paper are testimony to the advances and improvements of capture techniques and drugs. Biological information and capture experiences were noted for 75 immobilised giraffes, of which, knockdown time data were recorded for 43 individuals. Effective and safe immobilisation requires a competent team, proper planning, skill and knowledge. In this manuscript, we address procedures, techniques, ethical compliance, welfare and safety of the study animals. General experiences and lessons learned are also shared and should benefit future captures and immobilisations by limiting the risks involved. The sharing of experiences and information could influence and improve critical assessments of different capture techniques and can likely contribute to the success rate of immobilisation and translocation success for giraffes in the future.

ABSTRACT

The proximate composition of milk from fifteen free-ranging white rhinoceroses at different lactation stages is reported with detailed analysis of fatty acid composition and minerals. Lactose is the main component at 7.93 ± 0.53%, followed by 0.93 ± 0.19% protein, 1.76 ± 0.39% fat, 0.40 ± 0.18% ash, 0.05 ± 0.01% glucose, and 0.04 ± 0.02% non protein nitrogen The interindividual variation of all the components is high, showing no trend of change over lactation. The K and P content decreased over lactation. The fatty acid composition of rhinoceros milk is characteristic with a high saturated fatty acids content of 62%-84%, of which the medium chain fatty acids form the major portion. The C10:0, C12:0, C16:0, and C18:1c9 are the major fatty acids in the milk fat. The results are compared with the two other Perissodactylae families, the Equidae and Tapiridae. Differences in gross composition are small, but the milk of the Rhinocerotidae have the lowest gross energy, while the milk fats consist of the highest amounts of saturated fatty acids, while the low levels of C16:1c9 and C18:1c9 indicate the lowest mammary Δ9-desaturase activity.

ABSTRACT

Giraffes are an important tourist attraction, and human presence to wildlife is increasing. This has an impact on an animal's behavior and its endocrine correlates. Studies on other species show alterations in movement patterns, vigilance, and stress-related hormone levels in the presence of humans. Limited information is available on how anthropogenic activities alter giraffe's behavior, social structure, and related endocrine parameters. The purpose of this study was to obtain insight into anthropogenic influences on giraffe's behavior and adrenal activity. We used GPS devices mounted onto giraffes to compare the distance walked in the presence or absence of human observers. We also conducted behavioral observations to assess their vigilance and collected fecal samples to analyze their fecal glucocorticoid metabolite (fGCM) concentrations. Giraffes walked significantly further distances in the presence of humans, but the cumulative time that observers were present decreased the hourly distance walked with an observer present, suggesting that the giraffes were becoming habituated. The number of observers present significantly increased the percentage of time spent on observing an observer as well as the number of unhabituated individuals present in the herd. The percentage of time spent observing a human observer did not decrease with the increase of habituation. Last, fGCM concentrations increased with human presence but decreased when individuals became habituated to human presence. More research is needed to understand the effect of anthropogenic influences in different scenarios (e.g., tourism, vehicles, hunting, etc.).

ABSTRACT

The major nutrient and fatty acid composition of the milk of impala and tsessebe is reported and compared with other Bovidae and species. The proximate composition of impala milk was 5.56 ± 1.96% fat, 6.60 ± 0.51% protein, and 4.36 ± 0.94% lactose, and that of tsessebe milk was 8.44 ± 3.19%, 5.15 ± 0.49%, and 6.10 ± 3.85%, respectively. The high protein content of impala milk accounted for 42% of gross energy, which is typical for African Bovids that use a "hider" postnatal care system, compared to the 25% of the tsessebe, a "follower". Electrophoresis showed that the molecular size and surface charge of the tsessebe caseins resembled that of other Alcelaphinae members, while that of the impala resembled that of Hippotraginae. The milk composition of these two species was compared by statistical methods with 13 other species representing eight suborders, families, or subfamilies of African Artiodactyla. This showed that the tsessebe milk resembled that of four other species of the Alcelaphinae sub-family and that the milk of this sub-family differs from other Artiodactyla by its specific margins of nutrient contents and milk fat with a high content of medium-length fatty acids (C8-C12) above 17% of the total fatty acids.

ABSTRACT

Disorders of sexual development (DSD) in wild mammals are rarely described. A male South African giraffe (Giraffa camelopardalis giraffa) was identified with bilateral cryptorchidism. The testes were intra-abdominal, smaller and less ovoid than in normal male giraffes. The right testis was situated more cranially than the left and connected to a longer deferent duct with normal ampullae. One distended vesicular gland filled with mucoid material was identified. A short penis, situated in the perineal area, was directed caudally and presented hypospadias. Histologically, testicular hypoplasia was present; the epididymis tubules contained no spermatozoa and the deferent duct and vesicular gland were inflamed. The blood testosterone concentration was 16.27 nmol/L and oestrone sulphate concentration was 0.03 ng/mL. The aetiology of the abnormalities is unknown.

ABSTRACT

The study of animal locomotion can be logistically challenging, especially in the case of large or unhandleable animals in uncontrolled environments. Here we demonstrate the utility of a low cost unmanned aerial vehicle (UAV) in measuring two-dimensional running kinematics from free-roaming giraffes (Giraffa camelopardalis giraffa) in the Free State Province, South Africa. We collected 120 Hz video of running giraffes, and calibrated each video frame using metatarsal length as a constant object of scale. We tested a number of methods to measure metatarsal length. The method with the least variation used close range photography and a trigonometric equation to spatially calibrate the still image, and derive metatarsal length. In the absence of this option, a spatially calibrated surface model of the study terrain was used to estimate topographical dimensions in video footage of interest. Data for the terrain models were collected using the same equipment, during the same study period. We subsequently validated the accuracy of the UAV method by comparing similar speed measurements of a human subject running on a treadmill, with treadmill speed. At 8 m focal distance we observed an error of 8% between the two measures of speed. This error was greater at a shorter focal distance, and when the subject was not in the central field of view. We recommend that future users maximise the camera focal distance, and keep the subject in the central field of view. The studied giraffes used a grounded rotary gallop with a speed range of 3.4-6.9 ms-1 (never cantering, trotting or pacing), and lower duty factors when compared with other cursorial quadrupeds. As this pattern might result in adverse increases in peak vertical limb forces with speed, it was notable to find that contralateral limbs became more in-phase with speed. Considering the latter pattern and the modest maximal speed of giraffes, we speculate that tissue safety factors are maintained within tolerable bounds this way. Furthermore, the angular kinematics of the neck were frequently isolated from the pitching of the body during running; this may be a result of the large mass of the head and neck. Further field experiments and biomechanical models are needed to robustly test these speculations.

ABSTRACT

In a recent paper in Current Biology, Fennessy and colleagues [1] conclude that there are four species of giraffe and that their numbers are declining in Africa. Giraffes (Giraffa camelopardalis) are presently classified as one species, with nine subspecies, which are considered 'Vulnerable' on the IUCN Red List [2]. The present consensus of one species divided into nine subspecies has previously been questioned (Supplemental information), and Fennessy and colleagues [1] provide another viewpoint on giraffe taxonomy. The fundamental reason for different taxonomic interpretations is that they are based upon different datasets that adopt different statistical techniques and follow different criteria for nomenclature.

BACKGROUND: The current taxonomy of the African giraffe (Giraffa camelopardalis) is primarily based on pelage pattern and geographic distribution, and nine subspecies are currently recognized. Although genetic studies have been conducted, their resolution is low, mainly due to limited sampling. Detailed knowledge about the genetic variation and phylogeography of the South African giraffe (G. c. giraffa) and the Angolan giraffe (G. c. angolensis) is lacking. We investigate genetic variation among giraffe matrilines by increased sampling, with a focus on giraffe key areas in southern Africa. RESULTS: The 1,562 nucleotides long mitochondrial DNA dataset (cytochrome b and partial control region) comprises 138 parsimony informative sites among 161 giraffe individuals from eight populations. We additionally included two okapis as an outgroup. The analyses of the maternally inherited sequences reveal a deep divergence between northern and southern giraffe populations in Africa, and a general pattern of distinct matrilineal clades corresponding to their geographic distribution. Divergence time estimates among giraffe populations place the deepest splits at several hundred thousand years ago. CONCLUSIONS: Our increased sampling in southern Africa suggests that the distribution ranges of the Angolan and South African giraffe need to be redefined. Knowledge about the phylogeography and genetic variation of these two maternal lineages is crucial for the development of appropriate management strategies.
