ABSTRACT

This study aimed to investigate the effects of dietary semen vaccariae extracts (SVE) on the production performance, colostrum components, and relative gene expression related to mammogenesis of lactating sows. 48 pregnant sows were selected and randomly allocated into four groups, with six replicates and two sows per replicate. The first group was the control (CON), while the other groups received the same diet further supplemented with 1.5, 3.0 and 4.5 g SVE per kg (SV1, SV2 and SV3, respectively). Compared with the control group, (1) the average daily gain was increased (p < 0.05) in SV1, SV2, and SV3 during the 11-21 days and 1-21 days of lactation; (2) the serum insulin-like growth factor-1, insulin, prolactin, and estrogen contents in SV1, SV2, and SV3 were increased (p < 0.05) on the 1st and 21st day of lactation; (3) The plasma Lysine, Threonine, and Tryptophan concentrations were also higher (p < 0.05) in SV1, SV2, and SV3 on the 1st and 21st day of lactation; (4) The milk Lysine, Methionine, Threonine, and Tryptophan concentrations were higher (p < 0.05) in SV1, SV2, and SV3 on the 1st and 21st day of lactation; (5) The milk lactose ratio and milk protein content were increased (p < 0.05) in the groups treated with semen vaccariae on the 1st day of lactation, while the milkfat ratio and milk protein content were increased (p < 0.05) in SV2 and SV3 on the 21st day of lactation; (6) the immunoglobulin M, A, and G contents were increased (p < 0.05) in the groups treated with the semen vaccariae on the first day of lactation; and (7) the relative PRLR, STAT5a, FcRn, CSN2, and LALBA expressions were higher (p < 0.05) in the groups treated with the semen vaccariae on the 1st and 21st day of lactation. In this study, the optimum dosage was 3.0 g/kg semen vaccariae, which increased the average daily gain of piglets, total lactation yield, and serum hormone levels, improved the amino acid levels in plasma, and facilitated the milk quality, up-regulated the relative gene expressions in the mammogenesis.

ABSTRACT

The aims of this study were to test the effects of dietary probiotics and acidifiers on the production performance, colostrum components, serum antioxidant activity and hormone levels, and gene expression in the mammary tissue of lactating sows. Four treatments were administered with six replicates to 24 lactating sows. The control group (GC) received a basal diet, while the experimental groups received a basal diet with 200 mL/d probiotics (GP), 0.5% acidifiers (GA), and 200 mL/d probiotics + 0.5% acidifiers (GM), respectively. Compared with the GC, (1) the average weight of the piglets on the 21st day of lactation in the GM was higher (p < 0.05); (2) the colostrum fat ratio increased significantly (p < 0.05); (3) the malondialdehyde levels in GP and GM were lower (p < 0.05) on the 11th day; (4) on the 1st, 11th, and 21st days, the prolactin in GP and GM increased (p < 0.05); (5) on the 21st day, the relative expression levels of the prolactin receptor and fatty acid synthase were increased (p < 0.05). In summary, the basal diet mixed with 200 mL/d probiotics + 0.5% acidifiers could improve the production performance, colostrum components, serum antioxidant activity, and hormone levels of lactating sows.

ABSTRACT

Preference and performance trials were conducted to investigate the effects of extruded corn with different degrees of gelatinization on the feed preference, growth performance, nutrient digestibility, and fecal microbiota of weaning piglets. In the preference trial, 144 piglets who were 35 days old were weighed and allotted to six treatments with four replications per treatment. Piglets in each treatment group were allowed to choose two of the following four corn-supplemented diets: conventional corn (NC) or extruded corn with low (LEC; 41.82% gelatinization), medium (MEC; 62.60% gelatinization), or high (HEC; 89.93% gelatinization) degrees of gelatinization for 18 days. The results showed that the piglets preferred diets supplemented with a low degree of gelatinization of extruded corn. In the performance trial, 144 piglets who were 35 days old were weighed and allotted into four treatments with six replications per treatment. Piglets in each treatment were fed one of the four diets for 28 days. The results showed that LEC and MEC decreased the feed:gain ratio at 14-28 days and 0-28 days, respectively, and increased the apparent total tract digestibility (ATTD) of crude protein compared with NC. Meanwhile, LEC increased the total protein and globulin content in the plasma on day 14, and MEC increased the ATTD of ether extract (EE) compared with NC. Extruded corn with low and medium degrees of gelatinization increased the abundance of Bacteroidetes at the phylum level and Lactobacillus, Alloprevotella, Prevotellaceae_UCG-03, and Prevotella_2 at the genus level. The results showed that extruded corn can improve feed preference, increase growth performance and nutrient digestibility, and modify gut microbiota, and the ideal degree of gelatinization is approximately 41.82-62.60%.

ABSTRACT

This study was conducted to investigate the effects of different supplementation levels of soybean hulls and corn stalk in high-fibre gestation diet on the performance, colostrum composition and faecal microbiota of sows. Forty first-farrowing Danish Landrace sows were randomly assigned to five dietary treatment groups. The control (CON, 3.15% crude fibre) group was fed a normal diet, and the treatment groups were soybean hulls low-fibre (SHL, 6.00% crude fibre) group, soybean hulls high-fibre (SHH, 8.00% crude fibre) group, corn stalk low-fibre (CSL, 6.00% crude fibre) group and corn stalk high-fibre (CSH, 8.00% crude fibre) group. The weaning weight of the litter and the average daily feed intake of the lactating sows in the SHL, SHH and CSH groups were higher than those in the CON group (p < 0.05). The immunoglobulin A and G levels of the colostrum in the SHL, SHH, CSL and CSH groups were higher than those in the CON group (p < 0.05), and the immunoglobulin M levels in the SHL, SHH and CSH groups were higher than those in the CON group (p < 0.05). The abundance of Proteobacteria at the phylum level in the CON group was higher than that in the CSL, CSH and SHH groups (p < 0.05). The abundance of Lactobacillaceae at the family level in the SHH and CSL groups were higher than that in the CON group (p < 0.05). The abundance of Lactobacillus at the genus level in the SHH and CSL groups were higher than that in the CON group (p < 0.05). In conclusion, SHH group had the best effect, and the optimal crude fibre level in the gestation diet of sows is 8%.

ABSTRACT

The effects of dietary resveratrol (0, 300, 400 and 500 mg/kg) on the chemical composition, antioxidative capacity, meat quality and volatile compound concentrations of duck meat were investigated. The results showed that dietary resveratrol had no significant effects on the chemical composition. Dietary resveratrol supplementation increased superoxide dismutase, glutathione peroxidase, catalase activity, pH15 min, pH24 h and color, and reduced the malondialdehyde, and carbonyl contents and shear force, thereby improving water mobility and distribution (T2b, T21, T22), drip loss, cooking loss and volatile compounds concentration of duck meat. In conclusion, dietary resveratrol supplementation improved the meat quality of ducks by enhancing the antioxidant capacity, improving the color and shear force, and suppressing lipid and protein oxidation, and 500 mg/kg dietary resveratrol had the greatest effect in this study.

ABSTRACT

The objectives of this study were to investigate the effects of soybean isoflavone (SI) and astragalus polysaccharide (APS) mixture on the colostrum components, serum antioxidant, immune and hormone levels of lactating sows. A total of 72 healthy Yorkshire × Landrace lactating sows, were randomly divided into four treatments with six replicates and three lactating sows for each replicate. The control group was fed the basal diet, while the experimental groups were fed the basal diet with 100, 200 and 300 mg/kg SI and APS mixture in the form of powder, respectively. Compared with the control group, (a) the total lactation yield of the 200 mg/kg group was significantly higher (p < 0.05) at 21 days, (b) there was no significant difference in colostrum composition, (c) TG, CHO and MDA content in each treatment group were significantly decreased (p < 0.05), (d) IgA, GH, IGF-1, TNF-α and SOD contents in the 200 mg/kg group were significantly increased (p < 0.05). The SI and APS mixture could improve the average daily feed intake, lactation yield, serum antioxidant activities, immune function, and hormone levels of lactating sows, and the optimum dosage in this study was 200 mg/kg.

ABSTRACT

This study aimed to investigate the effects of different supplementation levels of stevia residues in high-fiber diets on the fecal microorganisms of pregnant sows. Forty-eight first-farrowing Danish Landrace sows with similar weight, age, and due date were randomly divided into four groups. The control group was fed a normal diet (CON; 3.15% crude fiber content), and the treatment groups were supplemented with 20% stevia residue (SRL), 30% stevia residue (SRM), or 40% stevia residue (SRH) stevia residue, with crude fiber content of 7.79%, 9.15%, and 10.68%, respectively. The test period was 135 days, and Illumina Miseq high-throughput sequencing was used to test the fecal bacteria of the pregnant sows on day 56. Compared to the control group, species diversity was greater in the 30% stevia residue group. At the phylum level, Firmicutes and Spirochaetes had the greatest relative abundances; Firmicutes was most abundant in the SRM group, and Spirochaetes was most abundant in the CON group. At the genus level, Lactobacillus, Streptococcus, and Lachnospiraceae_XPB1014_group, and Treponema_2 had the highest relative abundances in the stevia-residue treatments. Among them, Lactobacillus and Treponema_2 were more abundant in SRL, Streptococcus was more abundant in SRH, and Lachnospiraceae_XPB1014_group was more abundant in SRM. Linear discriminant analysis effect size (LEfSe) showed that the bacterial species differed among the stevia residue treatments. SRL mainly included g__Lactobacillus and g__Romboutsi, SRM mainly included g__Lachnospiraceae_XPB1014_group and g__Ruminococcaceae_UCG_014, SRH mainly included g__Streptococcus, and CON mainly included g__Treponema_2, f__Clostridiaceae_1, g__norank_f__Muribaculaceae, and g__norank_f__p_251_o5. Stevia-residue supplementation significantly increased the abundance of beneficial bacteria, such as g__Lachnospiraceae_XPB1014_group, g__Christensenellaceae_R-7_group, and g__Ruminococcaceae_UCG-005 (p < 0.05), and significantly reduced the abundance of harmful bacteria, such as Treponema_2 (p < 0.05). Moreover, compared with the control group, the stevia-residues treatment groups reduced the feed cost 8.33%, 12.50%, and 13.33% per sow per day, respectively. In conclusion, high-fiber diets can improve the intestinal balance and evenness index of the intestinal flora in pregnant sows, promote the relative abundance of beneficial bacteria, reduce the relative abundance of harmful bacteria, and reduce feed cost. The optimal supplementation level of the stevia residue was found to be 30%.

ABSTRACT

The objective of this study was to clarify whether formation of nitrosylmyoglobin (MbFeIINO) by Lactobacillus fermentum AS1.1880 in meat is due to nitric oxide synthase (NOS) activity. Confocal laser scanning microscopy exhibited strong green fluorescence in the L. fermentum sample treated with a nitric oxide (NO)-specific probe, directly indicating that NO was produced. Furthermore, determination of NOS activity based on the presence of NO metabolites indicated the existence of NOS in L.fermentum. A NOS inhibitor, NG-nitro-L-arginine methyl ester, significantly inhibited the activity of NOS in L.fermentum (P < 0.05). Futhermore, NOS protein was detected in L.fermentum by Western blot analysis. L-arginine addition largely increased the NOS activity of L.fermentum (P < 0.05). In meat batters, the redness of a sample inoculated with L.fermentum was higher than that of the control and colour was significantly improved with the addition of L-arginine (P < 0.05), indicating that more MbFeIINO was formed.

ABSTRACT

This study investigated the effect of ultrasound-assisted immersion freezing (UIF) at different ultrasonic power levels on the myofibrillar protein primary, secondary and tertiary structures of common carp (Cyprinus carpio). Furthermore, protein thermal stability, electrophoresis pattern, and microstructure of the muscle tissue were also studied. Compared with a control, an ultrasonic power of less than 175 W had no significant negative effect on protein primary structure (P > 0.05), including total sulfhydryl, reactive sulfhydryl, carbonyl groups, free amino groups, dityrosine content, and surface hydrophobicity. UIF at 175 W (UIF-175) minimized the changes in protein secondary and tertiary structures. There were no obvious changes in the SDS-PAGE patterns of the control and frozen sample proteins. Microstructure analysis showed that an appropriate ultrasonic power (UIF-175) promoted the formation of smaller and more uniform ice crystals, reduced the damage of muscle tissue by ice crystals, and maintained the sarcomere integrity. In addition, UIF-175 samples had higher protein thermal stability. Overall, ultrasound treatment at a proper power (UIF-175) effectively minimized the changes in protein structure and protected the protein thermal stability during freezing process.

ABSTRACT

This study investigated the effects of multiple freeze-thaw (F-T) cycles on water mobility, microstructure damage and protein structure changes in porcine longissimus muscle. The transverse relaxation time T2 increased significantly when muscles were subjected to multiple F-T cycles (P<0.05), which means that immobile water shifted to free water and the free water mobility increased. Multiple F-T cycles caused sarcomere shortening, Z line fractures, and I band weakening and also led to microstructural destruction of muscle tissue. The decreased free amino group content and increased dityrosine in myofibrillar protein (MP) revealed that multiple F-T cycles caused protein cross-linking and oxidation. In addition, the results of size exclusion chromatography, circular dichroism spectra, UV absorption spectra, and intrinsic fluorescence spectroscopy indirectly proved that multiple F-T cycles could cause protein aggregation and degradation, α-helix structure disruption, hydrophobic domain exposure, and conformational changes of MP. Overall, repeated F-T cycles changed the protein structure and water distribution within meat.

ABSTRACT

The objective of this study was to investigate the physicochemical and structural properties of composite gels prepared with porcine myofibrillar protein (MP) and lard, glycerolized lard (GL) or purified glycerolized lard (PGL). The gels prepared with MP and GL or PGL had significantly higher penetration force and water-holding capacity (WHC) than the gel with lard (P<0.05) and formed a more compact and orderly microstructure. Compared with the distributions of T2 relaxation times of the pure MP gel, T21 and T22 of the gels that were prepared with GL or PGL moved in the direction of slower relaxation time, which suggests that the water mobility in the gel system was restricted. The presence of lard, GL and PGL did not affect the participating proteins in composite gels. The presence of GL and PGL altered the secondary and tertiary structures of MP in composite gels, which changed the gel properties. In general, the composite gels that were prepared with MP and GL or PGL showed improved gel quality.

ABSTRACT

This study was conducted to evaluate the contributions of fat addition levels and storage duration at -18 °C to the oxidation and physicochemical changes of frozen pork dumpling filler. With an increase in the fat addition, thiobarbituric acid-reactive substances and carbonyl production increased (P < 0.05), the transition temperatures (Tmax) shifted to lower temperatures, and the total enthalpy (ΔH) of protein denaturation reduced (P < 0.05). Dynamic rheological measurements revealed a compromised viscoelastic network in filler protein gels with increased fat levels and storage time. Increasing the fat level also increased cooking loss and decreased breaking strength (P < 0.05). The sensory results showed that the dumplings with a high fat level had significantly higher texture, juiciness, and overall acceptability scores (P < 0.05). The results suggest that the dumplings with a high fat level stored for long periods enhanced oxidation and cooking loss and decreased breaking strength; however, it is important to add proper fat considering the palatability.

ABSTRACT

This experiment was conducted to assess the antioxidant efficacy of black currant (Ribes nigrum L.) extract (BCE) in raw pork patties during chilled storage. The extracting conditions of frozen BCE including ethanol concentrations (0-100%) and extracting times (0.25-12h) were studied. BCE extracted with 40% ethanol for 2h had the highest anthocyanin content, the strongest radical scavenging activities as well as the second strongest reducing power. BCE was condensed and added to pork patties at 5, 10 or 20 g/kg. Compared with the control, BCE treatments significantly decreased the thiobarbituric acid-reactive substance values and carbonyls formation and reduced the sulfhydryl loss of pork patties in a dose-dependent manner (P<0.05), which showed that the BCE significantly inhibited lipid and protein oxidation. The BCE-treated patties showed significantly higher redness (P<0.05) than the control. The findings demonstrated strong potential for BCE as a natural antioxidant in meat and meat products.
