ABSTRACT

BACKGROUND: The bidirectional relationships between metabolic syndrome (MetS) and major depressive disorder (MDD) were discovered, but the influencing factors of the comorbidity were barely investigated. We aimed to fully explore the factors and their associations with MetS in MDD patients. METHODS: The data were retrieved from the electronic medical records of a tertiary psychiatric hospital in Beijing from 2016 to 2021. The influencing factors were firstly explored by univariate analysis and multivariate logistic regressions. The propensity score matching was used to reduce the selection bias of participants. Then, the Bayesian networks (BNs) with hill-climbing algorithm and maximum likelihood estimation were preformed to explore the relationships between influencing factors with MetS in MDD patients. RESULTS: Totally, 4126 eligible subjects were included in the data analysis. The proportion rate of MetS was 32.6 % (95 % CI: 31.2 %-34.1 %). The multivariate logistic regression suggested that recurrent depression, uric acid, duration of depression, marriage, education, number of hospitalizations were significantly associated with MetS. In the BNs, number of hospitalizations and uric acid were directly connected with MetS. Recurrent depression and family history psychiatric diseases were indirectly connected with MetS. The conditional probability of MetS in MDD patients with family history of psychiatric diseases, recurrent depression and two or more times of hospitalizations was 37.6 %. CONCLUSION: Using the BNs, we found that number of hospitalizations, recurrent depression and family history of psychiatric diseases contributed to the probability of MetS, which could help to make health strategies for specific MDD patients.

ABSTRACT

The M × N port wavelength-selective switch (WSS) is a crucial device used for Reconfigurable Optical Add/Drop Multiplexors and optical switching nodes in optical communication systems. The primary function of an M × N port WSS is to simultaneously transmit and switch multiple input optical signals from input fiber ports to output fiber ports through spatial light coupling. The port array module in a WSS that is responsible for coupling the spatial beam with the fiber determines the important parameters of the M × N port WSS, such as the number of input/output ports and insertion loss. In this paper, VirtualLab Fusion software 2023.1 (Build 1.558), as a powerful physical optics simulation tool, is used to design and optimize a silicon micro-lens array that can achieve the high-precision coupling of a fiber array with a pitch of 1143 µm. Finally, the designed micro-lens is manufactured and experimentally demonstrates its good beam focusing ability with a 3 dB insertion loss. The designed micro-lens array coupling system, which delivers 28 focused spots of approximately 1mm in size (the beam has a 1/e2 diameter) after transmitting a distance of around 300 mm, effectively extends the number of WSS ports. This design method of the micro-lens array significantly amplifies the port count of the M × N port wavelength-selective switch, effectively expanding it to encompass an impressive 28 × 28 ports.

ABSTRACT

Atom economy is one of the main concerns for material synthesis. Here, the facile synthesis of Lewis acidic boron-containing porous organic polymers (B-POPs) via hydroboration polymerization reaction of commercially available borane dimethyl sulfide complex (BH3 ∙SMe2 ) with multi-alkynes under mild reaction conditions is presented. This new synthetic method for B-POPs has the advantage of high atom economy. The resulted porous alkenyl borane polymers (PABPs) have unique features such as high boron content, strong Lewis acidity, and high surface areas. Owing to the strong Lewis acid-base interactions, PABPs exhibit excellent adsorptive capacity toward triethylamine (up to 841 mg g-1 ) and pyridine (up to 1396 mg g-1 ) vapor.

ABSTRACT

Autophagy has been documented to participate in immune responses and inflammatory diseases, but the mechanistic actions of monocyte autophagy in sepsis remain largely unknown. This study intends to analyze the mechanism of autophagy of peripheral blood monocyte cells (PBMCs) in sepsis based on single-cell RNA sequencing (scRNA-seq). The scRNA-seq data of PBMC samples from sepsis patients were downloaded from the GEO database, followed by identification of cell marker genes, key pathways and key genes. The bioinformatics analysis showed that the PBMC samples of sepsis patients mainly contained 9 immune cell types, among which three types of monocytes showed significant changes in cell numbers in sepsis patients. Of note, the highest autophagy score was found in the intermediate monocytes. The Annexin signaling pathway was a key pathway for the communication between monocytes and other cells. More importantly, SPI1 was predicted as a key gene in the autophagy phenotype of intermediate monocytes, and SPI1 might suppress ANXA1 transcription. The high expression of SPI1 in sepsis was confirmed by RT-qPCR and Western blot analysis. Dual luciferase reporter gene assay verified that SPI1 could bind to the promoter region of ANXA1. Furthermore, it was found that SPI1 might affect monocyte autophagy in the mouse model of sepsis through regulation of ANXA1. In conclusion, we provide insight into the mechanism underlying the septic potential of SPI1, which enhances monocyte autophagy by inhibiting the transcription of ANXA1 in sepsis.

ABSTRACT

Objective: To explore the correlation of serum IL-18, BDNF, and IL-1ß with depression and prognosis after acute exacerbation of chronic obstructive pulmonary disease (COPD). Methods: By means of retrospective analysis, the data of 240 patients at the acute exacerbation of COPD treated in our hospital (February 2018-February 2021) were analyzed. All patients received conventional treatment 1 d after admission, patients' serological indicators were measured before treatment, and after 30 d of follow-up, the patients were divided into the survival group (SG) and death group (DG) according to their clinical outcomes, the Beck's Depression Inventory (BDI) scores of the surviving patients were investigated, the correlation of IL-18, BDNF, and IL-1ß levels with depression was analyzed by R analytics, and the correlation of IL-18, BDNF, and IL-1ß levels with prognosis was analyzed by ROC curve analysis. Results: The results of 30 d follow-up showed that 220 patients survived (91.7%) and 20 patients died (8.3%). Among the surviving patients, 95 patients had depression and 125 patients did not have depression; the BDI scores of the depressed subjects and the nondepressed subjects were 10.35 ± 1.25 points and 2.06 ± 0.76 points, respectively; significant differences in IL-18, BDNF, and IL-1ß levels between SG and DG were observed (P < 0.05); significant differences in IL-18, BDNF, and IL-1ß levels between the depressed subjects and the nondepressed subjects were observed (538.43 ± 19.02 vs. 515.32 ± 9.65, 7.54 ± 0.56 vs. 12.11 ± 2.41, and 8.70 ± 0.98 vs. 8.12 ± 0.87; P < 0.001); among the depressed patients, the IL-18 and IL-1ß levels were positively correlative with the BDI scores (r = 0.781, r = 0.2583, P < 0.001, P = 0.012), and the BDNF level was negatively correlative with the BDI scores (r = -0.3277, P = 0.001) before treatment; according to the ROC analysis, the AUC (95% CI) of IL-18, BDNF, and IL-1ß in predicting prognosis was 0.8770 (0.8281-0.9260), 0.7723 (0.6879-0.8567), and 0.7165 (0.6080-0.8250) (P < 0.05), respectively. Conclusion: In regard to the depression in COPD patients after acute exacerbation, IL18 and IL-1ß show positive correlation, and BDNF presents negative correlation. All three indicators have predictive value for patient outcome.

ABSTRACT

BACKGROUND: Novel inhibitor of histone acetyltransferase repressor (NIR), a corepressor with a novel inhibitor of histone acetyltransferase (INHAT) activity, has been reported to be a negative modulator of p53 and a regulator of the cell cycle in cancer cells. However, the role of NIR in the progression of breast cancer remains elusive. MATERIALS AND METHODS: Oncomine database was used to analyze the mRNA levels and prognosis value of NIR in breast cancer. We performed loss-of-function and gain-of-function studies using lentivirus expressing shRNA targeting NIR, enhancer of zeste homolog 2 (EZH2) and forkhead box O3 (FOXO3) or lentivirus expressing NIR or FOXO3, respectively. Cell proliferation and colony formation assays were performed. Co-immunoprecipitation (Co-IP) and immunoprecipitation (IP) were performed to identify the interaction between NIR and polycomb repressive complex 2 (PRC2) subunits. ChIP assay was used to identify the enrichment of NIR, EZH2, H3K27ac and H3K27me3 at the FOXO3 promoter region and the regulation of H3K27 modification at the FOXO3 promoter by NIR. RESULTS: High levels of NIR expression were correlated with poor prognosis in breast cancer patients. Knockdown of NIR suppressed the proliferation of breast cancer cells. Mechanically, NIR was recruited by EZH2 to the promoter vicinity of FOXO3 via direct protein-protein interaction. Silencing NIR increased H3K27ac and decreased H3K27me3 levels at the FOXO3 promoter, resulting in enhancing FOXO3 expression. In accordance with this, growth inhibition of breast cancer cells caused by silencing of NIR could be reversed by FOXO3 knockdown. CONCLUSION: NIR knockdown inhibited proliferation by switching the H3K27me3 and H3K27ac marks at the FOXO3 promoter to promote FOXO3 transcription, and this effect depends on the physical interaction between NIR and PRC2 in breast cancer cells. Our results suggest that NIR might be a potential target for breast cancer treatment.

ABSTRACT

Polyvinyl alcohol (PVA)/carboxyl methyl cellulose sodium (CMC)/Na2CO3 composite films with different contents of Na2CO3 were prepared by blending and solution-casting. The effect of Na2CO3 on the microstructure of PVA/CMC composite film was analyzed by Fourier-transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), differential scanning calorimetry (DSC), and atomic force microscopy (AFM). Its macroscopic properties were analyzed by water sorption, solubility, and dielectric constant tests. The results show that the microstructure of PVA/CMC/Na2CO3 composite films was different from that of PVA and PVA/CMC composite films. In addition, compared to PVA and PVA/CMC composite films, the water sorption of PVA/CMC/Na2CO3 composite films relatively increased, the solubility in water significantly decreased, and the dielectric properties significantly improved. All these results indicate that the hydrogen bonding interaction between PVA and CMC increased and the crystallinity of PVA decreased after the addition of Na2CO3. This was also a direct factor leading to increased water sorption, decreased solubility, and enhanced dielectric properties. The reaction mechanism of PVA, CMC, and Na2CO3 is proposed to further evaluate the effect of Na2CO3 on the microstructure and macroscopic properties of PVA/CMC/Na2CO3 composite films.

ABSTRACT

OBJECTIVE: To evaluate the clinical efficacy of warm moxibustion therapy in the recovery of quadriceps muscle strength in patients undergoing total knee arthroplasty (TKA) with analgesia of the femoral nerve block (FNB). METHODS: A total of 174 patients with KOA were randomized into a warm moxibustion group and a rehabilitation group, 87 cases in each group. In the warm moxibustion group, warm moxibustion combined with conventional quadriceps strength training were used. In the rehabilitation group, conventional quadriceps strength training was given. The warm moxibustion was applied at Liangqiu (ST 34) and Zusanli (ST 36), the treatment was given twice a day, 7 days for one course, with a total of 2 courses.The quadriceps muscle strength of the two groups was recorded and compared at 24 h before FNB, 24, 48, 72 and 96 h after surgery, and the resting and exercise VAS pain scores were also recorded at the same time point. And the first time for standing up and the first straight raising time in the two groups were compared, and the occurrence of adverse reactions in the two groups were observed. RESULTS: At 24, 48, 72 and 96 h after FNB, the quadriceps muscle strength in the warm moxibustion group was better than that in the rehabilitation group (P<0.05, P<0.01). At 72 h and 96 h after FNB, the resting and exercise VAS scores of the warm moxibustion group were lower than those of the rehabilitation group (both P<0.001). The average first straight leg raising time in the warm moxibustion group was postoperative (31.03±10.78) h, and the time in the rehabilitation group was postoperative (47.23±15.78) h. The difference was statistically significant (P<0.001). The average time of the first time for standing up in the warm moxibustion group was postoperative (25.76±7.00) h, and postoperative (33.12±11.18) h in the rehabilitation group. The difference was also statistically significant (P<0.001). No adverse reactions occurred in both groups. CONCLUSION: Warm moxibustion combined with conventional quadriceps strength training can improve the symptoms of quadriceps weakness in patients with femoral nerve block after total knee arthroplasty, and accelerate the recovery of joint function, which is superior to conventional quadriceps strength training.

ABSTRACT

Echelle grating is a kind of special diffraction grating. Working with high diffraction orders and big diffraction angle, which has the advantages of high resolution and full wave shining. It has been widely used in high-end spectrum instrument, which greatly promoted the development of aerospace, astronomy, medical, military, environment and other cutting-edge technology. However, professional scoring system needs to be customized, and the price is very expensive. The use of sophisticated ultra precision machining equipment to process in the ladder grating can greatly reduce the preparation cost of the mother plate of the ladder grating. Due to the bad straightness and high accumulative error of ultra precision single point diamond lathe, it can't satisfy the demand of preparation when preparing the echelle grating, casuing the bad diffraction wave front. In order to reduce the straightness error, this paper comes up with the error compensation for the single point diamond lathe. Firstly, we make the first compensation based on the accumulative error curve. When the compensation ratio is 0.75 to 0.85, the peak valley value (pv) of the diffraction wave front is about 400 nm, reaching its greatest effect of the first straightness compensation. Secondly, we make the straightness compensation according to the diffraction wave front curve of the blazed order. The pv of the diffraction wave front is about 83nm. The results show that the diffraction wave front is greatly improved which is beneficial to improve the quality of the grating, and has a guiding role in the actual grating characterization.

ABSTRACT

Fine particulate matter (PM2.5) is the major air pollutant in Beijing, posing serious threats to human health. Land use regression (LUR) has been widely used in predicting spatiotemporal variation of ambient air-pollutant concentrations, though restricted to the European and North American context. We aimed to estimate spatiotemporal variations of PM2.5 by building separate LUR models in Beijing. Hourly routine PM2.5 measurements were collected at 35 sites from 4th March 2013 to 5th March 2014. Seventy-seven predictor variables were generated in GIS, including street network, land cover, population density, catering services distribution, bus stop density, intersection density, and others. Eight LUR models were developed on annual, seasonal, peak/non-peak, and incremental concentration subsets. The annual mean concentration across all sites is 90.7 µg/m(3) (SD = 13.7). PM2.5 shows more temporal variation than spatial variation, indicating the necessity of building different models to capture spatiotemporal trends. The adjusted R (2) of these models range between 0.43 and 0.65. Most LUR models are driven by significant predictors including major road length, vegetation, and water land use. Annual outdoor exposure in Beijing is as high as 96.5 µg/m(3). This is among the first LUR studies implemented in a seriously air-polluted Chinese context, which generally produce acceptable results and reliable spatial air-pollution maps. Apart from the models for winter and incremental concentration, LUR models are driven by similar variables, suggesting that the spatial variations of PM2.5 remain steady for most of the time. Temporal variations are explained by the intercepts, and spatial variations in the measurements determine the strength of variable coefficients in our models.

ABSTRACT

In an attempt to develop potent and selective antitumor agents, a series of novel 1,4-disubstituted phthalazine derivatives was designed and synthesized. All the prepared compounds were screened for their cytotoxic activities against A549, HT-29 and MDA-MB-231 cell lines in vitro. Among them, seven compounds (7a-7e, 7j and 7i) displayed excellent selectivity for MDA-MB-231 cells with IC(50) values in the nM range, a desirable range for pharmacological testing. The most promising compound, 7a (IC(50) = 3.79 microM, 2.32 microM, 0.84 nM), was 5.6-, 10.8- and 6.9 x 10(4)- times more active than PTK-787 (IC(50) = 21.16 microM, 22.11 microM, 57.72 microM), respectively.
