ABSTRACT

The present study evaluated the viability of Toxoplasma gondii tissue cysts in dry-aged pork loins (m. longissimus) after 14, 21 and 28 days under controlled temperature (0 °C ±â€¯1 °C). The pigs (n = 9) were orally inoculated with 3,000 T. gondii oocysts. The right loin of each pig was aged for a predetermined period, and the left loin was kept unprocessed as a control. Two experiments were performed. In Experiment 1, the loins of three pigs were aged for 14 days and then bioassayed in both cats and mice. In Experiment 2, the loins of six pigs were bioassayed only in mice, and the ageing periods were 14, 21, and 28 days. Toxoplasma gondii tissue cysts remained viable in loins aged up to 14 days, as confirmed by bioassays in cats and mice. Viable T. gondii was not recovered by bioassays in mice from loins that were aged for 21 or 28 days. These results demonstrate that T. gondii remained viable in vacuum-packed dry-aged pork loins for 14 days at controlled temperature but not for 21 days or longer.

ABSTRACT

Neospora spp. are intracellular protozoa with worldwide distribution and closely related to Toxoplasma gondii, which can infect a variety of mammals including horses. From September 2013 to June 2014, 185 horses from northern, central and southern parts of mainland Portugal were randomly sampled and tested for detection of immunoglobulin (Ig) G antibodies to Neospora spp. using an indirect multi-species enzyme-linked immunosorbent assay (ELISA) commercial test (ID Screen® Neospora caninum Indirect Multi-species; ID.vet Innovative Diagnostics, Grabels, France). Two horses (1.1%; CI: 0.1-3.8%), one male and one female, were found to be seropositive for Neospora spp. Both seropositive animals were horses housed indoors but with access to outdoors, used for leisure activities and were apparently healthy, with good body condition and with no alterations at physical examination. This was the first serologic survey of antibodies to Neospora spp. carried out in horses from Portugal.

ABSTRACT

Toxoplasma gondii and Leishmania infantum are zoonotic protozoal parasites. Serum samples were obtained from 186 donkeys (Equus africanus asinus) from Portugal and assessed for antibodies to T. gondii by the modified agglutination test (MAT). For titration of antibodies to Leishmania spp. the direct agglutination test was used (DAT). Eleven donkeys were seropositive for T. gondii with titres of 20 (n = 7), 80 (n = 2), 640 (n = 1) and ≥ 2560 (n = 1). One donkey was seropositive for Leishmania spp. (titre of 800). Foram obtidas amostras de soro de 186 jumentos (Equus africanus asinus) e avaliadas para anticorpos anti-T. gondii pelo teste de aglutinação direta modificada (TADM), em Portugal. Para a titulação de anticorpos anti-Leishmania spp. foi usado o teste de aglutinação direta (TAD). Onze jumentos foram soropositivos para T. gondii com títulos de 20 (n = 7), 80 (n = 2), 640 (n = 1) e ≥ 2560 (n = 1). Um jumento foi soropositivo para Leishmania spp. (título de 800). Os jumentos em Portugal estão expostos e podem ser infectados com T. gondii e Leishmania spp.

Resumo Neospora spp. são protozoários intracelulares com distribuição mundial e estreitamente relacionados com Toxoplasma gondii, que podem infectar uma variedade de mamíferos, incluindo cavalos. De setembro de 2013 a junho de 2014, 185 cavalos de áreas do Norte, Centro e Sul de Portugal continental foram aleatoriamente amostrados e testados para a detecção de anticorpos imunoglobulinas (Ig) G anti-Neospora spp., utilizando-se um ensaio imunoenzimático (ELISA) indireto multi-espécies comercial (ID Screen® Neospora caninum Indirect Multi-species; ID.vet Innovative Diagnostics, Grabels, France). Dois cavalos (1,1%; IC: 0,1-3,8%), um macho e uma fêmea, foram detectados como seropositivos para Neospora spp. Ambos os animais seropositivos eram cavalos mantidos em cocheiras mas com acesso aos piquetes, eram utilizados para atividades de lazer e estavam aparentemente saudáveis, com boa condição corporal e sem alterações ao exame físico. Essa é o primeiro rastreio de anticorpos para Neospora spp. realizado em cavalos de Portugal.

ABSTRACT

Neospora caninum is protozoan parasite with domestic and wild dogs, coyotes and grey wolves as the definitive hosts and many warm-blooded animals as intermediate hosts. It was cultivated and named in 1988. Neosporosis is a major disease of cattle and has no public health significance. Since 1990's N. caninum has emerged as a major cause of abortion in cattle worldwide, including in Brazil. N. caninum also causes clinical infections in several other animal species. Considerable progress has been made in understanding the biology of N. caninum and there are more than 200 papers on this subject from Brazil. However, most of the reports on neosporosis from Brazil are serological surveys. Overall, little is known of clinical neosporosis in Brazil, particularly cattle. The few reports pertain to sporadic cases of abortion with no information on epidemics or storms of abortion. The objective of the present review is to summarize all reports from Brazil and suggest topic for further research, including prevalence of N. caninum oocysts in soil or in canine feces, and determining if there are additional definitive hosts, other than the domestic dog. There is need for a national survey in cattle using defined parameters. Future researches should focus on molecular characterization of N. caninum strains, possibility of vaccine production and relationship between wildlife and livestock epidemiology.

Resumo Neospora caninum é um protozoário parasita que possui os canídeos domésticos e selvagens, coiotes e lobos cinzentos como hospedeiros definitivos e vários animais de sangue quente como hospedeiros intermediários. Foi cultivado e nomeado em 1988. A neosporose é uma das principais doenças em bovinos e não tem significância em saúde pública. Desde 1990, N. caninum tem emergido como uma das principais causas de aborto em bovinos em todo o mundo, inclusive no Brasil. N. caninum também causa infecções clínicas em várias outras espécies animais. Consideráveis avanços foram feitos na compreensão da biologia desse parasita e há mais de 200 trabalhos sobre o assunto no Brasil. No entanto, a maioria dos relatos de neosporose do Brasil são relacionados a sorologia. Em geral, pouco se sabe sobre a neosporose clínica no Brasil, particularmente em bovinos. Os poucos relatos referem-se a casos esporádicos de aborto sem informações sobre epidemias ou surtos de aborto. O objetivo da presente revisão é resumir todos os relatos sobre N. caninum no Brasil e sugerir tópicos para pesquisas futuras, incluindo a prevalência de oocistos de N. caninum no solo ou em fezes caninas e determinar se há hospedeiros definitivos adicionais, exceto o cão doméstico no país. Uma pesquisa nacional em bovinos usando parâmetros definidos seria de grande importância. Pesquisas futuras deveriam ser focadas na caracterização de cepas de N. caninum, possibilidade de produção de vacinas e a relação epidemiológica entre a vida selvagem e o gado.

ABSTRACT

This is the first report of Toxoplasma gondii infection in black vultures (Coragyps atratus), which are obligate scavengers found throughout the Americas. Serum samples from 121 wild black vultures caught in urban areas of the city of São Paulo, Brazil, were tested for the presence of T. gondii antibodies using the modified agglutination test (MAT; cutoff point 1:5). T. gondii antibodies were found in 16 birds (13.2%), with titers of 1:5 (6 birds), 1:10 (8 birds), and 1:20 (2 birds).(AU)


Este é o primeiro relato de infecção por Toxoplasma gondii em urubus (Coragyps atratus) que são aves carniceiras obrigatórias, encontradas no continente americano. Amostras de soro de 121 urubus, capturados em área urbana da cidade de São Paulo, Brasil, foram testadas quanto à presença de anticorpos anti-T. gondii pelo teste de aglutinação modificada (MAT, ponto de corte de 1:5). Anticorpos foram encontrados em 16 (13,2%) aves com títulos de 1:5 (6 aves), 1:10 (8 aves) e 1:20 (2 aves).(AU)

We present a set of data on human and chicken Toxoplasma gondii seroprevalence that was investigated and analysed in light of groundwater vulnerability information in an area endemic for waterborne toxoplasmosis in Brazil. Hydrogeological assessment was undertaken to select sites for water collection from wells for T. gondii oocyst testing and for collecting blood from free-range chickens and humans for anti-T. gondii serologic testing. Serologic testing of human specimens was done using conventional commercial tests and a sporozoite-specific embryogenesis-related protein (TgERP), which is able to differentiate whether infection resulted from tissue cysts or oocysts. Water specimens were negative for the presence of viable T. gondii oocysts. However, seroprevalence in free-range chickens was significantly associated with vulnerability of groundwater to surface contamination (p < 0.0001; odds ratio: 4.73, 95% confidence interval: 2.18-10.2). Surprisingly, a high prevalence of antibodies against TgERP was detected in human specimens, suggesting the possibility of a continuous contamination of drinking water with T. gondii oocysts in this endemic setting. These findings and the new proposed approach to investigate and analyse endemic toxoplasmosis in light of groundwater vulnerability information associated with prevalence in humans estimated by oocyst antigens recognition have implications for the potential role of hydrogeological assessment in researching waterborne toxoplasmosis at a global scale.

ABSTRACT

Toxoplasma gondii is a protozoan parasite that infects a large spectrum of warm-blooded animals, including humans. Small rodents and marsupials play an important role in the epidemiology of T. gondii because they are sources of infection for domestic and feral cats. Serum samples from 151 rodents and 48 marsupials, captured in the Atlantic Forest, São Paulo State, southeastern Brazil, were analyzed for the presence of T. gondii antibodies. Antibodies detected by the modified agglutination test (MAT ≥ 25) were found in 8.6% (13/151) of the rodents and 10.4% (5/48) of the marsupials, with titers ranging from 25 to 6400 and from 25 to 3200, respectively for the rodents and marsupials. Three of the eight species of rodents (Akodon spp., Oligoryzomys nigripesand Rattus norvegicus), and one from the four marsupial species (Didelphis aurita) presented positive animals. T. gondii was described for the first time in the rodent Oligoryzomys nigripes.

BACKGROUND: Neospora caninum and Toxoplasma gondii are important pathogens of worldwide distribution. N. caninum is a major cause of abortion in cattle and dogs are main reservoirs because they excrete the environmentally resistant oocysts. Toxoplasmosis is a worldwide zoonosis and dogs are considered as sentinels for this parasite because of their close contact with people and cats; additionally dog meat is also used for human consumption in China. The aim of the present study was to assess the prevalence of N. caninum and T. gondii infection in dogs from China. A total of 425 countryside dog hearts in Jilin, Henan and Anhui provinces of the People's Republic of China were collected from slaughter houses in two batches; the first batch of 96 in October 2013, and the second batch of 329 in April 2014. Serum samples extracted from 96 dog hearts were tested for antibodies to N. caninum and from 425 dog hearts were tested for T. gondii antibodies in the modified agglutination tests (cut-off 1:25 for both), using respective antigens. RESULTS: Antibodies to N. caninum were 6 of 96 (6.25%) of dogs with titers of 1:25 in 2, 1:50 in 3, and 1:100 in 1. All seropositive dogs were more than 1 year old. Antibodies to T. gondii were found in 35 of 425 (8.24%) dogs with titers of 1:25 in 15, 1:50 in 14; and 1:100 in 6. CONCLUSION: The results of the present study indicated low prevalence of N. caninum and T. gondii antibodies in dogs of China, compared with Europe and America. Identification of the risk factors that underlie these differences may help prevention of neosporosis and toxoplasmosis. This is the first report of N. caninum infection in dogs from China.

Toxoplasmosis is an important cause of abortion in sheep and a zoonotic risk to humans, leading to significant hazards to health and to economic losses. This study examined the seroprevalence and associated risk factors for infection with Toxoplasma gondii in 379 sheep from 12 flocks in Rio de Janeiro state, Brazil. Using the modified agglutination test (MAT), 202 (53.3 %) of 379 were seropositive with titers of 1:25 in 65, 1:50 in 40, 1:100 in 23, 1:200 in 11, 1:400 in 36, 1:800 in 7, 1:1,600 in 1, and 1:3,200 or higher in 19 sheep. The most significant factors associated with T. gondii seropositivity in sheep were age, gender, and veterinary care. Finding of T. gondii antibodies in 97 of the 202 sheep in titers of 1:100 or higher is indicative of persistently infected animals. This high level of seropositivity requires urgent control measures to reduce impact on animal productivity and public health.

Alpacas are important to the economy of several countries. Little is known of Toxoplasma gondii infection in alpacas worldwide. In the present study, T. gondii was isolated and genetically characterized from alpacas for the first time. Alpacas (n = 16) and rams (n = 12) pastured on a farm in Virginia, USA, were examined at necropsy. Antibodies to T. gondii were determined by the modified agglutination test (MAT, 1:25) and found in 6 of 16 alpacas with titers of 1:100 (2 alpaca), 1:400 (2 alpacas), 1:800 (1 alpaca), and 1:1,600 (1 alpaca), and 5 of 12 rams in titers of 1:50 in one, 1:400 in one, 1:800 in one, 1:1,600 in one, and 1:3,200 in one. Tissues of all 16 alpacas were bioassayed in mice or in cats. Muscles (heart, skeletal muscle) of nine alpacas with MAT titers of 1:25 were fed to T. gondii-free cats; the cats did not shed oocysts. Viable T. gondii was isolated from tissues of two of six seropositive alpacas by bioassay in mice. Viable T. gondii was isolated from three of three seropositive sheep by bioassay in mice. Genotyping using cell-cultured tachyzoites revealed four genotypes, including one for ToxoDB PCR-RFLP genotype #2 (type III), one for genotype #3 (type II variant), one for genotype #170, and two for a new genotype designated as ToxoDB PCR-RFLP genotype #230. Thus, four of the five T. gondii isolates in the present study belonged to different genotypes. These results indicate a higher genetic diversity among T. gondii isolates circulating in the USA than previously realized.

ABSTRACT

In this study, serum samples of 53 wild animals from two different states from the southeast region of Brazil were analyzed for the presence of anti-Toxoplasma gondii antibodies by the modified agglutination test (MAT), with a cut-off of 1: 5 for birds and of 1: 25 for mammals. Out of the sampled animals, 27 were birds and 26 were mammals, and from this total, 83% (n = 44) were free-living animals. Antibodies were found in 13 mammals, from which 11 were free-living animals, and in five birds, all of which were free-living. In this study, T. gondii antibodies were detected in four bird species (crested seriema, Cariama cristata; buff-necked ibis, Theristicus caudatus; picazuro pigeon, Patagioenas picazuro; and burrowing owl, Athene cunicularia) and in a giant anteater (Myrmecophaga tridactyla) for the first time.

ABSTRACT

In this study, serum samples of 203 animals from different locations, from zoos and breeding facilities from the north and northeast regions of Brazil, were analyzed for the presence of anti-Toxoplasma gondii antibodies by the modified agglutination test (MAT) with a cutoff of 1:25. Of the sampled animals, 184 were adult mammals of both sexes and 19 were birds. Antibodies were found in 61 of 184 mammals, and no association between sex and age of the animals and the presence of T. gondii antibodies was observed (P < 0.05). Anti-T gondii antibodies were not found in birds. Toxoplasma gondii was detected in Brazilian tapir (Tapirus terrestris) for the first time.
