ABSTRACT

It is postulated that there is negative correlation between milk yield and reproductive performance. However, some studies definitely doubt this causality. The aim of our study was to investigate the relationship between milk production and fertility on three dairy farms. The production parameter was the milk yield (in kg), and fertility was expressed by the number of inseminations per conception (AI index), as well as by the length of the service period (in days). A total of 13 012 lactations from cows with their first three lactations completed were analysed. The number of inseminations was significantly correlated with the milk yield and with the studied farm (p < 0.0001), but its correlation with the lactation number was not significant (p = 0.9477). A similar relationship was found after evaluating the length of the service period. A multiplicative model showed that a 2000 kg milk increase extended the service period by 9% and increased the AI index by 13%. Thereafter, using quartiles of the cows, the service period of the highest-producing group rose by 41.5 days, and the AI index by almost 1, compared to the lowest quartile. Our results indicate a definitive decline in reproductive indicators parallel to an increase in milk production but did not prove an inevitable correlation.

ABSTRACT

Excessive use of azole fungicides in agriculture poses a potential threat to honeybees and other pollinator insects; however, the detailed effects of these molecules remain largely unclear. Hence, in the present study it was aimed to investigate the acute sublethal effects of tebuconazole on the redox homeostasis and fatty acid composition in the brain of honeybees. Our findings demonstrate that tebuconazole decreased total antioxidant capacity, the ratio of reduced to oxidized glutathione and disturbed the function of key antioxidant defense enzymes along with the induction of lipid peroxidation indicated by increased malondialdehyde levels, while it also altered the fatty acid profile of the brain. The present study highlights the negative impact of tebuconazole on honeybees and contributes to the understanding of potential consequences related to azole exposure on pollinator insects' health, such as the occurrence of colony collapse disorder.

ABSTRACT

Butyrate promotes rumen epithelium growth and function; however, the effect of prepartum butyrate supplementation on dairy cow productivity, health and their offspring has not been extensively studied. Furthermore, no studies have investigated the effect of magnesium butyrate (MgB), which is also a source of magnesium. A trial was performed to test the hypothesis that prepartum MgB supplementation (105 g/cow/day) would increase colostrum quality and improve calving, newborn calf vitality and cow health. Multiparous Holstein cows were randomly assigned to MgB supplemented (n = 107) and Control groups (n = 112). Colostrum yield and the total yield of IgG, protein and lactose were higher (p ≤ 0.05) in the supplemented group. The calving assistance rate was lower (p ≤ 0.012), and the neonatal vitality score was higher (p ≤ 0.001) in the MgB group. Improved parameters related to cow health and fertility were observed in the supplemented group. The MgB group also had higher milk yield during the first week of lactation (p ≤ 0.001), and a higher (p ≤ 0.05) body condition score from 3 to 9 weeks after calving. In conclusion, prepartum MgB supplementation provides a wide range of benefits for dairy cows, as well as their newborn calves.

ABSTRACT

BACKGROUND: Magnesium butyrate (MgB) supplementation of dairy cows during the three-week close-up period was tested for its effects on blood energy analytes, rumination time, inflammation, and lactation performance. METHODS: Daily milk yield was recorded and weekly milk samples collected for the first 70 days of lactation from MgB supplemented (MgB, n = 34), and unsupplemented (Control, n = 31) multiparous Holstein-Friesian cows. During a period from week 3 to week 10 postpartum, blood samples were taken and analyzed for various parameters, and ruminant activity was measured. RESULTS: The MgB group yielded 25.2% more milk than the Control during week 1, and had increased milk fat and protein concentrations over a longer duration. Somatic cell counts (SCC) were decreased in the MgB group independent of days in milk. No differences were observed between groups in terms of plasma non-esterified fatty acids, ß-hydroxybutyrate, glucose, or blood iCa levels. The MgB group had lower haptoglobin (Hp) levels during lactation relative to the Control group. Time spent ruminating increased after calving with MgB due to a shorter post calving rumination delay relative to the Control group. CONCLUSIONS: Prepartum MgB supplementation improved lactation performance without affecting blood energy analytes. The basis by which MgB also improved rumination activity remains to be determined, as DMI could not be assessed. As MgB lowered SCC and Hp concentrations, it is speculated that MgB may help minimize postpartum inflammatory processes.

ABSTRACT

Near-infrared (NIR) spectroscopy was employed to determine the differences between forage mixtures of winter cereals and Italian ryegrass and to evaluate fermentation characteristics of mixed silages. Forages were harvested on five phases (Cuts 1-5), with 1 week interval (n = 100). The yield of the last harvest (Cut 5) was ensiled and analyzed on four different days (D0, D7, D14, and D90) (n = 80). Principal component analysis based on the NIR data revealed differences according to the days of harvest, differences between winter cereals and Italian ryegrass forages, and differences in the fermentation stages of silages. The partial least square regression models for crude protein (CP), crude fiber (CF), and ash gave excellent determination coefficient in cross-validation (R2 CV > 0.9), while models for ether extract (EE) and total sugar content were weaker (R2 CV = 0.87 and 0.74, respectively). The values of root mean square error of cross-validation were 0.59, 0.76, 0.22, 0.31, and 2.36 %DM, for CP, CF, EE, ash, and total sugar, respectively. NIR proved to be an efficient tool in evaluating type and growth differences of the winter cereals and Italian ryegrass forage mixtures and the quality changes that occur during ensiling.

BACKGROUND: Sows are exposed to severe stress and hormonal challenges during their whole productive life. As polyunsaturated fatty acids play an important role in immune and reproductive functions, with a better understanding of their role in breeding sows' nutrition, improved performance and more sustainable pig production can be achieved. OBJECTIVES: In this study, we investigated the effects of omega-6 and omega-3 fatty acid supplementation on the antioxidant status and hormone levels of sows. METHODS: A total of 48 Danish Large white × Danish Landrace sows were supplemented either with sunflower oil (SO) as a control group or with fish oil (FO) as experimental group at the same dose of 10 g/kg feed. Blood samples were collected on day 14 of lactation, 5 days after weaning (insemination), and 30 days after insemination. To estimate antioxidant and reproductive effects, the amounts of reduced glutathione (GSH), thiobarbituric acid reactive substance, the activity of glutathione peroxidase (GPx), serum 17ß oestradiol (E2), progesterone (P4), and 6-keto prostaglandin F1α (6-keto PGF1α) levels were investigated. RESULTS: FO-based supplementation increased GPx activity on day 14 of lactation. Five days after weaning, the concentration of GSH in FO-fed sows was significantly higher than that in SO-fed sows. The E2 content of blood was significantly lower in the experimental group than in the control group for two of the three examined periods (day 14 of lactation and 30 days after insemination), whereas P4 levels were significantly higher in the experimental group 5 days after weaning. We found that 6-keto PGF1α levels were systematically lower in the experimental group throughout the trial. CONCLUSIONS: This study provides evidence of the major impact of omega-6 and -3 fatty acids on the tested hormone levels, which serve as precursors for the production of E2 and P4 but have an opposite effect on PGF2α production.

ABSTRACT

The aim of this study was metagenomics analyses of acquired antibiotic-resistance genes (ARGs) in the intestinal microbiome of two important food-animal species in Hungary from a One Health perspective. Intestinal content samples were collected from 12 domestic pigs (Sus scrofa) and from a common carp (Cyprinus carpio). Shotgun metagenomic sequencing of DNA purified from the intestinal samples was performed on the Illumina platform. The ResFinder database was applied for detecting acquired ARGs in the assembled metagenomic contigs. Altogether, 59 acquired ARG types were identified, 51 genes from domestic pig and 12 genes from the carp intestinal microbiome. ARG types belonged to the antibiotic classes aminoglycosides (27.1%), tetracyclines (25.4%), ß-lactams (16.9%), and others. Of the identified ARGs, tet(E), a blaOXA-48-like ß-lactamase gene, as well as cphA4, ampS, aadA2, qnrS2, and sul1, were identified only in carp but not in swine samples. Several of the detected acquired ARGs have not yet been described from food animals in Hungary. The tet(Q), tet(W), tet(O), and mef(A) genes detected in the intestinal microbiome of domestic pigs had also been identified from free-living wild boars in Hungary, suggesting a possible relationship between the occurrence of acquired ARGs in domestic and wild animal populations.

ABSTRACT

Short chain fatty acid (SCFA) butyrate has various beneficial effects on the gut microbiota as well as on the overall health status and metabolism of the host organism. The modulatory role of butyrate on gut barrier integrity reflected by tight junction protein expression has been already described in mammalian species. However, there is limited information available regarding chickens. Therefore, the main aim of this study was to monitor the effects of protected butyrate on claudin barrier protein and monocarboxylate transporter 1 abundance in different gastrointestinal segments of chickens as well as the growth performance of broiler chickens. The effect of protected butyrate on the caecal microbiota was monitored by quantifying the concentrations of total eubacteria and key enzymes of butyrate production. Furthermore, intestinal SCFA concentrations were also measured. Based on the data obtained, protected butyrate increased the overall performance as well as the barrier integrity of various gut segments. Protected butyrate also positively affected the SCFA concentration and composition. These findings provide valuable insight into the complex effects of protected butyrate on broiler gut health, highlighting the beneficial effects in improving intestinal barrier integrity and performance parameters.

ABSTRACT

The intestinal microbiome of dogs can be influenced by a number of factors such as non-starch polysaccharides as well as some non-digestible oligo- and disaccharides. These molecules are only decomposed by intestinal anaerobic microbial fermentation, resulting in the formation of volatile fatty acids (VFAs), which play a central role in maintaining the balance of the intestinal flora and affecting the health status of the host organism. In the present study, the effects of lactulose and psyllium husk (Plantago ovata) were investigated regarding their influence on concentrations of various VFAs produced by the canine intestinal microbiome. Thirty dogs were kept on a standard diet for 15 days, during which time half of the animals received oral lactulose once a day, while the other group was given a psyllium-supplemented diet (in 0.67 and in 0.2 g/kg body weight concentrations, respectively). On days 0, 5, 10 and 15 of the experiment, feces were sampled from the rectum, and the concentration of each VFA was determined by GC-MS (gas chromatography−mass spectrometry). Lactulose administration caused a significant increase in the total VFA concentration of the feces on days 10 and 15 of the experiment (p = 0.035 and p < 0.001, respectively); however, in the case of psyllium supplementation, the concentration of VFAs showed a significant elevation only on day 15 (p = 0.003). Concentrations of acetate and propionate increased significantly on days 5, 10 and 15 after lactulose treatment (p = 0.044, p = 0.048 and p < 0.001, respectively). Following psyllium administration, intestinal acetate, propionate and n-butyrate production were stimulated on day 15, as indicated by the fecal VFA levels (p = 0.002, p = 0.035 and p = 0.02, respectively). It can be concluded that both lactulose and psyllium are suitable for enhancing the synthesis of VFAs in the intestines of dogs. Increased acetate and propionate concentrations were observed following the administration of both supplements; however, elevated n-butyrate production was found only after psyllium treatment, suggesting that the applied prebiotics may exert slightly different effects in the hindgut of dogs. These findings can be also of great importance regarding the treatment and management of patients suffering from intestinal disorders as well as hepatic encephalopathy due to portosystemic shunt.

Influencing the endocrine metabolic regulation of chickens by nutritional factors might provide novel possibilities for improving animal health and productivity. This study was designed to evaluate the impact of dietary cereal type (wheat-based (WB) vs. maize-based (MB) diets), crude protein level (normal (NP) vs. lowered (LP)), and sodium (n-)butyrate (1.5 g/kg diet) supplementation (vs. no butyrate) on the responsiveness of hepatic glucagon receptor (GCGR), insulin receptor beta (IRß) and mammalian target of rapamycin (mTOR) in the phase of intensive growth of chickens. Liver samples of Ross 308 broiler chickens (Gallus gallus domesticus) were collected on day 21 for quantitative real-time polymerase chain reaction and Western blot analyses. Hepatic GCGR and mTOR gene expressions were up-regulated by WB and LP diet. GCGR and IRß protein level decreased in groups with butyrate supplementation; however, the quantity of IRß and mTOR protein increased in WB groups. Based on these data, the applied dietary strategies may be useful tools to modulate hepatic insulin and glucagon signaling of chickens in the period of intensive growth. The obtained results might contribute to the better understanding of glycemic control of birds and increase the opportunity of ameliorating insulin sensitivity, hence, improving the production parameters and the welfare of broilers.

There are several mathematical models and measurements to determine the efficiency of the digestibility of different feedstuffs. However, there is lack of information regarding the direct methods or measurement techniques used to analyse the physical response of the different parts of the gastrointestinal tract (GIT) of growing pigs to different diets. Smooth muscle electromyography (SMEMG) is a non-invasive method for the measurement of gastrointestinal myoelectrical activity. In the present study, SMEMG methodology has been adapted from laboratory rats to pigs, and the effects of feedstuffs with control (CTR) or experimentally increased (EXP) amounts of fibre were investigated on gastrointestinal tract motility. Nine barrow pigs ((Danish Landrace × Danish Yorkshire) × Danish Duroc) were used (30 ± 3 kg), and their CTR and EXP feedstuffs contained 29 and 49 g/kg crude fibre (CF), respectively. Myoelectric activities of the stomach, ileum and caecum were detected in the awake pigs by a pair of electrodes. The recorded myoelectric signals were analysed with fast Fourier transformation (FFT), and the spectra were expressed in GIT section-specific cycles per minutes (cpm) values and the maximum power spectrum density (PsDmax). A significant increase (P < 0.001) was observed in the value of the PsDmax of the small intestine (20-25 cpm) as a consequence of the EXP diet. The PsDmax values of the stomach (3-5 cpm) and large intestine (1-3 cpm) did not show any significant change in pigs fed the EXP diet. As a direct and non-invasive method, SMEMG is suitable for the rapid evaluation of the effects of diets with different fibre contents on the GIT of non-anaesthetised, free-moving pigs.

ABSTRACT

The Fusarium mycotoxins deoxynivalenol (DON) and zearalenone (ZEA) frequently contaminate grain crops, especially maize, the basis of poultry's feed. Mycotoxins enter the food chain and induce pathological changes in edible tissues. Milk thistle (Silybum marianum) has been used for the treatment of liver disease in humans because of its antioxidant and hepatoprotective effects, but its utility in veterinary use is poorly examined. To investigate possible protective properties against mycotoxin caused oxidative stress in poultry, pressed form of milk thistle seed (0.5%) was tested in white, female, Hungarian ducks over a feeding period of 47 days. Ducks were separated into 3 groups. The first group was fed with normal diet. The second group was fed with normal diet contaminated with DON (4.9 mg/kg) and ZEA (0.66 mg/kg). The third group received mycotoxin contaminated feed with milk thistle supplementation. Histological examination, markers of the redox status and metal element concentration measurements were carried out. The results showed alterations in the histological examination and in the redox homeostasis markers as a short-term effect by strengthening the antioxidant system. Acute exposure of mycotoxins caused an oxidative stress, which induced an effective antioxidant defensive response of the organism indicated by the free sulfhydryl group content (from 0.72 ± 0.06 to 0.77 ± 0.11) and the reducing power (0.49 ± 0.06 to 0.52 ± 0.08) elevation. The short-term free radical injury may be compensated by the liver resulting in decreased lipid peroxidation markers (malondialdehyde concentration: from 16.86 ± 0.49 to 0.94 ± 0.15, conjugated diene concentration: from 0.21 ± 0.07 to 0.17 ± 0.03). Silymarin further strengthtened the antioxidant defense by the elevation of sulfhydryl groups concentration and reducing power property resulted in decreased total scavenger capacity. However the concentration of lipid peroxidation markers were further elevated by the used antioxidant treatment (MDA: 5.2 ± 0.35, DC: 0.26 ± 0.08). In conclusion, the mycotoxin-contamination activated effectively the antioxidant system. The milk thistle supplementation has cytoprotective effects according to the histological findings, activated the antioxidant system, however the elevation of lipid peroxidation products need further explanation.

INTRODUCTION: Fatty liver can develop as a result of diseases, surgical procedures, medicaments, malnutrition or excessive alcohol consumption, however, fat and poor fiber feeding can be attributed as the primary cause. Non-alcoholic fatty liver can be found in 20-30% of the population. Generally, alimentary-induced fatty liver in early state is described as uncomplicated liver injury. AIM: The aim of our research was to study the effect of fat rich nutrition on the gut-liver axis by routine laboratory, analytical and histological methods in rats. METHODS: We also examined the redox parameters of the liver and of the bowel. Fatty acid composition and element content of liver were measured. RESULTS: Significant changes were found in parameters of redox homeostasis as well as alterations in liver enzymes and metabolites. The changes could be detected in the liver, blood and bowel parts. The development of fatty liver is associated with the decrease of transmethylation capacity. Fatty acid composition and metal ion homeostasis were also altered in liver. Histological examinations showed that hepatocytes were swollen in the central part of the liver lobules, showed droplets and pycnotic nuclei, which were characterized by fatty degeneration. Small and large bowel enterocytes were damaged, sometimes pushed from the surface, and sometimes inflammatory reactions in the mucous membrane were observed. CONCLUSION: Our results suggest that alimentary fatty liver in early state is not considered simply as a reversible alteration because it alters the entire body's redox homeostasis and establishes heart and serious metabolic diseases as well as hasten the development of gastrointestinal tumors. ABSTRACT

Red deer (Cervus elaphus) hinds ( n = 3 × 10 ) of identical initial body weight (BW, ca. 68 kg) were reared on a monocotyledonous grass (G group), on a grass-papilionaceous (GP group) or on pure papilionaceous pasture each of 2 ha (P group) for 219 d. At the end of the experiment carcass tissue composition was assessed by means of computer tomography, slaughter value and meat quality were characterized and tissue - longissimus thoracis et lumborum (LTL), thigh and liver - samples were taken for fatty acid composition analysis. The primary aim was to assess nutrition-driven differences. Hinds of group P provided higher final BW (101 kg vs. 90 and 91.9 kg in groups G and GP, respectively) and higher BW gain (32.6 kg during the total period vs. 22.4 and 22.1 kg). The carcass weight exceeded those of the other groups significantly (68.8 kg vs. 59.3 and 63.2 kg), while there was no difference among groups in the perirenal fat weight and red color tone ( a * ) of the LTL. Groups G and P differed significantly in the LTL weight (highest in P), its dripping loss (lowest in G), lightness (L; highest in P) and yellow color tone ( b * ). In the thigh muscle, LTL and liver the highest proportion of fatty acid CLA9c11t was reached on the G pasture, and the same trend was true for docosahexaenoic acid (DHA , C22:6 n3) in the muscles. The n6  /  n3 fatty acid ratio was the highest on the P pasture in the liver and both muscles. The liver incorporated the highest proportion of linoleic acid (C18:2 n6) and converted it rather effectively to arachidonic acid (C20:4 n6), coupled with the lowest α -linolenic acid presence. In conclusion, concerning muscle mass production, group P proved to be the most advantageous pasture; meanwhile LTL meat quality factors (dripping loss, DHA proportion, pH, color) were more favorable on the G pasture.

The short-chain fatty acid butyrate, either in unprotected or protected form, is widely applied as a growth-promoting feed additive in poultry nutrition; however, its possible effects on the carcass composition of broilers have not been fully elucidated. Further, lowering dietary crude protein (CP) levels is an important issue in poultry farming, contributing to ecologically beneficial lower nitrogen excretion. The main aims of this study were to test how unprotected and protected forms of butyrate and decreased dietary CP content with essential amino acid (lysine, methionine, threonine, tryptophan) supplementation ("LP-EAA" diet) affect carcass parameters and the chemical composition of muscles in broilers. Ross 308 chickens were randomized to seven groups ( n = 10 /group) receiving adequate CP-containing (normal protein, "NP") or LP-EAA diets, both supplemented with or without unprotected sodium butyrate, and NP diets with different forms of protected sodium butyrate. Carcass traits were measured, and the chemical composition of pectoral and femoral muscles was analyzed at the age of 6 weeks. Carcass weight was significantly increased by the LP-EAA diet and all protected butyrate types tested, while the relative breast meat yield was significantly higher in LP-EAA than NP groups and in both unprotected and protected butyrate-supplemented chickens compared to controls. The protein content of the femoral muscle was significantly decreased, but its lipid content was significantly elevated by the LP-EAA diet and by all types of butyrate addition. However, no changes were detected in the chemical composition of pectoral muscle. In conclusion, breast meat production can be effectively stimulated by dietary factors, such as by reducing dietary CP content with essential amino acid supplementation and by applying butyrate as a feed additive, while its chemical composition remains unchanged, in contrast to the femoral muscle. The aforementioned nutritional strategies seem to be the proper tools to increase carcass yield and to alter meat composition of broilers, contributing to more efficient poultry meat production.

Scarce studies have investigated the impact of fumonisin B1 (FB1) on the hepatic tissue fatty acid (FA) profile, and no study is available on piglets. A 10-day in vivo experiment was performed on seven piglets/group: control and FB1-fed animals (diet was contaminated with fungal culture: 20 mg FB1/kg diet). Independent sample t-test was carried out at p < 0.05 as the significance level. Neither growth, nor feed efficiency, was affected. The hepatic phospholipid (PL) fatty acids (FAs) were more susceptible for FB1, while triglyceride (TG) was less responsive. The impact of FB1 on hepatic PL polyunsaturated fatty acids (PUFAs) was more pronounced than on saturated fatty acids. Among all PUFAs, predominant ones in response were docosapentaenoicacid (DPA) (↓), docosahexaenoic DHA (↓) and arachidonic acids (↑). This led to a higher omega-6:omega-3 ratio, whereas a similar finding was noted in TGs. Neither total saturation (SFA) nor total monousaturation (MUFA) were affected by the FB1 administration. The liver showed an increase in malondialdehyde, as well as antioxidant capacity (reduced glutathione and glutathione peroxidase). The plasma enzymatic assessment revealed an increase in alkaline phosphatase (ALP), while alanine transaminase (ALT), aspartate transaminase (AST), lactate dehydrogenase (LDH), and gamma-glutamyltransferase (GGT) were not influenced. The microscopic sections provided evidence of vacuolar degeneration of the hepatocytes' cytoplasm, but it was not severe. Furthermore, the lung edema was developed, while the kidney was not affected. In conclusion, regarding FB1-mediated hepatotoxicity in piglets, the potential effect of slight hepatotoxicity did not compromise growth performance, at least at the dose and exposure period applied.

ABSTRACT

Male Wistar rats were intraperitoneally dosed with fumonisin B1 (FB1; 0, 20, 50 and 100 mg kg-1 dietary dose equivalent) for 5 & 10 days to assess dose- and time-dependent effects on renal and hepatic phosphatidylcholine (PC), phosphatidylinositol (PI) and phosphatidylethanolamine (PE) fatty acid (FA) profiles. Renal PC showed increasing FA saturation (SAT) after 5 days; after 10 days polyunsaturation (PUFA) decreased markedly (Σ n3 (total n3), Σ n6, PUFA, unsaturation index (UI) and average FA chain length (ACL)), mostly with linear dose response. In the PI FAs similar changes were observed, decreasing monounsaturated FA, PUFA, UI and ACL (5 & 10 days), while the PE fraction was responsive in Σ n6 (↓) and SAT (↑), but only after 5 days (without dose response for both PI & PE). Liver PC exhibited increasing saturation (C16:0), decreasing polyunsaturation (C20:3 n6 [dihomo-γ-linolenic acid, DGLA]; C20:3 n3); the PI FA profile showed similar alterations after 5 days. PC & PI FA failed to respond in a dose-dependent manner to FB1. In PE FA profile DGLA decreased, with a decrease of the total n6 FA proportion and dose-dependent increase of n3 FAs. Results revealed expressed renal sensitivity, supporting our earlier published results in terms of oxidative stress and histopathological modifications.

ABSTRACT

This study investigates the metabolic effects of maize- or wheat-based diets with normal (NP) and lowered (LP) dietary crude protein level [the latter supplemented with limiting amino acids and sodium (n-)butyrate at 1.5 g/kg diet] at different phases of broiler fattening. Blood samples of Ross 308 broilers were tested at the age of 1, 3 and 6 weeks. Total protein (TP) concentration increased in wheat-based and decreased in LP groups in week 3, while butyrate reduced albumin/TP ratio in week 1. Uric acid level was elevated by wheat-based diet in week 1 and by wheat-based diet and butyrate in week 3, but decreased in LP groups in weeks 3 and 6. Aspartate aminotransferase activity was increased by wheat-based diet in week 3, and creatine kinase activity was intensified by LP in weeks 3 and 6. Blood glucose level decreased in wheat-based groups in week 3; however, triglyceride concentration was augmented in the same groups in week 3. No change of glucagon-like peptide 1, glucose-dependent insulinotropic polypeptide and insulin concentration was observed. In conclusion, an age-dependent responsiveness of broilers to dietary factors was found, dietary cereal type was a potent modulator of metabolism, and a low crude protein diet supplemented with limiting amino acids might have a beneficial impact on the growth of chickens.

ABSTRACT

(1) Background and (2) Methods: A 14-day in vivo, multitoxic (pure mycotoxins) rat experiment was conducted with zearalenone (ZEA; 15 µg/animal/day), deoxynivalenol (DON; 30 µg/animal/day) and fumonisin B1 (FB1; 150 µg/animal/day), as individual mycotoxins, binary (FD, FZ and DZ) and ternary combinations (FDZ), via gavage in 1 mL water boluses. (3) Results: Body weight was unaffected, while liver (ZEA↑ vs. DON) and kidney weight (ZEA↑ vs. FDZ) increased. Hepatocellular membrane lipid fatty acids (FAs) referred to ceramide synthesis disturbance (C20:0, C22:0), and decreased unsaturation (C22:5 n3 and unsat. index), mainly induced by DON and to a lesser extent by ZEA. The DON-FB1 interaction was additive on C20:0 in liver lipids. In renal phospholipids, ZEA had the strongest effect on the FA profile, affecting the saturated (C18:0) and many n6 FAs; ZEA was in an antagonistic relationship with FB1 (C18:0) or DON (C18:2 n6, C20:1 n9). Hepatic oxidative stress was the most expressed in FD (reduced glutathione and glutathione peroxidase), while the nephrotoxic effect was further supported by lipid peroxidation (malondialdehyde) in the DON treatment. (4) Conclusions: In vivo study results refer to multiple mycotoxin interactions on membrane FAs, antioxidants and lipid peroxidation compounds, needing further testing.

ABSTRACT

Intestinal cytochrome P450 (CYP) enzymes play key role in the first pass metabolism of orally ingested xenobiotics, providing a primary metabolic barrier, being of special importance in maintaining animal health and production. This study was aimed to investigate how intestinal drug-metabolizing CYPs can be modulated by nutritional factors in broiler chicken. We investigated the effects of the natural growth promoter (n-)butyrate of different origin (feed supplementation of protected or non-protected forms and/or inducing caecal microbial production by supporting higher level of dietary non-starch polysaccharides [NSP]) on the activity of duodenal CYPs. To observe the connection between intestinal CYP activity and butyrate concentration, the distribution of differently originated butyrate was also assessed by measuring its concentration in various intestinal segments and different vessels of portal and systemic circulation. Butyrate of different origin showed varying distribution properties as being absorbed from different parts of the gastrointestinal tract. Intestinal CYP1A and CYP2H2 activities were increased by dietary butyrate supplementation and by the increased caecal microbial butyrate production, while CYP3A37 activity was minimally influenced by microbial butyrate only. The present study proved that both dietary and microbial butyrate could alter the activity of CYPs in the duodenal epithelium. Our findings suggest that intestinal CYPs could be induced not only by the intestinal luminal butyrate, but also from basolateral side, by the already absorbed butyrate. Such action of butyrate can be of special importance from food safety and pharmacotherapeutic point of view as it may modify the metabolism and intestinal kinetics of simultaneously applied xenobiotics.
