ABSTRACT

Introduction: The rutting period imposes a stressful condition on male camels, which results in elevated serum cortisol levels and alterations in their sexual behavior. Therefore, the current work was carried out to investigate the effect of mineral-vitamin premix supplementation on behavior, reproductive performance, hormones, serum oxidative stress profile, and other serum biochemical parameters of Camelus dromedarius during the breeding season. Methods: Fourteen mature, fertile male Camelus dromedarius were divided into two groups, a control group (n = 7) and a mineral-vitamin premix group (n = 7). The present study lasted for 95 days during the rutting period (1st February to 5th May). Each camel in the premix group received a daily diet of 50 g of mineral-vitamin premix throughout the whole rutting period, during which the frequencies and durations of the following behaviors: maintenance, posture, aggressiveness, and sexual activity were collected every 20 min. At the end of the study, blood samples were collected. Results: Results revealed that the premix group showed higher (P < 0.05) maintenance (feeding and rumination), standing, and overall sexual desire-related behavior frequency, besides more times (P < 0.001) for rumination, standing, walking, and lying while showing lower (P < 0.001) frequencies of overall aggressive behaviors than the control group. The serum concentration of malondialdehyde, nitric oxide, cortisol, blood glucose, and urea evidenced a significant decrease in the premix group compared with the control one, while significantly elevated levels of reduced glutathione, testosterone, total antioxidant capacity, triiodothyronine, and thyroxin, total protein, albumin, globulin, calcium, phosphorus, potassium, and magnesium were recorded in the premix group in comparison with the control. Conclusion: It could be concluded that daily dietary supplementation of 50 g of mineral-vitamin premix to male camels during the breeding season is necessary to overcome the oxidative stress and serum cortisol concentration with a subsequent decrease in aggressive behavior and improvement to testosterone level in blood, body condition score and body weight gain.

ABSTRACT

Egypt has several beaches, as well as the Nile River and a few lakes; therefore, it could compensate for the lack of protein in red meat with fish. Fish, however, may become a source of heavy metal exposure in humans. The current study was to assess the level of five toxic metals, lead (Pb), cadmium (Cd), mercury (Hg), arsenic (As), and aluminum (Al), in six species, namely, Oreochromis niloticus (O. niloticus), Mugil cephalus (M. cephalus), Lates niloticus (L. niloticus), Plectropomus leopardus (P. leopardus), Epinephelus tauvina (E. tauvina), and Lethrinus nebulosus (L. nebulosus), collected from the El-Obour fish market in Egypt. The residual concentrations of the tested toxic metals in the examined O. niloticus, M. cephalus, L. niloticus, E. tauvina, P. leopardus, and L. nebulosus species were found to be higher than the European Commission's maximum permissible limits (MPL) for Pb and Cd by 10 and 20%, 15 and 65%, 75 and 15%, 20 and 65%, 15 and 40%, and 25 and 5%. In contrast, 30% of L. niloticus exceeded the MPL for Hg. It was shown that the average estimated daily intake (EDI) and the target hazard quotient (THQ) in fish samples are below safety levels for human consumption and hazard index (HI < 1). From the human health point of view, this study showed that there was no possible health risk to people due to the intake of any studied species under the current consumption rate in the country.

ABSTRACT

With the increase in nanoparticles (NPs) products on the market, the possibility of animal and human exposure to these materials will increase. The smaller size of NPs facilitates their entrance through placental barriers and allows them to accumulate in embryonic tissue, where they can then be a source of different developmental malformations. Several toxicity studies with chemically synthesized titanium dioxide NPs (CTiO2 NPs) have been recently carried out; although there is insufficient data on exposure to biosynthesized titanium dioxide NPs (BTiO2 NPs) during pregnancy, the study aimed to evaluate the ability of an eco-friendly biosynthesis technique using garlic extract against maternal and fetal genotoxicities, which could result from repeated exposure to TiO2 NPs during gestation days (GD) 6-19. A total of fifty pregnant rats were divided into five groups (n = 10) and gavaged CTiO2 NPs and BTiO2 NPs at 100 and 300 mg/kg/day concentrations. Pregnant rats on GD 20 were anesthetized, uterine horns were removed, and then embryotoxicity was performed. The kidneys of the mothers and fetuses in each group were collected and then maintained in a frozen condition. Our results showed that garlic extract can be used as a reducing agent for the formation of TiO2 NPs. Moreover, BTiO2 NPs showed less toxic potential than CTiO2 NPs in HepG2 cells. Both chemically and biosynthesized TiO2 NP-induced genetic variation in the 16S rRNA sequences of mother groups compared to the control group. In conclusion, the genetic effects of the 16S rRNA sequence induced by chemically synthesized TiO2 NPs were greater than those of biosynthesized TiO2 NPs. However, there were no differences between the control group and the embryo-treated groups with chemically and biologically synthesized TiO2 NPs.

ABSTRACT

The aim of this study was to compare three on-farm commercial methods for the indirect detection of subclinical mastitis in dairy cows: the California mastitis test (CMT), the Porta side somatic cell count milk test (Porta SCC), and the DeLaval cell counter (DCC), with the Fossomatic cell count (FSCC), and to evaluate the relationship between the determined somatic cell count SCC and the occurrence of intramammary pathogens in the milk of dairy cows. A total of 284 sensory unchanged mixed milk samples, collected during the milking on a dairy farm, were analyzed in this study for somatic cell counts by the mentioned on-farm tests. Quarter milk samples (n = 583) from all the selected cows were cultured. The agreement, sensitivity, and specificity of the three indirect commercial diagnostic tests (the CMT, the Porta SCC, and the DeLaval cell counter) were calculated, and the FSCC was used as the gold standard. The results were analyzed statistically using the Pearson correlation test and the paired t-test. The CMT matched with the FSCC in 83.1% of the samples, with the Porta SCC in 80.6%, and with the DCC in 80.3% of the samples. The sensitivity and specificity reached 81.0% and 92.9% for the CMT, 79.4% and 90.7% for the Porta SCC, and 75.8% and 97.5% for the DCC, respectively. The correlation between the FSCC and the Porta SCC was 0.86 (p < 0.0001), and between the FSCC and the DCC, it was 0.92 (p < 0.0001). The differences between them were insignificant. Bacteria were detected in 130 (22.3%) quarter milk samples. The most prevalent bacteria were Enterococcus spp. (36.2%), followed by E. coli (20%), coagulase-negative staphylococci (13.1%), A. viridans (9.2%), Streptococcus spp. (9.2%), Proteus spp. (6.2%), and S. intermedius (3.9%). Contagious isolates (S. aureus) were detected in 3 quarter milk samples (2.3%). The agreement between the individual tests and the microbiological culture was as follows: 69.2% for the CMT; 73.7% for the Porta SCC; 71.6% for the DCC; and 76.5% for the FSCC. Higher SCCs were detected in the milk samples contaminated with bacteria than in the healthy milk (p < 0.001). No significance was found between the presence of individual species of intramammary pathogens and the different levels of SCCs. Based on the results, bacteria are the predominant cause of subclinical mastitis. The increased SCC of some milk samples with no presence of bacteria meant that the increase could have been caused by numerous other agents (viruses, fungi, or algae) or factors for mastitis in the dairy industry.

ABSTRACT

This study was conducted to evaluate the incidence of mastitis in 153 dairy cows and to evaluate the kinetics of adhesion of isolates obtained from surfaces and milk in comparison with the reference strain (RS), CCM 4223. The surfaces of the floor, teat cup, and cow restraints were aseptically swabbed in three replicates (n = 27). Of the total number of infected cows (n = 43), 11 samples were found to be positive for Staphylococcus aureus, 12 samples tested positive for non-aureus staphylococci, 6 samples tested positive for Streptococcus spp., and 11 samples tested positive for other bacteria (Escherichia coli, Pseudomonas spp.) or a mixed infection. The most represented pathogen in milk (11/43) and on surfaces (14/27) was S. aureus. The kinetics of adhesion of the reference strain and isolates of S. aureus on stainless steel surfaces were determined after 3, 6, 9, 12, 24, and 48 h, and 3, 6, 9, 12, and 15 days of incubation. All strains reached counts higher than 5 Log10 CFU/cm2 needed for biofilm formation, except RS (4.40 Log10 CFU/cm2). The isolates of S. aureus revealed a higher capability to form biofilm in comparison with RS during the first 3 h (p < 0.001). Thus, there is a significant difference between the occurrence of S. aureus on monitored surfaces-floor, teat cup, and cow restraints-and the frequency with which mastitis is caused by S. aureus (p < 0.05). This finding raises the possibility that if various surfaces are contaminated by S. aureus, it can result in the formation of biofilm, which is a significant virulence factor.

ABSTRACT

The aim of this study was to compare the effect of humic acid (HA) obtained by extraction from alginate on the incubation of roes and fry development in African cichlids, Labidochormis caeruleus, as well as their influence on the stabilization of the physicochemical parameters of water in an aquarium during artificial breeding. The roes were obtained by extruding from a female buccal cavity immediately after fertilization. For the experiment, 4 groups of 40 roes were formed in an incubator with an artificial hatchery. Groups 1-3 were exposed to 1%, 5%, and 10% concentrations of HA, respectively. The control group C was not exposed to HA. In all groups, the mortality and size differences of the fry, as well as the temperature, pH, hardness, nitrite, and nitrate levels in the tanks, were determined during a 30-day monitoring period until the resorption of the yolk sac. The results of this study indicated the ability of HA in 5% and 10% concentrations to reduce nitrite and nitrate levels in the aquatic environment, which significantly reduced the mortality of roes and the survivability of the fry. The determination of the morphological measurements of the fry revealed an increased body length in the groups exposed to 5% and 10% HA concentrations compared to the control group by the end of the monitored period. It was also noted that the yolk sac was resorbed two days earlier in the same groups than in the control. Thus, the results showed that HAs are suitable for use in the artificial aquarium incubation of roes and fry development, which are increasingly exposed to adverse environmental factors. The knowledge obtained in this study and its transfer into practice can allow even less experienced aquarists to successfully breed aquarium fish species that could not normally be bred under artificial conditions without the addition of HA.

ABSTRACT

Fish is one of the most valuable foods with high-quality animal protein. However, aquaculture, or ingesting contaminated food, allows organochlorine pesticides (OCPs) to enter the fish's body, and therefore, it negatively impacted public health. One-hundred and twenty random samples of Clupea harengus (C. harengus), Mugil cephalus (M. cephalus), Sardinella aurita (S. aurita), Oreochromis niloticus (O. niloticus), Neptunus pelagicus (N. pelagicus) and Sepia savigngi (S. savigngi) (n = 20 each) were collected from local markets in Mansoura city, Egypt. Samples were checked to see whether any residues of OCPs with the application of risk assessment due to their consumption by Mansoura citizens. The findings indicated that summation hexachlorocyclohexane (∑HCH) in examined seafood samples ranged from 0.27 ± 0.13 in N. pelagicus to 61.61 ± 52.03 µg.kg-1 in S. aurita. Also, the γ-HCH isomer was considered the more prominent among isomers. Hexachlorobenzene (HCB) was found in five different species, with mean values of 2.03 ± 1.85, 1.5.7 ± 1.17, 0.94 ± 0.87, 0.35 ± 0.06, and 0.18 ± 0.06 µg.kg-1 in C. harengus, S. aurita, M. cephlaus, O. niloticus, and S. savigngi. Moreover, summation of Heptachlors (∑HPTs) was 10.19 ± 7.63, 1.27 ± 0.26, 2.58 ± 0.11, 0.95 ± 0.12, 0.21 ± 0.11 and 0.32 ± 0.03 µg.kg-1 of wet weight in examined C. harengus, M. cephlaus, S. aurita, O. niloticus, N. pelagicus, and S. savigngi. Aldrin and dieldrin residues were 3.75 ± 1.31 and 4.86 ± 1.33 µg.kg-1 in C. harengu, meanwhile they were 1.61 ± 0.77 and 0.78 ± 0.04 µg.kg-1in M. cephalus. Dichlorodiphenyldichloroethylene (pp-DDE) was dominant in all examined species within different concentrations 5.08 ± 4.12, 0.98 ± 0.10, 3.07 ± 0.91, 0.93 ± 0.27, 0.08 ± 0.01 and 0.35 ± 0.02 µg.kg-1 in C. harengus, M. cephlaus, S. aurita, O. niloticus, N. pelagicus and S. savigngi, respectively. We concluded that all examined seafood samples were lower than the recommended maximum residue limit. Also, the estimated daily intake was less than the permitted daily intake. Non-carcinogenic indices of target hazard quotient and hazard index for OCPs in all examined species were less than 1.

In this study, we assessed the effect of humic substances, as an organic supplement in feed, on the fattening performance, meat quality and selected lipid and mineral parameters from the blood serum of rabbits. Three groups of the Giant Saris rabbit breed were used (one control and two experimental), with 16 animals per group. The animals in the control group were fed a standard pellet diet, the humic substances group received a basal diet supplemented with 5% humic substances, and the third group received a basal diet with 5% humic−fatty substances preparation during the entire experiment (from 35 to 120 days of age). There were 85 days of fattening; then, the rabbits were slaughtered. In the group supplemented with 5% humic−fatty substances addition, we noticed a higher final weight (p < 0.05) and higher average daily gains compared to the control group at the end of the fattening period, at 120 days of age. On the other hand, a slightly lower final weight (p > 0.05) in the group supplemented with humic substances was found compared to the control group. In the comparison of the individual parameters of the meat quality in rabbits, we observed a positive effect in the reduction in the intramuscular fat content and the lipid parameters as well as a lower total cholesterol from the blood serum in both supplemented groups. Regarding the mineral parameters, we observed elevated blood serum values of calcium and phosphorus in both experimental groups. The addition of humic−fatty substances appears to be the most effective way of supplementing rabbit feed due to the synergistic effect of humates and vegetable oils for their optimal growth development and the production of reduced-fat meat.

ABSTRACT

This study investigated 960 Slovak and Czech spotted cattle from four different conventional (non-organic) dairy herds located in Eastern Slovakia and Czechia during early lactation (14-100 days after calving). Dairy cows were examined clinically; milk from fore-stripping of each udder quarter was subjected to sensory examination and assessed by the California mastitis test (CMT), and laboratory analyses of bacterial pathogens in milk, including virulence factors, were conducted. Positive CMT scores (1-3) for one or more quarters were detected in 271 (28.2%) of the examined animals. Out of 230 infected milk samples, representing 24.0% of all dairy cows, staphylococci (59.1% of positive findings) were the most commonly isolated organisms, followed by E. coli (11.3%), streptococci Str. uberis (9.1%) and Str. agalactiae (3.4%), and enterococci (6.1%). From 136 isolates of S. aureus (38 isolates) and non-aureus staphylococci (NAS; 98 isolates), virulence factors and their resistance to 14 antimicrobials were detected using the disk diffusion method, with PCR detection of the methicillin resistance gene, mecA. An increased incidence of clinical and chronic forms of mastitis has been reported in mastitic cows in which staphylococci, especially S. aureus and NAS (S. chromogenes, S. warneri, and S. xylosus), have been detected and compared to other isolated udder pathogens. From those species, S. aureus and isolates of NAS mentioned above showed multiple virulence factors that are more likely to hydrolyze DNA, hemolysis, produce gelatinase and biofilm, and have multi-drug resistance as compared to other less virulent staphylococci. Generally, the isolated staphylococci showed 77.2% resistance to one or more antimicrobials, in particular to aminoglycosides, ß-lactams, macrolides, or cephalosporins. Isolates that showed the ability to form a biofilm were more resistant to more than one antimicrobial than isolates without biofilm production. Multi-drug resistance to three or more antimicrobial classes was recorded in 16 isolates (11.7%), and the presence of the mecA gene was also confirmed in two isolates of S. aureus and two species of NAS.
