ABSTRACT

BACKGROUND: Both genetic and environmental factors play crucial roles in the development of major depressive disorder (MDD) and suicide attempts (SA). However, the interaction between both items remains unknown. This study aims to explore the interactions between the genetic variants of the serotonin 2 A receptor (HTR2A) and the nitric oxide synthase 1 (NOS1) and environmental factors in patients who experience MDD and SA. METHODS: A total of 334 patients with MDD and a history of SA (MDD-SA) were recruited alongside 518 patients with MDD with no history of SA (MDD-NSA), and 716 healthy controls (HC). The demographic data and clinical characteristics were collected. Sequenom mass spectrometry was used to detect eight tag-single nucleotide polymorphisms (tagSNPs) in HTR2A (rs1328683, rs17068986, and rs3125) and NOS1 (rs1123425, rs2682826, rs3741476, rs527590, and rs7959232). Generalized multifactor dimensionality reduction (GMDR) was used to analyze the gene-environment interactions. RESULTS: Four tagSNPs (rs17068986, rs3125, rs527590, and rs7959232) exhibited significant differences between the three groups. However, these differences were not significant between the MDD-SA and MDD-NSA groups after Bonferroni correction. A logistic regression analysis revealed that negative life events (OR = 1.495, 95%CI: 1.071-2.087, P = 0.018), self-guilt (OR = 2.263, 95%CI: 1.515-3.379, P < 0.001), and negative cognition (OR = 2.252, 95%CI: 1.264-4.013, P = 0.006) were all independently associated with SA in patients with MDD. Furthermore, GMDR analysis indicated a significant interaction between HTR2A rs3125 and negative life events. Negative life events in conjunction with the HTR2A rs3125 CG + GG genotype were associated with a higher SA risk in patients with MDD when compared to the absence of negative life events in conjunction with the CC genotype (OR = 2.547, 95% CI: 1.264-5.131, P = 0.009). CONCLUSION: Several risk factors and a potential interaction between HTR2A rs3125 and negative life events were identified in patients with SA and MDD. The observed interaction likely modulates the risk of MDD and SA, shedding light on the pathogenesis of SA in patients with MDD.

ABSTRACT

Interferon-induced transmembrane proteins (IFITMs) play an important role in the innate immune response triggered by viral infection. Transmissible gastroenteritis virus (TGEV) causes severe diarrhea, vomiting and dehydration in piglets, resulting in huge economic losses to the swine industry. In this study, we showed that IFITM3 inhibits the replication of TGEV and interferes with the binding of TGEV to PK15 cells. Moreover, the inhibitory effect of IFITM3 on TGEV circumvents the upregulation of inflammatory cytokines. Subsequently, we found that the M22A mutant loses part of the antiviral effect of IFITM3 on TGEV; in contrast, the K24A mutant enhances the antiviral effect of IFITM3. Notably, our data shows a synergistic effect between IFITM3 and CQ, which further amplifies the antiviral effect against TGEV.

ABSTRACT

BACKGROUND: C-reactive protein (CRP) has been shown to predict antidepressant treatment outcomes in several trials, but they were limited to small-sample and strictly-restricted conditions. This study plans to verify if CRP can predict antidepressant efficacy in large samples in the real world. METHODS: 918 depressed patients who had tested CRP were included, then were followed up through their outpatient visits or by telephone to obtain information about their medication therapy (SSRIs, SNRIs, MT, NaSSA) and assess efficacy using the Clinical Global Impressions-Improvement scale (CGII). Efficacy was classified as effective and ineffective and CRP was separated into the low CRP group (CRP <1 mg/L, n = 709) and the high CRP group (CRP ≥1 mg/L, n = 209).The efficacy was compared in different groups. RESULTS: Using Kaplan-Meier survival analysis and Cox proportional regression model to analyze, it was discovered that SNRIs were more effective than SSRIs in treating patients with high CRP(HR = 1.652, p = 0.037,95 % CI:1.031-2.654), and SSRIs were more effective in treating patients with low CRP than those with high CRP (HR = 1.257, p = 0.047,95 % CI:1.003-1.574), while no difference in efficacy between the two groups was found in patients using SNRIs, MT, NaSSA. LIMITATIONS: Small amounts of MT and NaSSA were included, and some factors that may affect CRP value have not been controlled. CONCLUSION: CRP could predict the efficacy of SSRIs in the real world, depressed patients with high CRP may be more likely to respond poorly to SSRIs.

ABSTRACT

Recently, wireless camera sensor networks (WCSNs) have entered an era of rapid development, and WCSNs assisted by unmanned aerial vehicles (UAVs) are capable of providing enhanced flexibility, robustness and efficiency when executing missions such as shooting targets. Existing research has mainly focused on back-end image processing to improve the quality of captured images, but it has neglected the question of attaining quality images on the front-end, which is significantly influenced by the location and hovering time of the UAV. Therefore, in this paper, we conceive a novel shooting utility model to quantify shooting quality, which is maximized by simultaneously considering the UAV's trajectory planning, hovering time and shooting point selection. To expound further, we prove the submodularity of the utility function, whereby the original problem can be expressed as a submodular maximization problem with path constraints, and we propose a utility-cost ratio (UCR) algorithm to maximize shooting utility through two-level optimization. Then, by using the relaxation of the cost function, we analyze the gap between the proposed algorithm and the optimal algorithm (OPT) and prove that the UCR algorithm has a bi-criterion approximation ratio of 1-1/e/2. Simulation results show that the algorithm outperforms both the random algorithm (RAN) and the maximum shooting utility point selection algorithm (MSU) in terms of shooting utility and time utilization efficiency, improving shooting utility by 51% and 21% compared to the RAN and MSU algorithms, respectively, and achieving at least 88.2% of the OPT algorithm in terms of time utilization efficiency.

ABSTRACT

African swine fever (ASF) is a highly detrimental viral disease caused by African swine fever virus (ASFV). The occurrence and prevalence of this disease have become a serious threat to the global swine industry and national economies. At present, the detection volume of African swine fever is huge, more sensitive and accurate detection techniques are needed for the market. pp62 protein, as a protein in the late stage of infection, has strong antigenicity and a high corresponding antibody titer in infected pigs. In this study, the CP530R gene was cloned into expression vector pET-28a to construct a prokaryotic expression plasmid, which was induced by IPTG to express soluble pp62 protein. Western blot analysis showed that it had great reactivity. Using the purified recombinant protein as an antigen, an indirect ELISA method for detecting ASFV antibody was established. The method was specific only to ASFV-positive serum, 1:1600 diluted positive serum could still be detected, and the coefficients of variation (CV) of the intra assay and inter assay were both <10%. It turns out that the assays had excellent specificity, sensitivity, and repeatability. This provides an accurate, rapid, and economical method for the detection of ASFV antibody in clinical pig serum samples.

ABSTRACT

In optical wireless communication (OWC), the superimposed optical orthogonal frequency division multiplexing (O-OFDM), such as layered asymmetrically clipped O-OFDM (LACO-OFDM), can improve spectrum efficiency by appropriately combining multiple O-OFDM signals for simultaneous transmission. However, it suffers from increased receiver complexity and latency. Therefore, a novel architecture of hybrid pulse-amplitude-modulated discrete multitone modulation (HPAM-DMT) is proposed in this paper to support a spectrum-efficiency OWC link. In HPAM-DMT, a PAM-DMT signal is carefully designed by using the real parts of subcarriers and is then superimposed on the classic PAM-DMT signal for simultaneous transmission, which fully exploits the spectrum in terms of subcarriers. Moreover, thanks to the well-designed aritecture of the two superimposed PAM-DMT signals, the proposed HPAM-DMT achieves the same spectrum efficiency as the LACO-OFDM, with much lower complexity and latency. Notable improvement of the bit-error rate (BER) performance is observed for the proposed HPAM-DMT compared to LACO-OFDM under the corruption of the transmitter nonlinearity. Moreover, HPAM-DMT achieves a relatively higher power efficiency than conventional O-OFDM schemes, which makes it a competitive O-OFDM scheme for IM/DD-based OWC.
