ABSTRACT

OBJECTIVE: Emergency service vehicle incidents are a leading cause of firefighter fatalities and are also hazardous to civilian road users. Modifiable driving behaviors may be associated with emergency service vehicle incidents. The goal of this study was to use telematics to identify driving behaviors associated with crashes in the fire service. METHODS: Forty-three emergency service vehicles in 2 fire departments were equipped with telematics devices (12 in Department A and 31 in Department B). The devices collected vehicle coordinates, speed, and g forces, which were monitored for exceptions to driving rules established by the fire departments regarding speeding, harsh braking, and hard cornering. Fire department administrative reports were used to identify vehicles involved in crashes and merged with daily telematics data. Penalized logistic regression was used to identify driving rules associated with crashes. Least absolute shrinkage and selection operator (LASSO) regression was used to generate a telematics-based risk index for emergency service vehicle incidents. RESULTS: Nearly 1.1 million km of driving data and 44 crashes were recorded among the 2 departments during the study. Harsh braking was associated with increased odds of crash in Department A (odds ratio [OR] = 2.22; 95% confidence interval [CI], 1.09-4.51) and Department B (OR = 1.55; 95% CI, 1.12-2.15). For every kilometer of nonemergency speeding, the odds of crash increased by 35% in Department A (OR = 1.35; 95% CI, 1.03-1.77) and by over 2-fold in Department B (OR = 2.09; 95% CI, 1.19-3.66). In Department B, hard cornering (OR = 1.14; 95% CI, 1.03-1.26) and emergency speeding (OR = 1.65; 95% CI, 1.06-2.57) were also associated with increased odds of crash. The final LASSO risk index model had a sensitivity of 73% and specificity of 57%. CONCLUSIONS: Harsh braking and excessive speeding were driving behaviors most associated with crash in the fire service. Telematics may be a useful tool for monitoring driver safety in the fire service.

ABSTRACT

Surgical site infections (SSI) are an uncommon, but significant, consequence of surgical interventions. There are very few studies investigating SSI risk in veterinary medicine, and even fewer in cattle, despite the fact that major surgeries are commonly conducted on livestock. Furthermore, the suboptimal conditions under which such surgeries are frequently performed on livestock could be considered an important risk factor for the development of SSIs. With increasing public concern over the contribution of veterinary-prescribed antimicrobials to the emergence of antimicrobial-resistant bacteria in people, there is widespread scrutiny and criticism of antimicrobial use in livestock production medicine systems. While the causal link between antimicrobial resistance in livestock and people is heavily debated, it is clear that the prevalence of antimicrobial resistance, in any population, is closely correlated with the antimicrobial 'consumption' within that population. As the veterinary profession explores ways of addressing the emergence and selection of antimicrobial-resistant bacteria in food-producing animals, there is a need for veterinarians and producers to carefully consider all areas of antimicrobial use, and employ an evidence-based approach in designing appropriate clinical protocols. This paper aims to review current knowledge regarding the risk factors related to abdominal SSI in periparturient cows, and to encourage practitioners to judiciously evaluate both their standard operating procedures and their use of antimicrobials in these situations. In a second paper, to be published in a subsequent issue of Veterinary Record, these principles will be used to provide specific evidence-based recommendations for antimicrobial use in bovine abdominal surgery.

ABSTRACT

The development of acquired resistance to cyathostome challenge after 1 season's exposure to a cyathostome-contaminated pasture was investigated using 17 parasite-naive ponies, which were 2-3 yr of age. These were divided into 3 groups: 1 to graze a cyathostome-contaminated pasture for 4 mo (exposed ponies), 1 to graze a "clean" pasture not previously grazed by parasitized animals (nonexposed ponies), and 1 group to remain in the barn under helminth-free conditions (parasite-free ponies). After pasture exposure all ponies were housed in stalls in the barn dewormed with ivermectin (200 micrograms/kg) and oxibendazole (100 mg/kg), a treatment that eliminated most cyathostomes encysted in the mucosa as well as all luminal parasites, on the basis of necropsies of 5 animals, after 17 days. Remaining ponies were challenged with 100,000 cyathostome-infective third-stage larvae (L3) per os 3 wk after anthelmintic treatment. Necropsies were performed 7 wk after the challenge. Total cyathostome burdens (luminal plus encysted stages) were not significantly different among any of the groups. However, a significantly higher percentage of hypobiotic early L3 (EL3) and a lower percentage of adults were found in exposed ponies. This observation supports the hypothesis that resistance acquired through exposure promotes cyathostome hypobiosis. This increase in EL3 in exposed ponies was associated with a significant increase in weight of cecum and ventral colon biopsies.

ABSTRACT

This experiment assessed the effects of 12 mo of daily treatment of young horses with recombinant equine somatotropin (eST) on 1) carcass and internal organ traits at necropsy and 2) residual effects in live horses for 60 d after cessation of treatment. Seven horses received eST daily at 20 microg/kg BW; seven others received vehicle (controls). Four horses from each group were killed at the end of treatment. There were few effects of eST treatment on hematologic assessments or histopathologic evaluations of internal organs. Treatment with eST increased the weights of the right adrenal gland (P = 0.090), left (P = 0.085) and right (P = 0.013) kidneys, liver (P = 0.012), tended to inrease the weights of pancreas (P = 0.082), spleen (P = 0.008), and heart (P = 0.102), and decreased (P = 0.032) somatotropin (ST) content in the adenohypophysis. Loin-eye area at the 10th rib was also greater (P = 0.01) in eST-treated horses than in controls. There was no difference (P > 0.15) between groups in left adrenal, brain, parathyroid glands, or thyroid gland weights or in 10th-rib fat thickness. In the remaining two control and three eST-treated horses (one control horse died), plasma IGF-I concentrations were higher (P = 0.001) in treated animals through d 6 after cessation of treatment and then dropped precipitously. Insulin concentrations in treated animals tended to be elevated (P = 0.08) only on d 0. There was a treatment x day interaction (P = 0.04) for plasma urea nitrogen levels, which increased in treated horses. A decrease (P < 0.05) in BW in the treated animals was observed by 21 d after treatment. There was no difference (P > 0.15) in insulin or glucose response to glucose tolerance tests given on d 0 through 60 after cessation of treatment. Overall ST response to secretagogue was reduced (P < 0.05) in eST-treated horses compared with controls. In summary, long-term treatment of growing horses with eST decreased endogenous ST response to secretagogue and increased plasma IGF-I concentrations and many internal organ weights but had little effect on hematologic or histopathologic characteristics at necropsy. The effects on IGF-I concentrations were lost within 6 d, and BW in treated horses decreased within 3 wk after cessation of treatment.

ABSTRACT

Two trials were conducted to confirm the efficacy of ivermectin paste against endoparasites of horses. In these trials, 20 ponies were treated with ivermectin oral paste at 200 mcg x kg body weight once on Day 0, and 20 ponies served as unmedicated controls. The animals carried naturally acquired parasite infections as confirmed by pretrial fecal examination. The animals were necropsied for worm recovery on Days 14, 15 or 16. Parasites recovered were identified to species. Horses treated with ivermectin had significantly (P<0.05) fewer (>99.0% reduction) adult small strongyles (Coronocyclus spp including C. coronatus, C. labiatus, C. labratus; Cyathostomum spp including C. catinatum, C. pateratum; Cylicocyclus spp including C. ashworthi, C. elongatus, C. insigne, C. leptostomum, C. nassatus, C. radiatus; Cylicodontophorus bicoronatus; Cylicostephanus spp including C. asymetricus, C. bidentatus, C. calicatus, C. goldi, C. longibursatus, C. minutus; Gyalocephalus capitatus; Parapoteriostomum spp including P. euproctus, P. mettami; Petrovinema poculatum; Poteriostomum spp including P. imparidentatum, P. ratzii) and adult large strongyles (Strongylus edentatus, S. vulgaris; Triodontophorus spp including T. brevicauda, T. serratus; Craterostomum acuticaudatum) than the controls. Ivermectin was also highly effective (94% to >99%, P<0.05-0.01) against Gasterophilus intestinalis larvae, Habronema spp., Oxyuris equi, Parascaris equorum. The data from these two trials confirm that ivermectin paste administered to horses orally at 200mcg x kg(-1) continues to be highly effective for treatment and control of a broad range of small and large strongyle species as well as other species of gastrointestinal parasites.

ABSTRACT

One hundred twenty-four cows (92 multiparous and 32 primiparous) were used to evaluate the effect of grain supplements containing high crude protein [(22.8% CP, 5.3% rumen undegradable protein (RUP), dry matter basis], moderate CP (16.6% CP, 6.1% RUP), and moderate CP with supplemental RUP (16.2% CP, 10.8% RUP) on lactation performance of Holstein cows rotationally grazing annual ryegrass-oat pastures. Supplemental protein was provided by solvent extracted soybean meal in the high CP and moderate CP supplements and as a corn gluten meal-blood meal mixture (2.8:1) in the moderate CP, high RUP supplement. Cows were blocked according to previous mature milk equivalent production and calving date (partum group; 0 d in milk or postpartum group; 21 to 65 d in milk) and randomly assigned to dietary treatments. Grain was individually fed, at approximately a 1:3 grain to milk ratio, before a.m. and p.m milkings. The study was replicated during two grazing seasons that averaged 199 d. Cows had ad libitum access to bermudagrass hay while on pasture (dry matter intake = 1.3 kg/d). Protein supplementation had no effect on study long pasture dry matter (12.7 +/- 1.0 kg/d) or total dry matter (23.9 +/- 1.2 kg/d) consumption. Protein concentration did not affect actual milk yield of either calving group (high CP vs. moderate CP); however, postpartum group cows receiving high CP grain supplements maintained greater milk fat concentrations (3.34 vs. 3.11%), which led to higher fat-corrected milk (FCM) yields than control cows receiving moderate CP grain diets (30.3 vs. 28.9 kg/d). Crude protein concentration in milk of high CP-supplemented, postpartum group cows was also higher than moderate CP cows (3.42 vs. 3.27%). Additional RUP did not increase FCM yield above that generated by moderate CP grain diets for partum (34.3 vs. 32.9 kg/d) or postpartum-group cows (28.9 vs. 28.2 kg/d). Increasing CP concentration of grain supplement did not affect milk yield of Holstein cows grazing immature winter annual pastures. Supplementing additional RUP was without benefit, indicating that in this study energy deprivation may have been the major nutritional constraint for high-producing dairy cows grazing lush pastures.

An experiment was conducted over a 2-yr period to investigate the influence of grain crude protein (CP) and rumen undegradable protein (RUP) concentration on reproduction and energy status of dairy cows grazing annual ryegrass (Lolium multiflorum) and oats (Avena sativa). Holstein cows (n = 122) were blocked by calving group [partum (0 d postpartum) vs. postpartum (41 +/- 19 d postpartum at study initiation)] and assigned to grain supplements containing high CP [22.8% of dry matter (DM)], moderate CP (16.6%), or moderate CP (16.2%)] supplemented with RUP from blood meal and corn gluten meal. Postpartum condition loss was greater and first-service pregnancy rate was lower for partum-group cows receiving high CP grain supplements compared with control cows receiving moderate CP supplements. The RUP supplements reduced grain consumption, increased days to first estrus, and reduced first-service pregnancy rate of partum-group cows. The reproduction of postpartum group cows was unaffected by protein supplements. Plasma urea nitrogen was higher for cows fed high CP diets, but plasma ammonia nitrogen, glycated hemoglobin, nonesterified fatty acids, beta-hydoxybutyrate, glucose, and insulin concentrations were similar to cows fed moderate CP. Excess postpartum condition loss, coupled with inconsistent protein supplement effects on days to first service and first-service pregnancy rate, suggest that energy deprivation may have contributed to the low fertility experienced by grazing cows in this study.

ABSTRACT

Fourteen foals between 4 and 4.5 mo of age were used to determine the effects of 12 mo of daily treatment with equine somatotropin (eST) on growth, metabolic, and hormonal characteristics. The foals were paired by sex, type, and lineage, and one of each pair was administered eST daily at 20 microg/kg of BW. Body weights, body measurements, and assessments of glucose tolerance and feedback effects on endogenous somatotropin (ST) secretion were made routinely. Treatment with eST did not alter (P > 0.10) BW, height at withers, length of body, widths of chest and rump, heart girth, length of head, front or rear cannon lengths, front or rear cannon circumferences, gaskin circumference, or skin thickness, even though plasma IGF-I concentrations were doubled (P = 0.012). Glucose concentrations were higher (P = 0.03) in treated horses before glucose infusion; there was no difference (P > 0.10) in the glucose response to infusion. The insulin response to glucose infusion in the treated horses was generally higher (P = 0.0069) than in controls. Endogenous ST secretion in response to a ST secretagogue was reduced (P = 0.0001) in foals treated with eST in all months. The prolactin and thyroid-stimulating hormone responses to thyrotropin-releasing hormone on June 1 were not affected (P > 0.10) by treatment. In conclusion, daily treatment of growing horses with eST for 12 mo at the recommended dose altered the hormonal and metabolic characteristics known to be affected by ST but did not alter the growth characteristics of the animals.

ABSTRACT

Equine influenza virus remains a major health concern for the equine industry in spite of ongoing vaccination programmes. Previous work has shown that the immune system of horses can be affected by strenuous exercise. The possible adverse consequence of exercise-induced alterations in lymphocyte responses measured in vitro was unknown. Here we demonstrate that subjecting vaccinated ponies to a 5 day strenuous exercise programme results in a significant suppression of their T cell-mediated immune response to equine influenza virus as measured by decreased lymphoproliferation and gamma interferon production measured in vitro. These same ponies also demonstrated increased susceptibility to influenza disease following a challenge exposure to the same strain of virus. Rested ponies that had received the same vaccine and challenge were completely protected from disease. Our results demonstrate that exercise-induced suppression of the equine immune response to influenza virus can be associated with an increased susceptibility to disease.

ABSTRACT

Parasite-naive pony foals were used as sentinels to monitor transmission of gastrointestinal parasites of equids in Louisiana during 4 seasons of the year. Two annual periods were studied, 1988-1989 and 1992-1993. Two or 3 foals each season were turned out to graze a contaminated pasture along with resident parasitized mares and their foals. After a grazing period of 8 wk, sentinel ponies were held in a parasite-free box stall for a period of 6 wk to allow parasites to develop, thus enhancing the evaluation of hypobiotic stages. Following this holding period, necropsies were performed for complete parasite recoveries. Data show that transmission of large and small strongyles occurs during all seasons in southern Louisiana, with highest levels of transmission occurring in the winter and only minimal transmission taking place in the summer. Numbers of mucosal cyathostomes, as well as total cyathostome numbers, were highest in the winter, and luminal cyathostome numbers were highest in the spring. Transmission of Anoplocephala perfoliata and Parascaris equorum occurred during all seasons of the year, although numbers of P. equorum were reduced in spring 1989 and 1993. Gasterophilus intestinalis instars were recovered from fall and winter sentinels only. Oxyuris equi L4 were found all seasons 1 yr, but only during the fall and winter of the final year.

ABSTRACT

Cyathostome development and survival on pasture in subtropical climates of the US have yet to be completely defined and available data on seasonal transmission are minimal. In an attempt to study this phenomenon, a group of pony mares and their foals was maintained on a naturally contaminated pasture in southern Louisiana. Fecal egg counts (FEC) and numbers of infective third stage larvae (L3) kg(-1) dry herbage were recorded biweekly during two time periods, from January 1986 through December 1988, and September 1996 through October 1997. A FEC rise occurred during the late summer-early autumn which preceded the peak of L3 on pasture during the winter season. The numbers of cyathostome L3 were reduced during the hottest months of the year due mainly to daily minimum temperatures above 18 degrees C, and in winter during short freezing spells when daily minimum temperatures dropped below 0 degrees C. Tilling of the pasture reduced the number of cyathostome L3 during the early winter months but this is an efficacious measure only if horses are given an effective anthelmintic treatment prior to being returned to pasture. The data collected suggest that parasite reduction in southern Louisiana is possible using a treatment program with treatment beginning at the end of September and continuing through the end of March.

ABSTRACT

The primary purpose of this experiment was to assess the possible beneficial effects of recombinant equine somatotropin (reST) administration on wound healing in adult geldings. The effects of the 21-d reST treatment on carbohydrate and lipid metabolism and on endogenous ST characteristics were monitored as well. Single, full-thickness skin incisions (7.62 x 7.62 cm) were made in the pectoral region of all geldings on d 0. Treated geldings received reST at 20 microg/kg BW i.m., and control geldings received vehicle (10 mM sodium borate) at equivalent volumes daily from d 0 (immediately after surgery) through d 20. Tracings of the wounds were made with acetate transparencies, and wound areas were calculated via a digital analyzer. In addition to once-daily blood samples collected at specified days throughout the treatment period, an i.v. glucose tolerance test was performed on d 16, and three assessments of endogenous ST secretion were performed in the 2 d immediately following the end of treatment: epinephrine administration during the morning of d 21, an exercise test during the afternoon of d 21, and i.v. aspartic acid infusion on d 22. There was no effect (P > . 1) of reST treatment on wound healing as assessed by changes in wound areas. Daily plasma ST, IGF-I, glucose, and insulin concentrations were higher (P < .05) and urea-nitrogen concentrations were lower (P < .001) in geldings receiving reST relative to controls. Glucose, NEFA, and insulin concentrations were all higher (P < .01) in reST-treated geldings before glucose infusion on d 16, and the responses to glucose were greater (P < .05) as well. Epinephrine administration increased (P < .02) ST concentrations in control geldings on d 21 but not in reST-treated geldings; a similar suppressive effect of reST treatment was observed for the ST response to exercise (P < .001). After aspartic acid infusion on d 22, reST-treated geldings had a much smaller (P < .001) ST response than did control geldings. In conclusion, reST administered to geldings at 20 microg/kg BW i.m. caused hyperglycemia, hyperinsulinemia, insulin insensitivity, mobilization of fatty acids, and an apparent negative feedback on the pituitary's ST response to various stimuli known to induce ST secretion. However, there was no beneficial effect of reST treatment with the wound model used in this experiment.

ABSTRACT

This report describes two mares presented for evaluation of anorexia, fever of unknown origin, and weight loss. Clinical examination, laboratory findings, and transrectal ultrasonographic images suggested ovarian abscessation. One mare was successfully treated medically. Because of financial considerations, the second mare was euthanatized and a postmortem examination was performed. At necropsy, there was an enlarged right ovary with an adhesion to the large colon. Microscopic findings were characteristic of an ovarian abscess.

ABSTRACT

We conducted a study to determine the effects of excess dietary crude protein (CP) and rumen undegradable protein (RUP) on reproduction and lactation performance of Holstein cows. During each of three yearly replicates, cows were blocked by previous mature equivalent milk production and randomly assigned at calving (n = 47; partum group) or at 42 +/- 21 d postpartum (n = 134; postpartum group) to the following dietary treatments: 1) ryegrass pasture supplemented with a corn and soybean meal grain mix (high CP, moderate RUP); 2) ryegrass pasture mornings and corn silage evenings, supplemented with grain as in diet 1 (moderate CP, moderate RUP control diet), and 3) ryegrass pasture mornings and corn silage evenings, supplemented with a grain mix containing corn, soybean meal, corn gluten meal, and blood meal (moderate CP, high RUP). Dietary CP and RUP concentrations were approximately 23.1, 5.8; 17.7, 5.0; and 17.2, 6.8% of dry matter for diets 1 to 3, respectively. Plasma urea N concentrations were highest in cows fed diet 1 (25.0 mg/dl), intermediate in cows on diet 2 (20.1 mg/dl), and lowest in cows on diet 3 (18.5 mg/dl). Cows fed excess dietary protein (diet 1) exhibited lower first breeding pregnancy rates (24.1 vs. 41.0%) and lower overall pregnancy rates (53.4 vs. 75.4%) than did cows fed diet 2, increasing time nonpregnant by an average of 15.1 d per cow. Reproductive performance was similar between cows fed diets 2 and 3. Mean fat-corrected milk (FCM) yield was not affected by protein concentration (diet 1 vs. 2); however, partum group cows that received supplemental RUP (diet 3) produced more 3.5% FCM than controls in early lactation. Feeding grain diets that contained excess dietary protein impaired the reproductive performance of dairy cows grazing ryegrass.

ABSTRACT

In a trial designed to evaluate the efficacy of the recommended dosage of moxidectin 2% oral gel against the gastric stages of Gasterophilus spp., 14 ponies were selected from a herd on the basis of the inclusion criterion of the presence of Gasterophilus spp. eggs attached to their hair coats. After random allocation, the ponies were treated with 1 of 2 treatments, moxidectin 2% equine gel in a single dose at the commercial dosage of 400 microg moxidectin/kg body weight or placebo gel. The animals were necropsied 14 days posttreatment. Efficacies against second- and third-instar Gasterophilus intestinalis De Geer were 100% and 99.5%, respectively.

ABSTRACT

Eighteen mixed-breed, naturally infected ponies ranging in age from 1 to 16 yr and four cyathostome-naïve ponies reared and maintained under parasite-free conditions ranging in age from 1 to 4 yr were used in this study. Naturally-infected ponies were treated with 1 dose of ivermectin (IVM) at 200 micrograms kg-1, followed by a 5-day regimen of oxibendazole (OBZ) at 20 mg kg-1 to remove existing cyathostome burdens; cyathostome-naïve control ponies were treated with IVM alone. The naturally infected ponies were matched on age and gender, then randomly assigned to one of three treatment groups of six animals per group; the four cyathostome-naïve ponies constituted a fourth group. Following OBZ treatment, Group 1 ponies were treated with pyrantel tartrate (PT) in their pelleted ration; the remaining ponies received only the pelleted ration. Beginning on experiment Day 3, a daily challenge infection of 10(4) mixed cyathostome larvae was administered orally to ponies of Group 1, Group 2 and the cyathostome-naïve controls. Group 3 ponies served as unchallenged controls to determine residual parasite burdens following IVM/OBZ treatment. Necropsy examinations were performed on three Group 3 ponies on Day 1; the remainder of the necropsy examinations began on Day 41. Cyathostome burdens were evaluated by recovery of larvae and adults from the luminal contents, by digestions of the intestinal mucosa, and by mural transillumination of full-thickness intestinal sections. Differences in postchallenge clinical responses were also compared. Necropsy examinations included comparisons of grossly visible inflammation of the large bowel, weights of biopsy specimens from each region, and histologic evaluations of these biopsies. Parasite recoveries at necropsy indicated a strong protective effect derived from daily PT treatment. Mean weights of intestinal biopsies corresponded with worm burdens, but histological evaluation did not reveal architectural or cellular changes to account for the increase in weight; therefore, edema was suspected. A strong age-related resistance to challenge infection was apparent in both the PT-treated and control groups by virtue of the lower mean worm burdens found in older ponies compared to younger ponies of the same treatment group; however, daily PT treatment of older ponies reduced the variability of their worm burdens to a uniformly low level. Comparisons of luminal and mucosal parasite burdens of age stratified nontreated controls further suggest that the age related resistance, which is acquired, targets increasing numbers of parasite stages as this resistance matures. Further, there is no evidence for an immune mediated acquisition of hypobiotic L3.

ABSTRACT

Three groups of foals were raised under different management programs in this study: Group 1 (n = 6) and Group 2 (n = 6) were raised with their dams on pasture; Group 3 foals (n = 5) were raised under parasite-free conditions. Mares and foals of Group 1 received daily pyrantel tartrate (PT) treatment with their pelleted feed ration, whereas mares and foals of Groups 2 and 3 received only the pelleted ration. Pasture-reared foals were weaned and moved to a heavily contaminated pasture for 5 weeks. Group 1 foals continued to receive daily PT treatment whereas Group 2 foals received only the pelleted feed ration. Following this period, all foals were moved into box stalls. Half of each group was challenged with 10(3) Strongylus vulgaris infective third-stage larvae (L3), 5 x 10(3) Strongylus edentatus L3 and 10(5) mixed cyathostome L3; the remaining half served as unchallenged controls. Necropsy examinations were performed 6-week post-challenge for evaluation of parasite burdens and lesions. Daily PT treatment of Group 1 reduced the patent cyathostome infections of both mares and foals and was effective in reducing pasture burdens of infective larvae. Daily treatment of Group 1 foals during weaning continued to suppress EPG levels; however, it did not prevent large strongyle infections during the weaning period. Group 1 foals were more sensitive to challenge than Group 2 foals, which did not exhibit any post-challenge disturbances. Group 1 foals were equally susceptible to challenge as parasite-free foals.

Three fecal egg count reduction assays (FECR) and one critical trial were performed to determine the efficacy of pyrantel pamoate (PP) at 6.6 mg base kg-1 on a well managed stud farm in Louisiana where a loss of efficacy was suspected. Efficacy of PP based on FECR varied from 25% in mares to 83% in yearlings. Second treatments with PP 2 weeks following an initial treatment failed to reduce eggs per gram (EPG). A critical trial was performed to determine the cyathostome species resistant to PP. Three strongyle-naive ponies which acquired infections on the farm were used for this purpose. Following treatment with PP at the recommended dose, 11 species of cyathostomes remained in the intestine of the tracer ponies. Reduced efficacies (62%-88%) were noted for seven species. Resistance to oxibendazole (OBZ), which was > 90% effective on this farm in 1982, was also evaluated by FECR and found to exist. The results of one experiment indicate that dual resistance of parasites to PP and OBZ also exists.

ABSTRACT

Intense exercise affects various parameters of the immune system. The overall effect of exercise on immune function is dependent upon the physical condition of the subject, the intensity and duration of the exercise period, and the immune parameter assessed. Unconditioned horses subjected to a single bout of intensive exercise exhibit multiple alterations in immune function, including an augmentation of lymphokine activated killer (LAK) cell function. This increase in LAK cell activity is not due to an increase in circulating LAK precursors. While peripheral blood mononuclear cells from exercising horses exhibit greater responsiveness to IL-2, this is not due to an increase in IL-2 receptor expression. LAK cell generation in vitro is augmented by those catecholamines and neuropeptides which are produced during exercise, suggesting a direct effect of these compounds on LAK cell generation at a step post IL-2 receptor binding.

ABSTRACT

Two dosages of moxidectin oral gel were evaluated and compared to a therapeutic dose of ivermectin oral paste in the control of a spectrum of gastrointestinal parasites of ponies naturally infected in southern Louisiana or Mississippi. Thirty-two mixed-breed ponies ranging in age from one to 21 years were used in this controlled test. Eight weeks prior to the experiment, ponies grazing on contaminated pasture were moved to a paddock and fed a pelleted ration, thus reducing or eliminating the potential for additional infection and ensuring the existence of a population of encysted larvae. Ponies were then allocated to replicates of four animals based on values of fecal strongyle egg counts and percent strongyle larvae composition determined from Baermann sedimentations of fecal cultures. Members of replicates were allocated to one of four treatment groups: moxidectin oral gel administered at 300 micrograms kg-1 body weight, moxidectin oral gel at 400 micrograms kg-1, the oral gel vehicle as negative control, and ivermectin oral paste at 200 micrograms kg-1. Prior to treatment, ponies were confined in pairs to covered concrete runs by treatment group. Two weeks following treatment, necropsy examinations of all animals were performed. Parasites were recovered from the lumen of the stomach, the intestinal tract, the cranial mesenteric artery and its major branches, the peritoneal body wall and from pepsin digests of mucosal scrapings taken from the cecum and large colon. Encysted cyathostome larval burdens were also compared using mural transillumination of segments of the large colon for visualization of the encysted forms. Control ponies were not uniformly infected with the spectrum of parasites; however, moxidectin, at either dosage, compared favorably with ivermectin in the control of the adults of Strongylus vulgaris, Strongylus edentatus, Triodontophorus spp., Oesophagodontus robustus, Trichostrongylus axei, Oxyuris equi, Parascaris equorum, Habronema muscae, as well as both the adult and larval Cyathostominae recovered from the lumen. Moxidectin also appears as efficacious as ivermectin against migrating large strongyle larvae at the two weeks post-treatment evaluation. Moxidectin demonstrated a trend towards greater efficacy against encysted cyathostome larvae than a therapeutic dosage of ivermectin, but this difference was not statistically significant. Moxidectin was less effective than ivermectin against Gasterophilus intestinalis and was equally ineffective as ivermectin against Anoplocephala perfoliata.
