ABSTRACT

Propagation by somatic embryogenesis in Theobroma cacao has some issues to be solved, as many morphologically abnormal somatic embryos that do not germinate into plants are frequently observed, thus hampering plant production on a commercial scale. For the first time the methylome landscape of T. cacao somatic embryogenesis was examined, using whole-genome bisulfite sequencing technique, with the aim to understand the epigenetic basis of somatic embryo abnormalities. We identified 873 differentially methylated genes (DMGs) in the CpG context between zygotic embryos, normal and abnormal somatic embryos, with important roles in development, programmed cell death, oxidative stress, and hypoxia induction, which can help to explain the morphological abnormalities of somatic embryos. We also identified the role of ethylene and its precursor 1-aminocyclopropane-1-carboxylate in several biological processes, such as hypoxia induction, cell differentiation and cell polarity, that could be associated to the development of abnormal somatic embryos. The biological processes and the hypothesis of ethylene and its precursor involvement in the somatic embryo abnormalities in cacao are discussed.

ABSTRACT

Cocoa is a perennial and arboreal species intolerant to strong and frequent winds and, for this reason, is usually grown with windbreaks of trees. The mechanical alterations caused by the wind in the field have a great impact on the growth, development and productivity of cocoa. The present work had a main objective to understand the molecular mechanisms of responses to mechanical stress, caused by the action of constant wind flow in young plants of cocoa through alterations of the proteomic profile in young (YL) and mature leaves (ML). Plants were exposed to constant wind (CW) at a speed of 4.5 m s-1 for 12 h. There was a reduction in the accumulation of proteins in YL and a significant increase in ML submitted to CW in relation to the control. Differentially accumulated proteins, identified in YL and ML, belong to a broad functional group, related to energy production and carbon metabolism. Besides that, there was a higher efficiency in the protein relative abundance associated to energy production and the assimilation of carbon in the ML exposed to CW, in relation to the control. It was observed the appearance of new isoforms and, or post-transitional changes, which represent an acclimatization and tolerance response of these leaves to the stressor factor. In contrast, in YL, the energy production and the synthesis of gene products essential for their growth and development were affected by the mechanical stress caused by the wind, making them more intolerant.

ABSTRACT

Ornamental hybrids of passion flowers are thoroughly diffused in many countries and used in the decoration of houses and gardens. However, the cultivation of ornamental passion fruits practically unexploited in Brazil. This study aimed at evaluating the growth and blooming of F1 hybrids of Passiflora L. (P. sublanceolata J.M. MacDougal [ex P. palmeri var. sublanceolata Killip] vs. P. foetida var. foetida L.) cultivated in ceramic and concrete pots under different shading levels. The vegetative and flower evaluations were carried out weekly, in clonal cuttings 60 days after rooting. The height, leaf length and width, the number of internodes and leaves and stem diameter were evaluated using a randomized complete design in a factorial scheme which corresponded to two genotypes, two types of pot, three shading levels (25 %, 50 %, 75 %) and seven weeks of evaluation, with four replications. For the variable number of flowers, the same experimental design was adopted. However, the number of evaluations was modified for three periods, but this was not considered for the flower diameter and leaf area. The shading levels of 25 % and 50 % were the most favorable to the growth in height, whereas hybrid genotypes under 25 % shade had greater increase in the number of leaves, internodes and stem diameter, showing tolerance to moderate shade. The higher values for length, width and leaf area were observed at 75 % shade. The greatest number of flowers was verified at 25 % shadow in concrete pots. As for the types of pot, the ceramic ones were more favorable to the growth of hybrid plants during the first weeks of acclimatization to the treatments, and the concrete ones were more propitious to blooming. Thus, the use of hybrid plants in concrete pots for the ornamentation of internal environments is recommended, if they are well illuminated.
