ABSTRACT

Endocrine treatment, particularly tamoxifen, has shown significant improvement in the prognosis of patients with estrogen receptor-positive (ER-positive) breast cancer. However, the clinical utility of this treatment is often hindered by the development of endocrine resistance. Therefore, a comprehensive understanding of the underlying mechanisms driving ER-positive breast cancer carcinogenesis and endocrine resistance is crucial to overcome this clinical challenge. In this study, we investigated the expression of MICAL-L2 in ER-positive breast cancer and its impact on patient prognosis. We observed a significant upregulation of MICAL-L2 expression in ER-positive breast cancer, which correlated with a poorer prognosis in these patients. Furthermore, we found that estrogen-ERß signaling promoted the expression of MICAL-L2. Functionally, our study demonstrated that MICAL-L2 not only played an oncogenic role in ER-positive breast cancer tumorigenesis but also influenced the sensitivity of ER-positive breast cancer cells to tamoxifen. Mechanistically, as an estrogen-responsive gene, MICAL-L2 facilitated the activation of the AKT/mTOR signaling pathway in ER-positive breast cancer cells. Collectively, our findings suggest that MICAL-L2 could serve as a potential prognostic marker for ER-positive breast cancer and represent a promising molecular target for improving endocrine treatment and developing therapeutic approaches for this subtype of breast cancer.

ABSTRACT

Wheat plants with yellow stripes on their leaves were collected in the city of Tai'an (Shandong province, China). High-throughput sequencing analysis of the collected plants showed that they were coinfected with wheat leaf yellowing-associated virus (WLYaV) and an unidentified polerovirus. The genome of the unidentified virus, tentatively named "triticum yellow stripe virus" (TriYSV), comprises 5,595 nucleotides and contains seven open reading frames (ORFs), with a typical polerovirus genome structure. Analysis by sequence alignment showed that TriYSV had the highest sequence similarity to wheat yellow dwarf virus (WYDV, a tentative member of the genus Polerovirus), with 87.3% nucleotide sequence identity over the whole genome. Except for P3a and the coat protein (CP), all of the proteins encoded by TriYSV showed < 90% amino acid identity to those of other poleroviruses. Phylogenetic analysis based on RNA-dependent RNA polymerase and CP amino acid sequences and complete genome nucleotide sequences showed that the poleroviruses WYDV, cereal yellow dwarf virus RPS (CYDV-RPS), CYDV-RPV, and barley yellow dwarf virus GPV are the most closely related to TriYSV. Thus, TriYSV is proposed to be a new member of the genus Polerovirus.

ABSTRACT

To investigate the dynamic changes and influencing factors for the quality of life in nursing care in patients with lung cancer after resection. Totally, 136 patients undergoing lung cancer resection in our hospital from January 2019 to January 2022 were prospectively enrolled as subjects. The quality of life was measured before and 1 and 2 weeks and 1, 3, 6, and 12 months after the operation to analyze the dynamic changes in the quality of life in nursing care. Clinical data of patients were collected at the time of discharge. The patients were divided into high-quality and low-quality groups according to the median level of quality of life in nursing care at the final follow-up. The logistic regression equation was applied to analyze the influencing factors for the quality of life in nursing care after lung cancer resection. Of 136 patients receiving lung cancer resection, 32 were lost to follow-up until the final follow-up, so 104 patients were finally included. According to the median level of quality of life in nursing care at the final follow-up, the patients were divided into high-quality and low-quality groups (n = 52 per group). The quality of life in nursing care first decreased, then increased, and then stabilized after lung cancer resection. The comparison of clinical data between the two groups exhibited that albumin level was higher in the high-quality group than that in the control group. The age, proportion of living alone, S-AI score, and FoP-Q-SF score were lower in the high-quality group than those in the low-quality group (P < 0.05). Univariate logistic regression analysis demonstrated that high albumin (OR = 0.884) was a protective factor for the quality of life in nursing care after lung cancer resection (P < 0.05). Living alone (OR = 1.333), high S-AI score (OR = 1.211), high FoP-Q-SF score (OR = 1.221), and advanced age (OR = 1.209) were the risk factors for the quality of life in nursing care after lung cancer resection (P < 0.05). Multivariate logistic regression analysis demonstrated that high albumin (OR = 0.861) was a protective factor for the quality of life in nursing care after lung cancer resection (P < 0.05). Living alone (OR = 1.144), high S-AI score (OR = 1.170), high FoP-Q-SF score (OR = 1.161), and advanced age (OR = 1.181) were the risk factors for the quality of life after lung cancer resection (P < 0.05). The quality of life in nursing care first decreased, then increased, and then stabilized after lung cancer resection. Albumin, age, living alone, and S-AI and FoP-Q-SF scores were the influencing factors for the quality of life in nursing care after lung cancer resection. In the nursing care process after lung cancer resection, we should focus on elderly patients living alone who are affected by anxiety and fear of recurrence to improve the quality of life of these patients.

ABSTRACT

Potato aucuba mosaic virus (PAMV), a positive single-strand RNA virus, has one of the longest genomes of the viruses in the genus Potexvirus. In 2019, potato samples with mottle and crinkling symptoms from Huzhou, Zhejiang province, China, were identified to be infected with PAMV, potato virus X (PVX), and potato virus Y (PVY) by transcriptome sequencing. To study the effects of single infection by PAMV, the full-length sequence of PAMV from Huzhou (MT193476) was determined and an infectious full-length cDNA clone was constructed. This cDNA clone was infectious by agro-infiltration, leading to systemic symptoms in Nicotiana benthamiana, tomato, pepper, and potato.

ABSTRACT

The apoplast is the extracellular space for signalling, nutrient transport, and plant-microbe interactions, but little is known about how plant viruses use the foliar apoplast. Proteomic analysis of the apoplasts isolated from potato virus X (PVX)-infected Nicotiana benthamiana plants showed that the coat protein (CP) is the dominant viral component. The presence of the CP in the apoplast was confirmed by western blot, viral nucleic acid was detected by reverse transcription-PCR and northern blot, and viral particles were observed by transmission electron microscopy (TEM). The apoplast from infected leaves was infectious if rubbed onto healthy leaves but not when infiltrated into them. The exosomes were separated from the apoplast fluid by high-speed centrifugation and TEM showed that PVX particles were not associated with the exosomes. These results suggest that PVX virions are released to the N. benthamiana apoplast in a one-way manner and do not share the bidirectional transport of exosomes.

ABSTRACT

The complete genome sequence of a novel foveavirus identified in garlic (Allium sativum L.) in China was determined using RNA-seq, reverse transcription polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE) PCR. The entire genomic RNA (GenBank accession MT981417) is 8748 nucleotides long excluding the 3'-terminal poly(A) tail and contains five open reading frames (ORFs). These ORFs encode the viral replicase, a triple gene block, and a coat protein. The virus was tentatively named "garlic yellow stripe associated virus" (GarYSaV). Pairwise comparisons of protein sequences show that GarYSaV encodes proteins that share less than 47% identity with those of other foveaviruses, suggesting that it represents a new species in the genus. Phylogenetic analysis of amino acid sequences of the replicase and CP confirm that GarYSaV is a member of the genus Foveavirus. To our knowledge, this is the first report of a foveavirus in a monocot plant.

ABSTRACT

Systemic necrosis often occurs during viral infection of plants and is thought mainly to be the result of long-term stress induced by viral infection. Potato virus X (PVX) encodes the P25 pathogenicity factor that triggers a necrotic reaction during PVX-potato virus Ysynergistic coinfection. In this study, we discovered that NbALY916, a multifunctional nuclear protein, could interact with P25. When NbALY916 expression was reduced by tobacco rattle virus (TRV)-based virus-induced gene silencing, the accumulation of P25 was increased, which would be expected to cause more severe necrosis. However, silencing of NbALY916 reduced the extent of cell death caused by P25. Furthermore, we found that overexpression of NbALY916 increased the accumulation of H2 O2 and triggered more extensive cell death when coexpressed with P25, even though accumulation of P25 was itself reduced by the increased expression of NbALY916. Furthermore, transient expression of P25 specifically induced the expression of NbALY916 mRNA, but not the mRNAs of three other ALYs in Nicotiana benthamiana. In addition, we showed that silencing of NbALY916 or transient overexpression of NbALY916 affected the infection of PVX in N. benthamiana. Our results reveal that NbALY916 has an antiviral role that, in the case of PVX, operates by inducing the accumulation of H2 O2 and mediating the degradation of P25.

ABSTRACT

Pepper mild mottle virus (PMMoV) causes serious economic losses in pepper production in China. In a survey for viral diseases on pepper, two PMMoV isolates (named PMMoV-ZJ1 and PMMoV-ZJ2) were identified with different symptoms in Zhejiang province. Sequence alignment analysis suggested there were only four amino acid differences between the isolates: Val262Gly, Ile629Met and Ala1164Thr in the replicase, and Asp20Asn in the coat protein. Infectious cDNA clones of both isolates were constructed and shown to cause distinctive symptoms. Chlorosis symptoms appeared only on PMMoV-ZJ2-infected plants and the Asp20Asn substitution in the CP was shown to be responsible. Confocal assays revealed that the subcellular localization pattern of the two CPs was different, CP20Asp was mainly located at the cell periphery, whereas most CP20Asn located in the chloroplast. Thus, a single amino acid in the CP determined the chlorosis symptom, accompanied by an altered subcellular localization.

ABSTRACT

The ubiquitous Acinetobacter baumannii is an important and troublesome pathogen of nosocomial infection. Multidrug-resistant A baumannii meningitis is very difficult to treat, usually with high mortality. The disease is generally associated with craniotomy, but rarely with lumbar puncture. However, in our case, a 45-year-old male construction worker suffered multidrug-resistant A baumannii meningitis associated with lumbar puncture. The patient responded poorly to the initial treatment with an intravenous infusion of tigecycline. However, later treatment with an intrathecal injection of tigecycline (concentration 1 mg/mL) at a dose of 10 mg every 12 hours proved to be highly efficacious. Thus, intrathecal injection of tigecycline should be seriously considered as a treatment for multidrug-resistant A baumannii meningitis.

ABSTRACT

With an expanded electron-rich cavity and a fixed conformation, macrocycle H was found to encapsulate π-extended viologens G1-G4 to form the first case of pseudo[3]rotaxanes based on oxacalixarenes. The complexation was investigated in detail both in solution and in the solid state using NMR spectroscopy and X-ray crystallography. Due to the three-dimensional nonsymmetric structure of H, three orientational isomers of the pseudorotaxanes could be expected theoretically. However, as the crystal structure analysis revealed, only one of the three isomers was obtained with either G1 or G3. Moreover, with regard to the different lengths of the linkers in the guest molecules, completely opposite orientations of the macrocycles on the axles were observed, which could be explained by different complexation modes between the components in the pseudo[3]rotaxanes. Additionally, the complexation between the host and the guest could be reversibly switched on and off using a suitable acid and base. These results will provide us with the opportunity to design and elaborately regulate high-order molecular devices.

ABSTRACT

New triptycene-derived oxacalixarene H1 was efficiently synthesized by a template cyclization step, and anilino-substituted macrocycle H2 was subsequently afforded through straightforward nucleophilic displacement of the active chlorine atom in H1. Oxacalixarene H1 adopts a fixed boat-like 1,3-alternate conformation and shows moderate complexation abilities towards various bipyridinium salts. However, the affinities of H2 towards the guests were found to be substantially stronger, which could be tentatively attributed to the additional hydrogen-bonding site, π-π stacking site, and especially the increased electron richness of the host. Furthermore, the acid-base switchable complexation process between H1 and the bipyridinium salt was also realized, which could potentially facilitate the construction of high-level stimuli-responsive supramolecular structures based on the newly synthesized oxacalixarene.

ABSTRACT

It is widely acknowledged that the excessive reactive oxygen species (ROS) or reactive nitrogen species (RNS) induced oxidative stress will cause significant damage to cell structure and biomolecular function, directly or indirectly leading to a number of diseases. The overproduction of ROS/RNS will be balanced by nonenzymatic antioxidants and antioxidant enzymes. Polysaccharide or glycoconjugates derived from natural products are of considerable interest from the viewpoint of potent in vivo and in vitro antioxidant activities recently. Particularly, with regard to the in vitro antioxidant systems, polysaccharides are considered as effective free radical scavenger, reducing agent, and ferrous chelator in most of the reports. However, the underlying mechanisms of these antioxidant actions have not been illustrated systematically and sometimes controversial results appeared among various literatures. To address this issue, we summarized the latest discoveries and advancements in the study of antioxidative polysaccharides and gave a detailed description of the possible mechanisms.

ABSTRACT

The manipulation of supramolecular devices to carry out sophisticated and programmed tasks is bound up with the spatial allocation of their components, especially the threading direction of the guest, which controls the host-guest orientation in the device. However, insights are needed to probe more possibilities for steering the threading direction. We have developed a new system consisting of a three-dimensional nonsymmetric oxacalixarene (H) with a fixed comformation and (bi)pyridinium salts (G1-G3), in which we found that based on the intrinsic discrepancies between the two semi-cavities of H, the electron densities of the axles greatly affect the threading direction. This was unequivocally demonstrated by NMR spectra and single crystal structures. With elaborate design, unidirectional threading was achieved, resulting in an oriented rotaxane. Therefore, we describe a new approach in which the threading direction and final orientation may be finely controlled by adjustment of the structure of the guest.

ABSTRACT

To safely and effectively apply artemisinin sustained-release granules to control and prevent algal water-blooms, the effects of artemisinin and its sustained-release granules on freshwater alga (Scenedesmus obliquus (S. obliquus) and Microcystis aeruginosa (M. aeruginosa)), as well as the production and release of microcystins (MCs) were studied. The results showed that artemisinin sustained-release granules inhibited the growth of M. aeruginosa (above 95% IR) and S. obliquus (about 90% IR), with M. aeruginosa more sensitive. The artemisinin sustained-release granules had a longer inhibition effect on growth of pure algae and algal coexistence than direct artemisinin dosing. The artemisinin sustained-release granules could decrease the production and release of algal toxins due to the continued stress of artemisinin released from artemisinin sustained-release granules. There was no increase in the total amount of MC-LR in the algal cell culture medium.

ABSTRACT

The unsaturated fatty acid (linoleic acid) sustained-release microspheres were prepared with linoleic acid (LA) using alginate-chitosan microcapsule technology. These LA sustained-release microspheres had a high encapsulation efficiency (up to 62%) tested by high performance liquid chromatography with a photo diode array. The dry microspheres were characterized by a scanning electron microscope, X-ray diffraction measurement, dynamic thermogravimetric analysis and Fourier transform infrared spectral analysis. The results of characterization showed that the microspheres had good thermal stability (decomposition temperature of 236°C), stable and temperature independent release properties (release time of more than 40 d). Compared to direct dosing of LA, LA sustained-released microspheres could inhibit Microcystis aeruginosa growth to the non-growth state. The results of this study suggested that the LA sustained-release microspheres may be a potential candidate for algal inhibition.

ABSTRACT

A series of triptycene-derived homooxacalixarene analogues were conveniently synthesized by a one-pot approach starting from 2,7-dihydroxytriptycene and 1,3-bisbromomethylbenzene derivatives under mild reaction conditions. Similarly, two pairs of "basket-like" triptycene-derived homooxacalixarene analogues were also designed and synthesized. Structures of these macrocyclic molecules in both solution and solid state were studied by NMR experiments and X-ray crystallography. Because of the rigid triptycene units, the homooxacalixarene analogues showed large cavities and fixed conformations even up to 380 K. It was also found that these novel macrocycles could be served as efficient host molecules for complexation with fullerenes C(60) and C(70).

ABSTRACT

The binding properties of triptycene-derived oxacalixarenes containing 1,8-naphthyridine subunits 1a and 1b toward metal ions were investigated in detail by spectroscopic methods. A couple of new N,O-bridged diazadioxacalixarenes 2a, 2b were synthesized and their binding properties to metal ions were also evaluated. The results showed that the oxacalixarene 1a, a cis-isomer with a boat-like 1,3-alternate conformation and a symmetrical cavity structure, exhibited a highly selective fluorescence response toward Hg(2+) ions, while 1b, 2a and 2b did not show selective response towards any specific metal ions. Such selectivity can thus be controlled by the bridging heteroatoms and cavity structures of the macrocycles. Moreover, it was found that the fluorescence of 1a was considerably quenched upon the addition of Hg(2+), and a 1 : 2 stoichiometry host-guest complex was proposed on the basis of the Job plot and (1)H NMR titrations.

ABSTRACT

A pair of triptycene-derived oxacalixarenes, 1a and 1b, containing two naphthyridine subunits have been synthesized by a convenient one-pot approach. Macrocycles 1a and 1b are diastereomers, with 1a being a cis isomer with a boat-like 1,3-alternate conformation and 1b being a trans isomer with a curved boat-like conformation in the solid state. Both 1a and 1b have fixed conformations in solution at room temperature, as revealed by variable-temperature (1)H NMR spectroscopy experiments. It was found that oxacalixarenes 1a and 1b could both form 1:1 [2]pseudorotaxane-type complexes with paraquat derivatives with different functional groups in solution and in the solid state, which resulted in the further synthesis of a pair of novel isomeric [2]rotaxanes. Moreover, the controlled dethreading and rethreading processes of the [2]pseudorotaxanes were also easily achieved by acid-base stimuli or Hg(2+) association/dissociation.
