ABSTRACT

To explore the effects of seawater temperature and biological processes on aragonite saturation state (Ωarag), eight field surveys were conducted from December 2014 to November 2015 at A4HDYD station in the North Yellow Sea (NYS). Low regional yearly initial values of seawater Ωarag (1.26-1.76) were obtained in winter due to low seawater temperature. During spring to early autumn, surface-water Ωarag increased from 2.02 to 3.47 for the increasing seawater temperature and biological production, whereas bottom-water Ωarag remained as low as 1.44-1.79 due to low seawater temperature, net community respiration and the influence of the North Yellow Sea Cold Water Mass (NYSCWM). Following the collapse of stratification in late autumn, the seawater was well-mixed with an average Ωarag of 2.17. The annual cycle of seawater temperature dominated the seasonal variations of Ωarag, counteracted by biological production increasing Ωarag of 0.49 in surface water in March and strengthened by net community respiration decreasing Ωarag of 0.11-0.41 below the pycnocline during summer and autumn. It is, therefore, mid-to-high latitude coastal areas impacted by human activities are likely subject to ocean acidification due to the combined effects of low seawater temperature and net community respiration.

ABSTRACT

Liver ischemia/reperfusion (I/R) injury is a common injury after liver transplantation and hepatectomy. Skimmianine (Ski) has antibacterial, antiviral pharmacological effects. However, it is not clear whether Ski has a protective effect against liver I/R injury. In the present study, we established a mouse liver I/R model and an AML12 cell hypoxia-reoxygenation (H/R) model, both pretreated with different concentrations of Ski. Serum transaminase levels, necrotic liver area, cell viability, inflammatory factors, oxidative stress and apoptosis-related levels were measured to assess the protective effect of Ski against liver I/R injury. Western blotting was used to detect apoptosis-related proteins and PI3K-AKT pathway-related proteins. Mice and cells were also treated with PI3K inhibitor LY294002 to assess changes in indicators of liver injury. The results showed that Ski significantly reduced transaminase levels, liver necrosis area, oxidative stress, and apoptosis levels in mice with I/R. Ski also inhibited cell injury and apoptosis after H/R. Moreover, Ski activated phosphorylation of PI3K-AKT pathway-related proteins after liver I/R and cell H/R. Importantly, the PI3K inhibitor LY294002 effectively reversed the alleviation of I/R injury caused by Ski. These results confirm that Ski exerts a protective effect against liver I/R injury through activation of the PI3K-AKT pathway.

ABSTRACT

Different studies are filling the gaps in the distribution map of global marine microplastics. However, the data on the seasonal variation is relatively limited, which may lead to overestimation or underestimation of the distribution level of microplastics. To understand baseline data and seasonal variations of the microplastics in the surface seawater of the Yellow Sea, a survey over four seasons was conducted during 2017-2018. Microplastics were collected using a 330 µm manta net. It was found that the abundance of microplastics was 0.63 ± 0.57 particles/m3. The seasonal abundance variation was spring > summer ≈ winter > autumn. The main categories were foam, line, and fragments, accounting for 32 %, 19 %, and 19 % of the total amount, respectively, and the dominant components were polypropylene, polyethylene, and polyethylene terephthalate, accounting for 38 %, 22 %, and 22 % of particles, respectively. The factors affecting the variation included wind-induced mixing, river input, and environmental topography.

ABSTRACT

Chimeric antigen receptor (CAR) T cell therapy is a novel therapeutic approach that uses gene editing techniques and lentiviral transduction to engineer T cells so that they can effectively kill tumors. However, CAR T cell therapy still has some drawbacks: many patients who received CAR T cell therapy and achieve remission, still had tumor relapse and treatment resistance, which may be due to tumor immune escape and CAR T cell dysfunction. To overcome tumor relapse, more researches are being done to optimize CAR T cell therapy to make it more precise and personalized, including screening for more specific tumor antigens, developing novel CAR T cells, and combinatorial treatment approaches. In this review, we will discuss the mechanisms as well as the progress of research on overcoming plans. (AU)

OBJECTIVE: Low serum adiponectin level can predict hypertension development, and adiponectin gene (ADIPOQ) polymorphisms have been reported to be linked with hypertension risk. Whereas, the interaction between ADIPOQ polymorphisms and environmental factors on the susceptibility of hypertension remained unclear. The purpose of this study was to explore the relationship of ADIPOQ polymorphisms with hypertension risk and their interaction with lipid levels in coal miners. METHODS: A matched case-control study with 296 case-control pairs was performed in a large coal mining group located in North China. The participants were questioned by trained interviewers, and their ADIPOQ genotype and lipid levels were determined. Logistic regression, stratified analysis, and crossover analysis were applied to evaluate the effects of rs2241766, rs1501299, and rs266729 genotypes and gene-lipid interaction on hypertension risk. RESULTS: In this matched case-control study, the genotypes of rs2241766 TG+GG, rs1501299 GT+TT, and rs266729 CG+GG were marginally related to hypertension risk. Individuals with high total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) level were susceptible to hypertension (TC: odds ratio [OR] = 1.807, 95% confidence intervals [95%CI] = 1.266-2.581; LDL-C: OR = 1.981, 95%CI = 1.400-2.803; HDL-C: OR = 1.559, 95%CI = 1.093-2.223). Antagonistic interactions were detected between rs2241766 and TC, rs1501299 and TC, rs2241766 and LDL-C, and rs1501299 and HDL-C (rs2241766 and TC: OR = 0.393, 95%CI = 0.191-0.806; rs1501299 and TC: OR = 0.445, 95%CI = 0.216-0.918; rs2241766 and LDL-C: OR = 0.440, 95%CI = 0.221-0.877; rs1501299 and HDL-C: OR = 0.479, 95%CI = 0.237-0.967). Stratified analysis showed that hypertension risk was high for the subjects with rs2241766 TG+GG or rs1501299 GG under the low lipid level but low for those under the high lipid level. In the case group, the TC and LDL-C levels for rs2241766 TG+GG were lower than those for rs2241766 GG, and the TC and HDL-C levels for rs1501299 GT+TT were higher than those for rs1501299 GG. CONCLUSIONS: Although the effects of ADIPOQ polymorphisms alone were not remarkable, an antagonistic interaction was observed between ADIPOQ polymorphisms and lipid levels.

ABSTRACT

Six members of the gasdermin family are involved in various biological functions in malignant tumors. The present study aimed to perform a comprehensive analysis of gasdermin family genes in pan-cancer. Raw data was acquired from the genotype-tissue expression (GTEx) and the Cancer Genome Atlas. High inter-tumor heterogeneity in the expression between paracancerous and tumor tissues was observed across cancers. Survival analysis confirmed that the risk or protective effects of gasdermin family members on prognosis depended on the cancer types. The mutation frequency appeared to be high, and the mutation group had a worse prognosis. Besides, gasdermin family genes were associated with immune infiltrate subtypes, stromal and immune cell infiltration levels, TMB, MSI, immune checkpoint gene expression, and tumor stemness scores. Moreover, gasdermin family gene expressions affected the expressions of MMR genes and methyltransferases and could predict cancer cells sensitivity to chemotherapeutic drugs. Subsequently, the findings were double-checked in LIHC and PAAD. GSEA results indicated the gasdermin family genes mainly involved in tumor metabolism and immune microenvironment remodeling related signaling pathways. In conclusion, our findings confirmed that gasdermin family genes were potential therapeutic cancer targets in pan-cancer.

The RASSF family proteins have been implicated in the development of human cancers. To date, the expression pattern and biological significance of RASSF4 in colorectal cancers (CRC) have not been fully investigated. In the current study, we explored expression pattern of RASSF4 in 118 CRC specimens and 30 adjacent 'normal' colon tissues by immunohistochemistry. The results showed that RASSF4 was downregulated in CRC tissues compared with adjacent 'normal' tissues. RASSF4 downregulation significantly associated with advanced tumour-node-metastasis (TNM) stage, T status, positive node status and high Ki-67 index. Analysis of TCGA dataset also supported RASSF4 downregulation in CRC tissues. Ectopically expressed RASSF4 in LoVo cells inhibited cell growth, colony formation, cell cycle progression and increased the sensitivity to 5-FU treatment. Annexin V/PI apoptosis assay showed that RASSF4 overexpression increased 5-FU-induced apoptosis and downregulated the mitochondrial membrane potential. In addition, Western blot demonstrated that RASSF4 overexpression repressed YAP and Bcl-2 while upregulating p21 expression. YAP knockdown abolished the role of RASSF4 on Bcl-2. ChIP assay showed that TEAD4, a major YAP binding transcription factor, could bind to the promoter regions of Bcl-2. In conclusion, our data showed that RASSF4 was downregulated in human CRC. RASSF4 regulated malignant behaviour through YAP/Bcl-2 signalling in CRC cells.

ABSTRACT

Microplastics have attracted worldwide attention due to their potential threat to the marine ecosystem, with such pollutants even detected in the polar seas. Although in-depth research on microplastics has increased in recent years, studies in Antarctic waters remain relatively scarce compared with coastal waters and open oceans. In this study, microplastics in surface and subsurface Antarctic waters were investigated. The average microplastic abundance in the surface water was 0.10 ± 0.14 items/m3, with highest abundance in the Ross Sea, and the average microplastic abundance in the subsurface water was 1.66 ± 1.20 items/m3, with highest abundance in the Dumont d'Urville Sea. Polyester was the main microplastic in the surface waters (87.3%), while polypropylene (33.1%), polyester (28.7%), and polyethylene (22.8%) were the dominant microplastics in the subsurface waters. Results indicate that microplastic pollution in Antarctic waters may come from the Antarctic continent as well as southward transport from the ocean at mid- and low latitudes.

ABSTRACT

The pro-oncogene ETS-1 (E26 transformation-specific sequence 1) is a key regulator of the proliferation and invasion of cancer cells. The present work examined the correlation of the aberrant expression of ETS-1 with histological or clinical classification of astrocytoma: grade I (pilocytic astrocytoma), grade II (diffuse astrocytoma), grade III (anaplastic astrocytoma), and grade IV (glioblastoma multiforme). MicroRNA, miR-338-5p, was predicted by an online tool (miRDB) to potentially target the 3' untranslated region of ETS-1; this was confirmed by multi-assays, including western blot experiments or the point mutation of the targeting sites of miR-338-5p in ETS-1's 3'untralation region (3'UTR). The expression of miR-338-5p was negatively associated with that of ETS-1 in astrocytoma, and deficiency of miR-338-5p would mediate aberrant expression of ETS-1 in astrocytoma. Mechanistically, hypermethylation of miR-338-5p by DNA methyltransferase 1 (DNMT1) resulted in repression of miR-338-5p expression and the aberrant expression of ETS-1. Knockdown or deactivation of DNMT1 decreased the methylation rate of the miR-338-5p promoter, increased the expression of miR-338-5p, and repressed the expression of ETS-1 in astrocytoma cell lines U251 and U87. These results indicate that hypermethylation of the miR-338-5p promoter by DNMT1 mediates the aberrant expression of ETS-1 related to disease severity of patients with astrocytoma.

ABSTRACT

The study aimed to evaluate the prognostic significance of preoperative systemic inflammatory biomarkers including albumin to globulin ratio (AGR), neutrophil to lymphocyte ratio (NLR), lymphocyte to monocyte ratio (LMR), and platelet to lymphocyte ratio (PLR) and establish a nomogram in hepatocellular carcinoma (HCC) patients after microwave ablation (MWA). 192 HCC patients receiving MWA as initial therapy from the first ward of hepatobiliary surgery were classified as training cohort. Whereas, 84 patients from the second of hepatobiliary surgery were classified as validation cohort. Kaplan-Meier (KM) method and univariate analyses showed that AGR, NLR, LMR, and PLR were significantly associated with OS in the training cohort. Multivariate analysis including clinicopathologic features screened out independent predictors including ascites, tumor size, cancer embolus, AGR, and PLR. Based on those variables, a nomogram for predicting OS was established. The C-index was 0.794 in the training cohort and 0.772 in the validation cohort. Calibration plots identified the nomogram performed well with an ideal model. Compared with Barcelona Clinic Liver Cancer (BCLC) staging system and simple tumor size, the nomogram showed better predictive ability. Besides, the nomogram discovered the highest diagnostic accuracy in predicting postoperative clinical outcome than the combination of the present models with tumor size. In conclusion, the constructed nomogram could accurately predict individualized survival probability and might support clinician in individual treatment optimization and clinical decision-making.

ABSTRACT

Objective: To achieve multiple target fishing hook by efficiently grafting polymers containing benzophenone (BP) groups and photochemically coupling molecules in medicines onto magnetic nanoparticles (MNPs). Methods: MNPs attached carbonyl groups (Fe3O4-COOH) were prepared through a hydrothermal process. Then they were modified with DMSA, forming MNPs with thiol groups (Fe3O4-SH). Fe3O4-SH nanoparticles were grafted with polymer containing BP groups by surface-initiated condensation polymerization. Effects of monomer feed ratios and contents on the amounts of BP groups were investigated. The molecules in medicines were covalently coupled onto MNPs via photochemical reactions of BP groups. The contents of coupled molecules were determined by FT-IR and UV-Vis spectra analyses. Results: MNPs with average size of 100 nm were produced, modified with DMSA, and decorated by grafting polymer containing photosensitive BP groups. When the content of monomer containing BP groups was increased in grafting polymerizations, more BP groups were incorporated onto MNPs. This was conductive to the subsequent photo- coupling. FT-IR and UV-Vis spectra analyses confirmed the coupling of molecules in medicines. The active H and steric hinderance of the molecules affected their coupling. Conclusion: The resultant magnetic target fishing hook is ready as a probe for targets identification of traditional Chinese medicine (TCM).

BACKGROUND AND AIM: Preoperative systemic inflammatory biomarkers, including neutrophil to lymphocyte ratio (NLR), derived neutrophil to lymphocyte ratio (dNLR), platelet to lymphocyte ratio (PLR), and lymphocyte to monocyte ratio (LMR) have been developed to predict patient outcome in several types of carcinomas. The aim of this study was to investigate the potential prognostic value of NLR, dNLR, PLR, and LMR, and establish a prognostic nomogram in postoperative GBC patients who underwent radical cholecystectomy. METHODS: 169 GBC patients were retrospectively enrolled in the present study. ROC curve analysis was used to determine the optimal cut-off values of systemic inflammatory biomarkers. The prognostic value of those biomarkers was investigated according to the Kaplan-Meier method and Cox regression model. A relevant prognostic nomogram was established. RESULTS: Results showed that NLR, dNLR, PLR, and LMR were significantly associated with overall survival (OS); whereas, NLR and LMR were retained as independent indicators. Based on these independent predictors including tumor differentiation, T stage, N stage, CEA, NLR, and LMR, a nomogram was generated with an accuracy of 0.801. CONCLUSION: Based on our findings, the predictive nomogram could accurately predict individualized survival probability of postoperative GBC patients, and might support clinicians in treatment optimization and clinical decision-making.

Worldwide the seafloor has been recognized as a major sink for microplastics. However, currently nothing is known about the sediment microplastic pollution in the North Pacific sector of the Arctic Ocean. Here, we present the first record of microplastic contamination in the surface sediment from the northern Bering and Chukchi Seas. The microplastics were extracted by the density separation method from collected samples. Each particle was identified using the microscopic Fourier transform infrared spectroscopy (µFTIR). The abundances of microplastics in sediments from all sites ranged from not detected (ND) to 68.78 items/kg dry weight (DW) of sediment. The highest level of microplastic contamination in the sediment was detected from the Chukchi Sea. A negative correlation between microplastic abundance and water depth was observed. Polypropylene (PP) accounted for the largest proportion (51.5%) of the identified microplastic particles, followed by polyethylene terephthalate (PET) (35.2%) and rayon (13.3%). Fibers constituted the most common shape of plastic particles. The range of polymer types, physical shapes and spatial distribution characteristics of the microplastics suggest that water masses from the Pacific and local coastal inputs are possible sources for the microplastics found in the study area. In overall, our results highlight the global distribution of these anthropogenic pollutants and the importance of management action to reduce marine debris worldwide.

Based on a survey conducted from June to July 2013, aragonite saturation state variation and control in the river-dominated marginal BoHai and Yellow seas were investigated. Surface water Ωarag ranged from 2.0-3.8, whereas subsurface water Ωarag was generally lower than 2.0. Temperature changes had a strong influence on Ωarag through induced CO2 solubility changes in seawater. Riverine freshwater input decreased Ωarag in the Changjiang and Yalu river estuaries, but induced higher Ωarag in the Yellow River estuary. Biological processes had opposite effects on Ωarag, whereby elevated biological production led to the highest Ωarag in the South Yellow Sea surface water, whereas net community respiration/remineralization induced low Ωarag in subsurface water. Stratification affected the level and scale of low Ωarag in subsurface water. By the year 2100, surface water with Ωarag > 2.0 will disappear except for the Yellow River estuary, and most of the subsurface water will develop substantial aragonite undersaturation.

OBJECTIVE: The purpose of this retrospective study was to investigate the relationship between temperature-humidity index (THI), season, and conception rate (CR) of Holstein cows in central Taiwan. METHODS: The mean performance and number of observations were statistically evaluated for various parameters, including age at first service, number of days open, gestation length, CR, and calving interval for different parities. RESULTS: The results indicate that the mean age at first service was 493.2 days; the gestation length was similar across all cows of different parities, ranging from 275.1 to 280.7 days. The overall CR of all inseminations was significantly lower in multiparous cows (47.26%±0.22%) than in heifers (57.14%±0.11%) (p<0.05). At THI>72 and during the hot season (from June to November), CRs for multiparous cows were significantly reduced compared to that for heifers, while the ratio remained unchanged among heifers for all seasons. CONCLUSION: To achieve a high CR, lactating cows should be bred in winter and spring (from December to May) from the start of the seasonal breeding program, whereas the heifer should be allowed to breed in summer and fall under the subtropical climate in Taiwan.

Based upon surveys conducted in November 2012 and June 2013, the distribution and dynamics of aragonite saturation state (Ωarag) were investigated in the southern Yellow Sea (SYS) of China. In summer, surface water Ωarag ranged from 2.1-3.8 and enhanced biological production fueled by Changjiang River freshwater input increased Ωarag to 3.8 in the southern SYS. However, subsurface water Ωarag was <2.0 in the central SYS. During autumn, surface water Ωarag was 2.0-2.9, lower than that in summer due to ventilation between surface and low Ωarag (1.0-1.4) subsurface waters in the central SYS. Community respiration and/or aerobic remineralization dominated low Ωarag in subsurface waters, while water stratification influenced the level and scale of acidity accumulation. By the end of this century, waters with Ωarag>2.0 could disappear from the SYS with increasing atmospheric CO2, while bottom waters Ωarag may become undersaturated due to the impact of eutrophication.

ABSTRACT

Hydroxyl polycyclic aromatic hydrocarbons (OH-PAHs) are derived from hydroxylated PAHs as contaminants of emerging concern. They are ubiquitous in the aqueous and atmospheric environments and may exist in the polar snow and ice, which urges new insights into their environmental transformation, especially in ice. In present study the simulated-solar (λ > 290 nm) photodegradation kinetics, products and pathways of four OH-PAHs (9-Hydroxyfluorene, 2-Hydroxyfluorene, 1-Hydroxypyrene and 9-Hydroxyphenanthrene) in ice were investigated, and the corresponding implications for the polar areas were explored. It was found that the kinetics followed the pseudo-first-order kinetics with the photolysis quantum yields (Φs) ranging from 7.48 × 10(-3) (1-Hydroxypyrene) to 4.16 × 10(-2) (2-Hydroxyfluorene). These 4 OH-PAHs were proposed to undergo photoinduced hydroxylation, resulting in multiple hydroxylated intermediates, particularly for 9-Hydroxyfluorene. Extrapolation of the lab data to the real environment is expected to provide a reasonable estimate of OH-PAH photolytic half-lives (t1/2,E) in mid-summer of the polar areas. The estimated t1/2,E values ranged from 0.08 h for 1-OHPyr in the Arctic to 54.27 h for 9-OHFl in the Antarctic. In consideration of the lower temperature and less microorganisms in polar areas, the photodegradation can be a key factor in determining the fate of OH-PAHs in sunlit surface snow/ice. To the best of our knowledge, this is the first report on the photodegradation of OH-PAHs in polar areas.

ABSTRACT

BACKGROUND: Exercise is beneficial for blood glucose metabolism. However, whether moderate aerobic exercise could improve impaired fasting glucose is unknown. And the mechanism is also needed to investigate. METHODS: A cross-sectional research was performed and 120 participants with impaired fasting glucose (IFG) were randomly assigned into active and controlled groups. Briefly, participants in active group were required to take moderate aerobic exercise at least 30 min for five times per week, whereas in controlled group, participants were also advised to take exercise but not mandatorily required the same degree as that of active group. At baseline and 3 month's follow-up, laboratory and demographic variables were compared. RESULTS: At baseline, no significant between-group differences were observed. Generally, leukocyte ROCK2 activity in the active and controlled groups were 58.7 ± 6.0 mg/mL and 60.2 ± 7.3 mg/mL, and daily average exercise time at baseline in both groups was extremely little, with 5.2 ± 3.8 min and 5.9 ± 3.5 min, respectively. After 3 months' follow-up, 52 and 56 participants in the active and controlled groups completed the whole program. Compared to baseline, leukocyte ROCK2 activity and daily average exercise time were improved in both groups. Nonetheless, compared to the controlled group, leukocyte ROCK2 activity was reduced more profoundly and the daily average exercise time was longer in the active group (37.5 ± 6.3 min versus 18.3 ± 7.2 min, p < 0.05). Moreover, the percentage of IFG in the active group was decreased more prominently than the controlled group (76.9% versus 82.1%, p < 0.05). Multivariate regression analyses revealed that exercise time and leukocyte ROCK2 activity was significantly associated with IFG, with OR of 0.836 (active group versus controlled group, 95% CI 0.825-0.852, p < 0.05) in exercise time, and 1.043 (controlled group versus active group, 95% CI 1.021-1.069, p < 0.05) in leukocyte ROCK2 activity. In addition, exercise time was significantly associated with leukocyte ROCK2 activity, with OR of 0.822 (active group versus controlled group, 95% CI 0.818-0.843, p < 0.05). CONCLUSION: In subjects with IFG, increased daily average exercise time is beneficial for improving fasting blood glucose metabolism, and the mechanism may be associated with its effects on attenuating leukocyte ROCK2 activity.

ABSTRACT

The collision fault detection of a XXY stage is proposed for the first time in this paper. The stage characteristic signals are extracted and imported into the master and slave chaos error systems by signal filtering from the vibratory magnitude of the stage. The trajectory diagram is made from the chaos synchronization dynamic error signals E1 and E2. The distance between characteristic positive and negative centers of gravity, as well as the maximum and minimum distances of trajectory diagram, are captured as the characteristics of fault recognition by observing the variation in various signal trajectory diagrams. The matter-element model of normal status and collision status is built by an extension neural network. The correlation grade of various fault statuses of the XXY stage was calculated for diagnosis. The dSPACE is used for real-time analysis of stage fault status with an accelerometer sensor. Three stage fault statuses are detected in this study, including normal status, Y collision fault and X collision fault. It is shown that the scheme can have at least 75% diagnosis rate for collision faults of the XXY stage. As a result, the fault diagnosis system can be implemented using just one sensor, and consequently the hardware cost is significantly reduced.
